Two fluoroalcohols--1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and 1,1,1,3,3,3-hexafluoro-2-methyl-2-propanol (HFTB)--were evaluated for the first time as volatile buffer acids in the basic mobile phase for reversed-phase chromatography with electrospray ionization-mass spectrometric (LC-ESI-MS) detection of five antibiotics. Chromatographic separation as well as positive and negative ion ESI-MS intensities using these novel buffer components were compared to traditional buffer systems. Overall, the highest signal intensities and best chromatographic separation for the five antibiotics (ciprofloxacin, norfloxacin, ofloxacin, sulfadimethoxine and sulfamethoxazole) were achieved using 5 mM HFIP as the buffer acid to methanol : water mobile phase (pH of the aqueous component adjusted to 9.0 with ammonium hydroxide). Comparable results were achieved using 5 mM HFTB (pH adjusted to 9.0 with ammonium hydroxide). The suitability of HFIP for analysis of antibiotic residues in lettuce is demonstrated.  相似文献

The retention behavior of a large group of analytes (35) with varied properties (pKa and logP) was studied on eight hydrophilic interaction LC columns with different surfaces, stationary phase chemistries, and types of particles. The acetonitrile content (5–95%), buffer concentration (0.5–200 mM), and pH of the mobile phase (3.8 and 6.8) were evaluated for their effects on the retention behavior. The type of stationary phase had a significant impact on the selectivity and retention time of the tested analytes. Completely different selectivity was observed on the aminopropyl stationary phase. In this study, the influence of the buffer concentration was similar for all tested columns, except for the aminopropyl stationary phase. Increasing the buffer concentration led to decreased retention times for the basic compounds and increased retention times for the acidic compounds, while the inverse behavior was observed on the aminopropyl stationary phase. The selectivity of the individual stationary phases was evaluated at pH 3.8 and 6.8. Much lower selectivity differences between the stationary phases were observed at pH 6.8 than pH 3.8. Bare silica stationary phases were used in the comparison of the particles (fused‐core and fully porous particles of 3 and 1.7 μm) and the columns provided by different manufacturers.  相似文献

In this study, the retention behavior and selectivity differences of water‐soluble vitamins were evaluated with three types of polar stationary phases (i.e. an underivatized silica phase, an amide phase, and an amino phase) operated in the hydrophilic interaction chromatographic mode with ESI mass spectrometric detection. The effects of mobile phase composition, including buffer pH and concentration, on the retention and selectivity of the vitamins were investigated. In all stationary phases, the neutral or weakly charged vitamins exhibited very weak retention under each of the pH conditions, while the acidic and more basic vitamins showed diverse retention behaviors. With the underivatized silica phase, increasing the salt concentration of the mobile phase resulted in enhanced retention of the acidic vitamins, but decreased retention of the basic vitamins. These observations thus signify the involvement of secondary mechanisms, such as electrostatic interaction in the retention of these analytes. Under optimized conditions, a baseline separation of all vitamins was achieved with excellent peak efficiency. In addition, the effects of water content in the sample on retention and peak efficiency were examined, with sample stacking effects observed when the injected sample contained a high amount of water.  相似文献

The impact of the proportion of CO2 concentration in methanol-water-CO2 mobile phases on the separation of several substituted benzoic acids was explored by studying the variation of retention with mobile phase pH in these mixtures. As the amount of CO2 in methanol-aqueous buffer-CO2 mixtures increased, a more basic buffer was needed to control the dissociation of these acids. Differences in terms of retention, separation efficiency and peak asymmetry were shown for substituted benzoic acids with methanol-water-CO2 and methanol-aqueous buffer-CO2 mixtures. Variations of these chromatographic parameters with mobile phase pH were related to the dissociation of these acids and their interaction with methanol-aqueous buffer-CO2 mobile phases and the stationary phase. The addition of a buffer into methanol-aqueous solution-CO2 was an effective means to optimize separations of acidic analytes with high fluidity liquid mobile phases. The substituted benzoic acids had baseline separation in the least amount of time using the high fluidity liquid mobile phases.  相似文献

Anionic species with ion pair forming ability are commonly used to enhance the retention and efficiency of basic analytes in RPLC separations. However, little is known about the interactions between organic mobile phase modifiers and such ion pairing anions. In this work, we measured the magnitude of the retention increase of basic drugs and peptides upon addition of strong inorganic ion pairing anions (e.g. perchlorate) as a function of the volume fraction of modifier in acidic water-acetonitrile mobile phases on two different stationary phases. We found that the increase in retention upon addition of various salts depended strongly on the eluent strength. In general, larger retention increases upon addition of the anion were observed at higher organic fractions. Regression of retention against the volume fraction of organic modifier indicated that the ion pair forming anions substantially decreased S values while only slightly changing ln k'w values. The decrease in S is the major cause of the retention increase of basic drugs and peptides when such anions are added to the mobile phase.  相似文献

