A new dipyrroloquinone, zyzzyanone A 1, having a pyrrolo[3,2-f]indole-4,8(1H,7H)-dione skeleton, was isolated from the Australian marine sponge Zyzzya fuliginosa, along with the known pyrroloquinoline alkaloids, makaluvamines C, E, G, H, and L, and damirones A and B. The structure of 1 was determined by spectroscopic data. Zyzzyanone A 1 shows moderate cytotoxic activity against mouse Ehrlich carcinoma cells (IC50 25 μg/mL), inhibits the cell division of fertilized sea urchin eggs at a concentration of 25 μg/mL, and exhibits UV-A and UV-B absorbing activity.  相似文献

New glycosyl derived ligand and its complexes, with SnCl4·5H2O (1) and (CH3)2SnCl2(2) were synthesized and characterized by spectroscopic (IR, 1H, 13C, and 119Sn NMR, UV-vis, ESI-MS) and analytical methods. Interaction studies of 1 and 2 with CT DNA were studied by using various biophysical techniques, which showed high binding affinity of 2 with CT DNA. In vitro cytotoxicity of complexes 1 and 2 were evaluated against different human cancer cell lines of different histological origins by employing SRB Assay. The organotin(IV) complex 2 exhibited remarkable activity against DWD (oral cancer) cell lines with GI50 values <10 μg/ml. Complex 2 induced apoptosis of DWD cell line at a very low concentration of 1-4 μg/mL.  相似文献

The structures and biological properties of peptides produced by two genera of marine-derived fungi, an atypical Acremonium sp., and a Metarrhizium sp., were explored. The Acremonium strain was isolated from a marine sponge and has previously been shown by our group to produce peptides from the efrapeptin and RHM families. The isolation and structural elucidation of the new linear pentadecapeptides efrapeptins Eα (1) and H (2) and N-methylated octapeptides RHM3 (3) and RHM4 (4) were carried out through a combination of 1D and 2D NMR techniques and tandem MS. Additional known efrapeptins E, F, and G and the known syctalidamides A and B were also isolated. The absolute configurations of 1-4 are proposed to be the same as the original compound families. The marine sponge-derived Metarrhizium sp. was shown to produce destruxin cyclic depsipeptides including A, B, B2, desmethyl B, E chlorohydrin, and E2 chlorohydrin. Efrapeptins Eα (1), F, and G each displayed IC50s of 1.3 nM against H125 cells, and destruxin E2 chlorohydrin displayed an IC50 of 160 nM against HCT-116 cells. An initial therapeutic assessment suggested a continuous (168 h) exposure of at least 2 ng/mL, or a daily (24 h) exposure of at least 300 ng/mL for H125 cells treated with efrapeptin G, and a continuous (168 h) exposure of at least 190 ng/mL for HCT-116 cells treated with destruxin E2 chlorohydrin, will cause 90% tumor cell death in vitro.  相似文献

Fourteen new norditerpenoids (abiesanordines A-N, 1-14), including a novel podocarpene bearing a rare enolic structure (abiesanordine A, 1), were isolated from Abies georgei together with eight known ones. Their structures were determined mainly by detailed analysis of 1D and 2D NMR spectroscopic data including HSQC, DQF COSY, HMBC, and NOESY. All the isolates were tested for inhibitory activities against LPS-induced NO production in RAW264.7 macrophages, abiesanordine I (9) showed the strongest activity with the IC50 value of 17.0 μg/mL. Furthermore, it exhibited no cytotoxicity against RAW264.7 macrophages under the concentration of 50 μg/mL.  相似文献

One new modiolin, microsphaerodiolin (1), and seven new phthalides, microsphaerophthalides A-G (2-8), together with 12 known compounds were isolated from the endophytic fungus Microsphaeropsis arundinis PSU-G18. Their structures were elucidated by spectroscopic methods. The new 3-oxygenated phthalides are rare natural products. The known 1-(2,5-dihydroxyphenyl)-2-buten-1-one exhibited significant antifungal activity against Microsporum gypseum SH-MU-4 with an MIC value of 8 μg/mL, moderate antimalarial activity with an IC50 value of 9.63 μg/mL and strong radical scavenging potency with the IC50 value of 0.018 mg/mL. The new compounds 2 and 6 showed moderately antifungal activity against M. gypseum SH-MU-4 and Cryptococcus neoformans, respectively, with equal MIC values of 64 μg/mL.  相似文献

