The diluted aerosols from a cigarette (3R4F) and an e-cigarette (Vype ePen) were compared in two commercially available in vitro exposure systems: the Borgwaldt RM20S and Vitrocell VC10. Dosimetry was assessed by measuring deposited aerosol mass in the exposure chambers via quartz crystal microbalances, followed by quantification of deposited nicotine on their surface. The two exposure systems were shown to generate the same aerosols (pre-dilution) within analytically quantified nicotine concentration levels (p = 0.105). The dosimetry methods employed enabled assessment of the diluted aerosol at the exposure interface. At a common dilution, the per puff e-cigarette aerosol deposited mass was greater than cigarette smoke. At four dilutions, the RM20S produced deposited mass ranging 0.1–0.5 µg/cm2/puff for cigarette and 0.1–0.9 µg/cm2/puff for e-cigarette; the VC10 ranged 0.4–2.1 µg/cm2/puff for cigarette and 0.3–3.3 µg/cm2/puff for e-cigarette. In contrast nicotine delivery was much greater from the cigarette than from the e-cigarette at a common dilution, but consistent with the differing nicotine percentages in the respective aerosols. On the RM20S, nicotine ranged 2.5–16.8 ng/cm2/puff for the cigarette and 1.2–5.6 ng/cm2/puff for the e-cigarette. On the VC10, nicotine concentration ranged 10.0–93.9 ng/cm2/puff for the cigarette and 4.0–12.3 ng/cm2/puff for the e-cigarette. The deposited aerosol from a conventional cigarette and an e-cigarette in vitro are compositionally different; this emphasises the importance of understanding and characterising different product aerosols using dosimetry tools. This will enable easier extrapolation and comparison of pre-clinical data and consumer use studies, to help further explore the reduced risk potential of next generation nicotine products.

Summary Nicotine has been analyzed by gas chromatography nitrogen-phosphorus detection in tissue samples obtained by repeated minimal trauma tissue biopsies from human subcutaneous adipose tissue. For sample preparation, a single extraction step of the tissue samples with chloroform was performed at 30–45°C. Calibration curves generated with spiked porcine subcutaneous adipose tissue were linear over a concentration range from 0.20 to 100 μg g−1, therefore, the limit of quantification was fixed at 0.20 μg g−1. The limit of detection was found to be 0.05 μg g−1 adipose tissue. The recovery of nicotinespiked porcine subcutaneous adipose tissue by chloroform extraction was 100±8%. The performance of the assay was not affected by the complex lipid matrix. The method was employed for analysis in a clinical study on nicotine penetration from a transdermal delivery system through the skin of moderate smokers. Low‐rate dynamic contact angles of 12 liquids on a poly(methyl methacrylate/ethyl methacrylate, 30/70) P(MMA/EMA, 30/70) copolymer were measured by an automated axisymmetric drop shape analysis‐profile (ADSA‐P). It was found that five liquids yield nonconstant contact angles, and/or dissolve the polymer on contact. From the experimental contact angles of the remaining seven liquids, it is found that the liquid–vapor surface tension times cosine of the contact angle changes smoothly with the liquid–vapor surface tension (i.e., γl|Kv cos θ depends only on γl|Kv for a given solid surface or solid surface tension). This contact angle pattern is in harmony with those from other methacrylate polymer surfaces previously studied.45,50 The solid–vapor surface tension calculated from the equation‐of‐state approach for solid–liquid interfacial tensions14 is found to be 35.1 mJ/m2, with a 95% confidence limit of ± 0.3 mJ/m2, from the experimental contact angles of the seven liquids. © 1999 John Wiley & Sons, Inc. Considering the importance of alkaloids in tobacco smoke, their presence was evaluated in real work environments. Sampling was carried out with fixed pumps and eventual degradation of the alkaloids during collection and storage until analysis was tested. A quantification method with an alternative internal standard (quinaldine) was evaluated and proposed. Offices and leisure rooms with high smoking intensity were chosen in order to detect minor alkaloids. The correlation between the environmental tobacco smoke markers - nicotine, 3-ethenylpyridine and myosmine - and the minor compounds was evaluated and all R2 were higher than 0.538. Nicotine levels quantified in the office rooms ranged from 0.02 to 64.67pg/m3 and a maximum of 129.33pg/m3 was found in one leisure room with very high smoking intensity. Myosnline and nicotyrine were the most abundant minor alkaloids and very high contents were. quantified in the leisure room mentioned previously.  相似文献

