This paper proposes a new automated spectrophotometric method for the simultaneous determination of phosphate and arsenate without pre-treatment, which is faster, simpler, less expensive and hazardous than other well-known methods used with water samples. Such method is based on the different kinetic characteristics of complex formation of phosphate and arsenate with ammonium molybdate. A flow system was used in order to achieve good mixing and to provide precise time control. All the measurements were performed at the isosbestic point wavelength (885 nm). Chemical variables were optimized by factorial design (ammonium molybdate 0.015 mol L−1, potassium antimony tartrate 1 × 10−4 mol L−1, and sulphuric acid 0.7 mol L−1). An appropriate linear range for both analytes (0.50-8.00 μmol L−1), good inter-day reproducibility (4.9% [P] and 3.3% [P + As]) and a sample throughput of 6 h−1 were obtained. The detection limits are 0.4 μmol L−1 P and 0.19 μmol L−1 [P + As] (3.3 Sy/x). The method was validated.  相似文献

The common sweeteners aspartame, cyclamate, saccharin and acesulfame K were determined by capillary electrophoresis with contactless conductivity detection. In order to obtain the best compromise between separation efficiency and analysis time hydrodynamic pumping was imposed during the electrophoresis run employing a sequential injection manifold based on a syringe pump. Band broadening was avoided by using capillaries of a narrow 10 μm internal diameter. The analyses were carried out in an aqueous running buffer consisting of 150 mM 2-(cyclohexylamino)ethanesulfonic acid and 400 mM tris(hydroxymethyl)aminomethane at pH 9.1 in order to render all analytes in the fully deprotonated anionic form. The use of surface modification to eliminate or reverse the electroosmotic flow was not necessary due to the superimposed bulk flow. The use of hydrodynamic pumping allowed easy optimization, either for fast separations (80 s) or low detection limits (6.5 μmol L−1, 5.0 μmol L−1, 4.0 μmol L−1 and 3.8 μmol L−1 for aspartame, cyclamate, saccharin and acesulfame K respectively, at a separation time of 190 s). The conditions for fast separations not only led to higher limits of detection but also to a narrower dynamic range. However, the settings can be changed readily between separations if needed. The four compounds were determined successfully in food samples.  相似文献

The fluorescence intensity of methylene blue (MB) quenched by DNA in the pH range of 6.5-8.0 was studied with synchronous fluorescence technology. A novel method for detecting single-stranded and double-stranded DNA was developed. The decreased fluorescence intensity at 664 nm is in proportion to the concentration of DNA in the range of 0.28-11.0 μmol L−1 for ctDNA, 0.14-8.25 μmol L−1 for thermally denatured ctDNA and 0.28-8.25 μmol L−1 for hsDNA. The detection limits (S/N = 3) are 0.11, 0.04 and 0.04 μmol L−1, respectively. The method is rapid, selective, and the reagents are lower toxic. It has been used for the determination of DNA in synthetic samples with good satisfaction. In addition, the interaction modes between MB and ctDNA and the mechanism of the fluorescence quenching were also discussed in detail. The experimental results from absorption spectra and fluorescence polarization indicate that the possible interaction modes between MB and DNA are the electrostatic binding and the intercalation binding.  相似文献

