Chitosan (Cs) mats obtained by electrospinning are potentially ideal scaffolds for tissue engineering. This technique allows obtaining nanometric fibrous structures with preferred orientation, which in turn enable cells to align themselves and produce extracellular matrix along desired orientations. In this study, we fabricated aligned Cs electrospun nanofiber mats and investigated the role of the amino acid l-Arginine (L-Arg) as stabilizing agent. Morphological, chemical, mechanical and biological characterizations were performed on untreated and L-Arg treated nanofibrous mats showing the role of L-Arg as biomimetic stabilizer. L-Arg acts as chemical stabilizer of nanofibrous mats, providing improved wettability behavior, mechanical properties and stability even after 60 days in aqueous medium in comparison to untreated mats. Moreover, preliminary biological tests demonstrated favorable cell-material interactions implying physiological responses in terms of viability and proliferation. The proposed L-Arg-treated Cs mats can be considered as potential scaffolds for highly oriented tissue patterning.  相似文献

Chitosan or poly(N-acetyl-d-glucosamine-co-d-glucosamine) with a degree of deacetylation of about 85% was fabricated into nanofibrous membranes by electrospinning from 7% w/v chitosan solution in 70:30 v/v trifluoroacetic acid/dichloromethane solvent system. The obtained fibers were smooth without the presence of beads. The diameters of the individual fiber segments were 126 ± 20 nm. The potential for use of the electrospun chitosan nanofibrous membranes as substrates for cell/tissue culture was evaluated with four different cell types, i.e., Schwann cells, osteoblast-like cells, keratinocytes and fibroblasts, in terms of the attachment and the proliferation of the cells as well as the morphology of the seeded and the cultured cells. The results were compared with the corresponding solvent-cast films. Both types of the chitosan substrates supported the attachment and, at the same time, promoted the largest increase in the viability of the cultured keratinocytes. The viability of Schwann cells cultured on these substrates increased marginally well, but the attachment of the cells on the surfaces was relatively poor. Finally, both types of the chitosan substrates showed cytostatic property towards both osteoblast-like cells and fibroblasts, despite the convincingly good attachment of osteoblast-like cells on the surfaces.  相似文献

We report the fabrication of multiwalled carbon nanotube (MWCNT)-incorporated electrospun polyvinyl alcohol (PVA)/chitosan (CS) nanofibers with improved cellular response for potential tissue engineering applications. In this study, smooth and uniform PVA/CS and PVA/CS/MWCNTs nanofibers with water stability were formed by electrospinning, followed by crosslinking with glutaraldehyde vapor. The morphology, structure, and mechanical properties of the formed electrospun fibrous mats were characterized using scanning electron microscopy, Fourier transform infrared spectroscopy, and mechanical testing, respectively. We showed that the incorporation of MWCNTs did not appreciably affect the morphology of the PVA/CS nanofibers; importantly the protein adsorption ability of the nanofibers was significantly improved. In vitro cell culture of mouse fibroblasts (L929) seeded onto the electrospun scaffolds showed that the incorporation of MWCNTs into the PVA/CS nanofibers significantly promoted cell proliferation. Results from this study hence suggest that MWCNT-incorporated PVA/CS nanofibrous scaffolds with small diameters (around 160 nm) and high porosity can mimic the natural extracellular matrix well, and potentially provide many possibilities for applications in the fields of tissue engineering and regenerative medicine.  相似文献

Electrospinning has been extensively used to construct tissue‐engineered scaffolds because of its ability to provide the fibrous scaffold with structurally analogous to the naturally occurring protein in the extracellular matrix of native tissues. In addition, the modification of scaffolds with bioactive molecules is beneficial as this can create an environment that consists of biochemical cues to further promote cell adhesion, proliferation, and differentiation. In the present contribution, we prepared and investigated the potential used of aligned electrospun poly(3‐hydroxybutyrate) (PHB) scaffold immobilized with bioactive molecule to serve as nervous scaffold. Laminin was successfully immobilized on the surface using covalent binding between functional groups of modified scaffolds and protein. The ability to use for neural regeneration was evaluated in vitro towards murine neuroblastoma Neuro2a cell line and mouse brain‐derived neural stem cells. The surface modification with laminin immobilized on the PHB fibrous scaffolds supported the attachment and promoted the proliferation of Neuro2a very wells. Despite the good attachment and proliferation of Neuro2a and mouse brain‐derived neural stem cells were not able to proliferate on the neat PHB, hydrolyzed PHB and laminin immobilized on hydrolyzed PHB fibrous scaffold.  相似文献

