Abstract

A new cyclic pentapeptide, pentaminolarin (1), and a new cytochalasin, xylochalasin (2), along with thirteen known compounds (315) were isolated from the wood-decaying fungus Xylaria sp. SWUF08-37. The absolute configurations of 1 were determined by a combination of Marfey’s method and TDDFT ECD calculation and the absolute configurations of 2 were established by TDDFT ECD calculation. Compound 12 showed moderate cytotoxicity against HeLa (IC50?=?19.60?µg/mL), HT29 (IC50?=?17.31?µg/mL), HCT116 (IC50?=?14.28?µg/mL), MCF-7 (IC50?=?15.38?µg/mL), and Vero (IC50?=?24.97?µg/mL) cell lines by MTT assay. Compounds 1 and 2 showed slight cytotoxicity against all tested cancer cell lines.  相似文献

A new phenolic glucoside, abeliaside, along with four known compounds, 5,6,7,4′-tetrahydroxy flavones, caffeic acid, 4-O-caffeoylquinic acid and caffeic acid glucoside, was isolated from the leaves of Abelia triflora R. Br. (Caprifoliaceae). The structure of the new compound was elucidated by different spectroscopic techniques. Compounds 15 were assayed for their anticancer activities against two cancerous human cell lines, MCF-7 and PC-3 cells, and normal Vero cell line using the crystal violet staining method. From the results it could be seen that caffeic acid possessed the highest anticancer effect against MCF-7 (IC50: 17 μg/mL) and PC-3 (IC50: 20.1 μg/mL) compared to vinblastine sulphate as reference drug (IC50: 4.6, 2.8 μg/mL). The other compounds showed weak anticancer activity on both cell lines.  相似文献

Cinnamomum stenophyllum (Meisn.) Vattimo-Gil (Lauraceae) is a native and vulnerable Brazilian species restricted to the Atlantic Forest. The leaf essential oil obtained by hydrodistillation was characterized for the first time by two-dimensional gas chromatography with time-of-flight mass spectrometry (GC × GC-TOFMS). This analysis resulted in the tentatively identification of 80 compounds, showing the superior performance of this method in comparison to the seven compounds identified by GC–MS. The identified compounds included 8 ketones, 7 monoterpene hydrocarbons, 30 oxygenated monoterpenes, 4 sesquiterpene hydrocarbons and 23 oxygenated sesquiterpenes, showing that the C. stenophyllum oil contained mostly oxygenated mono and sesquiterpenes. The oil cytotoxicity was tested against two human cancer cell lines, colon adenocarcinoma (HCT-116) and breast cancer carcinoma (MCF-7), and the non-tumor retinal pigment epithelial cells (RPE) using the colorimetric MTT assay. Both cancer cell lines were sensible to leaf essential oil, with IC50 < 20 μg/mL (HCT, IC50 = 9.95 μg/mL and MCF-7, IC50 = 16.65 μg/mL), while there was no cytotoxicity against the non-tumor cells at tested concentrations (IC50 > 50 μg/mL), suggesting selectivity to cancer cells. The results showed that the C. stenophyllum leaf essential oil has a cytotoxic potential, presenting several compounds already known as biologically active against tumor cells.  相似文献

Three new xanthones, named calaxanthones A-C (13), along with 17 known xanthones (420) were isolated from the roots of Calophyllum calaba. Their structures were determined by spectroscopic analysis. All isolated compounds were evaluated for their cytotoxicity against five cancer cell lines (KB, HeLa S-3, HT-29, MCF-7 and HepG-2). Compound 3 showed potent cytotoxicity against all the five cancer cell lines with IC50 values in the range of 0.82–5.04 μM. Furthermore, compound 6 showed potent cytotoxicity against KB, HeLa S-3 and HepG2 cells with IC50 values of 7.06, 5.27 and 9.64 μM, respectively. Additionally, compound 7 showed potent cytotoxicity against KB cell with an IC50 value of 4.62 μM.  相似文献

