The synthesis and structure-activity-relationship (SAR) for a series of N-substituted piperazinyl carbamoyl 7-15 and piperazinyl acetyl 18-26 derivatives of tetrahydropapaverine have been carried out. The general synthetic methods of carbamoyl tetrahydropapaverine analogues involve N-substituted piperazines and carbamoyl imidazole tetrahydropapaverine as starting materials. Another route for synthesizing these compounds, involving the formation of carbamoyl imidazole piperazine has also been explored. Acylation of tetrahydropapaverine followed by substitution with various piperazinyl moities afforded the acetyl tetrahydropapaverine derivatives. Variously substituted piperazines have been used to monitor the effect of electron releasing and electron withdrawing substituents upon the antispasmodic activity of the molecules. Effect of varying electron densities on the antispasmodic activity, by altering the position of these groups on the benzene ring has also been monitored. Pharmacological methods involve the in vitro antispasmodic activity studies on a freshly removed guinea pig ileum using a force displacement transducer amplifier connected to a physiograph. Among the analogues synthesized in the present study, a promising compound 7, a potent muscle relaxant as compared to papaverine has been obtained.  相似文献

An efficient preparation of linear and curved bis- and branched tris-5-(2,2'-bipyridines) of nanoscopic dimensions possessing rigid conjugated bridges is presented. The synthesis, which avoids the need of protection/deprotection methodology, utilizes central bridge precursors which are outwardly di- and trifunctionalized with a 5-(2-chloropyridine) synthon via a chemoselective palladium-catalyzed Sonogashira or Negishi cross-coupling protocol to yield the bridged linear (5a-c, 5f,g) and curved (6, 7) bis- and branched (8) tris-5-(2-chloropyridines). Under more forcing conditions, the ethyne-bridged 5-(2-chloropyridines) undergo the Stille cross-coupling reacton with 2-trimethylstannylpyridines to afford the conjugatively bridged linear (1a,b, 1g-j) and curved (2a, b, 3a,b) bis- and branched, (4a,b) tris-5-(2,2'-bipyridines) in good overall yields. The phenyl- and biphenyl-bridged linear bis-5-(2, 2'-bipyridines) (1c-f) were best prepared from the bis-5-(2-bromopyridines) (5d,e) to ensure completion of the Stille cross-coupling reactions. The Stille cross-couplings showed a marked substituent effect in which the terminally phenylated bis- and tris-5-(2,2'-bipyridines) were formed in higher yields than the methyl-substituted analogues with the same bridge. The advantages of the methodology lie in its synthetic convenience and adaptibility for creating multitopic metal ion-binding scaffolds with a potentially very large variety of bridging units and substituents on the terminal pyridine rings. The bridged 5-(2-chloropyridines) may also serve as precursors for the fabrication of metal ion-coordinated conjugated polymers.  相似文献

Abstract

The four possible diastereoisomers of 4,5-diphenylcyclophosphamides (2–5) and their N-methyl analogues (6–9) have been synthesized via condensation of aminopropanols 10 and 11 (threo- and erythro) with bis-(2-chloroethyl)-phosphoramidic dichloride 12. Anticancer screening tests against L 1210 lymphoid leukaemia in mice did not show any therapeutic activity.  相似文献

Present studies were undertaken on the preparation of synthetic analogues of bis- or tetra-coumarins and their activity against HIV-1 integrase (HIV-1 IN). Among these coumarin analogues, compounds 14, 16 and 18 were found to be potent molecules against HIV-1 IN at IC50 values of 0.96, 0.58, and 0.49 microM, respectively. The results provided a tool for guiding the further design of more potent antiviral agents and for predicting the affinity of related compounds.  相似文献

