采用分光光度法研究了电子给体苯巴比妥钠与π电子受体茜素红的荷移反应,建立了荷移分光光度法测定苯巴比妥钠的方法。在水溶液中,苯巴比妥钠与茜素红荷移络合物的最大吸收波长为530 nm,该络合物的组成为1∶1,表观摩尔吸光系数ε为4.43×103L.mol-1.cm-1,稳定常数为2.30×105。药物质量浓度在5~40 mg/L范围内符合比尔定律,相关系数为0.9996。当苯巴比妥钠浓度为20 mg/L时,10次测定结果的相对标准偏差为1.3%。测定了针剂中的苯巴比妥钠,加标回收率在98.9%~105%之间。  相似文献

The interaction of carbamazepine and phenobarbital in rabbits was investigated. The drugs were administered to the rabbit orally as a single dose. By simultaneous administration the sequence of drugs was varied, with an interval between doses of 15 min. The doses of carbamazepine and phenobarbital were 100 and 25 mg, respectively. It was established that phenobarbital appears to be an inductor for carbamazepine independently sequence of administration of the drugs. Carbamazepine reveals inductive properties for phenobarbital in the case where phenobarbital enteres in the organism first. It was ascertained also that, by simultaneous administration, these drugs reduce absorption of each other in plasma.  相似文献

We prepared a phenobarbital (PB) solid dispersion (SD) with phosphatidylcholine (PC). PB was present in an amorphous state in SD if its mole fraction was under 0.75. An infrared (IR) spectra study suggested a hydrogen bond between NH in PB and phosphate in PC, with a ratio of about 1:1. When the mole fraction of PB was less than 0.50, differential scanning calorimetry (DSC) curves showed endothermic peaks at 57, 90 and 145 degrees C, and an exothermic peak at 60 degrees C. The IR spectrum and X-ray diffraction pattern changed after holding at 70 degrees C, so at this point it is considered that the metastable state of SD changed into a stable state, and extra energy was released. When the mole fraction of PB was high, PB also arranged near hydrophobic group because an endothermic peak was observed at 46-52 degrees C, which was lower than fully hydrated PC. PB is similar to indomethacin (IM) in molecular shape and to phenytoin (PHT) in chemical structure. Its DSC curve and IR spectra are similar to PHT, and the limit ratio of its amorphous state is the same as IM. It is considered that the chemical structure is an important factor in its interaction to PC, and also, the molecular shape is important to arrange into PC lattice.  相似文献

A new technique for determination of phenobarbital by homogeneous fluoroimmunoassay is described. Simultaneous determination of free and antibody-bound fluorescein-labelled phenobarbital is accomplished by making phase-resolved fluorescence measurements, in which selectivity is provided by the difference between the fluorescence lifetimes of the two species, as well as by the difference in fluorescence intensities. The technique requires no specialized reagents other than those inherent in immunoassay (antibody, antigen and labelled antigen). Problems due to non-specific binding of labelled antigen to sample matrix components can be alleviated by addition of a relative excess of albumin.  相似文献

A new liquid chromatographic procedure for the determination of phenobarbital in plasma samples is described. The proposed system uses a Spherisorb octadecyl-silane ODS-2 C(18) analytical column, a guard column of similar characteristics, and a 0.03 M CTAB-3% 1-propanol at pH 7 mobile phase. The UV detector was set at 250 nm. Butabarbital was used as internal standard. Sample preparation only required the addition to the plasma samples of a 0.1 M SDS solution at pH 3 and centrifugation before injection into the chromatographic system. The limit of detection was 0.83 microg/mL of phenobarbital in plasma samples. The coefficients of variation were lower than 7. 5%.  相似文献

An analytical procedure is described for determining residues of sodium phenobarbital in animal chow at levels as low as 0.14 ppm. The methanol extract is subjected to a liquid-liquid cleanup at pH 13 and 1, further cleaned up on a silica gel column and assayed by high-pressure liquid chromatography by using an ultraviolet absorption detector at 210 nm. Data concerning extraction efficiency, partition values and stability of the chemical in animal chow are also presented.  相似文献

The functionally excellent flash-heater methylation, ethylation and butylation techniques have been extended to include the higher alkyl homologs. Phenobarbital and diphenylhydantoin have been alkylated using the tetraalkylammonium hydroxides in which the alkyl group ranges from methyl to hexyl. The dissolution of phenobarbital (PB) from solid dispersion with phosphatidylcholine (PC) was studied. PB was present in an amorphous state in solid dispersion (PB-PC) if the mole fraction of PB was under 0.75. Thus, supersaturation was observed when an excess amount of PB-PC was dispersed in pH 1.2 and 6.8 media. The degree of supersaturation was largest when the mole fraction of PB was 0.25, although it was only 1.3-fold of the PB solubility in this case. Dissolution from PB-PC was rapid and complete in both pH 1.2 and 6.8 media regardless of the mole fraction of PB, above 90% within 5 min. Bioavailability after the oral administration of PB-PC to rabbits with a dose of 15 mg/kg equivalent to PB was compared with that of PB crystals. The area under the plasma concentration curve was bigger, but not significant. The maximum concentration was significantly higher, and the time to maximum concentration was significantly faster. These results indicate that the absorption rate became high with PB-PC because the dissolution was rapid.  相似文献

The effect of -cyclodextrin (-CD) on the disposition of hexobarbital (HBA) and phenobarbital (PhBA) after intravenous administration, with the simultaneous administration with -CD, was investigated in mice.In the case of HBA, the whole blood concentration was slightly heightened, the brain and liver concentrations were significantly lowered, and the kidney concentration was significantly heightened. Moreover, -CD also influenced the disposition and formation of 3-hydroxy- and 3-keto-HBA. On the other hand, in the case of PhBA, the whole blood concentration was slightly lowered, the time at maximum brain concentration (t-max) was prolonged about 30 to 60 min, the kidney concentration was significantly heightened at the initial stage, while the liver concentration did not show a clear difference by the simultaneous administration with -CD. These results suggest that the disposition of drugs might be modified by the use of CD.  相似文献