Electrospun nanofibers are of the same length scale as the native extracellular matrix and have been extensively reported to facilitate adhesion and proliferation of cells and to promote tissue repair and regeneration. With a primary focus on tissue repair and regeneration using electrospun scaffolds, only a few studies involved electrospun nanofiber scaffolds directing cell behaviors have been reported. In this study, we prepared electrospun nanofiber scaffolds with distinct fiber configurations, namely, random and aligned orientations of nanofibers, as well as oriented yarns, and investigated their effects on cell behaviors. Our results showed that these scaffolds supported good proliferation and viability of murine fibroblasts. Fiber configuration profoundly influenced cell morpho-logy and orientation but showed no effects on cell proliferation rate. The yarn scaffold had comparable total protein accumulation with the random and aligned scaffolds, but it supported a greater pro-liferation rate of fibroblasts with significantly elevated collagen de-position due to its porous fibrous configuration. Cell-seeded yarn scaffolds showed a greater Young's modulus compared with cell-free controls as early as 1 week. Together with its unique fiber configuration similar to the native extracellular matrix of the myocardium, the yarn scaffold might be a suitable matrix material for modeling cardiac fibrotic disorders.  相似文献

In the development of tissue engineering scaffolds, the interactions between material surface and cells play crucial roles. The biomimetic 3‐D scaffolds absolutely provide better results for fulfilling requirements such as porosity, interconnectivity, cell attachment and proliferation. In this study, 3‐D electrospun scaffolds were prepared by using an electrospinning technique. Photo cross‐linkable polyvinyl alcohol was used as a polymeric matrix. During the electrospinning, the nanofibers were cross‐linked with in situ ultraviolet radiation. The crosslinked polymer fibers were achieved in a simple process at a single step. Nanofiber surface was modified with collagen by a chemical approach. The chemical structures were proven by attentuated total reflectance Fourier transform infrared spectroscopy and proton nuclear magnetic resonance. The surface morphology of the nanofibers was characterized by scanning electron microscope (SEM). Morphological investigations show that the resulting nanofibrous matrix has uniform morphology with a diameter of 220–250 nm. In vitro attachment and growth of 3T3 mouse fibroblasts and human umbilical vein endothelial cells (ECV304) cells on polyvinyl alcohol‐based nanofiber mats were also investigated. Cell attachment, proliferation, and methylthiazole tetrazolium cytotoxicity assays indicated good cell viability throughout the culture time, which was also confirmed by SEM analysis. Inducing the formation of new blood vessels (angiogenesis) is an essential requirement for successful tissue engineering. Approaches have been proposed to enhance angiogenesis using growth factors and other biomolecules; however, this approaches present drawbacks in terms of high cost and patient safety. Copper is known to effectively regulate angiogenesis and can offer a more cost‐effective alternative than the direct use of growth factors. With this study, a strategy to incorporate copper in electrospun fibrous scaffolds with pro‐angiogenic properties is presented. Polycaprolactone (PCL) and copper(II)‐chitosan are electrospun using benign solvents. The morphological and physicochemical properties of the fiber mats are investigated through scanning electron microscopy (SEM), static contact angle measurements, energy dispersive X‐ray, and Fourier‐transform infrared spectroscopies. Scaffold stability in phosphate buffered saline at 37 °C is monitored over 1 week. A bone marrow stromal cell line (ST‐2) is cultured for 7 days and its behavior is evaluated using SEM, fluorescence microscopy and a tetrazolium salt‐based colorimetric assay. Results confirm that PCL/copper(II)‐chitosan is suitable for electrospinning. The fiber mats are biocompatible and favor cell colonization and infiltration. Most notably, the angiogenic potential of PCL/copper(II)‐chitosan blends is confirmed by a three‐fold increase in VEGF secretion by ST‐2 cells in the presence of copper(II)‐chitosan.  相似文献