A polystyrene-supported phosphine oxide-catalysed Beckmann rearrangement of ketoximes in 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) has been developed. We have investigated the effect of several common buffers (10-mM formic acid, 10-mM ammonium acetate, and 100-mM ammonium acetate) on the ionization of a series of model compounds that are amenable to negative atmospheric pressure chemical ionization to determine the extent of ionization quenching that can occur. In addition, we have compared the sensitivity of these standard mobile phases to a mobile phase that does not contain an acidic buffer component, but rather a base (N-methylmorpholine). The results showed that, as expected, the sensitivity for the test analytes was greatest in the mobile phase that lacked acidic components. In general, ionization of analytes that contained a single, more weakly acidic functional group was inhibited to a greater degree by more strongly acidic buffer components. In some cases, ionization was quenched completely by acidic buffer components, Ionization of compounds that were more strongly acidic was quite good in all mobile phases tested. Differences in the ionization efficiencies of the analytes in each mobile phase were correlated with the gas-phase reagent ions present. As a point of reference, each of the analytes also was analyzed in the positive ion mode and the signal intensities were compared to those obtained in the negative ion mode. In addition, the utility of mobile phases that contained N-methylmorpholine for chromatographic separations was demonstrated.  相似文献

Seven new stationary phases with different number of proline units and/or different linkage to silica gel were prepared and evaluated in order to improve the performance of proline chiral stationary phases. The average separation factor achieved with the 53 analytes increases with the number of proline units in the stationary phases. When the proline peptides are directly attached to the 3-methylaminopropyl silica gel without using the 6-methylaminohexanoic acid linker, the stationary phases perform better overall. For decaproline chiral stationary phase 8, the separation also depends on the mobile phase system used. For this stationary phase, the CH2Cl2/hexanes/2-propanol system significantly outperforms the 2-propanol/hexanes system. For the 53 analytes tested, the separation factors achieved with this stationary phase compare well with those for three commercial columns.  相似文献

Among numerous solvents available for chemical transformations, 1,1,1,3,3,3-hexafluoro-2-propanol (popularly known as HFIP) has attracted enough attention of the scientific community in recent years. Several unique features of HFIP compared to its non-fluoro analogue isopropanol have helped this solvent to make a difference in various subdomains of organic chemistry. One such area is transition metal-catalyzed C–H bond functionalization reactions. While, on one side, HFIP is emerging as a green and sustainable deep eutectic solvent (DES), on the other side, a major proportion of Pd-catalyzed C–H functionalization is heavily relying on this solvent. In particular, for distal aromatic C–H functionalizations, the exceptional impact of HFIP to elevate the yield and selectivity has made this solvent irreplaceable. Recent research studies have also highlighted the H-bond-donating ability of HFIP to enhance the chiral induction in Pd-catalyzed atroposelective C–H activation. This perspective aims to portray different shades of HFIP as a magical solvent in Pd-catalyzed C–H functionalization reactions.

A new generation of extremely acid stable "hyper-crosslinked" (HC) phases have been developed with good plate counts for basic drug separations. In our previous work, we successfully developed an approach for synthesizing HC stationary phases on silica substrates using aluminum trichloride catalyzed Friedel-Crafts (F-C) chemistry to improve the stability of silica-based RPLC stationary phases at low pH. However, the performance of basic analytes on these HC phases under acidic conditions was unusually poor compared to that of conventional silica-based C18 phases. The effects of the specific F-C catalysts used and the specific silica substrate on the chromatographic properties of HC phases have been studied. Modified synthetic strategies that give both good observed plate counts for basic analytes under acidic conditions and very good low pH stability without compromising other chromatographic properties of the hyper-crosslinked phases have been developed. Replacement of aluminum trichloride with tin tetrachloride as the catalyst for the F-C chemistry and use of a very high purity silica result in significantly improved plate counts for basic analytes. In formic acid buffered mobile phases, which are highly compatible with electrospray ionization LC-MS, basic analytes showed much better performance on the tin tetrachloride catalyzed HC phases than on any conventional commercial phase tested. The tin tetrachloride catalyzed HC phase is as stable as the original aluminum trichloride catalyzed HC phases, and much more stable than the bench mark acid stable commercial phase.  相似文献

A new single‐urea‐bound chiral stationary phase based on 3,5‐dimethylphenylcarbamoylated β‐cyclodextrin was prepared through the Staudinger reaction of mono (6A‐azido‐6A‐deoxy)‐per(3,5‐dimethylphenylcarbamoylated) β‐cyclodextrin and 3‐aminopropyl silica gel under CO2 atmosphere. The new phase exhibited good enantioseparation performance for 33 analytes using normal‐phase HPLC conditions; 19 of them were baseline separated. Effects of structure of analytes, alcoholic modifiers, and acidic/basic additives on separation performances of this new cyclodextrin chiral stationary phase have been studied in detail. The results showed that the retention and resolution of acidic and basic analytes on the CSP were greatly affected by the additives. Peak symmetry for some analytes could be improved by simultaneously adding acidic and basic additives to the mobile phase. This work expands the potential applications of the cyclodextrin‐based chiral stationary phases in the normal‐phase HPLC.  相似文献

