A divergent chemoenzymaytic approach for the preparation of core‐fucosylated and core‐unmodified asymmetrical N‐glycans from a common advances precursor is described. An undecasaccharide was synthesized by sequential chemical glycosylations of an orthogonally protected core fucosylated hexasaccharide that is common to all mammalian core fucosylated N‐glycans. Antennae‐selective enzymatic extension of the undecasaccharide using a panel of glycosyl transferases afforded core fucosylated asymmetrical triantennary N‐glycan isomers, which are potential biomarkers for breast cancer. A unique aspect of our approach is that a fucosidase (FucA1) has been identified that selectively can cleave a core‐fucoside without affecting the fucoside of a sialyl LewisX epitope to give easy access to core‐unmodified compounds.  相似文献

The asymmetric total synthesis of natural azasugars (+)‐castanospermine, (+)‐7‐deoxy‐6‐epi‐castanospermine, and synthetic (+)‐1‐epi‐castanospermine has been accomplished in nine to ten steps from a common chiral building block (S)‐ 8 . The method features a powerful chiral relay strategy consisting of a highly diastereoselective vinylogous Mukaiyama‐type reaction with either chiral or achiral aldehydes (≥95 % de; de=diastereomeric excess) and a diastereodivergent reduction of tetramic acids, which allows formation of three continuous stereogenic centers with high diastereoselectivities. The method also provides a flexible access to structural arrays of 5‐(α‐hydroxyalkyl)tetramic acids, such as 17/34 , and 5‐(α‐hydroxyalkyl)‐4‐hydroxyl‐2‐pyrrolidinones, such as 18 and 25/35 a . The method constitutes the first realization of the challenging chiral synthons A and D and thus of the conceptually attractive retrosynthetic analysis shown in Scheme 1 in a highly enantioselective manner.  相似文献

Macrolines constitute a class of natural products that has more than 100 members and displays diverse biological activities. These compounds feature a cycloocta[b]indole scaffold that represents an interesting target structure for biology‐oriented synthesis (BIOS). We have presented a solid‐phase synthesis of isomerically pure cycloocta[b]indoles by employing the Pictet–Spengler reaction and the Dieckmann cyclization as key steps. The scope of this reaction sequence was investigated in more detail by using various additional diversification procedures, such as Pd‐catalyzed Sonogashira or Suzuki couplings on a solid phase, thus allowing, for example, the generation of 10‐substituted cycloocta[b]indole derivatives. Finally, solution‐phase decoration of the cycloocta[b]indole skeleton by reduction and saponification was evaluated, thereby further extending the scope of the solid‐phase synthesis.  相似文献

The preparation of a series of copolymers of Nt‐butylacrylamide (NTBAM) with acrylamide (AM) is reported. The insolubility of NTBAM in water led to the testing of methanol, t‐butanol, and mixtures of these solvents with water to obtain effective copolymerization. Several of these polymerizations produced nonhomogeneous product mixtures. Samples of the components were separated and characterized by photoacoustic Fourier transform infrared spectroscopy and 13C NMR spectroscopy. Hydrodynamic volumes of the products were obtained from solution‐viscosity measurements, gel permeation chromatography, and multi‐angle laser light scattering methods. The NTBAM‐co‐AM copolymers had degrees of polymerization and molecular weights in the 4.1–5.9 × 104 monomer units and 3.25–4.5 × 106 g/mol range, respectively. This article extends the preparative details of a series of nonionic copolymers of acrylamide with N,N‐dimethylacrylamide, methacrylamide, and Nt‐butylacrylamide to the synthesis of cationic derivatives of these new copolymers. The described procedures gave products with cationicities of 14–26 mol %. We measured the mean squared radii of gyration and intrinsic viscosities of aqueous solutions of these products at several different pHs and NaCl concentrations to compare these values with those determined for the nonionic precursors and related commercial cationic polymers. Because the molecular weights of the examples measured varied widely, it was difficult to establish definite trends. However, the large values obtained for the mean squared radii of gyration and intrinsic viscosities, relative to the nonionic precursors of these polymers, demonstrated that the charged groups had a qualitatively greater effect on polymer extension than the nonpolar bulky groups. © 2001 John Wiley & Sons, Inc. A convergent and efficient transition‐metal‐free catalytic synthesis of 2‐aryl‐indoles has been developed. The interception of a highly reactive and transient aza‐ortho‐quinone methide by an acyl anion equivalent generated through N‐hetereocyclic carbene catalysis is central to this successful strategy. The first asymmetric total synthesis of (?)‐ophiodilactone A and (?)‐ophiodilactone B, isolated from the ophiuroid (Ophiocoma scolopendrina), is reported. The key features of the synthesis include the highly stereocontrolled construction of the structurally congested γ‐lactone/δ‐lactone skeleton through an asymmetric epoxidation, diastereoselective iodolactonization, and intramolecular epoxide‐opening with a carboxylic acid, and biomimetic radical cyclization of ophiodilactone A to ophiodilactone B.  相似文献

Divergent total syntheses of the enmein‐type natural products (?)‐enmein, (?)‐isodocarpin, and (?)‐sculponin R have been achieved in a concise fashion. Key features of the strategy include 1) an efficient early‐stage cage formation to control succeeding diastereoselectivity, 2) a one‐pot acylation/akylation/lactonization to construct the C‐ring and C8 quarternary center, 3) a reductive alkenylation approach to construct the enmain D/E rings and 4) a flexible route to allow divergent syntheses of three natural products.  相似文献

Short and highly efficient stereoselective syntheses provide machaeriols and cannabinoids in a divergent approach starting from a common precursor, commercially available (S)‐perillic acid. Key features of the novel strategy are a stereospecific palladium‐catalyzed decarboxylative arylation and a one‐pot sequence comprising a stereoselective hydroboration followed by oxidation or reduction of the corresponding intermediary boranes. The divergent approach is convincingly demonstrated by the five‐step syntheses of (+)‐machaeriol B, (+)‐machaeriol D, and related analogues, and the four‐step synthesis of (+)‐Δ8‐THC and an analogue.  相似文献