Fibrous scaffolds, which can mimic the elastic and anisotropic mechanical properties of native tissues, hold great promise in recapitulating the native tissue microenvironment. We previously fabricated electrospun fibrous scaffolds made of hybrid synthetic elastomers (poly(1,3‐diamino‐2‐hydroxypropane‐co‐glycerol sebacate)‐co‐poly (ethylene glycol) (APS‐co‐PEG) and polycaprolactone (PCL)) to obtain uniaxial mechanical properties similar to those of human aortic valve leaflets. However, conventional electrospinning process often yields scaffolds with random alignment, which fails to recreate the anisotropic nature of most of the soft tissues such as native heart valves. Inspired by the structure of native valve leaflet, we designed a novel valve leaflet‐inspired ring‐shaped collector to modulate the electrospun fiber alignment and studied the effect of polymer formulation (PEG amount [mole %] in APS‐co‐PEG; ratio between APS‐co‐PEG and PCL; and total polymer concentration) in tuning the biaxial mechanical properties of the fibrous scaffolds. The fibrous scaffolds collected on the ring‐shaped collector displayed anisotropic biaxial mechanical properties, suggesting that their biaxial mechanical properties are closely associated with the fiber alignment in the scaffold. Additionally, the scaffold stiffness was easily tuned by changing the composition and concentration of the polymer blend. Human valvular interstitial cells (hVICs) cultured on these anisotropic scaffolds displayed aligned morphology as instructed by the fiber alignment. Overall, we generated a library of biologically relevant fibrous scaffolds with tunable mechanical properties, which will guide the cellular alignment.  相似文献

Nanofibers based on natural polymers have recently been attracting research interest as promising materials for use as skin substitutes. Here, we prepared photocrosslinked nanofibrous scaffolds based on methacrylated chitosan (MACS) by photocrosslinking electrospun methacrylated chitosan/poly (vinyl alcohol) (PVA) mats and subsequently removing PVA from the nanofibers. We comprehensively investigated the solution properties of MACS/PVA precursors, the intermolecular action between MACS and PVA components, and the morphology of MACS/PVA nanofibers. Results indicated that the fiber diameter and morphology of the photocrosslinked methacrylated chitosan-based nanofibrous scaffolds were controlled by the MACS/PVA mass ratio and showed highly micro-porous structures with many fibrils. In vitro cytotoxicity evaluation and cell culture experiments confirmed that MACS-based mats with micro-pore structure were biocompatible with L929 cells and facilitated cellular migration into the 3D matrix, demonstrating their potential application as skin replacements for wound repair.  相似文献

Degradation profiles and surface wettability are critical for optimal application of electrospun fibrous mats as drug carriers, tissue growth scaffolds and wound dressing materials. The effect of surface morphologies and chemical groups on surface wettability, and the resulting matrix degradation profiles were firstly assessed for electrospun poly(d,l-lactide) (PDLLA) and poly(d,l-lactide)-poly(ethylene glycol) (PELA) fibers. The air entrapment between the fiber interfaces clarified the effects of various surface morphologies on the surface wettability. Chemical groups with lower binding energy were enriched on the fiber surface due to the high voltage of the electrospinning process, and a surface erosion pattern was detected in the degradation of electrospun PDLLA fibers, which was quite different from the bulk degradation pattern for other forms of PDLLA. Contributed by the hydrophilic poly(ethylene glycol) segments, the degradation of electrospun PELA fibers with hydrophobic surface followed a pattern different from surface erosion and typical bulk degradation.  相似文献

This study was aimed at investigating emulsion electrospinning to prepare biodegradable fibrous mats with encapsulation of human-nerve growth factor (NGF). One of the best methods for fabricating a bio-functional tissue engineering scaffold is to load bioactive agent into the scaffold. In this work, the feasibility of incorporating NGF into poly(l-lactide-co-caprolactone) fibers by emulsion electrospinning has been studied. The release behavior of encapsulated bovine serum albumin (BSA) was investigated. The bioactivity of NGF released from fibrous mats was verified by testing the neurite outgrowth of rat pheochromocytoma cells (PC12). Furthermore, the process of fiber forming during emulsion electrospinning was discussed. The results demonstrate that emulsion electrospun fibers can successfully encapsulate proteins and release them in a sustained manner. The bioactivity of NGF released from emulsion electrospun fibers was confirmed by PC12 bioassays.  相似文献