Two new (1 and 2) and three known (35) carbamidocyclophanes were isolated from a cultured freshwater cyanobacterium Nostoc sp. (UIC 10274) obtained from a sample collected at Des Plaines, Illinois. Their planar structures and stereoconfigurations were determined by extensive spectroscopic analysis including 1D/2D NMR experiments, HRESIMS as well as CD spectroscopy. Carbamidocyclophane F (1) showed potent anti-Mycobacterium tuberculosis activity in the microplate Alamar blue assay and low-oxygen-recovery assay with MIC values of 0.8 and 5.4 μM, respectively. Carbamidocyclophane F (1) also displayed antimicrobial activities against the gram positive bacteria Staphylococcus aureus and Enterococcus faecalis with MIC values of 0.1 and 0.2 μM, respectively. Carbamidocyclophane F (1) and Carbamidocyclophane G (2) both showed antiproliferative activity against MDA-MB-435 and HT-29 human cancer cell lines with IC50 values in the range from 0.5 to 0.7 μM.  相似文献

The yttrium chloride with the bridged bis(amidinate) L (L = Me3SiNC(Ph)N(CH2)3NC(Ph)NSiMe3) LYCl(DME) (2) was synthesized and structurally characterized. Treatment of LLnCl(sol)x (Ln = Yb, sol = THF, x = 2 1; Ln = Y, sol = DME, x = 1 2) with the dilithium salt Li2L(THF)0.5 afforded the novel bimetallic lanthanide complexes supported by three ligands, Ln22-L)3 · DME (Ln = Yb 3, Y 4; DME = dimethylether), instead of the designed complex LLn(μ2-L)LnL via the ligand redistribution reaction. Complexes 3 and 4 were fully characterized including X-ray analysis and 1H NMR spectrum for 4. Reaction of LnCl3 (Ln = Yb, Y) with 2 equiv. of Li2L(THF)0.5 gave the anionic complexes [Li(DME)3][L2Ln] (Ln = Yb 5, Y 6), which were confirmed by a crystal structure determination. The further study indicated that complexes 3 and 4 can also be synthesized by reaction of LnCl3 (Ln = Yb, Y) with 1.5 equiv. of Li2L(THF)0.5 or reaction of 1 and 2 with anionic complexes 5 and 6. Complexes 3, 4, 5 and 6 were found to be high active catalysts for ring-opening polymerization of ε-caprolactone (CL).  相似文献

Kehokorins A (1)-C (3), three novel dibenzofurans, have been isolated from field-collected fruit bodies of the myxomycete, Trichia favoginea var. persimilis, and their structures were elucidated by spectral data. Kehokorin A (1) was a α-l-rhamnopyranoside of kehokorin B (2), while kehokorin C (3) was a 1-demethoxy analog of 2. Kehokorin A (1) was cytotoxic against HeLa cells with an IC50 value of 1.5 μg/mL.  相似文献

The 2-imino-1,10-phenanthroline ligands, 1,10-C12H7N2-2-CRN(2,6-i-Pr2-4-R1-C6H2) [R = R1 = H (L1); R = H, R1 = Br (L2); R = H, R1 = CN (L3); R = H, R1 = i-Pr (L4); R = Me, R1 = H (L5); R = Me, R1 = i-Pr (L6)], have been prepared in high yield from the condensation reaction of 1,10-C12H7N2-2-CRO (R = H, Me) with one equivalent of the corresponding 4-substituted 2,6-diisopropylaniline. The molecular structures of L2, L5 and L6 reveal the imino nitrogen atoms to adopt a transoid configuration with respect to the phenanthrolinyl nitrogen atoms. Treatment of Lx with one equivalent of CoCl2 in n-BuOH at 90 °C gives the high spin complexes, (Lx)CoCl2 [Lx = L1 (1a), L2 (1b), L3 (1c), L4 (1d), L5 (1e), L6 (1f)], in which the metal centres exhibit distorted square pyramidal geometries. Activation of 1a-1f with excess methylaluminoxane (MAO) gives catalysts that are modestly active for the oligomerisation of ethylene affording mainly linear α-olefins along with some degree of internal olefins. While the donor capability of the 4-position of the N-aryl group does not appear to affect the activity of the catalyst, it does have an influence on the ratio of α-olefins to internal olefins. Single crystal X-ray diffraction studies have been performed on L2, L5, L6, 1a, 1c and 1f.  相似文献