A simple and precise square-wave adsorptive cathodic stripping voltammetry (SW-AdCSV) method has been described for simultaneous determination of Mn(II), Cu(II) and Fe(III) in water samples using a carbon paste electrode. In 0.1 mol L−1 acetate buffer (pH 5) containing 50 μmol L−1 of 2-(5′-bromo-2′-pyridylazo)-5-diethylaminophenol (5-Br-PADAP), Mn(II), Cu(II) and Fe(III) were simultaneously determined as metal-complexes with 5-Br-PADAP following preconcentration onto the carbon paste electrode by adsorptive accumulation at +1.0 V (vs. Ag/AgCl/3 M KCl). Insignificant interference from various cations (K+, Na+, Mg2+, Ca2+, Al3+, Bi3+, Sb3+, Se4+, Zn2+, Ni2+, Co2+, Cd2+, Pb2+, V5+, Ti4+ and NH4+), anions (HCO3, Cl, NO3−, SO42− and PO43−) and ascorbic acid was noticed. Limits of detection of 0.066, 0.108 and 0.093 μg L−1 and limits of quantitation of 0.22, 0.36 and 0.31 μg L−1 Mn(II), Cu(II) and Fe(III), respectively, were achieved by the described method. The described stripping voltammetry method was successfully applied for simultaneous determination of Mn(II), Cu(II) and Fe(III) in ground, tap and bottled natural water samples.  相似文献

In this paper, we presented a novel, rapid and highly sensitive sensor for glutathione (GSH), cysteine (Cys) and histidine (His) based on the recovered fluorescence of the carbon quantum dots (CQDs)–Hg(II) system. The CQDs were synthesized by microwave-assisted approach in one pot according to our previous report. The fluorescence of CQDs could be quenched in the presence of Hg(II) due to the coordination occurring between Hg(II) and functional groups on the surface of CQDs. Subsequently, the fluorescence of the CQDs–Hg(II) system was recovered gradually with the addition of GSH, Cys or His due to their stronger affinity with Hg(II). A good linear relationship was obtained from 0.10 to 20 μmol L−1 for GSH, from 0.20 to 45 μmol L−1 for Cys and from 0.50 to 60 μmol L−1 for His, respectively. This method has been successfully applied to the trace detection of GSH, Cys or His in human serum samples with satisfactory results. The proposed method was simple in design and fast in operation, which demonstrated great potential in bio-sensing fields.  相似文献

This work reports the preparation of a molecularly imprinted polymer (MIP) for selective catalytic detection of serotonin (5-hydroxytryptamine, 5-HT). The process is based on the synthesis of polymers with hemin introduced as the catalytic center to mimic the active site of peroxidase. The copolymer MIP, containing artificial recognition sites for 5-HT, has been prepared by bulk polymerization using methacrylic acid (MAA) and hemin as the functional monomers, and ethylene glycol dimethacrylate (EGDMA) as the cross-linker. For the determination of 5-HT, a flow injection analysis system coupled to an amperometric detector was optimized using multivariate analysis. The effects of different parameters, such as pH, buffer flow rate, buffer nature, peroxide concentration and sample volume were evaluated. After optimizing the experimental conditions, a linear response range from 1.0 up to 1000.0 μmol L−1 was obtained with a sensitivity of 0.4 nA/μmol L−1. The detection limit was found to be 0.30 μmol L−1, while the precision values (n = 6) evaluated by relative standard deviation (R.S.D.) were, respectively, 1.3 and 1.7% for solutions of 50 and 750 μmol L−1 of 5-HT. No interference was observed by structurally similar compounds (including epinephrine, dopamine and norepinephrine), thus validating the good performance of the imprinted polymer. The method was applied for the determination of 5-HT in spiked blood serum samples.  相似文献

A new microflow injection chemiluminescence (μFI-CL) system was described for the determination of cisplatin in human serum. By using the microchip with double spiral channel configuration, the sensitivity was greatly enhanced due to more efficient mixing of the analyte and reagent solutions. Experimental results revealed that common ions in human serum, such as Mn2+, Co2+, Fe3+, Cu2+, Zn2+, Ni2+, Na+, K+, Ca2+, Cl, NO3, Ac, CO32−, PO43−, SO42− did not cause interference with the detection of Pt(II) by using 1,10-phenanthroline as the masking agent. Under the optimized conditions, a linear calibration curve (R2 = 0.998) over the range 2.0 × 10−8 to 2.0 × 10−6 mol L−1 was obtained with the detection limit of 1.24 × 10−9 mol L−1. The relative standard deviation was found to be 3.46% (n = 12) for 2.0 × 10−7 mol L−1. The sample consumption was only 2 μL with the sample throughput of 72 h−1. It had been used for trace platinum determination in cisplatin injection and human serum samples after the dosage of cisplatin. The recovery varied from 97.6 to 103.9%. The results proved that the proposed μFI-CL system had the advantages of high sensitivity and precision, low sample and reagents consumption, and high analytical throughput.  相似文献

