In this paper, an improved recovery method for target ssDNA using amino-modified silica-coated magnetic nanoparticles (ASMNPs) is reported. This method takes advantages of the amino-modified silica-coated magnetic nanoparticles prepared using water-in-oil microemulsion technique, which employs amino-modified silica as the shell and iron oxide as the core of the magnetic nanoparticles. The nanoparticles have a silica surface with amino groups and can be conjugated with any desired bio-molecules through many existing amino group chemistry. In this research, a linear DNA probe was immobilized onto nanoparticles through streptavidin conjugation using covalent bonds. A target ssDNA(I) (5′-TMR-CGCATAGGGCCTCGTGATAC-3′) has been successfully recovered from a crude sample under a magnet field through their special recognition and hybridization. A designed ssDNA fragment of severe acute respiratory syndrome (SARS) virus at a much lower concentration than the target ssDNA(I) was also recovered with high efficiency and good selectivity.  相似文献

A novel, highly selective DNA hybridization assay has been developed based on surface-enhanced Raman scattering (SERS) for DNA sequences related to HIV. This strategy employs the Ag/SiO2 core-shell nanoparticle-based Raman tags and the amino group modified silica-coated magnetic nanoparticles as immobilization matrix and separation tool. The hybridization reaction was performed between Raman tags functionalized with 3′-amino-labeled oligonucleotides as detection probes and the amino group modified silica-coated magnetic nanoparticles functionalized with 5′-amino-labeled oligonucleotides as capture probes. The Raman spectra of Raman tags can be used to monitor the presence of target oligonucleotides. The utilization of silica-coated magnetic nanoparticles not only avoided time-consuming washing, but also amplified the signal of hybridization assay. Additionally, the results of control experiments show that no or very low signal would be obtained if the hybridization assay is conducted in the presence of DNA sequences other than complementary oligonucleotides related to HIV gene such as non-complementary oligonucleotides, four bases mismatch oligonucleotides, two bases mismatch oligonucleotides and even single base mismatch oligonucleotides. It was demonstrated that the method developed in this work has high selectivity and sensitivity for DNA detection related to HIV gene.  相似文献

The preparation,characterization,and application of silica-coated magnetic nanoparticles for papain immobilization is reported.Papain was covalently attached onto the(3-chloropropyl) trimethoxysilane-modified silica-coated magnetic nanoparticles. The enzyme-immobilized nanoparticles were characterized by Fourier transform infrared spectroscopy,X-ray powder diffraction,scanning electron microscopy,and vibrating sample magnetometry techniques.Response surface methodology combined with statistical analyses using Minitab were employed to evaluate optimum operating conditions to immobilize papain on the magnetic nanoparticles.The optimum conditions were: temperature = 27.3℃,pH of the enzyme solution = 7.1,concentration of papain = 3.3 mg/mL,and immobilization time = 10 h.Compared with the free papain,the immobilized papain displayed enhanced enzyme activity,better tolerance to variations in the medium pH and temperature,improved storage stability,and good reusability.Both the free and immobilized enzymes were effective for the clarification of pomegranate juice.  相似文献

A pyridinecarboxaldimine grafted to silica-coated magnetic nanoparticles was prepared. The structure and magnetic properties of the functionalized magnetic silica nanoparticles were identified by TEM, FT-IR, XRD, elemental analysis, and vibrating sample magnetometer (VSM). The supported pyridinecarboxaldimine as chelating ligand in combination with CuBr2 and 2,2,6,6-tetramethyl-1-piperadoxyl (TEMPO) exhibited efficient catalytic performance in the aerobic oxidation of primary alcohols to aldehydes. The functionalized magnetic silica nanoparticles could be easily recovered using an external magnetic field and reused for at least 6 times with low reduction in its performance in the aerobic oxidation of benzyl alcohol.  相似文献

A new method was developed for the speciation of inorganic arsenic in environmental water by using selective magnetic solid-phase extraction followed by inductively coupled plasma mass spectrometry. It is found that As(V) selectively adsorbed on amino-modified silica-coated magnetic nanoparticles (MNPs) in the pH range from 3 to 8, while As(III) is not be retained. The As(V)-loaded MNPs can be separated easily from the aqueous sample solution by simply applying an external magnetic field. The adsorbed As(V) was quantitatively recovered from the MNPs using using 1 M nitric acid. Total inorganic As was extracted after the permanganate oxidation of As(III) to As(V). Parameters affecting the separation were investigated systematically, and the optimal separation conditions were established. Under the optimal conditions, the limit of detection is 0.21 ng L?1, and the precision is 6.8% (at 10 ng L?1, for n?=?7). The method was applied to the speciation of inorganic arsenic in environmental water of tobacco growing area.