A novel class of potent human gastric lipase inhibitors, bis-2-oxo amide triacylglycerol analogues, was developed. These analogues of the natural substrate of lipases were prepared starting from 1,3-diaminopropan-2-ol. They were designed to contain the 2-oxo amide functionality in place of the scissile ester bond at the sn-1 and sn-3 position, while the ester bond at the sn-2 position was either maintained or replaced by an ether bond. The derivatives synthesized were tested for their ability to form stable monomolecular films at the air/water interface by recording their surface pressure/molecular area compression isotherms. The inhibition of human pancreatic and gastric lipases by the bis-2-oxo amides was studied using the monolayer technique with mixed films of 1,2-dicaprin containing variable proportions of each inhibitor. The nature of the functional group (ester or ether), as well as the chain length, at the sn-2 position influenced the potency of the inhibition. Among the compounds tested, 2-[(2-oxohexadecanoyl)amino]-1-[[(2-oxohexadecanoyl)-amino]methyl]ethyl decanoate was the most potent inhibitor, causing a 50% decrease in HPL and HGL activities at 0.076 and 0.020 surface molar fractions, respectively.  相似文献

During further chemical and biological investigations of Picrasma quassioides BENNET, four new bis-β-carboline alkaloids, quassidines E-H (1-4), and three new β-carboline alkaloids, canthin-16-one-14-butyric acid (5), 3-(1,1-dimethoxylmethyl)-β-carboline (6), and 6,12-dimethoxy-3-formyl-β-carboline (7), were isolated from its anti-inflammatory CHCl(3)-soluble fraction. Structures of new compounds were elucidated and characterized by MS and NMR analysis. A plausible biogenetic pathway for quassidine E (1), the first bis-β-carboline alkaloid in which a canthin-6-one moiety and a β-carboline moiety were connected together by a single carbon-carbon bond from the nature, was proposed. Quassidines E-G (1-3) showed potent inhibitory activity on the production of nitric oxide (NO), tumor necrosis factor α (TNF-α), or interleukin 6 (IL-6) in mouse monocyte-macrophage RAW264.7 cells stimulated by lipopolysaccharide (LPS). Analysis of anti-inflammatory activity of all β-carboline and bis-β-carboline alkaloids from P. quassioides showed that the carbonyl groups or double carbon-carbon bonds at C-14 for β-carbolines and C-14' for bis-β-carbolines were bioactive groups for their in vitro anti-inflammatory activity. Structure-activity relationship of these compounds on inhibitory activity of the three inflammatory cytokines was discussed.  相似文献

A series of p-tert-butylcalix[4]arene derivatives containing 1,4(1,5)-benzodiazepinone fragments as substituents were prepared. The biological activity of the compounds, namely, their antispasmodic activity in mice, was evaluated by antagonism with a spasmodic agent, Corazol, upon transdermal administration. According to the data obtained, benzodiazepine derivatives of calixarenes show higher antispasmodic activity compared to 3-hydroxyphenazepam, taking into account their considerably higher molecular weight.  相似文献

Two monomeric, five-coordinate lanthanide complexes, [bis-5,5'-(1,3-propanediyldiimino)-2,2-dimethyl-4-hexene-3-onato]samarium[2,6-bis(tert-butyl)-4-methylphenoxide] and [bis-5,5'-(1,3-propanediyldiimino)-2,2-dimethyl-4-hexene-3-onato]erbium[2,6-bis(tert-butyl)-4-methylphenoxide], were isolated from the reactions of 2,6-bis(tert-butyl)-4-methylphenol with [bis-5,5'-(1,3-propanediyldiimino)-2,2-dimethyl-4-hexene-3-onato]lanthanide[bis(trimethylsilyl)amido] (lanthanide = Er(3+) and Sm(3+)). The purified phenoxides were recovered in excellent yields and analytical purity, and the reactions proceeded cleanly without Schiff-base degradation or cluster formation. Analogously, [bis-3,3'-(1,3-propanediyldiimino)-1-phenyl-2-butene-1-onato]erbium[bis(trimethylsilyl)amido] was also directly converted to [bis-3,3'-(1,3-propanediyldiimino)-1-phenyl-2-butene-1-onato]erbium[2,6-bis(tert-butyl)-4-methylphenoxide]; however, a less sterically demanding alcohol (i.e., ethanol) yielded a neutral trinuclear oxo alkoxide species with each dianionic Schiff base asymmetrically bridging through micro-oxo interactions. In this polynuclear cluster, each symmetry-related, seven-coordinate erbium(III) ion exhibits monocapped trigonal prismatic geometry, which assembles by sharing triangular capped faces. Single-crystal X-ray diffraction revealed square-pyramidal metal coordination in each five-coordinate lanthanide ion with varied S(4) ruffling of the "square base" donor atoms and the six-membered propylene diamine chelate ring adopting the boat conformation. To contrast the effect of subtle ligand changes, we also report the synthesis and characterization of [bis-5,5'-(2,2-dimethyl-1,3-propanediyldiimino)-2,2-dimethyl-4-hexene-3-onato]samarium[bis(trimethylsilyl)amido], having gem-dimethyl substituents appended to the propylene bridge central carbon. The six-membered diamine chelate ring in this compound adopts the chair conformation without metal-hydrocarbon interaction. Also presented are qualitative activity observations and polymerization data for the polymerization of rac-lactide and epsilon-caprolactone using the five-coordinate lanthanide amidos and phenoxides.  相似文献