A high-pressure microwave digestion was applied for microwave-assisted extraction (MAE) of mercury species from sediments and zoobenthos samples. A mixture containing 3 mol L−1 HCl, 50% aqueous methanol and 0.2 mol L−1 citric acid (for masking co-extracted Fe3+) was selected as the most suitable extraction agent. The efficiency of proposed extraction method was better than 95% with R.S.D. below 6%. A preconcentration method utilizing a “homemade” C18 solid phase extraction (SPE) microcolumns was developed to enhance sensitivity of the mercury species determination using on-column complex formation of mercury-2-mercaptophenol complexes. Methanol was chosen for counter-current elution of the retained mercury complexes achieving a preconcentration factor as much as 1000. The preconcentration method was applied for the speciation analysis of mercury in river water samples. The high-performance liquid chromatography-cold vapour atomic fluorescence spectrometric (HPLC/CV-AFS) method was used for the speciation analysis of mercury. The complete separation of four mercury species was achieved by an isocratic elution of aqueous methanol (65%/35%) on a Zorbax SB-C18 column (4.6 mm × 150 mm, 5 μm) using the same complexation reagent (2-mercaptophenol). The limits of detection were 4.3 μg L−1 for methylmercury (MeHg+), 1.4 μg L−1 for ethylmercury (EtHg+), 0.8 μg L−1 for inorganic mercury (Hg2+), 0.8 μg L−1 for phenylmercury (PhHg+).  相似文献

A novel chemiluminescence gas-diffusion flow injection system for the determination of arsenic(III) in aqueous samples is described. The analytical procedure involves injection of arsenic(III) samples and standards into a 0.3 mol L−1 hydrochloric acid carrier stream which is merged with a reagent stream containing 0.2% (w/v) sodium borohydride and 0.015 mol L−1 sodium hydroxide. Arsine, generated in the combined carrier/reagent donor stream, diffuses across the hydrophobic Teflon membrane of the gas-diffusion cell into an argon acceptor stream and then reacts with ozone in the flow-through chemiluminescence measuring cell of the flow system. Under optimal conditions, the method is characterized by a wide linear calibration range from 0.6 μg L−1 to 25 mg L−1, a detection limit of 0.6 μg L−1 and a sample throughput of 300 samples per hour at 25 mg L−1 and 450 samples per hour at 25 μg L−1.  相似文献

A compact, reliable and low cost flow injection chemiluminescence system is described. The flow system consists of a set of solenoid micro-pumps that can dispense reproductive micro-volumes of solutions. The luminometer was based on a coiled cell constructed from polyethylene tubing that was sandwiched between two large area photodiodes. The whole equipment costs about US$ 750 and weights ca. 3 kg. Equipment performance was evaluated by measuring low concentrations of hydrogen peroxide by oxidation of luminol and for the determination of ammonium, based on its inhibition of the luminescence provided by the reaction of luminol and sodium hypochlorite. Linear responses were achieved within 1.0-80 μmol L−1 H2O2 and 0.6-60 μmol L−1 NH4+ with detection limits estimated as 400 nmol L−1 H2O2 and 60 nmol L−1 NH4+ at the 99.7% confidence level. Coefficients of variation were 1.0 and 1.8%, estimated for 20 μmol L−1 H2O2 and 15 μmol L−1 NH4+ (n = 20), respectively. Reagent consumption of 55 μg luminol, effluent volume of 950 μL per determination and sampling rate of 120 samples per hour were also achieved.  相似文献

