A simple and rapid high-performance liquid chromatographic (HPLC) method for determination of residual penicillin G (benzylpenicillin, PCG) in milk was developed. The sample preparation was performed by stirring with ethanol and reacting with 5 M 1,2,4-triazole-mercury (II) chloride solution at 65 degrees C for 10 min followed by an ultra centrifugation step. The HPLC separation was carried out using a Mightysil RP-4GP column, a mobile phase of acetonitrile and 0.1 M phosphate buffer (pH 6.5) (35:65, v/v) and a photo-diode array detector. The average recoveries from spiked PCG (0.004, 0.01, 0.05 and 0.1 microgram/mL) were above 86% with coefficients of variation between 1.2 and 4.5%. The limit of detection was 0.004 microgram/mL. This value corresponds to the maximum residue limit (MRL) in milk (0.004 microgram/mL, EU and Japan). The total time required for the analysis of one sample was below 40 min.  相似文献

A method for the determination/identification of residual sulfadimidine (SDD) in milk and eggs by high-performance liquid chromatography (HPLC) with a photo-diode array detector was developed. The sample preparation was performed by shaking with a mixture of 20% (w/v) trichloroacetic acid-methanol (4:1, v/v) followed by ultra-filtration using Molcut II®. A LiChrospher® 100 RP-8 (e) column and a mobile phase of 4% (v/v) acetic acid solution-acetonitrile (6:4, v/v) were used. The average recoveries from spiked SDD samples were 80.8–88.0% with coefficients of variation of 2.8–5.5%. The limits of detection in milk and eggs were 0.01 μg/mL and 0.01 μg/g, respectively. The total time required for the analysis of one sample was less than 20 min.  相似文献

A sensitive and selective gas chromatography with mass spectrometry method was developed for the simultaneous determination of three organophosphorus pesticides, namely, chlorpyrifos, malathion, and diazinon in three different food commodities (milk, apples, and drinking water) employing solid‐phase extraction for sample pretreatment. Pesticide extraction from different sample matrices was carried out on Chromabond C18 cartridges using 3.0 mL of methanol and 3.0 mL of a mixture of dichloromethane/acetonitrile (1:1 v/v) as the eluting solvent. Analysis was carried out by gas chromatography coupled with mass spectrometry using selected‐ion monitoring mode. Good linear relationships were obtained in the range of 0.1–50 μg/L for chlorpyrifos, and 0.05–50 μg/L for both malathion and diazinon pesticides. Good repeatability and recoveries were obtained in the range of 78.54–86.73% for three pesticides under the optimized experimental conditions. The limit of detection ranged from 0.02 to 0.03 μg/L, and the limit of quantification ranged from 0.05 to 0.1 μg/L for all three pesticides. Finally, the developed method was successfully applied for the determination of three targeted pesticides in milk, apples, and drinking water samples each in triplicate. No pesticide was found in apple and milk samples, but chlorpyrifos was found in one drinking water sample below the quantification level.  相似文献

In this study, a sensitive HPLC‐UV assay was developed and validated for the determination of LASSBio‐1736 in rat plasma with sodium diclofenac as internal standard (IS). Liquid–liquid extraction using acetonitrile was employed to extract LASSBio‐1736 and IS from 100 μL of plasma previously basified with NaOH 0.1 M. Chromatographic separation was carried on Waters Spherisorb®S5 ODS2 C18 column (150 × 4.6 mm, 5 μm) using an isocratic mobile phase composed by water with triethylamine 0.3% (pH 4), methanol and acetonitrile grade (45:15:40, v/v/v) at a flow rate of 1 mL/min. Both LASSBio‐1736 and IS were eluted at 4.2 and 5 min, respectively, with a total run time of 8 min only. The lower limit of quantification was 0.2 μg/mL and linearity between 0.2 and 4 μg/mL was obtained, with an R2 > 0.99. The accuracy of the method was >90.5%. The relative standard deviations intra and interday were <6.19 and <7.83%, respectively. The method showed the sensitivity, linearity, precision, accuracy and selectivity required to quantify LASSBio‐1736 in preclinical pharmacokinetic studies according to the criteria established by the US Food and Drug Administration and European Medicines Agency. An isolating method using a solid-phase extraction (SPE) ISOLUTE® C8 endcapped syringe-column for routine monitoring of residual tetracyclines (TCs) (oxytetracycline (OTC), tetracycline (TC), chlortetracycline (CTC), and doxycycline (DC)) in cow's milk is presented. In the simplest and most environmentally harmless method, milk samples could be applied directly to the SPE column, following which all TCs were eluted with water. No organic solvents were used at all. The purified sample was injected into a high-performance liquid chromatography (HPLC) with a photo-diode array detector (PDAD). For the HPLC determination/identification, a LiChrospher® 100 RP-8 endcapped column and a mobile phase of acetonitrile −7% (v v−1) acetic acid solution (in water) (35:65, v v−1) with a PDAD was used. The total time required for the analysis of one sample was <40 min. Average recoveries (spiked 0.1-1.0 μg ml−1 each drug) and their standard deviations were >80 and <5%, respectively.  相似文献

