Methods for the biological monitoring of aromatic hydrocarbons and their metabolites in the human blood and urine are reviewed. For the determination of the unchanged aromatic hydrocarbon in blood, gas chromatographic head-space analysis is recommended. The metabolites can be monitored by photometric, thin-layer chromatographic, high-performance liquid chromatographic and gas chromatographic methods. For the assessment of health risks caused by aromatic hydrocarbons, reference values and occupational limit values, expressed as biological tolerance values and biological exposure indices, have to be considered.  相似文献

Chlorotestosterone and its metabolites were determined in urine samples from bovine animals treated with chlorotestosterone acetate by oral and intramuscular routes. Sample preparation, involving enzymatic deconjugation and solid-phase extraction, was optimised. The effect of different enzyme preparations, pH, and time of incubation were studied. An extraction/clean-up procedure based on solid-phase extraction (C18 cartridge) and liquid-liquid clean-up was developed. Determination of chlorotestosterone and its metabolites was by enzyme immunoassay and GC-MS. Metabolites were converted into their TMS-enol-TMS-ether and TMS-oxime-TMS-ether forms before GC-MS (EI) analysis.  相似文献

A gas chromatographic-mass spectrometric analysis has been developed for the determination of debrisoquine and its metabolites in the urine of healthy individuals (controls) and patients with chronic renal failure. The sensitive and specific assay comprises selected-ion monitoring of the drug and the metabolites 4-hydroxydebrisoquine and 8-hydroxydebrisoquine using guanoxan as the internal standard. The limit of detection is ca. 0.2 microgram/ml. The clinical study shows that the healthy individuals and patients with chronic renal failure can be divided in two groups of extensive metabolizers and poor metabolizers, respectively. The extensive metabolizers excreted large amounts of 4-hydroxydebrisoquine and minor amounts of 8-hydroxydebrisoquine. The poor metabolizers excreted small amounts of 4-hydroxy metabolite, and no 8-hydroxydebrisoquine was detected in the urine.  相似文献

Andrographolide is one of principal components of a famous traditional Chinese herbal medicine Andrographis paniculate (BURM) NEES. Four new metabolites of andrographolide were isolated from human urine. All of them were characterized as sulfate and one of them also as a cysteine S-conjugate. The structures were determined to be andrographolide-3-O-sulfate (M-1), isoandrographolide-3-O-sulfate (M-2), 14-deoxyandrographolide-3-O-sulfate (M-3), 14-deoxy-12-(cysteine-S-yl)-andrographolide-3-O-sulfate (M-4), respectively, based on chemical evidence and spectroscopic analyses.  相似文献

A method for the determination of ethotoin and its p-hydroxylated and dealkylated metabolites in urine has been developed. Ethotoin and the metabolites were extracted from acidified urine with ethyl acetate and silylated before injection into a combined gas chromatograph--mass spectrometer. Four partly identified metabolites were recorded, but their exact quantitation was not possible as pure reference substances were not available. The limit of sensitivity was far below the amounts of ethotoin and of its metabolites found in urine from patients treated with therapeutic doses of ethotoin.  相似文献

A high-performance liquid chromatographic method for the determination of low levels (less than 1 microgram/ml) of the basic metabolite of bezitramide, 1-(4-piperidinyl)-1,3-dihydro-2H-benzimidazol-2-one, in human urine is described. A method for the determination of malotilate (I), the corresponding monocarboxylic acid (II) and its decarboxylated product (III) in plasma is described. Plasma was extracted with chloroform spiked with internal standard. The residue, dissolved in methanol, was chromatographed on a reversed-phase column with a mobile phase of 60% acetonitrile and 1% acetic acid in water. The sensitivity limit for I, II and III was 50, 25 and 100 ng/ml of plasma, respectively. Compound I in the same plasma extract was also analysed by gas chromatography--electron-impact mass spectrometry. The base peaks m/z 160 for I and m/z 162 for internal standard (IV) were monitored; the sensitivity limit for I was 2.5 ng/ml of plasma. The determination of the metabolites of I, II and its conjugate (V), and isopropyl-hydrogen malonate (VI) in urine by high-performance liquid chromatography is also described. The limit of quantification for VI was 2.0 micrograms/ml, and the overall coefficient of variation of VI was 4.7%. The limit of quantification for II in urine was 0.5 micrograms/ml and that for V was 1.0 micrograms/ml as total II (II + V). The overall precision of the method was satisfactory. The method was used to determine plasma and urine concentrations in four dogs orally dosed with 100, 200 or 400 mg of malotilate.  相似文献