A simple and sensitive spectrophotometric flow method for determination of low concentrations of the flotation collector O-ethyldithiocarbonate (ethyl xanthate, CH3CH2-O-CS2) in solutions is described. The method is based on ethyl xanthate detection at 301 nm in medium of NaOH 50 mmol L−1. By injection of 200 μL of sample, the analytical method shows linear response for the ethyl xanthate concentration from 0.5 up to 500 μmol L−1. Successive injections of 4 μmol L−1 ethyl xanthate (n = 23) show a coefficient of variation lower than 0.6%, denoting high repeatability. The detection limit is 0.3 μmol L−1. At a flow rate of 2.0 mL min−1, a frequency of 120 injections/h of ethyl xanthate can be attained. By introduction of a tangential dialysis cell in the FIA system, the manual sample filtration step with 0.22 μm filter was eliminated and the residual interference of suspended material, was completely overcome even for unfiltered sludge suspension samples, an important advantage that compensates for the frequency reduction to 25 injections/h elevation and detection limit elevation to 2 μmol L−1, still outreaching for many applications. Potential applications of the method embrace the at line determination of ethyl xanthate in the ore processing industry, control of the concentration at its optimal level during the flotation process, as well as monitoring of residues in the effluents.  相似文献

The interaction between mercaptoacetic acid (MA)-capped CdTe QDs, MA-capped CdTe/ZnS QDs or glutathione (GSH)-capped CdTe QDs with As(III) was studied using fluorescence spectrometry. As (III) has a high-affinity to reduced-GSH to form As(SG)3, and the emission of the GSH-capped CdTe QDs (λem. = 612 nm) is quenched effectively. Thus, a novel fluorescence spectrometric method was developed for As (III) determination by using GSH-CdTe QDs. Under optimal conditions, the quenched fluorescence intensity (F0/F) increased linearly with the concentration of As (III) ranging from 5.0 × 10−6 to 25 × 10−5 mol L−1. The limit of detection (3σ) for As (III) was found to be 2 × 10−8 mol L−1. This method is potentially useful in visual detection of As (III) under irradiation of the ultraviolet light.  相似文献

A fluorometric method for quantity analysis of biothiols was developed using a graphene oxide (GO)-based “molecular beacon”-like probe, which consisted of FITC labeled thymine (T)-rich single-stranded DNA (ssDNA), GO and Hg2+ ions. The labeled ssDNA containing T–T mismatches would self-hybridize to duplex in the presence of Hg2+, which can avoid its adsorption on GO and the fluorescence of this GO-based probe was recovered. The fluorescence of the probe quenched after the addition of biothiols such as glutathione (GSH) and cysteine (Cys) owing to thiol groups can selectively competitive ligation of Hg2+ ions with T–T mismatches. In the present work, the GO-based probe was used for the determination of GSH and Cys. Under the optimal conditions, a linear correlation was established between fluorescence intensity ratio I0/I and the concentration of GSH in the range of 2.0 × 10−9–5.0 × 10−7 mol L−1 with a detection limit of 1.0 × 10−9 mol L−1. The linear range for Cys is from 5.0 × 10−9 to 4.5 × 10−7 mol L−1 with a detection limit of 2.0 × 10−9 mol L−1. The proposed method was applied to the determination of GSH in human serum and cell extract samples with satisfactory results.  相似文献