This work evaluates the influence of electrospinning process parameters on the mean diameter and standard deviation of fiber diameters in electrospun poly(ethyl methacrylate) (PEMA). Processing conditions were selected using Taguchi’s statistical method. Oriented and unoriented electrospun mats with good mechanical properties were produced and demonstrated with tensile stress–strain diagrams. Differential scanning calorimetry (DSC) experiments showed that the polymer chains were forced into non-equilibrium conformations due to electrospinning. Enthalpic recovery during a heating scan is shown by an endothermic peak in the initial DSC heating scan that disappears in subsequent heating scans. An increase in the glass transition temperature with respect to PEMA films shows that the polymer is not equilibrated by heating above glass transition. Cell attachment was tested with MC3T3-E1 pre-osteoblastic cells cultured for short time periods on the electrospun mats. It is shown that the cells present less extended morphology with more diffuse perimetral focal adhesions than cells cultured on flat substrates. A tendency of cells to align in the direction of the substrate fibers in oriented electrospun membranes was also found.  相似文献

In this study, multiwalled carbon nanotubes (MWCNTs) were used to encapsulate a model anticancer drug, doxorubicin (Dox). Then, the drug-loaded MWCNTs (Dox/MWCNTs) with an optimized drug encapsulation percentage were mixed with poly(lactide-co-glycolide) (PLGA) polymer solution for subsequent electrospinning to form drug-loaded composite nanofibrous mats. The structure, morphology, and mechanical properties of the formed electrospun Dox/PLGA, MWCNTs/PLGA, and Dox/MWCNTs/PLGA composite nanofibrous mats were characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy, and tensile testing. In vitro viability assay and SEM morphology observation of mouse fibroblast cells cultured onto the MWCNTs/PLGA fibrous scaffolds demonstrate that the developed MWCNTs/PLGA composite nanofibers are cytocompatible. The incorporation of Dox-loaded MWCNTs within the PLGA nanofibers is able to improve the mechanical durability and maintain the three-dimensional structure of the nanofibrous mats. More importantly, our results indicate that this double-container drug delivery system (both PLGA polymer and MWCNTs are drug carriers) is beneficial to avoid the burst release of the drug and able to release the antitumor drug Dox in a sustained manner for 42 days. The developed composite electrospun nanofibrous drug delivery system may be used as therapeutic scaffold materials for post-operative local chemotherapy.  相似文献

Biodegradable cell‐incorporated scaffolds can guide the regeneration process of bone defects such as physiological resorption, tooth loss, and trauma which medically, socially, and economically hurt patients. Here, 0, 5, 10, and 15 wt% fluoridated hydroxyapatite (FHA) nanoparticles containing 25 wt% F? and 75 wt% OH? were incorporated into poly(ε‐caprolactone) (PCL) matrix to produce PCL/FHA nanocomposite scaffolds using electrospinning method. Then, scanning electron microscopy (SEM), X‐ray diffraction (XRD) pattern, and Fourier transform infrared spectroscopy (FTIR) were used to evaluate the morphology, phase structure, and functional groups of prepared electrospun scaffolds, respectively. Furthermore, the tensile strength and elastic modulus of electrospun scaffolds were investigated using the tensile test. Moreover, the biodegradation behavior of electrospun PCL/FHA scaffolds was studied by the evaluation of weight loss of mats and the alternation of pH in phosphate buffer saline (PBS) up to 30 days of incubation. Then, the biocompatibility of prepared mats was investigated by culturing MG‐63 osteoblast cell line and performing MTT assay. In addition, the adhesion of osteoblast cells on prepared electrospun scaffolds was studied using their SEM images. Results revealed that the fiber diameter of prepared electrospun PCL/FHA scaffolds alters between 700 and 900 nm. The mechanical assay illustrated the mat with 10 wt% FHA nanoparticles revealed the highest tensile strength and elastic modulus. The weight loss alternation of mats determined around 1% to 8% after 30 days of incubation. The biocompatibility and cell adhesion of mats improved by increasing the amounts of FHA nanoparticles.  相似文献