A method based on ultrasound‐assisted liquid–liquid extraction and high‐performance liquid chromatography has been optimized for the determination of six polybrominated diphenyl ether congeners. The optimal condition relevant to the extraction was first investigated, more than 98.7 ± 0.7% recovery was achieved with dichloromethane as extractant, 5 min extraction time, and three cycles of ultrasound‐assisted liquid–liquid extraction. Then multiple function was employed to optimize polybrominated diphenyl ether detection conditions with overall resolution and chromatography signal area as the responses. The condition chosen in this experiment was methanol/water 93:7 v/v, flow rate 0.80 mL/min, column temperature 30.0°C. The optimized technique revealed good linearity (R2 > 0.9962 over a concentration range of 1–100 μg/L) and repeatability (relative standard deviation < 6.3%). Furthermore, the detection limit (S/N = 3) of the method were ranged from 0.02 to 0.13 μg/L and the quantification limit (S/N = 10) ranged from 0.07 to 0.35 μg/L. Finally, the proposed method was applied to spiked samples and satisfactory results were achieved. These results indicate that ultrasound‐assisted liquid–liquid extraction coupled with high‐performance liquid chromatography was effective to identify and quantify the complex polybrominated diphenyl ethers in effluent samples.  相似文献

A sensitive and selective method for simultaneous determination of 29 toxic alkaloids in human blood and 31 in urine using high-performance liquid chromatography–electrospray ionization-tandem mass spectrometry was developed and validated. The samples were diluted with 0.1 mol L−1 HCl, and the target alkaloids were purified by solid phase extraction. The separation of 31 alkaloids was carried out on a C18 column using a gradient mobile phase with 10 mmol L−1 ammonium formate in water with 0.1% formic acid and methanol at the rate of 0.25 mL min−1. The triple-quadrupole mass spectrometer equipped with an electrospray source in the positive mode was set up in the dynamic multiple reactions monitoring mode (dynamic MRM) to detect the ion transitions of 31 alkaloids. The calibration curves were linear over a range of 0.5–400, 1–400, or 4–400 μg L−1 for target alkaloids in human blood and urine, and the correlation coefficients (r 2) was higher than 0.9943. The limit of determination and limit of quantification were 0.2–1 and 0.5–4 μg L−1 for blood and urine, respectively. The only exceptions were sanguinarine and chelerythrine in human blood. All the target alkaloids were stable under the test condition. In addition, the solvent effect and reconstituted solution were investigated. The method was validated and proved to be accurate and precise over the studied concentrations and suitable for poisoning diagnosis and forensic toxicology.

For the rational utilization and the quantitative quality control of the Stephania yunnanensis Lo, an HPLC‐DAD method was developed for the quantitative and simultaneous determination of five alkaloids in rat plasma (stepharine, sinomenine, palmatine, isocorydine and tetrahydropalmatine), which were the main active chemical constituents of this plant and belong to four kinds of isoquinoline‐type alkaloids (protoberberine, morphine, aporphine and protaporphine alkaloids). The contents of five alkaloids ranged from 0.09 to 2.32% (w/w). The method validation was tested for the linearity (r2 > 0.9975), precision (intra‐day RSD < 4.8% and inter‐day RSD < 4.9%), extraction recovery (85.49 ± 2.29% to 99.21 ± 1.48%) and stability (98.5 ± 5.3% to 101.2 ± 3.4%). We developed an HPLC‐DAD method to simultaneously measure these alkaloids in rat plasma after oral administration of the extract of this plant to rats. The results supported the hypothesis that isoquinoline alkaloids were the compounds responsible for the main pharmacological activities for anti‐inflammatory and analgesic.  相似文献

A simple HPLC‐UV method was developed and validated for the quantification of pterostilbene (3,5‐dimethoxy‐4'‐hydroxy‐trans‐stilbene), a pharmacologically active phytoalexin in rat plasma. The assay was carried out by measuring the UV absorbance at 320 nm. Pterostilbene and the internal standard, 3,5,4'‐trimethoxy‐trans‐stilbene eluted at 5.7 and 9.2 min, respectively. The calibration curve (20–2000 ng/mL) was linear (R2 > 0.997). The lower limits of detection and of quantification were 6.7 and 20 ng/mL, respectively. The intra‐ and inter‐day precisions in terms of RSD were all lower than 6%. The analytical recovery ranged from 95.5 ± 3.7 to 103.2 ± 0.7% while the absolute recovery ranged from 101.9 ± 1.1 to 104.9 ± 4.4%. This simple HPLC method was subsequently applied in a pharmacokinetic study carried out in Sprague–Dawley rats. The terminal elimination half‐life and clearance of pterostilbene were 96.6 ± 23.7 min and 37.0 ± 2.5 mL/min/kg, respectively, while its absolute oral bioavailability was 12.5 ± 4.7%. A rapid and cost‐effective method based on microwave‐assisted extraction followed by capillary electrophoresis was developed for simultaneous quantification of seven alkaloids in Corydalis decumbens for the first time. The main parameters affecting microwave‐assisted extraction and capillary electrophoresis separation were investigated and optimized. The optimal microwave‐assisted extraction was performed at 40°C for 5 min using methanol/water (90:10, v/v) as the extracting solvent. Electrophoretic separation was achieved within 15 min using an uncoated fused‐silica capillary (50 μm internal diameter and 27.7 cm effective length) and a 500 mM Tris buffer containing 45% v/v methanol (titrated to pH* 2.86 with H3PO4). The developed method was successfully applied to the quantification of seven alkaloids in Corydalis decumbens obtained from different regions of China. The combination of microwave‐assisted extraction with capillary electrophoresis was an effective method for the rapid analysis of the alkaloids in Corydalis decumbens .  相似文献