Bi-functional nanoparticles (NPs) that consist of silica-coated magnetic cores and luminescent lanthanide (Ln) ions anchored on the silica surface via organic linker molecules are reported. Compared to individual Ln ions, the hybrid NPs show a drastically enhanced photoluminescence due to the efficient ligand-to-metal energy transfer in the Ln-loaded NPs: the new bi-functional NPs could be used in a variety of biological applications involving magnetic separation and optical detection.  相似文献

We report the synthesis, characterization and relaxometric study of ferrofluids based on iron oxide, with potential for use as magnetic resonance imaging (MRI) contrast agents (CAs). The effect of different cost-effective, water-based surface modification approaches which can be easily scaled-up for the large scale synthesis of the ferrofluids has been investigated. Surface modification was achieved by silanization, and/or coating with non-toxic commercial dispersants (a lauric polysorbate and a block copolymer with pigment affinic groups, namely Tween 20 and Disperbyk 190) which were added after or during iron oxide nanoparticle synthesis. It was observed that all the materials synthesized functioned as negative contrast agents at physiological temperature and at frequencies covered by clinical imagers. The relaxometric properties of the magnetic nanoparticles were significantly improved after surface coating with stabilizers compared to the original iron oxide nanoparticles, with particular reference to the silica-coated magnetic nanoparticles. The results indicate that the optimization of the preparation of colloidal magnetic ferrofluids by surface modification is effective in the design of novel contrast agents for MRI by enabling better or more effective interaction between the coated iron oxide nanoparticles and protons present in their aqueous environment.  相似文献

Abstract

A novel, selective, and sensitive magnetic-mimetic enzyme fluorescence immunoassay method for antigen detection has been developed by taking advantage of a magnetic separation process and the amplification feature of the hemin label. This method is based on a twice amplified fluorescence signal. The signal is first amplified due to the ultrasmall size and the high surface-to-volume ratio of the silica-coated magnetite nanoparticles, which enable the nanoparticles to carry much more antibodies. Second, the mimetic enzyme (hemin) as a labeling reagent catalyzes the reaction of p-hydroxyphenyl acetic acid and H2O2 can further amplify the fluorescence signal. This protocol was also evaluated for a sandwich-type immunoassay of human IgG, and the calibration graph for human IgG was linear over the range of 0–100 ng mL?1 with a detection limit of 9.8 ng mL?1. This method can easily separate magnetic nanoparticles from the solution, which simplified the process and played a promising role for various applications in immunoassay.  相似文献

We report on a single-step coating process and the resulting colloidal stability of silica-coated spindle-type hematite nanoparticles (NPs) decorated with a layer of poly(acrylic acid) (PAA) polyelectrolyte chains that are partially incorporated into the silica shell. The stability of PAA coated NPs as a function of pH and salt concentration in water was compared to bare hematite particles and simple silica-coated hematite NPs, studying their electrophoretic mobility and the hydrodynamic radius by dynamic light scattering. Particles coated with this method were found to be more stable upon the addition of salt at pH 7, and their aggregation at the pH of the isoelectric point is reversible. The hybrid coating appears to increase the colloidal stability in aqueous media due to the combination of the decrease of the isoelectric point and the electrosteric stabilization. This coating method is not limited to hematite particles but can easily be adapted to any silica-coatable particle.  相似文献

Maghemite nanoparticles were synthesized by the coprecipitation method. Silica was coated to the maghemite nanoparticles and amino silane was modified to the surface of the silica magnetic nanoparticles. We use the biofunctional magnetic nanoparticles as a general agent to immobilize and separate the proteins in a broad range from different traditional Chinese medicines. The transmission electron microscopy results showed that the average diameter of the well-dispersed silica-coated nanoparticles was about 60 nm. The Fourier transform infrared spectrum indicated that the amino group had been successfully coupled to the surface of the maghemite particles. And the protein immobilization effect was characterized by the microplate reader. The characterization results proved that the synthesized functional magnetic nanoparticles could effectively immobilize and separate the proteins from traditional Chinese medicines.  相似文献

通过反向微乳液法, 在油溶性量子点表面包裹二氧化硅外壳, 使油溶性量子点水溶性化, 再利用3-氨丙基三乙氧基硅烷(APTES)在已形成的二氧化硅纳米颗粒表面进行氨基化改性, 制备富含氨基的二氧化硅包裹的量子点荧光纳米球. 通过透射电子显微镜(TEM)、粒径分析、zeta电位检测、紫外-可见分光光度、荧光分光光度和红外光谱等手段对产品进行了表征. 结果表明, 所制备的二氧化硅量子点纳米球(45 nm)具有单分散性、水溶性好及光化学稳定性强等优点. 通过静电作用, 所制备的单分散氨基化二氧化硅量子点对肿瘤细胞表面膜电荷进行了初步标记显像.  相似文献

