In pH 4.9 Britton-Robinson buffer solution, methotrexate (MTX) reacted with thallium(III) to form a 3∶1 chelate. This resulted in great enhancement of second-order scattering (SOS) spectra and frequency doubling scattering (FDS) spectra and appearance of new SOS and FDS spectra. Their maximum wavelengths were located at 520 and 390 nm, respectively. The increments of scattering intensities (ΔI) were directly proportional to the concentrations of MTX in the ranges of 0.022—2.0 μg•mL-1 (SOS method) and 0.008—2.5 μg•mL-1 (FDS method). The methods exhibited high sensitivities. The detection limits for MTX were 7.4 ng•mL-1 (SOS method) and 2.3 ng•mL-1 (FDS method), respectively. The optimum conditions of the reaction, the influencing factors and the effects of coexisting substances were investigated. A highly sensitive, simple and fast method for the determination of MTX has been developed. The method can be applied satisfactorily to the determination of MTX in human serum samples. In this work, the charge distribution of MTX was calculated by a CNDO quantum chemistry method. In addition, the reaction mechanism was discussed.  相似文献

The interaction between anionic surfactants (AS) and 1‐hexadecyl‐3‐methylimidazolium bromide [C16mim]Br was studied by using resonance light scattering (RLS) technique, UV‐Vis spectrophotometry and fluorometric methods. In Britton Robinson (BR) buffer (pH 6.0), [C16mim]Br reacted with AS to form supermolecular complex which resulted in enhancement in RLS intensity. Their maximum RLS wavelengths were all at 390 nm. Some important interacting experimental variables, such as the solution acidity, [C16mim]Br concentration, salt effect and addition order of the reagents, were investigated and optimized. Under the optimum conditions, quantitative determination ranges were 0.001–7 μg·mL?1 for dodecyl sodium sulfate (SDS), 0.001–6 μg·mL?1 for sodium dodecylbenzene sulfonate (SDBS) and 0.005–7 μg·mL?1 for sodium lauryl sulfonate (SLS), respectively, while the detection limits were 1.3 ng·mL?1 for SDS, 1.0 ng·mL?1 for SDBS and 5.1 ng·mL?1 for SLS, respectively. Based on the ion‐association reaction, a highly sensitive, simple and rapid method has been established for the determination of AS.  相似文献

In pH 1.0 acidic medium, double-charged triaminotriphenylmethane dyes such as methyl green (MEG) and iodine green (IG) react with potassium ferrocyanide to form 2 : 1 ion-association complexes by virtue of electrostatic forces and hydrophobic interaction. It results in the change of absorption and the great enhancement of resonance Rayleigh scattering (RRS) and the appearance of new RRS spectra. Two systems have similar spectral characteristics and their maximum RRS wavelengths are all located at 276 nm and smaller peaks are located at 332 and 457 nm, respectively. The intensity of RRS is directly proportional to the concentration of [Fe(CN)6]^4- in the range of 0.03-5.7 μg·mL^-1 (MeG system) or 0.04-5.9 μg·mL^-1 (IG system). The RRS method has high sensitivity and the detection limit (3σ) for potassium ferrocyanide is 9.3 ng·mL^-1 (MeG system) or 11.2 ng·mL^-1 (IG system). The optimum conditions, influencing factors and effects of foreign substances are investigated. The method also has a good selectivity. A sensitive, rapid and simple RRS method for the determination of potassium ferrocyanide in salinized food and table salt has been developed.  相似文献

