In the present study we report on the optimization and validation of a sensitive high performance liquid chromatography atmospheric pressure chemical ionization mass spectrometry (HPLC–APCI–MS) method for the determination of 8 bioactive amines (histamine, tyramine, tryptamine, 2-phenylethylamine, cadaverine, putrescine, spermidine and spermine) in donkey milk samples. The method involves donkey milk pre-treatment to remove proteins and pre-column dansylation of the amines. HPLC in reversed phase mode has been used for bioactive amines separation and the operating condition of the APCI–MS system proved to be powerful and very efficient for peak assignment. The separation was accomplished in a short time with an excellent resolution for all the amine peaks. Quantification was carried out by monitoring the characteristic [M+H]+ ion of each amine derivative. The method sensitivity, linearity and repeatability were assayed with satisfactory results. The detection limits of the analysed amines ranged from 0.5 μg L−1 to 15 μg L−1; the highest LOD was for spermine. Also remarkably good recovery values were obtained; at the lowest spiking level (1 μg L−1) the percent mean recoveries ranged from 77.7 to 109.7. Furthermore, as the investigations relate to a complex matrix as donkey milk, suitable studies on matrix effect were performed. Finally, the developed and validated method was applied to analyse 13 donkey milk samples. Among the identified bioactive amines, putrescine, spermine and spermidine proved to be the main amines in donkey milk. Their concentration levels in the present study were lower than the values determined in mature human, cow and sow milk.  相似文献

This paper presents a new LC method with evaporative light scattering detection (ELSD), for the separation and determination of the biogenic amines (histamine, spermidine, spermine, tyramine, putrescine and β-phenylethylamine) which are commonly present in cheese, as their presence and relative amounts give useful information about freshness, level of maturing, quality of storage and cheese authentication. The LC-ELSD method is validated by comparison of the results with those obtained through LC-UV determination, based on a pre-column dansyl chloride derivatisation step. The obtained data demonstrate that both methods can be interchangeably used for biogenic amines determination in cheese. The new LC-ELSD method shows good precision and permits to achieve, for standard solutions, limit of detection (LOD) values ranging from 1.4 to 3.6 mg L−1 and limit of quantitation (LOQ) values ranging from 3.6 to 9.3 mg L−1. The whole methodology, comprehensive of the homogenization-extraction process and LC-ELSD analysis, has been applied in the analysis of a typical Calabria (Southern Italy) POD cheese, known as Caciocavallo Silano. The most aboundant amine found was histamine, followed, in decreasing order, by tyramine, spermine, putrescine, β-phenylethylamine and spermidine, for a total amount of 127 mg kg−1. This value does not represent a possible risk for consumer health, according to the toxicity levels reported in literature and regarded as acceptable.  相似文献

Biogenic amines (BAs) are used for identifying spoilage in food. The most common are tryptamine (TRY), 2-phenylethylamine (PHE), putrescine (PUT), cadaverine (CAD) and histamine (HIS). Due to lack of chromophores, chemical derivatization with dansyl was employed to analyze these BAs using high performance liquid chromatography with a diode array detector (HPLC-DAD). However, the derivatization reaction occurs with any primary or secondary amine, leading to co-elution of analytes and interferents with identical spectral profiles, and thus causing rank deficiency. When the spectral profile is the same and peak misalignment is present on the chromatographic runs, it is not possible to handle the data only with Multivariate Curve Resolution and Alternative Least Square (MCR-ALS), by augmenting the time, or the spectral mode. A way to circumvent this drawback is to receive information from another detector that leads to a selective profile for the analyte. To overcome both problems, (tri-linearity break in time, and spectral mode), this paper proposes a new analytical methodology for fast quantitation of these BAs in fish with HPLC-DAD by using the icoshift algorithm for temporal misalignment correction before MCR-ALS spectral mode augmented treatment. Limits of detection, relative errors of prediction (REP) and average recoveries, ranging from 0.14 to 0.50 µg mL−1, 3.5–8.8% and 88.08%–99.68%, respectively. These are outstanding results obtained, reaching quantification limits for the five BAs much lower than those established by the Food and Agriculture Organization of the United Nations and World Health Organization (FAO/WHO), and the European Food Safety Authority (EFSA), all without any pre-concentration steps. The concentrations of BAs in fish samples ranged from 7.82 to 29.41 µg g−1, 8.68–25.95 µg g−1, 4.76–28.54 µg g−1, 5.18–39.95 µg g−1 and 1.45–52.62 µg g−1 for TRY, PHE, PUT, CAD, and HIS, respectively. In addition, the proposed method spends less than 4 min in an isocratic run, consuming less solvent in accordance with the principles of green analytical chemistry.  相似文献