A hydrophilic chiral capillary monolithic column for enantiomer separation in CEC was prepared by coating cellulose tris(3,5-dimethylphenyl-carbamate) (CDMPC) on porous hydrophilic poly(acrylamide-co-N,N'-methylene-bisacrylamide) (poly(AA-co-MBA)) monolithic matrix with confine of a fused-silica capillary. The coating conditions were optimized to obtain a stable and reproducible chiral stationary phase for CEC. The effect of organic modifier of ACN in aqueous mobile phase for the enantiomer separation by CEC was investigated, and the significant influence of ACN on the enantioresolution and electrochromatographic retention was observed. Twelve pairs of enantiomers including acidic, neutral, and basic analytes were tested and nine pairs of them were baseline-enantioresolved with acidic and basic aqueous mobile phases. A good within-column repeatability in retention time (RSD = 2.4%) and resolution (RSD = 3.2%) was obtained by consecutive injections of a neutral compound, benzoin, on a prepared chiral monolithic column, while the between-column repeatability in retention time (RSD = 6.4%) and resolution (RSD = 9.6%) was observed by column-to-column examination. The prepared monolithic stationary phase showed good stability in either acidic or basic mobile phase.  相似文献

Complex analyses of polar compounds, especially basic ones, require more selective stationary phases. The present paper describes a stationary phase prepared by thermal immobilization of poly(methyltetradecylsiloxane) onto chromatographic silica (PMTDS-SiO(2)). This stationary phase presents hydrophobic and ion-exchange interactions that confer both high retention and unique selectivities for basic solutes. The influence of ion-exchange interactions is confirmed by the increase in retention factors of basic solutes when the mobile-phase pH changes from acidic to neutral and by the decrease in retention factors when the mobile-phase pH changes from neutral to alkaline. The ion-exchange properties of the stationary phase are enriched in neutral mobile phase (pH 7-7.5) using soft Lewis bases such as tricine and tris as buffers but are suppressed in both acidic (pH 2.5-6) and highly alkaline mobile phases (pH≤10). Increasing both temperature and flow rate permits more rapid separations while maintaining the selectivity. The stability of the stationary phase is evaluated with acid, neutral and alkaline mobile phases.  相似文献

A novel stationary phase prepared by the thermal immobilization of poly(dimethylsiloxane) onto the surface of silica (PDMS–SiO2) has been described, evaluated and compared with 229 commercially available RP-LC stationary phases using the Tanaka column classification protocol. The phase exhibited many unique chromatographic properties and, based on the phases in the database, was most similar to the fluoroalkylated phases (aside from the obvious lack of fluoro selectivity imposed by the C–F dipole). The phase exhibited classic reversed-phase behaviour in acid mobile phase conditions and mixed-mode reversed-phase/cation-exchange retention behaviour in neutral mobile phase conditions. The phase exhibited acceptable stability at both low and intermediate pH, conditions which should impart optimum chromatographic selectivity to the phase. Retention of basic analytes was shown to occur by a “three site model” as proposed by Neue. This new PDMS–SiO2 stationary phase is extremely interesting in that the dominancy of its hydrophobic and ion-exchange interactions can be controlled by the influence of mobile phase pH, buffer type and concentration. The PDMS–SiO2 stationary phase may provide a complementary tool to reversed-phase and HILIC stationary phases. The present results highlight the fact that the type of buffer, its concentration and pH can not only affect peak shape but also retention, selectivity and hence chromatographic resolution. Therefore, in method development and optimization strategies it is suggested that more emphasis should be given to the evaluation of these mobile phase operating parameters especially when basic solutes are involved.  相似文献

To develop photo-tunable chromatography, several azobenzene derivatives were immobilized or modified on silica gels or incorporated into polymers. The supports prepared were used as stationary phases in semi-micro HPLC, and the photo-dependence of their retention evaluated. On these stationary phases, the retention measured with 2-propanol in hexane was increased by UV irradiation; in contrast, the retention was decreased when methanol was used as mobile phase. It was assumed that the polarity of the stationary phase increases when the molecular structure of the azobenzene moieties changes from the trans to the cis form. On irradiation of the column with visible light after UV irradiation and with methanol as mobile phase, the change of retention proved to be reversible. However, this reverse change was less dramatic than the initial change. Such retention behavior is attributable to photo-isomerization of the azobenzene moieties. These findings will be exploited in our development of new photo-tunable stationary phases and new photo-controlled separation systems.  相似文献

Variations of a thermal immobilization procedure using poly(methyltetradecilsiloxane) and silica produced fourteen stationary phases with carbon contents of 4-18%. The stationary phases were chromatographically evaluated with the Engelhardt, SRM 870 and Tanaka tests. Classifications using USP and Euerby procedures indicate that the new immobilized phases are different from most commercial phases although there was some similarity with phases that have high ion-exchange interactions. The retention mechanism involved in the separation of basic solutes on several of the new stationary phases was studied by varying pH, type of Lewis base and the ionic strength of the eluent. The separations are strongly influenced by the chemistry of the accessible free silanols. The stationary phases present good selectivity at intermediate pH where the basic analytes were protonated, suggesting use of intermediate pH for these separations. Stability tests show that the stationary phases have poor stability at very high pH, even at 23°C, but good stability in acidic mobile phases, even at 75°C, as expected for an immobilized polymer stationary phase.  相似文献