A simple, sensitive, and selective stability indicating high performance liquid chromatographic method has been developed and validated for quantitative analysis of carprofen (CPF) in presence of its degradation products. All degradation products in acid hydrolysis and photolysis were separated, identified by mass spectroscopic method and probable structures were elucidated. The forced degradation studies were performed on a bulk sample of CPF by using various methods like 0.1 M hydrochloric acid, 0.1 M sodium hydroxide, 0.33% hydrogen peroxide (H(2)O), heating at 60°C and exposure to UV light at 254 nm. A 5 μm particle octa desyl silane (ODS) column (150 mm × 4.6 mm) was used with acetonitrile-ammonium acetate (100 mM, pH-6.7) 40:60 (v/v) as a mobile phase at flow rate of 1.2 mL/min. Column oven temperature was maintained at 30°C and quantitation was achieved at 239 nm on the basis of peak area. The linear range and correlation coefficient (r(2)) was found 0.5-60 μg/mL and 0.9999 respectively. The limit of detection (LOD) and limit of quantitation (LOQ) were obtained 0.066 μg/mL and 0.20 μg/mL respectively . The proposed method was found to be suitable and accurate for quantitative analysis, stability study and characterisation of degradation product of CPF.  相似文献

建立了奶粉和液态奶中三聚氰胺的高效液相色谱(HPLC)检测方法。经阳离子交换固相萃取柱净化后的样品采用HPLC测定。优化的色谱条件:C18柱(4.6 mm ×200 mm,5 μm),流动相为乙腈-0.01 mol/L庚烷磺酸钠(pH 3.3)(体积比为10∶90),流速为1.0 mL/min,检测波长为236 nm,柱温为40 ℃,进样量为20 μL。方法的线性范围为1~500 mg/L,检出限为0.2 mg/kg (S/N=3),定量限为1 mg/kg (S/N=15),回收率为81.4%~83.7%,相对标准偏差为3.3%~8.5%(n=6)。  相似文献

建立了鲫鱼肌肉中残留的辛硫磷的基质固相分散-高效液相色谱-二极管阵列检测(MSPD-HPLC-DAD)的分析方法。通过优化样品处理条件,确定选取0.50 g鲫鱼肌肉样品与1.5 g弗罗里硅土、0.5 g无水硫酸钠混合研磨,并采用丙酮-正己烷溶液(体积比为40:60)为洗脱剂,洗脱剂用量为25 mL。优选的最佳色谱条件为:ODS色谱柱(250 mm×4.6 mm,5 μm),流动相为甲醇-水(体积比为50:50),流速0.6 mL/min,检测波长270 nm,进样量为20 μL。在上述条件下,辛硫磷质量浓度在0.01~10 mg/L范围内与响应信号呈良好的线性关系(r20.9994),检出限为3.3 μg/kg;相对标准偏差为1.1%~6.3%(n7);3个添加水平(0.05,0.1,1 mg/kg)下得到的回收率为88%~112%。该方法操作简单,耗时少,精密度高,符合农残分析的要求。  相似文献