A novel oil‐in‐salt liquid‐phase microextraction was developed and introduced for the extraction and concentration of the trace levels of active alkaloids in Coptis chinensis prior to being analyzed by high‐performance liquid chromatography with ultraviolet detection. Also, the oil‐in‐salt extraction mechanism was analyzed, the enrichment factor and extraction recovery were redefined, and the proposed method was compared with other methods. In the approach, the mixed solvent of pentanol/octanol (6:4, v/v) and NaCl (20% w/v) are immobilized on the permutite surface in turn to form oil‐in‐salt double membranes, through which the target analytes can be molecularized though salting‐out effect and be extracted by organic solvent. The main parameters affecting the approach were investigated and optimized. Under the optimized conditions, the enrichment factors of the analytes were 30–117, the linear ranges were 0.002–2 μg/mL for jatrorrhizine, coptisine, and palmatine, and 0.001–3 μg/mL for berberine (r 2 ≥ 0.9923). The limits of detection were less than 1 ng/mL. Satisfactory recoveries (84.3%–120.3%) and precision (0.9%–7.5%) were also obtained. These results confirm that the approach is a simple and reliable sample pretreatment procedure and allows for the quantification of active alkaloids in C. chinensis at actual concentration levels.  相似文献

A sensitive high‐performance liquid chromatography–positive ion electrospray tandem mass spectrometry (LC‐MS/MS) method was developed and validated for the quantification of methyllycaconitine (MLA) in rat plasma and brain tissue. Following acetonitrile protein precipitation, the analyte was separated using a gradient mobile phase on a reversed‐phase column and analyzed by MS/MS in the multiple reaction monitoring mode using the respective [M + H]+ ions, m/z 683–216 for MLA and m/z 260–116 for the internal standard. The assay exhibited a linear dynamic range of 0.5–250 ng/mL for MLA in rat plasma and brain tissue. The lower limit of quantification was 0.5 ng/mL. Acceptable precision (<12%) and accuracy (100 ± 6%) were obtained for concentrations over the standard curve range. Nicotine (NC) and its related compounds (cotinine (CN), nornicotine (NN), anatabine (AT) and anabasine (AB)) were simultaneously enantioseparated by CE using a capillary with amino groups and sulfated β‐CD as a chiral selector. The optimum running conditions were found to be 30 mM acetate buffer (pH 5.0) containing 8% sulfated β‐CD with an applied voltage of +15 kV at 30°C using direct detection at 260 nm. Using a capillary coated with amino groups, the EOF migrates toward the positive pole. However, when sulfated β‐CD was added to the BGE, it was found that the EOF migrated toward the negative pole due to ionic adsorption of sulfated β‐CD to amino groups on the capillary inner wall. All the cationic analytes migrated as anions, suggesting that they formed stable anionic complexes with sulfated β‐CD. With this system and a simple pretreatment with mini‐cartridges, NC alkaloids in five cigarette samples were enantioseparated. As a result, each of the compounds except for CN was detected. In the case of NC, only (S)‐NC was detected (more than 99.9%), but in the case of NN, AT and AB, the ratios of (S)‐isomer to total isomers were in the ranges 58–70, 81–85 and 59–65%, respectively. On the other hand, only NC was detected in cigarette smoke and the ratio of (S)‐ and (R)‐NCs was 96:4. The amounts of NC alkaloids in cigarettes suggest that the production of (R)‐NC resulted from racemization due to the high temperature/burning of the cigarette.  相似文献