The chloroform fraction of methanol (MeOH) extract of the aerial parts of Launaea mucronata was in vitro investigated for cytotoxicity against HCT116, HepG2 and MCF-7 cell lines, and resulted with IC50 = 20.0, 18.6 and 14.30 μg/mL, respectively. The chloroform fraction of the MeOH extract was subjected to further fractionation, which led to the isolation of two new coumarin compounds (6-isobutyl coumarin and 6-isobutyl-7-methyl- coumarin). The structures of the new compounds were elucidated by high field 1D and 2D NMR and ESI-MS spectroscopies. Low polar fractions revealed the isolation of other known triterpene compounds which were identified according to its spectral data and comparison with the literature. New coumarin compounds show high cytotoxicity against MCF-7, HCT116 and HepG2 cell lines.  相似文献

Abstract

The pigment was extracted from Penicillium aculeatum, purified and characterized as Ankaflavin by spectroscopic analysis. The stability of the pigment was determined under various conditions and was found to possess high stability. The cytotoxicity property of the purified pigment was determined by MTT assay in MCF-7, HCT116 and PC-3 and the studies were compared with its activity in CHOK1 cells. In MCF-7 and in CHOK 1 cells, the pigment exhibited very less toxicity. However, significant cytotoxicity was observed in HCT116 and PC-3 cells with IC50 of 162?μg mL?1 and 85?μg mL?1 for HCT116 and PC-3 cells respectively. In vitro toxicity was tested by haemolysis assay and MTT assay in HEK 293 cells. The pigment showed least cytotoxicity (<5%) at 160 and 320?μg mL?1 concentrations HEK 293 cells and negligible (<5%) toxicity on human erythrocytes at 160 and 320?μg mL?1, the highest concentrations tested.  相似文献

The EtOAc and MeOH extracts of the roots of Toddalia asiatica Lam. were investigated for the roots’ chemical constituents. Two new compounds including 2′R-acetoxytoddanol (1) and 8S-10-O-demethylbocconoline (3) as well as 15 known compounds were isolated. Compound 10 showed strong cytotoxicity against KB cells with an IC50 value of 2.60 μg/mL, which is nearly equal to the ellipticine standard, but showed no activity against Vero cells. Alkaloid 3 displayed weak cytotoxicity against the KB cell line with an IC50 value of 21.69 μg/mL.  相似文献

Cancer cases are alarmingly increasing worldwide, and newer chemotherapeutic agents are needed. Recent analogs of cisplatin (carboplatin, lobaplatin, nedaplatin and oxaliplatin) and their marketing as advanced chemotherapeutic drugs have furthered the interest in metal-based anti-cancer drugs. In the current study, two new polymeric organotin(IV) carboxylate complexes (1 and 2) have been synthesized and characterized. Spectroscopic studies showed that coordination took place via carboxylates. Furthermore, X-ray crystallographic study on 1 indicated that it possesses a monomeric structure and exists in polymeric formation due to additional Sn–N coordination, assigning seven coordinations to each metal ion. Both the complexes were tested against three cancerous (human colon cancer, HCT 116; breast cancer, MCF-7; and leukemia, K562) and one non-cancerous (3T3-L1) cell lines. Complex 1 showed exceptional cytotoxicity against cancerous cell lines (IC50 = 1.0 μM for HCT 116; 258.7 nM for MCF-7; and 46.7 nM K562) and remained comparatively non-toxic against normal cells (IC50 = 37.0?μM). This shows that both complexes have selective cytotoxicity against cancer cells.  相似文献

Abstract

Twenty-four compounds were isolated from the roots of Polygonatum cyrtonema Hua, including a new octopamine dimer, named trans-bis(N-feruloyl)octopamine (1). The structure was established on the basis of spectroscopic and chemical methods. All the extracts and compounds were evaluated for cytotoxic and antioxidant activities by using MTT and chemiluminescence assay. The extracts showed activity against MCF-7 and HepG-2 cell lines from IC50 0.30 to 1.01 mg mL?1. Compound 3 exhibited activity against HepG-2 cell lines with IC50 8.99?μM. Compound 7 exhibited activity against Hela cell lines with IC50 2.53?μM and BGC-823 cell lines with IC50 7.77?μM. Moreover, compound 7 showed antioxidant with IC50 12?µM compared to the positive control with IC50 77?µM. Compound 16 exhibited activity against HepG-2 cell lines with IC50 1.05?μM and MCF-7 cell lines with IC50 1.89?μM. These results indicated that this plant might be potential in natural medicine and healthy food.  相似文献