A high-performance liquid chromatography method is described for the simultaneous determination of the biogenic amines tryptamine, 2-phenylethylamine, putrescine, cadaverine, histamine, tyramine, spermidine, and spermine in cheese. The optimization of the procedure for the extraction of amines from the matrix is described. The separation of dansyl derivatives of the amines was achieved by reversed-phase liquid chromatography with gradient elution, followed by UV detection at 254 nm. The mobile phase was acetonitrile-0.01 M phosphate buffer (pH 7)-water. Under these conditions, rapid elution of the amines in less than 13 min was obtained. Validation of the method included calibration experiments, addition of standard amines for the determination of amine recoveries and repeatability tests.  相似文献

Hollow fibre liquid-phase microextraction with in situ derivatization using dansyl chloride has been successfully developed for the high-performance liquid chromatography-ultraviolet (HPLC-UV) determination of the biogenic amines (tryptamine, putrescine, cadaverine, histamine, tyramine, spermidine) in food samples. Parameters affecting the performance of the in situ derivatization process such as type of extraction solvent, temperature, extraction time, stirring speed and salt addition were studied and optimized. Under the optimized conditions (extraction solvent, dihexyl ether; acceptor phase, 0.1 M HCl; extraction time, 30 min; extraction temperature, 26 °C; without addition of salt), enrichment factors varying from 47 to 456 were achieved. Good linearity of the analytes was obtained over a concentration range of 0.1–5 μg mL−1 (with correlation coefficients of 0.9901–0.9974). The limits of detection and quantification based on a signal-to-noise ratio of 3–10, ranged from 0.0075 to 0.030 μg mL−1 and 0.03 to 0.10 μg mL−1, respectively. The relative standard deviations based on the peak areas for six replicate analysis of water spiked with 0.5 μg mL−1 of each biogenic amine were lower than 7.5%. The method was successfully applied to shrimp sauce and tomato ketchup samples, offering an interesting alternative to liquid–liquid extraction and solid phase extraction for the analysis of biogenic amines in food samples.  相似文献

Biogenic amines in wine usually are analyzed by high-performance liquid chromatography after direct derivatization. A method of isolation based on solid-phase extraction (SPE) with mixed-mode resins (Oasis MCX, reverse-phase and ion exchange) was developed. The different stages of the isolation process (loading, elution and washing) were optimized to obtain a simple procedure that yields a clean chromatogram. The relative standard deviation (%RSD) of the retention times and relative areas was less than 0.3% and 6%, respectively. Limits of quantification were lower than 0.16 mg L−1 for all the amines and the linear range of concentration was 0.16–8 mg L−1 for putrescine, cadaverine and tyramine, and up to 10 mg L−1 for histamine.  相似文献