The present study describes the development and validation of a selective liquid chromatographic (LC) method for the analysis of tenofovir disoproxil fumarate (TDF) and its related substances. The gradient method uses a base deactivated C18 column (Hypersil BDS column; 25 cm×4.6 mm I.D.) maintained at a temperature of 30°C. The mobile phases consist of acetonitrile, tetrabutylammonium/phosphate buffer pH 6.0 and water: (A; 2:20:78 v/v/v) and (B; 65:20:15 v/v/v). The flow rate is 1.0 mL/min and UV detection is performed at 260 nm. Good separation of TDF and 21 impurities was achieved. A system suitability test (SST) to check the quality of separation is also specified. The developed method was further validated with respect to robustness, precision, sensitivity and linearity. The method is proved to be robust, precise, sensitive and linear between 0.1 μg/mL and 0.15 mg/mL. The limit of detection and limit of quantification are 0.03 and 0.1 μg/mL, respectively. The method was successfully applied to the quantification of related substances and assay of commercial TDF samples (bulk substances and tablets).  相似文献

A simple and sensitive stability-indicating chiral HPLC method has been developed and validated per International Conference on Harmonization guidelines for the determination of enantiomeric purity of eluxadoline (Exdl). The impact of different mobile phase compositions and chiral stationary phases on the separation of Exdl enantiomer along with process- and degradation-related impurities has been studied. Homogeneity of Exdl and stable results of Exdl enantiomer in all degraded samples reveal the fact that the proposed method was specific (stability indicating). Amylose tris(3,5-dichlorophenyl carbamate) stationary phase column Chiralpak IE-3 (150 × 4.6 mm, 3 μm) provided better resolution with polar organic solvents than cellulose derivative, crown ether, and zwitterion stationary phases and nonpolar solvents. The mobile phase consisted of acetonitrile, tetrahydrofuran, methanol, butylamine, and acetic acid in the ratio of 500:500:20:2:1.5 (v/v/v/v/v). Isocratic elution was performed at a flow rate of 1.0 mL/min, column temperature of 35°C, injection volume of 10 μL, and UV detection of 240 nm. The United States Pharmacopeia (USP) resolution of the Exdl enantiomer was found to be more than 4.0 within a 65-min run time. Exdl enantiomer detector response linearity over the concentration range of 0.859–4.524 μg/mL was found to be R2 = 0.9985. The limit of detection, limit of quantification, and average percentage recovery values were established as 0.283 μg/mL, 0.859 μg/mL, and 96.0, respectively.  相似文献

A comparison between C18 silica and multi‐walled carbon nanotubes (MWCNTs) in the extraction of Chlorpyrifos and Phosalone in environmental water samples was carried out using HPLC. Parameters affecting the extraction were type and volume of elution solvent, pH and flow rate of sample through the adsorbent. The optimum conditions obtained by C18 cartridge for adsorption of these pesticides were 4 mL dichloromethane as elution solvent, sample pH of 5, flow rate of 1 mL/min, and those for MWCNT cartridge were 3 mL dichloromethane, pH of 5 and flow rate of 10 mL/min, respectively. Optimized mobile phase for separation and determination of these compounds by HPLC was methanol/water (80:20 v/v) with pH=5 (adjusted with phosphate buffer). Under optimal chromatographic and SPE conditions, LOD, linear range and precision (RSD n=8) were 3.03×10?3, 0.01–5.00 μg/mL and 2.7% for Chlorpyrifos and 4.03×10?4, 0.01–5.00 μg/mL and 2.3% for Phosalone, in C18 cartridge, respectively. These values for MWCNT were 4.02×10?6, 0.001–0.500 μg/mL and 1.8% for Chlorpyrifos and 1.02×10?6, 0.001–0.500 μg/mL and 1.5% for Phosalone, respectively.  相似文献

A simple, sensitive, stability-indicating HPLC method was developed and validated for the quantitative determination of the vasoprotective drug, naftazone in presence of its degradation products. The analysis was carried out on a Nucleosil 100-5 phenyl column (250 mm × 4.6 mm, 5 μm) using a mobile phase consisting of methanol-0.02 M sodium dihydrogen phosphate mixture (60:40, v/v) of pH 6.0. The analyses were performed at ambient temperature with a flow rate of 1.0 mL/min and UV detection at 270 nm. The method showed good linearity over the concentration range of 0.1-10.0 μg/mL with a lower detection limit of 0.032 and quantification limit of 0.096 μg/mL. The suggested method was successfully applied for the analysis of naftazone in its commercial tablets. Moreover, it was utilized to investigate the kinetics of alkaline, acidic and oxidative degradation of the drug. The apparent first-order rate constants, half-life times, and activation energies of the degradation process were calculated. The pH-rate profile curve was derived. Furthermore, the proposed method was successfully applied to the content uniformity testing of naftazone tablets.  相似文献