A sensitive and selective method for the paraoxon detection based on enzyme inhibition and fluorescence quenching was presented in this study. Under the catalytic effect of acetylcholinesterase (AChE), acetylthiocholine (ATCh) hydrolysis released thiocholine (TCh) which could react with N-(7-dimethylamino-4-methylcoumarin-3-yl) maleimide (DACM) to produce a blue fluorescence compound. Subsequently, AChE catalytic activity was inhibited with the addition of paraoxon, which caused TCh decreased, leading to a significant decrease of the blue fluorescent compound. Meanwhile, p-nitrophenol, the hydrolysis product of paraoxon, would lead to a quenching of the fluorescence. Therefore, fluorescence intensity of the system would decrease dramatically by a combined effect of enzyme inhibition and fluorescence quenching. Under optimal experimental conditions, an excellent linear relationship between the decrease of fluorescence intensity and paraoxon concentration over the range from 5.5 × 10−12 to 1.8 × 10−10 mol L−1 was obtained. Fluorescence background caused by nonenzymatic hydrolysis of ATCh or other matters was relatively low, the proposed approach offered adequate sensitivity for the detection of paraoxon at 3.5 × 10−12 mol L−1.  相似文献

A novel method of first derivative synchronous fluorescence was developed for the rapid simultaneous analysis of trace 1-hydroxypyrene (1-OHP), 1-naphthol (1-NAP), 2-naphthol (2-NAP), 9-hydroxyphenanthrene (9-OHPe) and 2-hydroxyfluorene (2-OHFlu) in human urine. Only one single scan was needed for quantitative determination of five compounds simultaneously when Δλ = 10 nm was chosen. In the optimal experimental conditions, there was a linear relationship between the fluorescence intensity and the concentration of 1-OHP, 1-NAP, 2-NAP, 9-OHPe and 2-OHFlu in the range of 1.75 × 10−9 to 4.50 × 10−6 mol L−1, 3.64 × 10−8 to 2.20 × 10−4 mol L−1, 8.18 × 10−9 to 1.20 × 10−4 mol L−1, 3.26 × 10−9 to 8.50 × 10−5 mol L−1 and 4.88 × 10−9 to 5.50 × 10−6 mol L−1, respectively. The limits of detection (LOD) were found to be 5.25 × 10−10 mol L−1 for 1-OHP, 1.10 × 10−8 mol L−1 for 1-NAP, 2.46 × 10−9 mol L−1 for 2-NAP, 9.77 × 10−10 mol L−1 for 9-OHPe and 1.46 × 10−9 mol L−1 for 2-OHFlu. The proposed method is reliable, selective and sensitive, and has been used successfully in the determination of traces of 1-OHP, 1-NAP, 2-NAP, 9-OHPe and 2-OHFlu in human urine samples, whose results were in good agreement with those gained by the HPLC method.  相似文献

A novel approach of using a gold disc microelectrode to analyze sweat samples for copper ions by anodic square wave stripping voltammetry (SW stripping voltammetry) is described. Sweat was collected from the lower back of four subjects after physical exercise and the sample volume required for the determinations was 100 μL. Under the optimized conditions, the calibration plot was linear over the range 1-100 μmol L−1 Cu(II) with a limit of detection of 0.25 μmol L−1. The precision was evaluated by carrying out five replicate measurements in a 1 μmol L−1 Cu(II) solution and the standard deviation was found to be 1.5%. Measurements were performed by inserting the microelectrode into sweat drops and Cu(II) concentrations in the analyzed samples ranged from 0.9 to 28 μmol L−1. Values obtained by the proposed voltammetric method agreed well with those found using graphite furnace atomic absorption spectroscopy (GFAAS).  相似文献

A gas chromatography–tandem mass spectrometric (GC–MS/MS) method has been established for the determination of cyanide in surface water. This method is based on the derivatization of cyanide with 2-(dimethylamino)ethanethiol in surface water. The following optimum reaction conditions were established: reagent dosage, 0.7 g L−1 of 2-(dimethylamino)ethanethiol; pH 6; reaction carried out for 20 min at 60 °C. The organic derivative was extracted with 3 mL of ethyl acetate, and then measured by using GC–MS/MS. Under the established conditions, the detection and quantification limits were 0.02 μg L−1 and 0.07 μg L−1 in 10-mL of surface water, respectively. The calibration curve had a linear relationship relationship with y = 0.7140x + 0.1997 and r2 = 0.9963 (for a working range of 0.07–10 μg L−1) and the accuracy was in a range of 98–102%; the precision of the assay was less than 7% in surface water. The common ions Cl, F, Br, NO3, SO42−, PO43−, K+, Na+, NH4+, Ca2+, Mg2+, Ba2+, Mn4+, Mn2+, Fe3+, Fe2+ and sea water did not interfere in cyanide detection, even when present in 1000-fold excess over the species. Cyanide was detected in a concentration range of 0.07–0.11 μg L−1 in 6 of 10 surface water samples.  相似文献