在pH为5.0-5.4的乙酸-乙酸钠缓冲溶液中,克林霉素(Clin)与钯(Ⅱ)形成螯合阳离子,它能进一步与二碘荧光素(DIF),赤藓红(Ery),曙红Y(EY)等卤代荧光素类染料反应形成1:1:1的三元离子缔合物,此时将引起吸收光谱变化和荧光猝灭,同时还导致共振瑞利散射(RRS)的急剧增强并产生新的RRS光谱,钯(Ⅱ)-克林霉素与DIF,Ery和EY形成产物的最大散射波长分别位于285,287,32 1nm处,另外还有些较弱的散射峰存在。散射增强(ΔI)与克林霉素浓度在一定范围内成正比,可用于克林霉素的定量测定。对于DIF,Ery和EY体系的线性范围和检出限分别为0.025-2.1μg•mL-1和7.8 ng•mL-1,0.053-2.4μg•mL-1和16.0 ng•mL-1;以及0.038-2.4μg•mL-1和11.0 ng•mL-1。本文研究了适宜的反应条件,考察了共存物质的影响,表明方法有较好的选择性,基于三元离子缔合物的RRS光谱,发展了一种高灵敏、简便快速测定克林霉素的新方法。文中还对离子缔合物的组成,结构和反应机理,以及离子缔合物对吸收,荧光和RRS光谱的影响进行了讨论。  相似文献

A solid-phase extraction method for preconcentration of silver and consequent determination by atomic absorption spectrometry is described. The method is based on the retention of silver on sulfur modified with 2-mercaptobenzoxazole. The retained silver is eluted from the column with a thiourea solution and determined by flame atomic absorption spectrometry. The preconcentration conditions such as pH, amount of reagent loaded on sorbent, type of eluent and its volume, flow rate and interfering ions were investigated. The calibration graph was linear in the range of 3–200 ng mL?1 of Ag+ in the initial solution with r = 0.9985. The limit of detection based on 3Sb was 1.0 ng mL?1. The relative standard deviation for ten replicate measurements of 50 and 150 ng mL?1 of Ag+ was 4.1 and 1.4 %, respectively. The method was applied to the determination of silver in radiology film and water samples.  相似文献

Abstract

A simple, rapid, selective, and sensitive method for the derivative spectrophotometric determination of Hg(II) and its simultaneous determination in the presence of Zn(II) using 2‐(5‐bromo‐2‐pyridylazo)‐5‐diethylaminophenol in the presence of cetylpyridinium chloride, a cationic surfactant, has been developed. The molar absorption coefficient and analytical sensitivity of the 1∶1 Hg(II) complex at 558 nm (λmax) are 5.78×104 L mol?1 cm?1 and 0.67 ng mL?1, respectively. The detection limit of Hg(II) is 1.40×10?2 ng mL?1, and Beer's law is valid in the concentration range 0.05–2.40 µg mL?1. Overlapping spectral profiles of Hg(II) and Zn(II) complexes in zero‐order mode interfere in their simultaneous determination. However, 0.10–2.00 µg mL?1 of Hg(II) and 0.065–0.650 µg mL?1 of Zn(II), when present together, can be simultaneously determined at zero cross point of the derivative spectrum, without any prior separation. The relative standard deviation for six replicate measurements of solutions containing 0.134 µg mL?1 of Hg(II) and 0.620 µg mL?1 of Zn(II) is 1.72 and 1.47%, respectively. The proposed method has successfully been evaluated for trace level simultaneous determination of Hg(II) and Zn(II) in environmental samples.  相似文献

A method for flow injection with chemiluminescence (CL) detection has been developed for the determination of proteins. It is based on the luminescence of the N-bromosuccinimide-fluorescein-protein system, where fluorescein is used as an energy transfer reagent in alkaline medium. The CL of the system is strongly enhanced by hexadecyl trimethyl ammonium bromide. Optimum conditions and possible mechanisms have been investigated. Under optimum experimental conditions, the linear range is from 0.4 to 40 µg·mL?1 for egg albumin, 0.2 to 20 µg·mL?1 for bovine serum albumin, and from 1 to 100 µg·mL?1 for bovine hemoglobin. The detection limits are 37, 62, and 240 ng·mL?1, respectively.  相似文献