The interfacial behavior of aqueous solutions of newly synthesized bis- and tris-ammonium salts (i.e., bis[2-hydroxy-3-(dodecyldimethylammonio)propyl]alkylamine dichlorides and bis[2-hydroxy-3-(dodecyldimethylammonio)propyl]dialkylammmonium trichlorides, respectively) was analyzed, both experimentally and theoretically. The dynamic and equilibrium surface tension of multiple ammonium salt solutions was measured by using a pendant drop shape analysis method. The determined surface tension isotherms indicated the lack of significant differences in surface activity between bis- and tris-ammonium salts, contrary to the expectations for divalent and trivalent surfactant ions. That effect was explained by assuming the formation of multiple surfactant ion-counterion associates. Taking into account the association process, a good correlation between experimental data and theoretical predictions was obtained by means of the "surface quasi two-dimensional electrolyte" (STDE) model of ionic surfactant adsorption. The degree of association necessary to explain the lack of difference in surface activity between bis- and tris-ammonium salts was in quantitative agreement with the results of measurements of the concentration of free chloride anions in the surfactant solution.  相似文献

A series of novel mono-1,2,3-triazole and bis-1,2,3-triazole acyclonucleoside analogues of 9-(4-hydroxybutyl)guanine was prepared via copper(I)-catalyzed 1,3-dipolar cycloaddition of N-9 propargylpurine, N-1-propargylpyrimidines/as-triazine with the azido-pseudo-sugar 4-azidobutylacetate under solvent-free microwave conditions, followed by treatment with K(2)CO(3)/MeOH, or NH(3)/MeOH. All compounds studied in this work were screened for their antiviral activities [against human rhinovirus (HRV) and hepatitis C virus (HCV)] and antibacterial activities against a series of Gram positive and negative bacteria.  相似文献

The Ullmann amination reaction was utilized to provide access to a number of fluorene analogues from common intermediates, via facile functionalization at positions 2, 7, and 9 of the fluorene ring. Through variation of amine or iodofluorene derivative, analogues bearing substitutents with varying electron-donating and electron-withdrawing ability, e.g., diphenylamino, bis-(4-methoxyphenyl)amine, nitro, and benzothiazole, were synthesized in good yield. The novel fluorene derivatives were fully characterized, including absorption and emission spectra. Didecylation at the 9-position afforded remarkably soluble derivatives. Target compounds 4, 5, and 9 are potentially useful as fluorophores in two-photon fluorescence microscopy. Their UV-vis spectra display desirable absorption in the range of interest suitable for two-photon excitation by near-IR femtosecond lasers. Preliminary measurements of two-photon absorption indicate the derivatives exhibit high two-photon absorptivity, affirming their potential as two-photon fluorophores. For example, using a 1,210 nm femtosecond pump beam, diphenylaminobenzothiazolylfluorene 4 exhibited nondegenerate two-photon absorption, with two-photon absorptivity (delta) of ca. 820 x 10(-50) cm(4) s photon(-1) molecule(-1) at the femtosecond white light continuum probe wavelength of 615 nm.  相似文献