Summary The fluorogenic reagent 2-naphthyloxycarbonyl chloride (NOC-Cl) has been used for the automated precolumn derivatization of biogenic amines (BAs) at ambient followed by liquid-chromatographic separation of the derivatives formed. For optimized derivatization samples in 0.5 M borate buffer (pH 9.0) were derivatized with 5 mM NOC-Cl in acetonitrile (MeCN) for 3 minutes. Excess of reagent was scavenged by addition of 20 mM glycine in water. For HPLC a Superspher? RP-18e column and gradient elution using 0.1 M sodium acetate buffer (pH 4.4) and MeCN were used. The NOC-derivatives were detected by fluorescence at an emission wavelength of 335 nm at an excitation wavelength of 274 nm. This method allows the detection of BAs (2-phenylethylamine, putrescine, histamine, cadaverine, tyramine, spermidine, spermine) found in food and beverages (fruit juices, wines, various vinegars, fermented cabbage juice, and salmon). Detection limit of BAs are approximately 49–113 μg kg−1 with the exception of histamine (747 μg kg−1) (injected amounts: 18–41 pg histamine 267 pg), at a signalto-noise ratio of 3:1. The limits of determination are approximately 82–189 μg kg−1 (histamine 1245 μg kg−1) at a signal-to-noise ratio of 5:1. The correlation coefficients of linearity are 0.9910–0.9976. A fast and reliable ultra-performance liquid chromatography (UPLC™) method for the determination of biogenic amines (ethanolamine, methylamine, agmatine, histamine, dimethylamine, ethylamine, octopamine, pyrrolidine, dopamine, isopropylamine, propylamine, tyramine, putrescine, butylamine, cadaverine, tryptamine, 2-phenylethylamine, 3-methylbutylamine, spermidine, spermine) in cheese was established. After pre-column derivatization with 6-aminoquinolyl-N-hydroxy-succinimidyl carbamate (AQC), 20 primary and secondary biogenic amines were separated on an Acquity™ UPLC™ column (BEH C18, 1.7 μm; 2.1 mm × 50 mm) within 9 min. Limits of detection (mg/100 g cheese) ranged from 0.04 (ethanolamine) to 1.62 (spermine), and limits of quantification were between 0.16 (ethanolamine) and 6.09 (spermine). The UPLC™ method was applied to the analysis of 58 cheese samples as retailed in Austria. About 13.8% of samples had a histamine content above 10 mg/100 g, and 22.4% had a tyramine content above 10 mg/100 g. Moreover, 8.6% of samples had a putrescine or cadaverine content higher than 10 mg/100 g. The total concentration of biogenic amines in two cheese samples was about 194 mg/100 g. Thus, obligatory monitoring of biogenic amines should be considered to ensure quality of cheese in future.  相似文献

Summary Biologically active amines (putreanine sulphate, N-acetyl putrescine, putrescine, cadaverine, histamine, agmatine, N-acetyl spermidine, spermidine, spermine) were separated and quantified in cereal flour and cereal products by a liquid chromatographic method. The method consists of the separation of ion pairs formed between biologically active amines and octanesulphonic acid on a reversed-phase column, postcolumn derivatization with o-phtalaldehyde-2-mercapthoethanol and spectrofluorometric detection. Results of the reliability study were satisfactory. The method was linear for each amine at 1–10 mg L−1. Putrescine and spermidine were the only amines always detected in cereal flour and cereal products, ranging from 2.45 to 47.83 mg kg−1 for putrescine and 3.27 to 37.14 mg kg−1 for spermidine. The most important differences among types of samples were found in polyamine derivatives. This work describes a methodology for Cd and Pb determination in sewage sludge slurry samples using thermospray flame furnace atomic absorption spectrometry (TS-FF-AAS). The TS-FF-AAS system was equipped with a Ni tube atomizer placed on an oxidizing air/acetylene flame. Two multivariate calibration models based on partial least squares (PLS) were proposed using total peak profiles (recorded during 57 s). Metals concentration in the sewage sludge samples were from 1.87 to 6.26 mg kg−1 for Cd and from 101 to 327 mg kg−1 for Pb. The limits of detection and quantification were, respectively, 0.2 and 0.7 μg kg−1 for Cd and 8 and 26 μg kg−1 for Pb. These values were three times lower than the limits found when these metals were calibrated using linear calibration with aqueous standard solutions.  相似文献