A structurally unique polyketide, mellisol (1) and 1,8-dihydroxynaphthol 1-O-α-glucopyranoside (3), were isolated from the fungus Xylaria mellisii (BCC 1005). The relative stereostructure of 1 was determined on the basis of X-ray crystallographic data. Compounds 1 and 3 exhibited activity against herpes simplex virus-type 1 with IC50 values of 10.50 and 8.40 μg/mL, respectively. They also showed cytotoxic activity against vero cells at the concentration of 40-50 μg/mL.  相似文献

Cyclodepsipeptides SCH 217048 (1), SCH 218157 (2), and a new analog, pleosporin A (3), were isolated from cultures of an unidentified elephant dung fungus of the family Pleosporaceae. The structure of 3 was elucidated on the basis of detailed spectroscopic interpretation. The absolute configurations of 13 were determined by chiral column HPLC analysis and Marfey’s method. Cyclodepsipeptides 13 exhibited antimalarial activity against Plasmodium falciparum K1 with respective IC50 values of 1.6, 6.4, and 1.6 μg/mL, while they did not show cytotoxicity against KB, MCF-7 and NCI-H187 cell-lines or non-cancerous Vero cells at 50 μg/mL.  相似文献

Three new polysulfur alkaloids, lissoclibadins 1 (1)-3 (3), were isolated from the ascidian Lissoclinum sp. (cf. L. badium Monniot, F. and Monniot, C., 1996). The structures of 1-3 were assigned on the basis of their spectral data, and the computational modeling study was utilized for 1. Compound 1 had a trimeric structure of three identical aromatic anime moieties connected through two sulfide and a disulfide bonds. Compounds 2 and 3 were dimeric structures of the same unit as that of 1 connected through a sulfide and disulfide bonds (2) and two sulfide bonds (3). Compounds 1 and 2 inhibited the growth of the marine bacterium Ruegeria atlantica (15.2 mm at 20 μg/disk and 12.2 mm at 5 μg/disk, respectively) and 2 showed antifungal activity to Mucor hiemalis (13.8 mm at 50 μg/disk). Compounds 1-3 were cytotoxic against HL-60 (IC50=0.37, 0.21, and 5.5 μM, respectively).  相似文献

Two unusual cembranoids, sarcofuranocembrenolides A (1) and B (2), were isolated from a soft coral Sarcophyton sp. together with five known cembranoids (3-7). Compound 1 had a unique carbon skeleton of 8,19-bisnorfuranocembrenolide. Compound 2 was a furanocembrenolide, but a C1 unit (C-20) was attached to C-10 instead of C-12 of the ordinary cembrenolides. These rearrangements are unique in the biosynthesis of cembranoid diterpenes. Lobohedleolide (5), (7Z)-lobohedleolide (6), and denticulatolide (7) inhibited the colony formation of V79 cells at ED50 values of 4.6 (abt. 1.52), 3.7 (1.22), and 3.6 (1.40) μM (μg/mL), respectively, and reduced TNF-α production from LPS-stimulated RAW264.7 cells at 3.0-10.0 μM.  相似文献

From an antimicrobial gram-positive actinomycete strain of Saccharomonospora azurea (MTCC11714) isolated from high altitude soil of Kargil (J&K, India), a new isocoumarin saccharonol B (2) along with two known compounds viz. saccharonol A (1) and piericidin A3 (3) was isolated and characterized. The structure of the new compound was established based on extensive 2D-NMR data. Saccharonol B (2) exhibited mild antimicrobial activity against a standard panel of microorganisms Staphylococcus aureus ATCC 29213, Candida albicans ATCC 90028, and Aspergillus fumigatus MTCC 1811 with MIC values in the range of 128–248 μg/mL. Saccharonol B (2) and piericidin A3 (3) showed selective cytotoxic activity against human pancreatic carcinoma cell line (MIAPaCa-2) with IC50 values of 9 and 8 μM, respectively. Mechanistic studies indicated that saccharonol B (2) arrests S-phase of the cell cycle and causes dose-dependent loss of mitochondrial potential in MIAPaCa-2 cells.  相似文献