A method for the extraction and quantitation of histamine and its metabolites from relatively small volumes of urine is described. It employs primarily ion-exchange and thin-layer chromatography and allows for quantitation in the mug range. Studies of atopic man and dog that employed this procedure yielded values comparable to those reported with gas chromatographic methods.  相似文献

A simple and sensitive high-performance liquid chromatographic method with ultraviolet detection is described for the simultaneous determination of lansoprazole and its metabolites in human serum and urine. The analytes in serum or urine were extracted with diethyl ether-dichloromethane (7:3, v/v) followed by evaporation, dissolution and injection into a reversed-phase column. The recoveries of authentic analytes added to serum at 0.05-2 micrograms/ml or to urine at 1-20 micrograms/ml were greater than 88%, with the coefficients of variation less than 7.1%. The minimum determinable concentrations of all analytes were 5 ng/ml in serum and 50 ng/ml in urine. The method was successfully applied to a pharmacokinetic study of lansoprazole in human.  相似文献

A method is described for the determination of aldicarb and its metabolites (the sulphoxide and sulphone) in urine by gas chromatography with flame photometric detection (GC-FPD). The sample was concentrated with a column containing activated charcoal and Florisil, and then eluted with dichloromethane-acetone (1:1, v/v). The aldicarb and aldicarb sulphoxide in the eluate solution were oxidized to aldicarb sulphone and the total sulphone concentration was determined by GC-FPD after extraction with dichloromethane and clean-up with an activated charcoal column. The detection limit was 0.0024 mg/l. The mean recoveries from spiked urine in the range 0.04-0.12 mg/l were 90.9%, 86.6%, 92.6% for aldicarb, aldicarb sulphoxide and aldicarb sulphone, respectively.  相似文献

A sensitive, selective and precise high-performance liquid chromatographic method for simultaneous determination of tauromustine and its demethylated metabolites in plasma and urine has been developed. It is based on solid-phase extraction on C18 sorbent and separation on a semipolar column. The analytical procedure is described in detail. The method has been validated with respect to linearity, recovery, selectivity, precision and detection limit. We have developed an analytical method by using liquid chromatography-mass spectrometry (LC-MS) to determine ketamine and its metabolites in urine. The ionization efficiency between two ionization modes (ESI and APCI) of LC-MS was compared to each other. An easy and simple sample preparation of urine samples was made by passing samples through a 0.22 μm PVDF syringe filter. The results indicated that the ionization efficiency of positive APCI mode is better than positive ESI mode for determination of trace ketamines. A wide linearity range of the research is from 5 to 250 ng mL−1 and the detection limits for ketamine, norketamine and dehydronorketamine were 0.95, 0.48 and 0.33 ng mL−1, respectively. The proposed method was tested by analyzing ketamine and metabolites in the urines of volunteers. The concentrations of ketamine, norketamine and dehydronorketamine are ranged of 5.4-131.0, 12.5-74.1 and 22.8-278.9 ng mL−1, respectively and the ketamines concentration profiles in human urine were also determined. The results demonstrate the suitability of liquid chromatography-mass spectrometry approach to analyze trace amount of ketamine and its metabolites in urine.  相似文献

Using gas chromatography/electron impact-mass spectrometry (GC/EI-MS) and high performance liquid chromatography with atmospheric pressure chemical ionization tandem mass spectrometry (HPLC/APCI-MS/MS), the structures of cyclofenil metabolites in human urine have been assigned. The hydroxyl metabolites liberated from the glucuronide conjugates after acid hydrolysis were characterized as the trimethylsilyl (O-TMS) derivatives using GC/MS. The conjugate glucuronide forms were detected without hydrolysis by HPLC/MS. Cyclofenil was not observed in urine. Tentative structures for the two metabolites are proposed.  相似文献