Biogenic amines, and putrescine and cadaverine in particular, have significant importance in the area of food quality monitoring, and are also potentially important markers of infection, for cancer, diabetes, arthritis and cystic fibrosis. A thermal desorption-gas chromatograph-heated differential mobility spectrometer was constructed and the significant effect of interactions between cell temperature and dispersion field strength on the observed responses studied. The experiment design was a Box-Wilson central composite design (CCD) over the levels of 10-24 kV cm−1 for dispersion field strength and 100-130 °C for cell temperature. The optimum values were estimated to be 16.22 kV cm−1 and 116 °C for putrescine and 14.78 kV cm−1 and 112 °C for cadaverine, respectively with an ammonia dopant at 19 mg m−3.An amine test atmosphere generator was constructed and produced stable concentrations of putrescine (7 mg m−3) and cadaverine (4 mg m−3) vapours at 50 ± 0.5 °C. Tenax TA-Carbotrap adsorbent tubes were used to sample putrescine and cadaverine vapour standards and a linear response function over the range of sample masses 5-20 ng was obtained at 15.0 kV cm−1 115 °C, with a R2 of 0.99 for both putrescine and cadaverine. The sample mass at the limit of detection was estimated to be 3 ng for putrescine and cadaverine. Preliminary data from sampling the headspace of chicken meat revealed a 62% increase in the recovered masses of putrescine from 0.84 to 1.36 ng in the sampled air.  相似文献

The paper summarises the findings of recent studies carried out to assess the levels of pesticide residues in water, sediment, soil and some biota collected from different parts of Tanzania. Although the intention is to cover the whole country, so far the studies have focused on areas with known large-scale pesticide use (Southern Lake Victoria and its basin, TPC sugar Plantations in Kilimanjaro region, Dar es Salaam coast, Mahonda-Makoba basin in Zanzibar) and a former pesticide storage area at Vikuge Farm in Coast region). Analysis of the cleaned extracts in GC-ECD/NPD revealed the dominance of organochlorines in all samples. Generally, low levels of residues were found in areas associated with agricultural pesticide use but the levels in the former storage areas were substantially high. DDT and HCH were dominant in all the studied areas. In the former areas, levels of ∑DDT in water, sediments and soil were up to 2 μg L−1, 700 μg kg−1 and 500 μg kg−1, respectively, while those of ∑HCH were up to 0.2 μg L−1, 132 μg kg−1 and 60 μg kg−1, respectively. The levels in aquatic biota were much higher than those in the water most likely due to bioaccumulation. In the former storage area at Vikuge the levels of pesticides in the topsoil were alarmingly high. Their concentrations were up to 282,000 mg kg−1 dry weight for ∑DDT and up to 63,000 mg kg−1 for ∑HCH. A herbicide, pendimethalin [N-(1-ethylpropyl)-2,6-dinitro-3,4-xylidine], was also found at concentrations up to 41,000 mg kg−1 dry weight. Thus the total pesticide content in the soil was almost 40%. Following these findings the area is now earmarked to be a demonstration site for a proposed GEF project ‘Bioremediation of POPs impacted soils in East Africa’.  相似文献

In the present article we report our results on the development of a selective automated method for the determination of histamine in seafood using the concept of zone fluidics. The method is based on the sequential on-line reaction of the analyte with o-phthalaldehyde in the absence of a nucleophilic reagent, followed by acidification. The careful selection of the chemical and instrumental variables enabled the determination of the analyte with adequate sensitivity at the low micromolar level and with specificity against other biogenic amines and amino acids such as histidine. The LOD was 0.05 μmol L−1 (0.6 mg kg−1) and linearity was obeyed in the range of 0.5–15 μmol L−1 (5.5–170 mg kg−1). The proposed method offers a satisfactory sampling rate of 15 h−1 and adequate accuracy and precision for the analysis of seafood products after minimum sample preparation and without employing a separation technique.  相似文献