Multifunctional colloidal core-shell nanoparticles of magnetic nanocrystals (of iron oxide or FePt) or gold nanorods encapsulated in silica shells doped with the fluorescent dye, Tris(2,2′-bipyridyl)dichlororuthenium(II) hexahydrate (Rubpy) were synthesized. The as-prepared magnetic nanocrystals are initially hydrophobic and were coated with silica using a microemulsion approach, while the as-prepared gold nanorods are hydrophilic and were coated with silica using a Stöber type of process. Each approach yielded monodisperse nanoparticles with uniform fluorescent dye-doped silica shells. These colloidal heterostructures have the potential to be used as dual-purpose tags—exhibiting a fluorescent signal that could be combined with either dark-field optical contrast (in the case of the gold nanorods), or enhanced contrast in magnetic resonance images (in the case of magnetic nanocrystal cores). The optical and magnetic properties of the fluorescent silica-coated gold nanorods and magnetic nanocrystals are reported.  相似文献

We present a facile and efficient method for modifying the surface of silica-coated Fe3O4 magnetic nanoparticles (MNPs) with bis(pyrazolyl) triazine ruthenium(II) complex [ MNPs@BPT–Ru (II) ] . Field emission-scanning electron microscopy, thermogravimetric/derivative thermogravimetry analysis, X-ray powder diffraction, Fourier-transform infrared spectroscopy, vibrating sample magnetometry, and energy-dispersive X-ray spectrometry analyses were employed for characterizing the structure of these nanoparticles. MNPs@BPT–Ru(II) nanoparticles proved to be a magnetic, reusable, and heterogeneous catalyst for the hydrogen transfer reduction of ketone derivatives. In addition, highly pure products were obtained with excellent yields in relatively short times in the presence of this catalyst. A comparison of this catalyst with those previously used for the hydrogen transfer reactions proved the uniqueness of MNPs@BPT–Ru(II) nanoparticle which is due to its inherent magnetic properties and large surface area. The presented method also had other advantages such as simple reaction conditions, eco-friendliness, high recovery ability, easy work-up, and low cost.  相似文献

A novel magnetically recoverable organic hydride compound was successfully constructed by using silica-coated magnetic nanoparticles as a support. An as-prepared magnetic organic hydride compound, BNAH (1-benzyl-1,4-dihydronicotinamide), showed efficient activity in the catalytic reduction of α,β-epoxy ketones. After reaction, the magnetic nanoparticle-supported BNAH can be separated by simple magnetic separation which made the separation of the product easier.  相似文献

12‐Tungstophosphoric acid supported on aerosil silica and silica‐coated γ‐Fe2O3 nanoparticles was prepared and characterized using transmission electron microscopy,scanning electron microscopy,and inductively coupled plasma atomic emission spectroscopy.The catalytic activity of the two prepared catalysts was compared in the synthesis of 1,8‐dioxo‐9,10‐diaryldecahydroacridines in water.12‐Tungstophosphoric acid was highly dispersed on the silica‐coated γ‐Fe2O3 nanoparticles and showed higher activity and a higher reuse number compared with the acid supported on aerosil silica.The catalyst could be recovered simply by using an external magnetic field and could be reused several times without appreciable loss of its catalytic activity.  相似文献

Highly monodisperse particles composed of a magnetic silica core and fluorescent polymer shell were synthesized with a combined technique of heterocoagulation and soap-free emulsion polymerization. Prior to heterocoagulation, monodisperse, submicrometer-sized silica particles were prepared with the Stober method, and magnetic nanoparticles were prepared with a modified Massart method in which a cationic silane coupling agent of N-trimethoxysilylpropyl- N, N, N-trimethylammonium chloride was added just after coprecipitation of Fe (2+) and Fe (3+). The silica particles with negative surface potential were heterocoagulated with the magnetic nanoparticles with positive surface potential. The magnetic silica particles obtained with the heterocoagulation were treated with sodium silicate to modify their surfaces with silica. In the formation of a fluorescent polymer shell onto the silica-coated magnetic silica cores, an amphoteric initiator of 2,2'-azobis[ N-(2-carboxyethyl)-2-2-methylpropionamidine] (VA-057) was used to control the colloidal stability of the magnetic cores during the polymer coating. The polymerization of St in the presence of a hydrophobic fluorophore of pyrene could coat the cores with fluorescent polymer shells, resulting in monodisperse particles with a magnetic silica core and fluorescent polymer shell. Measurements of zeta potential for the composite particles in different pH values indicated that the composite particles had an amphoteric property originating from VA-057 initiator.  相似文献