A method of vortex‐assisted dispersive liquid–liquid extraction coupled with gas chromatography and tandem mass spectrometry for the determination of nicotine and cotinine in urine was developed. Response surface methodology was applied to obtain the optimum extraction conditions. In this method, Plackett–Burman design was utilized to evaluate the impact of five selected factors on pretreatment procedure. Then, three main factors were optimized using a Box–Behnken design. The optimized method showed good linearities at 1–2000 μg/L with correlation coefficients of 0.9998 for nicotine and 0.9986 for cotinine. Recovery was 91.4–106 and 91.7–108% for nicotine and cotinine, respectively. The intraday relative standard derivations of determination were 1.47–4.06% for nicotine and 0.41–3.16% for cotinine, and interday relative standard derivations were 3.03–6.70% for nicotine and 1.64–6.38% for cotinine. The method detection limits for nicotine and cotinine were 0.33 and 0.34 μg/L, respectively. A total of 87 urine samples from smokers and nonsmokers were tested with the proposed method. Urinary nicotine and cotinine were 23.0–6.67 × 103 and 18.4–4.17 × 103 μg/(g·cr) for smokers and 1.31–286 and 1.39–131 μg/(g·cr) for nonsmokers, respectively. The method is sensitive, suitable and reliable for the determination of nicotine and cotinine in urine and meets the requirements for evaluating short‐term tobacco exposure.  相似文献

Norisoboldine (NIB) is one of the main bioactive isoquinoline alkaloids in Linderae Radix. A rapid, selective and sensitive method using UPLC‐ESI/MS was first developed for simultaneous determination of NIB and norisoboldine‐9‐Oα‐glucuronide (NIB‐Glu), its major metabolite in rat plasma. A one‐step protein precipitation with methanol was employed as sample preparation technique. Chromatographic separation was carried out on an Acquity UPLC BEH C18 column (50 × 2.1 mm, i.d. 1.7 µm) with a gradient mobile phase consisting of acetonitrile and water containing 0.1% formic acid. Detection and quantification were performed using a quadrupole mass spectrometer by selective ion reaction‐monitoring mode. Good linearity was achieved using weighted (1/x2) least squares linear regression over the concentration ranges 0.01–2 µg/mL for NIB and 0.025–25 µg/mL for NIB‐Glu. The lower limit of quantification of NIB and NIB‐Glu was 0.01 and 0.025 µg/mL, respectively. The intra‐ and inter‐day precisions (relative standard deviations) of the assay at all three quality control levels were 4.6–14.1% for NIB, and 5.0–12.2% for NIB‐Glu. The accuracies (relative error) were −13.5–8.1% for NIB and −12.8–7.6% for NIB‐Glu, respectively. The effects of ammonium sulfate aerosols on the kinetics of the hydroxyl radical reactions with C1–C6 aliphatic alcohols have been investigated using the relative rate technique. P‐xylene was used as a reference compound for the C2–C6 aliphatic alcohols study, and ethanol was used as a reference compound for the methanol study. Two different aerosol concentrations that are typical of polluted urban conditions were tested. The total surface areas of aerosols were 1400 μm2 cm?3 (condition I) and 3400 μm2 cm?3 (condition II). Results indicate that ammonium sulfate aerosols promote the ethanol/OH radical and 1‐propanol/OH radical reactions as compared to the p‐xylene/OH radical reaction. The relative rate of the ethanol/·OH reaction versus the p‐xylene/·OH reaction increased from 0.19 ± 0.01 in the absence of aerosols to 0.24 ± 0.01 and 0.26 ± 0.02 under aerosol conditions I and II, respectively. The relative rate of the 1‐propanol/·OH reaction versus the p‐xylene/·OH reaction increased from 0.45 ± 0.03 in the absence aerosols to 0.56 ± 0.02 and 0.55 ± 0.03 under aerosol conditions I and II, respectively. However, significant changes in the relative rates of the 1‐butanol/·OH, 1‐pentanol/·OH, and 1‐hexanol/·OH reactions versus the p‐xylene/·OH reaction were not observed for either aerosol concentration. The relative rates of the methanol/·OH reaction versus the ethanol/·OH reaction were identical in the absence and presence of aerosols. These results indicate that ammonium sulfate aerosols promote the methanol/·OH reaction as much as the ethanol/·OH reaction (as compared to the p‐xylene/·OH reaction). © 2001 John Wiley & Sons, Inc. Nicotine was selected as the target molecule for monitoring of the smoking process by multicommutation Fourier Transform Infrared spectroscopy (FTIR). The method involved the use of CHCl3 for on-line extraction of nicotine from tobacco, cigarette filters and tobacco ash from NH4OH alkalinized samples, and absorbance measurement of the characteristic band at 1316 cm−1 in the stopped-flow mode, by obtaining the peak area in the range between 1334 and 1300 cm−1. Under the best operational conditions, the procedure developed provided a detection limit of 0.05 mg mL−1 nicotine, corresponding to 0.5 mg g−1 in the solid sample, a relative standard deviation less than 2.5%, and a sampling frequency of 12 determinations h−1. It can be concluded that nicotine migrates in the smoke mainstream towards the filter during the smoking process. The smoking of cigarettes and cigars is different. Nicotine is retained weakly by both tobacco and filter in the case of cigarettes, and strongly by the unburned tobacco in cigars. The incomplete smoking of cigars and cigarettes reduces nicotine intake and thus reduces the additive effect.  相似文献