This study was carried out to determine the cytotoxic effect of seven plant extracts and the isolated compounds – syringin and 4-methoxycinnamyl alcohol – on cancerous and non-cancerous cells. The ethanol extract of Foeniculum vulgare was found to exhibit the most significant toxicity with an IC50 value of 19.97 μg/mL on HeLa cells. Bioassay-guided fractionation led to the isolation of two compounds, syringin (1) and 4-methoxycinnamyl alcohol (2). Both compounds showed toxicity against MCF-7, HeLa and DU145 cancer cell line. The results showed that compound 2 showed high toxicity against all the cancer cell lines with IC50 values of 14.24, 7.82 and 22.10 μg/mL, respectively. 4-Methoxycinnamyl alcohol also showed no apoptotic effect in cell cycle analysis after 48 h at a concentration of 10 μg/mL. However, DNA fragmentation study revealed that necrosis took place at a concentration of 10 μg/mL after 48 h exposure.  相似文献

Chemical investigation of the soft coral Sarcophyton glaucum collected from the Red Sea led to isolation of 11 isoprenoidal metabolites (111). A new sesquiterpenoid, 6-oxo-germacra-4(15),8,11-triene (1), a new natural cembranoid, sarcophinediol, along with two known sesquiterpenoids (2 and 3) and seven known cembranoids (511) was obtained. The structures of the compounds were established based on their NMR, MS, IR and UV spectral data. All compounds were evaluated for their cytotoxicity employing three cancer cell lines (HepG2, MCF-7 and HCT116). Compounds 4 and 6 showed significant cytotoxicity towards HepG2 with IC50 values of 18.8 ± 0.07 and 19.9 ± 0.02 μM; respectively. Compounds 57 exhibited potent cytotoxicity against MCF-7 cells with IC50 values of 9.9 ± 0.03, 2.4 ± 0.04 and 3.2 ± 0.02 μM, respectively. Compounds 1, 4 and 5 showed significant activities towards HCT116 cells with IC50 values of 29.4 ± 0.03, 19.4 ± 0.02 and 25.8 ± 0.03 μM, respectively.  相似文献

Abstract

Two new ceanothane triterpenes, 3,7-O,O-dibenzoyl ceanothic acid methylester (1) and 3-O-acetyl-7-O-benzoyl ceanothic acid methylester (2), along with nine known compounds (311), were isolated from the roots of Colubrina asiatica. The isolated compounds were identified by spectroscopic evidence. Compounds 1 and 2 showed antimalarial activity against Plasmodium falciparum with IC50 values of 4.67 and 3.07?µg/mL, respectively. Compound 2 also showed antimycobacterial activity against Mycobacterium tuberculosis (MIC 6.25?µg/mL). In addition, compounds 1, 2, 10 and 11 showed cytotoxicity against three cancer cell lines (KB, NCI-H187 and MCF-7) with IC50 values ranging from 8.32 to 46.72?µg/mL.  相似文献

The crude extracts of tubers of Aconitum spicatum (Bruhl) Stapf were investigated for in vitro antileishmanial activity against Leishmania major. The dichloromethane extract at pH 2.5 showed antileishmanial activity with IC50 value of 27.10 ± 0.0 μg/mL. Chromatographic purification of the dichloromethane extract led to isolation of three C-19 norditerpenoid alkaloids indaconitine (1), chasmaconitine (2) and ludaconitine (3). Compounds 3 and 2 showed antileishmanial activity with IC50 = 36.10 ± 3.4 and 56.30 ± 2.1 μg/mL, respectively. Compound 1 was less effective (IC50 > 100 μg/mL). The cytotoxicity of compounds 1, 2 and 3 studied against MCF7, HeLa and PC3 cancer cell lines and 3T3 normal fibroblast cell line did not show cytotoxicity at 30 μM.  相似文献

