A scalable and environmentally benign route to the free base of branaplam, a small molecule splicing modulator, was identified and developed. This alternative approach circumvented the inherent risk of dioxin formation associated with ortho-halo phenol derivatives present in the previous route. A Friedel–Crafts approach provided a reliable access to the key intermediate, subsequently followed by a Suzuki–Miyaura cross-coupling. Multiple process aspects of the synthetic approach were evaluated, and a robust process for its large-scale synthesis was developed and successfully demonstrated.  相似文献

The crystal structure of 2-(2-((4-(pyridin-2-yl)pyrimidin-2-yl)disulfanyl)pyrimidin-4-yl)pyridinium hexafluorophosphate was obtained by X-ray single-crystal diffraction. This molecule crystallizes in monoclinic P2/c space group with a=6.219(1), b=13.103(2), c=13.059(2) ?, β=97.567(2)°, Z=2, the final R=0.0525 and w R=0.1434. The title compound was prepared successfully from 1,2-bis(4-(pyridin-3-yl)pyrimidin-2-yl)disulfane(BPPD). TD-DFT computational studies on energy gaps and UV-Vis spectra of BPPD and the title compound have been carried out for comparisons. The resultant HOMO-LUMO gap of the BPPD compound is 4.45 e V, while that for the corresponding hexafluorophosphate salt is much smaller at 2.46 e V, which are in agreement with their UV-Vis absorptions.  相似文献

Condensation of 3-methyl-2-azafluorene with 2-formylfluorene results in the formation of a compound containing a polyconjugated system and two acidic CH centers, namely, 1-(fluoren-2-yl)-2-(2-azafluoren-3-yl)ethene. The conversions of this compound upon treatment with alkali have been studied by electronic absorption spectroscopy. 9-o- and 9-m-carboranylcarboxylic acids were used to synthesize σ-(o-carboran-9-yl)- and σ-(m-carboran-9-yl)-π-cyclopentadienyldicarbonyliron. The latter complexes, in reactions with bromine, undergo rearrangement with the cleavage of the BFe σ-bond, involving migration of the 9-o- and 9-m-carboranyl groups into the cyclopentadienyl ring, to give π-(o-carboran-9-yl)cyclopentadienyl- and π-(m-carboran-9-yl)cyclopentadienyl-dicarbonyliron bromides, respectively. A simple method to obtain these acids by the oxidation of 9-alkyl-o- and 9-alkyl-m-carboranes with CrO3 in CH3COOH has been found.  相似文献

Self-assembly of the ligand with Zn(II) in dichloromethane and methanol offers a dinuclear dimeric complex with a ligand:metal ratio of 2:2. X-Ray crystal structure analysis reveals two ligands bound through a head-to-tail pattern to two zinc centers to form a severely distorted helical conformation, which has the shape of a rectangle.  相似文献

Based on a computer-assisted analysis of the three-dimensional structure of the binary complex of E.coli dihydrofolate reductase (DHFR) with methotrexate, 5-(N-arylnortropan-3-yl)- and 5-(N-arylpiperidin-4-yl)-2,4-diaminopyrimidines 2 and 4 were designed as inhibitors of DHFR. Syntheses of the designed compounds have been carried out. The most potent compound 2a inhibited E. coli DHFR with Ki = 0.49.10?9M. The activities within the series of compounds synthesized could be rationalized by molecular-modelling experiments which served as the basis of this work. Several compounds within the presented series exhibit antimalarial activities in vitro and in vivo.  相似文献

The condensation of 3-(furan-2-yl)- and 3-(thiophen-2-yl)prop-2-enals with nitro-substituted CH acids, namely ethyl nitroacetate, nitroacetone, nitroacetophenone, and nitroacetonitrile, afforded a series of geminally activated nitro dienes, 4-(furan-2-yl)- and 4-(thiophen-2-yl)-1-nitrobuta-1,3-dienes. The product structure was confirmed by NMR and IR spectroscopy.

Cycloaddition to 1-(4,6-dimethylpyrimidin-2-yl)- and 1-(4,6-dimethoxy-s-triazin-2-yl)-3-oxidopyridinium betaines across the 2,6-positions of the pyridine rings with indene, acenaphthylene and ethyl cinnamate gave substituted 8-aza[3.2.1]bicycIooct-3-en-2-ones, whereas the [6π + 4π] cycloaddition reaction with 6,6-dimethylfulvene gave a tricyclo[6.3.1.02.6]dodeca-2,(6),4,9-trien-11-one. 近20年来,真菌感染特别是深部真菌感染日益引起人们的广泛关注[1]。三唑类抗真菌药物对真菌的选择性较高,代谢稳定,体内动态、安全性都较好,已成为目前抗真菌药物研究开发的重点[2]。三唑类抗真菌药物是细胞色素P450 14α 去甲基酶的抑制剂,可影响该酶所催化的真菌麦角甾醇的生物合成,导致真菌细胞膜破损而死亡[3,4]。1998年,SynPhar实验室及Taiho制药公司[5]研制开发的化合物Syn 2869采用了取代哌嗪作为侧链。体外实验表明,该化合物抗菌谱广,对多种念珠菌、隐球菌及曲莓菌均有效,体内实验中,与目前临床上治疗真菌感染的主要用药伊曲…  相似文献

利用Pummerer反应,以有机酸为反应原料、溶剂和催化剂,简便、高效地合成了一系列2-(2,6-二卤苯基)-3-(4,5,6-三甲基-2-嘧啶基)-5-酯酰氧基-1,3-噻唑烷-4-酮(7a~7g和8a~8g),产率为62%~85%.X衍射确证化合物主要异构体的构型为噻唑烷-4-酮环C-5与C-2位取代基处于反式构型,顺反异构体比例通过1H NMR确定.化合物7a和8a具有中等强度的抗HIV-RT活性.  相似文献