The supramolecular interaction of gemfibrozil with β-cyclodextrin (β-CD) was studied by spectrofluorimetry. The mechanism of the inclusion was discussed by spectrofluoremetry, infrared spectrum and ^1H NMR spectrum. The results showed that a 1 : 1 (β-CD : gemfibrozil) complex was formed with an apparent association constant of 3.844 × 10^3 L·mol^-1. Based on the enhancement of the fluorescent intensity of gemfibrozil, a spectrofluorimetric method for the determination of gemfibrozil in bulk aqueous solution in the presence of β-CD was developed. The linear range was 3.30 ng·mL^- 1 -6.00 ug·mL^-1 with the detection limit of 0.980 ng·mL^-1. There was no interference from the excipients normally used in tablet composition and the serum main compositions. The proposed method was then successfully applied to the determination of gemfibrozil in capsules and serum.  相似文献

Abstract

A simple, rapid, and sensitive analytical method has been developed for the determination of two fluoroquinolones, danofloxacin and marbofloxacin, in bovine milk samples. Separation and quantification were performed by micellar liquid chromatography with fluorescence detection (MLC?FD), using sodium dodecyl sulfate (SDS) as a surfactant. The influence of the principal factors, namely, the micelle concentration, the amount of organic modifier, tail‐reducing agents, the pH, and the temperature were studied. The suitable condition was found to be 75 mM SDS?10 mM phosphate buffer–18 mM tetrabutylammonium bromide/3% (v/v) 1‐propanol at pH 3.0 for the separation of marbofloxacin, danofloxacin, and tosufloxacin (internal standard) in about 20 min. The linear concentration range of application was 1.8–30.0 ng · mL?1 for danofloxacin and 16–120 ng · mL?1 for marbofloxacin, and the relative standard deviation ranged between 4.9 and 2.7%. The limit of detection found for danofloxacin was 0.5 ng · mL?1 and 5 ng · mL?1 for marbofloxacin. These values were lower than the maximum residue limits (MRLs) established by the European Union for these compounds in bovine milk. It was applied to check the eventual existence of these compounds above these limits on commercial milk samples. The validation method was completed with spiked milk samples. Recovery levels obtained were 90.3–108.2%.  相似文献

A new solid substrate-room temperature phosphorescence (SS-RTP) quenching method for the determination of trace As(V) has been developed, based on the facts that 9-hydroxy-2,3,4,9-tetrahydro-1,10-anthraquinone (R) can emit intense and stable SS-RTP on solid substrate, and α,α'-dipyridyl can activate As(V) catalysis of the reaction of H2O2 oxidizing R to non-phosphorescence compound R', which can cause the sharp quenching of SS-RTP. Under the optimum condition, the relationship between the ΔIp of the emitting intensity and 1.60-160 fg·spot^-1 As(V) (corresponding concentration: 0.0040-0.40 ng·mL^-1, sample volume: 0.4 μL·spot^-1) conformed to Beer' law. The regression equation of working curve can be expressed as ΔIp= 20.46+0.5492CAs(v) fig·spot^-1) (r= 0.9995, n = 6). The limit detection (LD) is 0.27 fg·spot^-1 [As(V) corresponding concentration: 6.8 × 10^-13 g·mL^-1, n=11]. The samples containing 0.0040 and 0.40 ng·mL^-1 As(V) were repeatedly determined for 11 times. RSD are 3.0% and 2.7% respectively. The SS-RTP mechanism was also discussed. R was synthesized in this paper. Meanwhile, the structure was determined by NMR, IR, mass spectra and elemental analysis.  相似文献

A simple, sensitive, and precise high performance liquid chromatographic method for the analysis of pantoprazole, rabeprazole, esomeprazole, domperidone and itopride, with ultraviolet detection at 210 nm, has been developed, validated, and used for the determination of compounds in commercial pharmaceutical products. The compounds were well separated on a Hypersil BDS C18 reversed-phase column by use of a mobile phase consisting of 0.05 M, 4.70 pH, potassium dihydrogen phosphate buffer - acetonitrile (720:280 v/v) at a flow rate of 1.0 mL min?1. The linearity ranges were 400–4,000 ng mL?1 for pantoprazole, 200–2,000 ng mL?1 for rabeprazole, 400–4,000 ng mL?1 for esomeprazole, 300–3,000 ng mL?1 for domperidone and 500–5,000 ng mL?1 for itopride. Limits of detection (LOD) obtained were: pantoprazole 147.51 ng mL?1, rabeprazole 65.65 ng mL?1, esomeprazole 131.27 ng mL?1, domperidone 98.33 ng mL?1 and itopride 162.35 ng mL?1. The study showed that reversed-phase liquid chromatography is sensitive and selective for the determination of pantoprazole, rabeprazole, esomeprazole, domperidone and itopride using single mobile phase.  相似文献