To investigate the potential application of microcrystal cellulose (MCC) and cellulose whisker (CW) in the electrospun vascular tissue scaffolds, cellulose acetate (CA) and cellulose composite scaffolds containing MCC and CW were electrospun from CA solutions and deacetylation. Structure and morphology of MCC, CW and the fibrous composite scaffolds were investigated using FT-IR, SEM, TEM and AFM. The wettability of the scaffolds was evaluated by water contact angle analysis. The effect of MCC and CW on the biocompatibility of the scaffolds for vascular smooth muscle cells (VSMC) was assayed by MTT test, fluorescent imaging and SEM. The biocomposite scaffolds displayed multi-scaled structure and morphology. The scaffolds containing MCC and CW simultaneously exhibited significantly higher cell viability compared to those with only MCC or CW and no filler. Cell viability and morphology within the scaffolds become better with increasing content of MCC and CW. The composite scaffolds with both micro- and nano-scale organization could mimic the native extracellular matrix more closely, and further produce synergistic enhancement on VSMC viability, adhesion and proliferation. This study provides the potential applications of renewable cellulose-based particulates in biomedical field.  相似文献

A poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) fibrous surface with various bead-on-string structures was fabricated by electrospinning. PHBV was electrospun at various concentrations and then CF4 plasma treatment was employed to further improve the hydrophobicity of the PHBV fiber surfaces. The surface morphology of the electrospun PHBV mats was observed by scanning electron microscopy (SEM). The surface properties were characterized by water contact angle (WCA) measurements and X-ray photoelectron spectroscopy (XPS). The surface morphology of the electrospun PHBV fibrous mats with the bead-son-string structure varied with the solution concentration. The WCA of all of the electrospun PHBV mats was higher than that of the PHBV film. In particular, a very rough fiber surface including porous beads was observed when PHBV was electrospun from the solution with a concentration of 26 wt%. Also, its WCA further increased from 141 degrees to 158 degrees after CF(4) plasma treatment for 150 s. PHBV can be rendered superhydrophobic by controlling the surface morphology and surface energy, which can be achieved by adjusting the electrospinning and plasma treatment conditions.  相似文献

Studies investigate the electrospinnability of poly(ε-caprolactone)/protein blends to produce fibers for tissue engineering applications. However, no reports show that zein can improve the scaffolding capacity toward stem cells and promote antiadhesive and bactericidal properties to the poly(ε-caprolactone)/zein fibers. We create fibers with average diameters ranging from 200 to 400 nm from the electrospinning of poly(ε-caprolactone)/protein mixtures. Poly(ε-caprolactone)/zein blends are electrospinnable at zein concentration between 20 and 40 wt% in a 70/30 formic acid/acetic acid mixture. Water contact angle measurements indicate that zein increases fiber hydrophilicity. The water contact angle decreases from 118° (pure poly(ε-caprolactone) fiber) to 73° for the scaffold containing 40 wt% zein. The zein (40 wt%) significantly increases Young's modulus from 260 MPa (pure poly(ε-caprolactone) fibers) to 980 MPa (poly(ε-caprolactone)/zein fibers) with no substantial influence on elongation at break (ε ≥ 125%) and tensile strength (≥0.040 MPa). The electrospun scaffolds containing zein also promote cell adhesion, proliferation, and spreading of adipose-derived human mesenchymal stem cells for at least 7 days of culture. The zein on poly(ε-caprolactone)/zein fibers can prevent the attachment and proliferation of Escherichia coli and Staphylococcus aureus. We propose these materials for wound healing and skin repair.  相似文献

The effectiveness of a multifunctional scaffold produced by the electrospinning of emulsions composed of organic PLGA and aqueous collagen-like protein (denoted as Fol-8Col) solutions is demonstrated. The resultant Fol-8Col/PLGA fibrous scaffolds with homogeneous morphology have mean fiber diameters from 600 to 2,000 nm. A uniform distribution of encapsulated Fol-8Col in the fibers is observed by fluorescence microscopy. TEM is used to clarify the representative core/sheath structure of emulsion electrospun Fol-8Col/PLGA fibers. Preliminary release assessment of encapsulated Fol-8Col shows results of sustained release for more than one month from the Fol-8Col/PLGA fibrous mats. The cytocompatibility of fibroblast cell line L929 with the fibrous composite seems promosing.  相似文献