The aim of the present study was to develop a method based on gas chromatography–tandem mass spectrometry (GC–MS/MS) to determine and quantify the d ‐limonene in mouse plasma and tissue samples. This new method was validated for the quantification of d ‐limonene with the linearity ranges 1.0–1000.0 ng/mL (r2 > 0.9952) for plasma samples and 5.0–5000.0 ng/g (r2 > 0.9940) for tissue samples. The intra‐ and inter‐day assay of precisions in plasma and tissues were <13.4% and the accuracies were within 91.1–105.8%. In the oral/inhalation administration pharmacokinetics and tissue distribution studies, the main pharmacokinetic parameters were the peak concentration = (97.150 ± 34.450)/(4336.415 ± 1142.418) ng/mL, the area under the curve = (162.828± 27.447)/(2085.721 ± 547.787) h ng/mL and the half‐life = (3.196 ± 0.825)/(0.989 ± 0.095) h. The tissue distribution of d ‐limonene in mice after oral/inhalation administration demonstrated a decreasing tendency in different tissues (liver > kidney > heart > lung > spleen).  相似文献

N,N–dimethylacetamide is an excipient used in intravenous busulfan formulations, a drug used in hematopoietic stem cell transplantation conditioning. The aim of this study was to develop and validate a liquid chromatography-tandem mass spectrometry method for simultaneous quantification of N,N-dimethylacetamide, and its metabolite N-monomethylacetamide in plasma from children receiving busulfan. A 4 μl aliquot of patient plasma was extracted using 196 μl 50% methanol solution and quantified against calibrators prepared in the extraction solvent given negligible matrix effects across three concentrations. 9[H2]-N,N-dimethylacetamide was used as an internal standard. Separation of N,N-dimethylacetamide and N-monomethylacetamide was achieved using a Kinetex EVO C18 stationary phase (100 mm × 2.1 mm × 2.6 μm) running an isocratic mobile phase of 30% methanol containing 0.1% formic acid at a flow of 0.2 ml/min over 3.0 min. The injection volume was 1 μl. Calibration curves for N,N-dimethylacetamide and N-monomethylacetamide were linear up to 1200 and 200 μg/L, respectively, with a lower limit of quantification 1 μg/L for both analytes. Calibrator accuracy and precision were within ± 10% of the test parameters across four concentration levels. Analytes were stable over 14 days at three different storage conditions. This method was successfully applied to measure N,N-dimethylacetamide and N-monomethylacetamide concentrations in a total of 1265 plasma samples from 77 children.  相似文献

A simple, rapid, sensitive and reliable liquid chromatographic method for the quantification of BP-1107 in rat plasma has been established. Plasma samples were prepared by extraction with tert-butyl methyl ether, and troglitazone was used as an internal standard. The analytical separation was performed on a C18 column using acetonitrile–0.3% phosphoric acid in water (pH 4.00 adjusted with triethylamine) (75:25, v/v) as a mobile phase. A detailed validation of the method was performed as per USFDA guidelines. For BP-1107 at the concentrations of 2.42, 16.11 and 32.22 μg mL?1 in rat plasma, the extraction recoveries were 114.14 ± 9.75, 95.37 ± 12.06 and 90.00 ± 6.46%, respectively. The mean recovery for internal standard was 91.96 ± 2.51%. The lower limit of quantitation of BP-1107 was 16 ng. The linear quantification range of the method was 0.81–53.70 μg mL?1 in rat plasma with a correlation coefficient greater than 0.999. The intra-day and inter-day accuracy for BP-1107 at 2.42, 16.11 and 32.22 μg mL?1 levels in rat plasma fell between 97.10–110.02 and 97.52–108.04%. The intra-day and inter-day precision were in the ranges of 1.91–5.63 and 4.43–6.28%, respectively. The method was successfully applied to a pharmacokinetic study of BP-1107 in rats after an intravenous administration.  相似文献