A new procedure for determination of biogenic amines (BA): histamine, phenethylamine, tyramine and tryptamine, based on the derivatization reaction with 2-chloro-1,3-dinitro-5-(trifluoromethyl)-benzene (CNBF), is proposed. The amines derivatives with CNBF were isolated and characterized by X-ray crystallography and 1H, 13C, 19F NMR spectroscopy in solution. The novelty of the procedure is based on the pure and well-characterized products of the amines derivatization reaction. The method was applied for the simultaneous analysis of the above mentioned biogenic amines in wine samples by the reversed phase-high performance liquid chromatography. The procedure revealed correlation coefficients (R2) between 0.9997 and 0.9999, and linear range: 0.10–9.00 mg L−1 (histamine); 0.10–9.36 mg L-1 (tyramine); 0.09–8.64 mg L−1 (tryptamine) and 0.10–8.64 mg L−1 (phenethylamine), whereas accuracy was 97%–102% (recovery test). Detection limit of biogenic amines in wine samples was 0.02–0.03 mg L−1, whereas quantification limit ranged 0.05–0.10 mg L−1. The variation coefficients for the analyzed amines ranged between 0.49% and 3.92%. Obtained BA derivatives enhanced separation the analytes on chromatograms due to the inhibition of hydrolysis reaction and the reduction of by-products formation.  相似文献

Rapid analytical method for the simultaneous separation and determination of amines and organic acids is a vital interest for quality control of citrus and their products. In the present study, a simultaneous high performance liquid chromatography (HPLC) method for the rapid separation of three amines and two organic acids was developed. Chromatographic separation of compounds was achieved using Xbridge C18 column at ambient temperature, with an isocratic mobile phase of 3 mM phosphoric acid at a flow rate of 1.0 mL min−1. A photodiode array (PDA) detector was used to monitor the eluent at 223 nm and 254 nm with a total analysis time of 10 min. Extraction of amines and organic acids from citrus juice was optimized. The method was validated by tests of linearity, recovery, precision and ruggedness. The limit of detection (LOD) and limit of quantification (LOQ) for amines and ascorbic acid were determined to be 5 ng and 9.8 ng, respectively. All calibration curves showed good linearity (R2 ≥ 0.9999) within the test ranges. The recoveries of the amines and organic acids ranged between 84% and 117%. The identity of each peak was confirmed by mass spectral (MS) analysis. The developed method was successfully applied to analyze the content of amines and organic acids in six different species and two varieties of citrus. Results indicate that mandarin and Marrs sweet orange contain high level of amines, while pummelo and Rio Red grapefruit had high content of ascorbic acid (137-251 μg mL−1) and citric acid (5-22 mg mL−1). Synephrine was the major amine present in Clementine (114 μg mL−1) and Marrs sweet orange (85 μg mL−1). To the best of our knowledge, this is the first report on simultaneous separation and quantification of amines and organic acids in Marrs sweet orange, Meyer lemon, Nova tangerine, Clementine, Ugli tangelo and Wekiwa tangelo.  相似文献

Summary In the last few years special attention has been paid to the pre-column derivatization of biogenic amines with dabsyl chloride because proper experimental conditions for this reaction are very important. In this study, an experimental design (Doehlert design) was used to optimize the variables involved in the dabsylation of the following amines: histamine, tyramine, phenylethylamine, tryptamine, cadaverine, putrescine, spermidine, and spermine. The optimum experimental conditions for forming the dabsyl derivatives are: reagent concentration, 1.75.10−3 M; pH, 8.2; temperature, 70°C; heating time (t h ), 21 min. Under these conditions good chromatographic repeatability is obtained.  相似文献