Two new p-terphenyls, neonambiterphenyls A and B (12), a new benzoquinone, neonambiquinone A (3), together with six known sesquiterpenes (49), were isolated from the bioluminescent mushroom Neonothopanus nambi PW3. The isolated compounds were identified by mass, IR and spectroscopic analyses (1D and 2D NMR). Compounds 13 and 57 showed cytotoxicity against cancer cell lines such as KB, NCI-H187 and MCF-7 with IC50 values ranging from 1.45 to 49.31?µg/mL. Using various chromatographic separations, sixteen compounds, including one new triterpene saponin named aegicoroside A (1), were isolated from the leaves of the Vietnamese mangrove Aegiceras corniculatum. Their structures were determined by spectroscopic methods such as 1D and 2D NMR and HR-ESI-MS. The cytotoxic activities of the isolated compounds against MCF7 (breast), HCT116 (colon), B16F10 (melanoma), and A549 (adenocarcinoma) cancer cell lines were also evaluated. Strong cytotoxicity was observed for sakurasosaponin (2) against all four cancer cell lines and for sakurasosaponin methyl ester (3) against MCF7, A549, and HCT116 cell lines with IC50 values ranging from 2.89 ± 0.02 to 9.86 ± 0.21 μM.  相似文献

Two new neoclerodane diterpenoids, barbatin F (1), barbatin G (2) together with four known compounds, scutebata A (3), scutebata B (4), scutebata C (5) and scutebata P (6) were isolated from the whole plant of Scutellaria barbata D.Don. Their structures were elucidated on the basis of spectroscopic studies. Six unknown cyathane diterpenes, named cyathinins A – E, and 10-hydroxyerinacine S, were isolated from Cyathus subglobisporus BCC44381 along with six known compounds, striatoid C, striatin C, striatals A, C, D, and glochidone. The chemical structures were determined by means of high resolution mass spectrometry (HRESI-MS) and nuclear magnetic resonance (NMR) spectroscopy. The absolute configurations were confirmed by X-ray diffraction analysis. The isolated active compounds exhibited antimalarial against Plasmodium falciparum (IC50 0.88–7.51?μM), antibacterial activities against Mycobacterium tuberculosis (MIC 25–50?μg/mL), Gram-positive bacteria of Bacillus cereus and Enterococcus faecium (MIC 0.78–50?μg/mL) and Gram-negative bacteria of Escherichia coli, Acinetobacter baumannii and Klebsiella pneumoniae in a presence of phenylalanine-arginine β-naphthylamide (MIC 3.13–50?μg/mL), antifungal activity against Candida albicans (IC50 8.6–80.3?μM) activities, and cytotoxic activity against both cancerous (MCF-7, KB, NCI-H187, IC50 0.36–28.32?μM) and non-cancerous (Vero, IC50 0.13–91.50?μM) cells.  相似文献

Three new bufadienolides 14β, 16β-dihydroxy-3β-[β-D-glucopyranosyl-(1→6)-(β-D-glucopyranosyl)oxy]-5α-bufa-20, 22-dienolide (1), 14β-hydroxy-3β-[β-D-glucopyranosyl-(1→4)-(β-D-glucopyranosyl)oxy]-5α-bufa-20, 22-dienolide (2) and hellebrigenin-3-O-β-D-glucosyl-(1→4)-β-D-glucoside (3), together with eight known bufadienolides (411) were isolated from the roots and rhizomes of Helleborus thibetanus. Their structures were elucidated by extensive spectroscopic methods and acid hydrolysis. Compounds 17 were evaluated for their cytotoxic activity against HCT116, A549 and HepG2 tumor cell lines. Compound 1 exhibited moderate cytotoxicity against HepG2 cells with IC50 value of 15.1?±?1.72?μM. Abstract

A series of new pyrido-cyclopenta[1,2-b]indole derivatives were synthesized via Knoevenagel reaction and followed by reflux with dimethylformamide dimethyl acetal. Their structures were investigated by spectral techniques and elemental analysis. In vitro antibacterial assessment against seven selected microorganisms evidenced that the compounds with halogen substituent have strong inhibitory action than that of the reference drugs. The antioxidant results were apparent that the compounds 5b, 5c, and 6c manifested explicit activity when compared with Butylhydroxyanisole and Vitamin-C. Cytotoxic activity analysis toward HeLa and MCF-7 cell lines was also assessed. Analogs 6c (IC50 values 15.1?μM and 18.6?μM) and 6d (IC50 values 17.4?μM and 20.7?μM) illustrated the interesting cytotoxicity activity. Molecular docking studies against p38 MAP kinase displayed a potential binding affinity with the receptor. Furthermore, in silico pharmacokinetic studies articulated the drug-likeness nature of the target compounds.  相似文献