A sensitive differential pulse polarographic assay was developed for the determination of phenobarbital or diphenylhydantoin in blood. The assay involves the selective extraction of the compound into chloroform from whole blood buffered to pH 7.0. After suitable “clean-up” of the sample, each compound is nitrated in 10% potassium nitrate in sulfuric acid at 25° for 1 h. The nitro-derivatives are extracted into ethyl acetate, and the residues are dissolved in 1 M phosphate buffer (pH 7.0) or 0.1 M sodium hydroxide for phenobarbital and diphenylhydantoin, respectively; the solutions are deoxygenated, and analyzed by differential pulse polarography. The overall recovery of phenobarbital and diphenylhydantoin from blood was 72.3% ±6.5 (sr) and 76.7 ±2.3 (sr) respectively. The sensitivity limit is 1–2 μg ml-1 of blood for both compounds. A modified assay for the determination of both compounds in blood with t.l.c. separation was also developed.  相似文献

A microchip electrophoresis method with laser induced fluorescence detection was developed for the immunoassay of phenobarbital. The detection was based on the competitive immunoreaction between analyte phenobarbital and fluorescein isothiocyanate (FITC) labeled phenobarbital with a limited amount of antibody. The assay was developed by varying the borate concentration, buffer pH, separation voltage, and incubation time. A running buffer system containing 35 mM borate and 15 mM sodium dodecyl sulfate (pH 9.5), and 2800 V separation voltage provided analysis conditions for a high-resolution, sensitive, and repeatable assay of phenobarbital. Free FITC-labeled phenobarbital and immunocomplex were separated within 30s. The calibration curve for phenobarbital had a detection limit of 3.4 nM and a range of 8.6-860.0 nM. The assay could be used to determine the phenobarbital plasma concentration in clinical plasma sample.  相似文献

The preferential solvation parameters of phenobarbital in aqueous binary mixtures of 1,4-dioxane, t-butanol, n-propanol, ethanol, propylene glycol and glycerol were derived from solution thermodynamic properties by using the IKBI method. This drug is sensitive to preferential solvation effects in all these mixtures. The preferential solvation parameter by the cosolvent (δx1,3) is negative in almost all the water-rich mixtures but positive in mixtures with similar proportions of solvents and cosolvent-rich mixtures, except in 1-propanol + water mixtures, where negative values are also found in mixtures with x1 ≥ 0.70. Hydrophobic hydration around the non-polar ethyl and phenyl groups of this drug in water-rich mixtures could play a relevant role in drug solvation. Otherwise, in mixtures of similar solvent compositions and in cosolvent-rich mixtures the preferential solvation by cosolvent could be due to the acidic behaviour of the drug.  相似文献

In this paper, a high-performance thin-layer chromatography (HPTLC) method combined with densitometry has been described. Chromatography was performed on silica gel Si 60F254 plates using dichloromethane-ethyl acetate-formic acid (9.5 +/- 0.5 +/- 0.1, v/v) mobile phase. This method has been successfully applied for the determination of phenobarbital in pharmaceuticals. Obtained results were comparable with traditionally used column high-performance liquid chromatography (HPLC) methods. For the proposed procedure, linearity (r > 0.999), sensitivity (limit of detection 0.4 microg/spot), recovery (97.8-102.1%), and repeatability were found to be satisfactory. The HPTLC-densitometry method has many advantages, such as simplicity, reasonable sensitivity, rapidity, and low cost, and it can be successfully used in routine quality control of multidrug preparations containing barbiturates.  相似文献

The use of multivariate spectrophotometric calibration for the simultaneous determination of the active components of antiepileptic tablets is presented. The resolution of binary mixtures of phenobarbital and phenytoin has been accomplished by using partial least squares (PLS-1) regression analysis. Although the components show an important degree of spectral overlap, they have been simultaneously determined with high accuracy, with no interference from tablet excipients. A comparison is presented with the related multivariate method of classical least squares (CLS) analysis, which is shown to yield less reliable results due to the severe spectral overlap presented by the studied compounds. A statistical measure for the spectral overlap is proposed.  相似文献

The formation constants of the binary complexes Cu(CM)2+ and Cu(CM) 2 2+ as well as those of the ternary complexes Cu(CM)L + and Cu(CM)2 L + (CM=Cimetidine=N-Cyano-N-methyl-N[(5-methyl-1H-imidazol-4-yl)methyltioethyl]-guanidine; HL=Phenobarbital=5-ethyl-5-phenyl-barbituric acid) have been determined in 0.1 and 1.0 mol dm–3 NaClO4 methanol solutions at 25±0.2°C. The values of logX, log stat.., and logK confirm the stability of the ternary complexes.

Zusammenfassung Die Stabilitätskonstanten der binären Komplexe Cu(CM)2+ und Cu(CM) 2 2+ sowie die der ternären Komplexe Cu(CM)L + und Cu(CM)2 L + (CM=Cimetidin=N-Cyan-N-methyl-N-[(5-methyl-1H-imidazol-4-yl)methylthioethyl]-guanidin; HL=Phenobarbitalum=5-Ethyl-5-phenyl-barbitursäure) wurden in 0.1 und 1.0M Lösungen von NaClO4 in Methanol bei 25±0.2°C bestimmt. Die Werte von logX, log stat. und logK bestätigen die Stabilität der ternären Komplexe.