The reaction of pyrylium salt with primary amine has been utilized to synthesize 4,4'-bipyri-dinium salts and pyridinium polymers. Seven new 4,4'-(1,4-phenylene-) bis-[N-alkyl(aryl)-2,6-diphenyl-] pyridinium perchlorates 4 and five corresponding bipyridinium polymers 5 were preparedwith good yields from 4,4'-(1,4-phenylene-) bis-(2,6-diphenyl-) pyrylium perchlorate 3. Two bis-(6-chloropurines) bridged by conformationally restricted tethers were synthesized as potential DNA bis-intercalating agents. Reduction of 4,6-dichloro-5-nitropyrimidine ( 1 ) afforded 5-amino-4,6-dichloropyrimidine ( 2 ) which was then used as the starting material. Reaction of 2 with 4,4′-diaminodiphenylmethane ( 3 ) and bis-(4-aminophenyl) ether ( 4 ) yielded bis-[4-(N-5-amino-4-chloro-6-pyrimidyl)aminophenyl]methane ( 5 ) and bis-[4-(N-5-amino-4-chloro-6-pyrimidyl)aminophenyl] ether ( 6 ), respectively. Acid-catalyzed condensation of the above pyrimidines, 5 and 6 , with triethyl orthoformate in N,N-dimethylacetamide gave bis-[4-(6-chloro-9-purinyl)phenyl]methane ( 7 ) and bis-[4-(6-chloro-9-purinyl)phenyl] ether ( 8 ). The spectral data on the new compounds will be discussed.  相似文献

[structures: see text] The absolute configuration of 1,2-, 1,3-, 1,4-, and 1,5-diols formed by two secondary (chiral) hydroxy groups can be deduced by comparison of the NMR spectra of the corresponding bis-(R)- and bis-(S)-MPA esters. The correlation between the NMR spectra of the bis-ester derivatives and the absolute stereochemistry of the diol involves the comparison of the chemical shifts of the signals for substituents R1/R2 and for the hydrogens attached to the two chiral centers [H(alpha)(R1) and H(alpha)(R2)] in the bis-(R)- and the bis-(S)-ester and is expressed as delta deltaRS. Theoretical calculations [energy minimization by semiempirical (AM1), ab initio (HF), DFT (B3LYP), and Onsager methods, and aromatic shielding effect calculations] and experimental data (NMR and CD spectroscopy) indicate that in these bis-MPA esters, the experimental delta deltaRS values are the result of the contribution of the shielding/deshielding effects produced by the two MPA units that combine according to the actual stereochemistry of the diol. The reliability of these correlations is demonstrated with a wide range of diols of known absolute configuration derivatized with MPA and 9-AMA as auxiliary reagents. A simple graphical model that allows the simultaneous assignment of the two asymmetric carbons of a 1,n-diol by comparison of the NMR spectra (delta deltaRS signs) of its bis-(R)- and bis-(S)-AMAA ester derivatives is presented.  相似文献

Hydrolysis of the asymmetric pyridine- and phenol-containing ligand HL (1) (2-hydroxy-4-6-di- tert-butylbenzyl-2-pyridylmethyl)imine) led to the use of bis-(3,5-di -tert-butyl-2-phenolato-benzaldehyde)copper(II), [Cu (II)(L (SAL)) 2] ( 1) as a precursor for bis-(2,4-di- tert-butyl-6-octadecyliminomethyl-phenolato)copper(II), [Cu (II)(L (2)) 2] ( 3), bis-(2,4-di- tert-butyl-6-octadecyl aminomethyl-phenolato)copper(II), [Cu (II)(L (2A)) 2] ( 3'), and bis-(2,4-di- tert-butyl-6-[(3,4,5-tris-dodecyloxy-phenylimino)-methyl]-phenolato)copper(II), [Cu (II)(L (3)) 2] ( 4). These complexes exhibit hydrophilic copper-containing head groups, hydrophobic alkyl and alkoxo tails, and present potential as precursors for redox-responsive Langmuir-Blodgett films. All systems were characterized by means of elemental, spectrometric, spectroscopic, and electrochemical techniques, and their amphiphilic properties were probed by means of compression isotherms and Brewster angle microscopy. Good redox activity was observed for 3 with two phenoxyl radical processes between 0.5 and 0.8 V vs Fc (+)/Fc, but this complex lacks amphiphilic behavior. To attain good balance between redox response and amphiphilicity, increased core flexibility in 3' and incorporation of alkoxy chains in 4 were attempted. Film formation with collapse at 14 mN.m (-1) was observed for the alkoxy-derivative but redox-response was seriously compromised. Core flexibility improved Langmuir film formation with a higher formal collapse and showed excellent cyclability of the ligand-based processes.  相似文献