Square wave voltammetry automated by sequential injection analysis was applied to determine the Freundlich adsorption coefficients for the adsorption of atrazine onto a clay rich soil. The detection limit in soil extracts was between 0.18 and 0.48 μmol L−1, depending on the medium used to prepare the extracts (0.010 mol L−1 KCl, CaCl2 or HNO3 and 0.0050 mol L−1 H2SO4), all of them conditioned in 40 mmol L−1 Britton-Robinson buffer at pH 2.0 in presence of 0.25 mol L−1 NaNO3. Also in soil extracts the linear dynamic range was between 1.16 and 18.5 μmol L−1 (0.25-4.0 μg mL−1), with a sampling frequency of 190 h−1. The Kf Freundlich adsorption coefficient was 3.8 ± 0.2 μmol1−1/n Ln kg−1 in medium of 0.010 mol L−1 KCl or CaCl2, but increased to 7.7 ± 0.1 and 9.0 ± 0.3 μmol1−1/n Ln kg−1 in 0.010 mol L−1 HNO3 and 0.0050 mol L−1 H2SO4, respectively. The increase of Kf was related to the decrease of pH from 6.4-6.7 in KCl and CaCl2 to 3.7-4.0 in presence of HNO3 or H2SO4, which favors protonation of atrazine, facilitating electrostatic attractions with negative charges of the clay components of the soil. The 1/n parameters were between 0.76 and 0.86, indicating that the isotherms are not linear, suggesting the occurrence of chemisorption at specific adsorption sites. No statistically significant differences were observed in comparison to the adsorption coefficients obtained by HPLC. The advantage of the proposed SI-SWV method is the great saving of reagent because it does not use organic solvent as in the case of HPLC (50% (v/v) acetonitrile in the mobile phase). Additionally the start up of SI-SWV is immediate (no column conditioning necessary) and the analysis time is only 19 s.  相似文献

A sensitive optode consisting of highly lead-selective ionophore (Lead IV), proton-selective chromoionophore (ETH 5294) and lipophilic anionic sites (KTpClPB) in plasticized polyvinyl chloride (PVC) membrane was fabricated. The optode membranes were used for determination of Pb2+ by absorption spectrophotometry in batch and flow-through systems. The influence parameters such as pH, type of buffer solution, response time and concentration of regenerating solution were optimized. The membrane responded to Pb2+ by changing its color from blue to pinkish purple in Tris buffer containing different concentration of Pb2+ at pH 7.0. The optode provided the response range of 3.16 × 10−8 to 5.00 × 10−5 mol L−1 Pb2+ with the detection limit of 2.49 × 10−8 mol L−1 in the batch system within the response time of 30 min. The dynamic range of 1.26 × 10−8 to 3.16 × 10−5 mol L−1 Pb2+ with detection limit of 8.97 × 10−9 mol L−1 were obtained in the flow-through system within the response time of 15 min. Moreover, the proposed optode sensors showed good selectivity towards Pb2+ over Na+, K+, Mg2+, Cd2+, Hg2+ and Ag+. It was successfully applied to determine Pb2+ in real water samples and the results were compared with well-established inductively coupled plasma optical emission spectrometry (ICP-OES). No significant different value (tcritical = 4.30 > texp = 1.00-3.42, n = 3 at 95% of confidence level) was found.  相似文献