A fast and sensitive HPLC method for analysis of cosmetic creams for hydroquinone, phenol and six preservatives has been developed. The influence of sample preparation conditions and the composition of the mobile phase and elution mode were investigated to optimize the separation of the eight studied components. Final conditions were 60% methanol and 40% water (v/v) extraction of the cosmetic creams. A C18 column (100 mm × 2.1 mm) was used as the separation column and the mobile phase consisted of methanol and 0.05 mol/L ammonium formate in water (pH=3.0) with gradient elution. The results showed that complete separation of the eight studied components was achieved within 10 min, the linear ranges were 1.0-200 μg/mL for phenol, 0.1-150 μg/mL for sorbic acid, 2.0-200 μg/mL for benzoic acid, 0.5-200 μg/mL for hydroquinone, methyl paraben, ethyl paraben and propyl paraben, butyl paraben, and good linear correlation coefficient (≥0.9997) were obtained, the detection limit was in the range of 0.05-1.0 μg/mL, the average recovery was between 86.5% and 116.3%, and the relative standard deviation (RSD) was less than 5.0% (n=6). The method is easy, fast and sensitive, it can be employed to analyze component residues in cosmetic creams especially in a quality control setting.  相似文献

In this work, a novel analytical method based on hollow fiber liquid phase microextraction (HF-LPME) and high performance liquid chromatography (HPLC) was developed for the analysis of melamine in fresh milk. The conditions of the HF-LPME were investigated and optimized. As a result, a supported liquid membrane containing 6-undecanone and di-2-ethylhexyl phosphoric acid (D2EHPA) was selected. The extractions were made from 25 mL aqueous donor phase (prepared from milk) with pH 5.0 to a more acidic acceptor phase (36 µL 1 M HCl) and the mass transfer was driven by the proton gradient between these phases. Other optimum conditions of the HF-LPME were 60 min extraction time at 360 rpm stirring rate and an extraction factor of 21 times (extraction efficiency 3%) was obtained. The C8 column was operated at 1 mL/min at room temperature and the UV detection wavelength was 240 nm for HPLC. The mobile phase was 10 mM sodium n-octanesulfonate (pH 3.0) mixed with acetonitrile (85:15, v/v). The relative recovery of melamine for milk samples spiked with 0.5–25 mg/kg was in the range of 89.1–120.6% with the RSDs (n = 4) of 4.0–8.5%. It was found that the proposed method provided a linear range from 0.1 to 50 mg/kg (r 2  = 0.9993), method detection limit (MDL) of 0.003 mg/kg and method quantification limit (MQL) of 0.01 mg/kg. The obtained results demonstrated that HF-LPME combined with HPLC is a simple and cheap method for the determination of melamine in fresh milk.  相似文献

Abstract

A simple and sensitive HPLC method was developed for the determination of naproxen in human plasma. The assay employs a microbore column packed with a C18 reversed-phase material (5 μm ODS Hypersil) with an isocratic mixture of acetonitrile and 10 mM phosphate buffer, pH 2.5 (40:60, v/v) as the mobile phase. The mobile phase was pumped at a flow rate of 0.5 ml/min. For sample analysis 200 μl of acetonitrile containing internal standard (flurbiprofen) was added to 100 μl of plasma. After centrifugation 10 mM phosphate buffer, pH 7.4 (200 μl) was added to the tube, then vortexed and centrifuged. The supernatant (20 μl) was injected onto the HPLC column. The chromatographic separation was monitored by a fluorescence detector at an emission wavelength of 350 nm with an excitation wavelength of 225 nm. The direct precipitation of plasma protein using acetonitrile gave a good recovery for both naproxen and the internal standard. The detection limit was 0.1 μg/ml for naproxen. The intra- and inter-assay coefficients of variation at different concentrations evaluated were less than 10%.  相似文献