Chromatographic fractionation of methanol extract from roots of the Piper chaba Hunter resulted in the isolation of four new dimeric alkaloids, chabamide H (1), I (2), J (3), K (4) together with 11 known compounds (5-15). Their chemical structures and relative stereochemistry were determined on the basis of the comprehensive spectroscopic techniques (IR, Mass, and NMR) and further confirmed by comparison of the data with those reported in literature. In addition, cytotoxic activities of all the dimeric amides (1-7) along with their monomers (8-10) were evaluated against cervical (HELA), breast (MCF-7), liver (HEPG2), colon (HT-29), and colon (COLO-205) cancer cell lines. Among the tested isolates, 5 and 7 exhibited potent cytotoxic activity against COLO-205 cell line with IC50 value of 3.10 μg/mL and 0.018 μg/mL, respectively. To prove biogenesis of the newly isolated compounds, biomimetic synthesis has also been carried out via Diels-Alder reaction by using copper(II) salts in aqueous medium.  相似文献

One monocyclofarnesol-derived sesquiterpenoid (1), four new meroterpenoids (25) as well as two isolation artifacts (67) and a known sesquiterpenoid (8) were isolated from the marine sponge Ircinia sp. Their chemical structures were elucidated via analyses of spectroscopic data, which included NMR and HRMS data. Two terpenoids, 1 and 4, showed PPARδ agonistic activities, with EC50 values of 18 and 30 μg/mL, respectively.  相似文献

Six new compounds, an N-hydroxypyridone glucoside, orbiocrellin A (1), its aglycone orbiocrellin B (2), chromone glucosides 3 and 4, a dihydrochromone 5a/5b, and a chromone 6, were isolated from the scale-insect pathogenic fungus Orbiocrella sp. BCC 33248. Orbiocrellin A (1) exhibited antimalarial activity against Plasmodium falciparum K1 (IC50 3.1 μg/mL) while it was non-cytotoxic. In contrast, orbiocrellin B (2) showed both antimalarial (IC50 2.1 μg/mL) and cytotoxic (NCI-H187 cells, IC50 0.70 μg/mL) activities.  相似文献

A series of organotin (IV) complexes with 6-amino-1,3,5-triazine-2,4-dithiol of the type [(RnSnCl4−n)2 (C3H2N4S2)] (n = 3: R = Me 1, n-Bu 2, PhCH23, Ph 4; n = 2: R = Me 5, n-Bu 6, PhCH27, Ph 8) have been synthesized. All the complexes 1-8 have been characterized by elemental analysis, IR, 1H and 13C NMR spectra. Among them complexes 1, 4, 5 and 8 have also been characterized by X-ray crystallography diffraction analyses, which revealed that the tin atoms of complexes 1, 4, 5 and 8 are all five-coordinated with distorted trigonal bipyramid geometries.  相似文献

A straightforward synthesis of (1S,2R,3R,4R)-4-(6-aminopurin-9-yl)-2-hydroxymethylcyclopentane-1,3-diol (2), an isomer of aristeromycin, and its 2′-deoxy derivative 3 from readily available disubstituted cyclopentenes is presented. An antiviral analysis of 2 showed it to have significant activity versus Epstein-Barr virus (IC50 0.62 μg/mL in the Elisa assay) and to be free of cytotoxicity effects against the host cells. In a much less comprehensive antiviral analysis, 3 also was active towards Epstein-Barr (IC50 7.58 μg/mL in the Elisa assay) but this was accompanied by cellular toxicity.  相似文献

Five-, six-, and seven-coordinate volatile butyltin(IV) heterobimetallic derivatives, respectively of the types, [BuSn{(μ-OPri)2Al(OPri)2}Cl2] (1), [BuSn{(μ-OPri)2Al(OPri)2}2Cl] (2), and BuSn{(μ-OPri)2M(OPri)x − 2}3 (3:M = Al (x = 4); 4:M = Ga (x = 4); 5:M = Nb (x = 6)) have been synthesized by the reactions of BuSnCl3 with potassium tetraisopropoxoaluminate in 1:1, 1:2, and 1:3 molar ratios. Replacement reactions of chloride in (1) and (2) with appropriate alkoxometallate (tetraisopropoxoaluminate, tetraisopropoxogallate, or hexaisopropoxoniobate) ligands result in the formation of novel BuSn(IV) heterotri- and tetra-metallic derivatives. All of these derivatives have been characterized by elemental analyses, molecular weight measurements, and spectroscopic (IR, 1H, 27Al, and 119Sn NMR) studies. Based on these studies, plausible structures for the new derivatives involving bidentate ligation of the alkoxometallate ligands have been suggested.  相似文献