A kind of 9 nm gold nanoparticles was prepared with the trisodium citrate and used to label goat anti-human IgG to obtain an IgG immunoresonance scattering spectral probe. In pH 5.8 buffer solution and in the presence of polyethylene glycol (PEG), the immune reaction between gold-labeled goat anti-human IgG and IgG took place, and the resonance scattering intensity at 580 nm (I580nm) was enhanced greatly. The enhanced intensity AIRS is pro- portional to the IgG concentration from 1.3 to 1.5 X 10^3 ng.mL^-1, with a detection limit of 0.78 ng.mL ^-1. This assay showed high sensitivity and good selectivity for quantitative determination of IgG in human serum, with satisfactory results.  相似文献

Simple, rapid and reliable method for the determination of albendazole (ABZ) was described. This includes the utility of some Π‐acceptors such as 2,3‐dichloro‐5,6‐dicyano‐p‐benzoquinone (DDQ) and 3,6‐dichloro‐2,5‐dihy‐ droxy‐p‐benzoquinone (p‐CLA) for estimation of ABZ drug (act as donor). The experimental conditions were optimized and the system obeys Beer's law for 7.50–80 and 10.00–85.00 µg·mL?1 of ABZ using DDQ and p‐CLA, respectively. The molar absorptivity and Sandell sensitivity were calculated to be 1.83×103 and 1.12×103 L·mol?1·cm?1, and 2.60 and 3.40 ng·cm?2 using DDQ and p‐CLA, respectively. The limits of detection and quantification were calculated to be (7.42 and 6.73) and (9.94 and 4.13) µg·mL?1 using DDQ and p‐CLA, respectively. The proposed methods were successfully applied to the determination of ABZ in commercially available dosage forms. The reliability of the assays was established by parallel determination by the official method and recovery studies. The chemical structures of the solid charge‐transfer (CT) complexes formed via reaction between ABZ under study and Π‐acceptors, have been elucidated using elemental analyses (C, H and N), IR, 1H NMR and mass spectra.  相似文献

Abstract

A effective and simple determination of poisonous trace element thallium(I) by means of kinetic catalytic reaction is proposed. The method is based on a catalytic effect of thallium(I) on a luminol-hydrogen peroxide system. Three different kinds of surfactants, cetrimonium bromide (CTMAB), sodium dodecyl sulphate (SDS), and Tween-80, are also investigated to improve the detection sensitivity. In optimum conditions, a highly selective and sensitive method for detecting trace thallium(I) has been established. The detection limit is 0.0073 µg · mL?1, the relative standard deviation for six determinations of 0.04 µg · mL?1 thallium(I) is less than 4.0%, and the linear range of determination is 0.02–0.1 µg · mL?1.  相似文献

A sensitive, simple, and accurate method for determination and pharmacokinetic study of ferulic acid and isoferulic acid in rat plasma was developed using a reversed-phase column liquid chromatographic (RP-LC) method with UV detection. Sample preparations were carried out by protein precipitation with the addition of methanol, followed by evaporation to dryness. The resultant residue was then reconstituted in mobile phase and injected into a Kromasil C18 column (250 × 4.6 mm i.d. with 5 μm particle size). The mobile phase was methanol-1% formic acid (33:67, v/v). The calibration plots were linear over the range 5.780–5780 ng·mL?1 for ferulic acid and 1.740–348.0 ng·mL?1 for isoferulic acid. Mean recoveries were 85.1% and 91.1%, respectively. The relative standard deviations (RSDs) of within-day and between-day precision were not above 15% for both of the analytes. The limits of quantification were 5.780 ng·mL?1 for ferulic acid and 1.740 ng·mL?1 for isoferulic acid. This RP-LC method was used successfully in pharmacokinetic studies of ferulic acid and isoferulic acid in rat plasma after intravenous injection of Guanxinning Lyophilizer.  相似文献