A rapid stability indicating reversed-phase high-performance liquid chromatography (HPLC) method is developed for the determination of the electrochemically controlled risperidone release from a novel drug delivery system, based on the intrinsically conducting polymer (ICP), polypyrrole. The chromatographic separation was carried out on a C(18) column using acetonitrile-potassium dihydrogen phosphate (45:55, v/v, pH 6.5; 0.05 M) as the mobile phase. The isocratic flow is at 1.0 mL/min, with a runtime of 6 min, and the UV detection is at 237 nm. This provided a calibration curve linear over the range of 1-100 μg/mL. Intra-day and inter-day accuracy range between 98.4% and 102.6%, and the RSD for precision is <1.43%. The limit of detection and quantitation were determined to be 0.001 μg/mL and 0.01 μg/mL, respectively. The analytical method confirmed risperidone is stable for the oxidizing electric potential and the acidic environment involved during the manufacture and operation of the novel drug delivery system. The rate of risperidone release from polypyrrole depended on electrical stimulation applied to the polymer. This HPLC method is significantly faster than previously published methods and is the first to investigate the effect of an oxidizing potential on risperidone stability, which is essential for the evaluation of controlled delivery from an ICP-based system.  相似文献

A new HPLC method was developed and validated for the determination of asymmetric and symmetric dimethylarginines and l ‐arginine in human plasma. After SPE and evaporation of the eluate, the samples were derivatised with an o‐phthaldialdehyde reagent containing 3‐mercaptopropionic acid. The derivatives formed were analysed by isocratic RP‐HPLC with electrochemical detection at +320 mV. The mobile phase consisted of 50 mM phosphate buffer (pH 6.1) containing 10% v/v acetonitrile, the flow rate was 1 mL/min. The retention times of all compounds including monomethylarginine (internal standard) were <24 min. The LODs (S/N 3:1) were 0.012 μM for both dimethylarginines and 0.013 μM for l ‐arginine; the linearity of the method was from 0.1 to 20 μM for both dimethylarginines and from 1 to 200 μM for l ‐arginine. Absolute extraction recoveries measured for all analytes ranged from 85 to 88%.  相似文献

A fast liquid chromatography-tandem mass spectrometry (LC-MS-MS) method is developed to determine lincomycin (LM) in honey, muscle, milk, and egg. Samples are cleaned-up at pH 4.7 using Strata-X-C mixed-mode polymeric strong cation exchange solid-phase extraction (SPE) cartridges, which could selectively adsorb the lincomycin from matrices under the acidic condition. LM is separated on the recently introduced Kinetex XB core-shell type HPLC column using isocratic elution mode with a mobile phase containing 0.1% formic acid in water/acetonitrile (93/7, v/v, pH 2.6) at a flow rate of 0.7 mL/min. The subsequent MS/MS detection has decreased ion effect, which allows the limit of detection (LOD) of LM for honey to be 0.05 μg/kg for honey and 0.5 μg/kg for muscle, milk, and egg. These LODs are much lower than those reported previously. The other main advantage of the developed method is the analysis time of only 3.5 min, which is about three times shorter than other reported LC-MS-MS methods. Recoveries varies between 94.2% and 125.2% and in-house reproducibility ranges from 3.7% to 28.7%. The developed method is validated according to European Union (EU) Commission Decision 2002/657/EC using a matrix-comprehensive validation strategy. All studied analytical parameters fulfills the EU guidelines.  相似文献

A simple, rapid and sensitive high-performance liquid chromatography (HPLC) method has been developed for the determination of triptolide. Triptolide was separated from skin endogenous and blank matrices on a 5 μm LiChrospher RP-C18 column by a mobile phase of methanol-water (65:35, v/v). The permeation samples were injected directly without pretreatment. The limit of quantitation (LOQ) and detection (LOD) for triptolide in permeation samples were far below (0.01 and 0.005 μg/mL, respectively). The method was linear over the range of 0.1-104.2 μg/mL with r2 = 0.9999. This HPLC assay is promising for measuring in vitro percutaneous penetration of triptolide through mice skins and also can be performed in the triptolide-loaded microemulsions formulation screening.  相似文献
