Methods are described of synthesizing various 3-carboxy-4-vinyl pyrrolidines, versatile building blocks for our drug discovery efforts. The 1,3-dipolar cycloaddition between activated olefins and nonstabilized azomethine ylide is a known method for synthesizing pyrrolidines in a stereospecific manner. Steric and electronic effects on the chemoselectivity of the 1,3-dipolar cycloaddition between azomethine ylide and α,β,γ,δ-unsaturated carboxylates have been explored.  相似文献

A nonstabilized azomethine ylide reacts with a wide range of substituted isatoic anhydrides to afford novel 1,3-benzodiazepin-5-one derivatives, which are generally isolated in high yield. The transformations involve 1,3-dipolar cycloaddition reactions of the ylide with the anhydrides to give transient, and in a representative case spectroscopically observable, oxazolidine intermediates that undergo ring-opening-decarboxylation-ring-closing reaction cascades to yield the 1,3-benzodiazepin-5-one products.  相似文献

Reactions of 3-nitro-2-trifluoro(trichloro)methyl-2H-chromenes, including 2-unsubstituted derivatives, with N-alkyl-α-amino acids (sarcosine, proline) and paraformaldehyde proceed diastereoselectively to give 1-benzopyrano[3,4-c]pyrrolidines in good yields as a result of a 1,3-dipolar cycloaddition of the intermediate nonstabilized azomethine ylide at the Δ3-bond of the chromene system.  相似文献

Various new structural entities related to x-azatricyclo[m.n.0. 0(a)()(,)(b)()]alkanes are constructed by the intramolecular [3 + 2] dipolar cycloaddition of nonstabilized cyclic azomethine ylides. Chromones bearing electron-withdrawing substituents at the 2- or 3-position react with nonstabilized azomethine ylides to produce 1-benzopyrano[2,3-c]pyrrolidines in good yields. Reactions of 3-cyanochromones proceed diastereoselectively to give 1-benzopyrano[2,3-c]pyrrolidines and tetrahydro-1H-spiro[chromeno[2,3-c]pyrrol-9,5′-oxazolidine]-9a-carbonitriles, depending on the reactant ratio, as a result of a 1,3-dipolar cycloaddition of the azomethine ylide at the double bond and carbonyl group of the chromone system. The latter undergoes demethylenation and recyclization into a novel hexahydrochromeno[2,3-c:3,4-c′]dipyrrole tetracyclic system on heating with hydrochloric acid.  相似文献

A simple and efficient method for the synthesis of tricyclic imidazolines derivatives via [3 + 2] 1,3-dipolar cycloaddition between nonstabilized azomethine ylide generated in situ with unactivated cyclic imines is reported here. The method provided easy and mild access to various fused tricyclic hexahydroimidazo[5,1-a]isoquinolines in excellent yields (up to 96%). This protocol is simple and easy to handle.  相似文献

Benzaldehydes react smoothly with the nonstabilized azomethine ylide derived from sarcosine and formaldehyde to form 5-aryloxazolidines, which undergo ring-opening to give 2-(indol-3-ylmethylamino)-1-arylethanols in 69–79% yields on reaction with indoles in acetic acid. Their subsequent acid-catalyzed cyclization into 4-aryl-2-methyl-1,2,3,4-tetrahydro-γ-carbolines was performed in polyphosphoric acid in moderate yields. The latter can also be prepared directly from 5-aryloxazolidines and indoles in polyphosphoric acid.  相似文献

1,3-Dipolar cycloaddition to Cyo of an azomethine ylide,which was generated in situ from refluxing a mixture of cyclohexanone and DL-valine in chlorobenzene under N2 afforded a monoadduct and a bisadduct.Even for longer reaction time,however,only monoadduct was obtained from the reaction of C70 with an azomethine ylide generated from refluxing a mixture of 2-undecanone and 2-aminoisobutyric acid in chlorobenzene under N2 The electrochemical properties of all the products were studied by cyclic voltammetry.  相似文献

Catalytic amounts of a protic reagent such as tert-butyl alcohol promote the isomerization of a stable amino-aryl-carbene into a transient azomethine ylide. Deprotonation of an alkyl-aldiminium salt also leads to a transient azomethine ylide, but labeling experiments rule out the transient formation of the corresponding amino-alkyl-carbene. An efficient access to 4-aryl imidazole scaffolds is described in two steps and one operation. Cyanoaryl-imidazolines are easily obtained by a (3?+?2) heterocyloaddition involving cyanobenzenes as dipolarophiles and a non-stabilized azomethine ylide as an electron-rich 1,3-dipole. Oxidation of the crude mixture affords an efficient two-steps access to the corresponding imidazoles.  相似文献

N-(Porphyrin-2-ylmethyl)glycine was synthesized and used as precursor of azomethine ylide, which was trapped with several dipolarophiles. The reaction of that azomethine ylide with dimethyl fumarate afforded the expected adduct. However, with 1,4-benzo- and 1,4-naphthoquinones only dehydrogenated adducts were isolated. Also, the reaction of that ylide with meso-tetrakis(pentafluorophenyl)porphyrin and tetraazaporphine allowed access to novel porphyrin-chlorin and porphyrin-tetraazachlorin dyads.  相似文献

Practical stereoselective synthetic routes to the antihistaminic drug olopatadine and its E-isomer have been developed, the key steps being a trans stereoselective Wittig olefination using a nonstabilized phosphorus ylide and a stereoselective Heck cyclization. The BF3OEt2 catalyzed one-pot 1,3-dipolar cycloaddition reaction of benzylamines, isatins and dimethyl acetylenedicarboxylate in dry methylene dichloride afforded the functionalized dihydrospiro[indoline-3,2'-pyrroles] in moderate to good yields and with high diastereoselectivity. The reaction was accomplished by the tandem 1,3-dioplar cycloaddition of in situ generated azomethine ylide with acetylenedicarboxylate and the nucleophilic addition of pyrrole ring to second molecular acetylenedicarboxylate.  相似文献