A method combining immunoaffinity chromatography with gas chromatography–mass spectrometry (GC–MS) has been established for determination of ractopamine residues in swine liver and urine. After clean-up on an immunoaffinity chromatography column, GC–MS analysis revealed recovery from blank swine liver and urine fortified at 2.5–20 ng g?1 (ng mL?1 for urine), respectively, was 68.2–78.6 and 76.2–83.1%. The limits of detection and quantification were 0.5 ng g?1 (or ng mL?1) and 2.0 ng g?1 (or ng mL?1), respectively. The procedure was used for analysis of ractopamine residues in samples of swine liver and urine in which the levels were unknown. The amounts detected were 9–216 ng g?1 (ng mL?1).  相似文献

Dispersive liquid–liquid microextraction based on solid formation without a disperser combined with high-performance liquid chromatography has been developed for the determination of 4-tert-butylphenol, 4-n-nonylphenol, and 4-tert-octylphenol. This method is rapid, easy, and uses only 10 µL of a low toxicity organic solvent (1-hexadecanethiol) for the extraction solvent and no disperser solvent. The extraction time and centrifugation time require less than 10 min. The linear range was 1–500 ng mL?1 for 4-tert-butylphenol, 2–1000 ng mL?1 for 4-tert-octylphenol, and 5–500 ng mL?1 for 4-n-nonylphenol with r2 ≥ 0.9986. The detection limits were between 0.2 and 1.5 ng mL?1. The recoveries of lake and river water samples were in the range of 79% to 108%, and the relative standard deviations were 5% to 10%.  相似文献

A gradient liquid chromatography-tandem mass spectrometry method has been developed and validated for the determination of gastrodin and ligustrazine hydrochloride in rat plasma and brain dialysates. Zolpidem was used as internal standard. For plasma samples, solid-phase extraction was used and the brain dialysates were collected from freely moving rats using brain microdialysis. Both were followed by HPLC separation and positive electrospray ionization tandem mass spectrometry detection (ESI–MS–MS). Chromatographic separation was achieved on a Symmetry RP-18 column using gradient elution with methanol and water containing 0.5% formic acid and 2 mM ammonium formate. Selected reaction monitoring (SRM) mode was used for quantitation. Good linearities were obtained in the range of 0.05–100 and 0.01–50 μg mL?1 for gastrodin and ligustrazine hydrochloride in rat plasma, and 0.05–1,000 ng mL?1 for both in dialysate. The lower limit of quantitation was 0.01 ng mL?1 for gastrodin and 0.05 ng mL?1 for ligustrazine. The method is precise and reliable and can be applied to pharmacokinetic studies.  相似文献

A simple and sensitive method was developed for the determination of three nonsteroidal anti-inflammatory drugs (NSAIDs)—ibuprofen, naproxen and fenbufen in human plasma. The method involved in column liquid chromatographic separation and chemilumenescence (CL) detection based on the CL reaction of NSAIDs, potassium permanganate (KMnO4) and sodium sulfite (Na2SO3) in sulfuric acid (H2SO4) medium. The chromatographic separation was carried out using a reversed-phase C18 column, which allowed the selective determination of the three medicines in the complicated samples. The special features of the CL detector provided lower LOD for determination than that of existing chromatographic alternatives. The results indicated that the linear ranges were 0.01–10.0 μg mL?1 for ibuprofen, 0.001–1.0 μg mL?1 for naproxen, and 0.01–10.0 μg mL?1 for fenbufen. The limits of detection were 0.5 ng mL?1 for ibuprofen, 0.05 ng mL?1 for naproxen and 0.5 ng mL?1 for fenbufen (S/N = 3). All average recoveries were in the range of 90.0–102.3%. Finally, the method had been satisfactorily applied for the determination of ibuprofen, naproxen and fenbufen in human plasma samples.  相似文献