建立了一种利用胶束电动毛细管色谱同时检测鱼肉中组胺、腐胺、2-苯乙基胺、尸胺、色胺、亚精胺及精胺7种生物胺的方法。样品经6%过氯酸萃取后,由苯甲酰氯衍生化,以含0.06 mol/L脱氧胆酸钠的0.02 mol/L硼酸(pH 9.2)-甲醇(体积比为95∶5)混合液为电泳介质,电泳电压25 kV,温度25 ℃,检测波长214 nm,在12 min内实现了7种生物胺的完全分离。7种生物胺的浓度与其峰面积在一定的范围呈良好的线性关系,检出限除组胺为15 μg/g外,其余均为5 μg/g。迁移时间和峰面积的日内、日间相对标准偏差均小于5%。该法用于海鱼中7种胺类物质含量的测定,结果令人满意。  相似文献

This study aims at the possibility of single structured paramagnetic microparticles (PMPs), composed of maghemite (γ-Fe2O3) core modified with chitosan called MAN8, or tetraethyl orthosilicate covered with Dowex called MAN35, to be helpful for isolation of biogenic amines prior to their further analysis. Primarily, we synthesized and characterized PMPs. To obtain the information about bead morphology, scanning electron microscopy was employed. Furthermore, X-ray fluorescence was employed to carry out the elemental composition analyses. To obtain further insight into interaction between PMP surface and biogenic amines, scanning electron microscope was employed. It was shown that binding of biogenic amines causes increase of relative current response of deprotonated microparticles. We tested the specificity of PMPs to bind biogenic amines on histamine, tyramine, spermine, spermidine, putrescine, and cadaverine. We found that two types of our PMPs were able to selectively bind spermidine, cadaverine, and histamine in the case of MAN35; and histamine, tyramine, and putrescine in the case of MAN8. Finally, we carried out the analyses of real samples obtained from patients suffering from prostate carcinoma, where histamine was determined as the most abundant biogenic amine (10.456–13.654 µg mL−1). The prepared PMPs were able to isolate the biogenic amines from real samples, and thus they may be helpful in construction of biosensors, or Lab-on-a-Chip platforms, enabling less painful, and more rapid diagnosis of prostate cancer.

A sorbent material based on a newly synthesized hydrazone ligand, 4-hydroxy-N′-[(E)-(2-hydroxyphenyl)methylidene]benzohydrazide was prepared by immobilizing the ligand into a silica sol-gel matrix. The capability of the sorbent material for the extraction of seven biogenic amines (BAs), i.e., tryptamine (TRY), β-phenylethylamine (PEA), putrescine (PUT), cadaverine (CAD), histamine (HIS), tyramine (TYR), and spermidine (SPD) was studied. Under the adopted conditions, the sorbent showed good selectivity towards PUT, CAD, HIS and SPD (% extraction (%E) > 96) while %E for TYR, TRY and PEA were 82.0, 78.9 and 46.4%, respectively. The sorbent could be used up to six extraction cycles for SPD, CAD and PUT and was applied to the determination of food samples (“budu”, ketchup, orange juice, soy sauce) that were spiked with 20 mg L−1 of the BAs. The extracted analytes were derivatized with dansyl chloride before the HPLC determination. With the exception of HIS and TYR in “budu” sample, reasonable recoveries were found for the other analytes in all the tested food samples.  相似文献

The need to monitor biogenic amines levels is essential for many areas of the food industry for two main reasons: the caustic nature and potential toxicity of these amines, and the potential to use amine levels as markers for freshness and quality in foodstuffs. Optimised analysis conditions used for the determination of biogenic amines derivatised with 2-napthyloxycarbonyl chloride has been applied to different pet food samples to assess the effectiveness of this method for complex sample matrices. Further to this, the use of high-resolution mass spectrometry has enabled the previously unconfirmed derivatised form of seven biogenic amines to be established. The derivatised forms identified include as mono substituted (tryptamine and histamine), bisubstituted (putrescine, cadaverine and tyramine), trisubstituted (spermidine) and tetrasubstituted (spermine). The methodology of biogenic amine determination was performed successfully to a range of pet food products highlighting the applicability to a variety of complex sample matrices.  相似文献
