Silver sol surface-enhanced Raman spectroscopy (SERS) was considered as a technique in the quantitative analysis of low-concentration thymine. Because of the poor stability and reproducibility of SERS signal, a polymer of polyacrylic acid sodium was selected as a stable medium to add into silver sol in order to obtain a surface-enhanced Raman spectroscopy signal. Assignments of Raman shift for solid thymine, SERS of thymine, and SERS of thymine containing stable medium were given. The comparison of Raman peaks between them showed that the addition of stable medium had a little influence on the SERS of thymine and is suitable for the quantitative analysis of low-concentration thymine.  相似文献

We have demonstrated surface-enhanced Raman spectroscopy (SERS) on arrays of Au nanorods aligned in line by a dynamic oblique deposition technique. For light polarized along the major axes of the nanorods, the plasma resonance of the Au nanorods has been tuned to a wavelength suitable for Raman spectroscopy. Raman scattering on the discrete nanorods is significantly enhanced compared with that on semicontinuous Au films. Since the preparation process is physically bottom-up, it is robust in its selection of the materials and is useful for providing SERS sensors at low cost.  相似文献

Resonance Raman (RR) spectroscopy has several advantages over the normal Raman spectroscopy (RS) widely used for in situ characterization of solid catalysts and catalytic reactions. Compared with RS, RR can provide much higher sensitivity and selectivity in detecting catalytically-significant surface metal oxides. RR can potentially give useful information on the nature of excited states relevant to photocatalysis and on the anharmonic potential of the ground state. In this critical review a detailed discussion is presented on several types of RR experimental systems, three distinct sources of so-called Raman (fluorescence) background, detection limits for RR compared to other techniques (EXAFS, PM-IRAS, SFG), and three well-known methods to assign UV-vis absorption bands and a band-specific unified method that is derived mainly from RR results. In addition, the virtues and challenges of surface-enhanced Raman spectroscopy (SERS) are discussed for detecting molecular adsorbates at catalytically relevant interfaces. Tip-enhanced Raman spectroscopy (TERS), which is a combination of SERS and near-field scanning probe microscopy and has the capability of probing molecular adsorbates at specific catalytic sites with an enormous surface sensitivity and nanometre spatial resolution, is also reviewed (300 references).  相似文献

This tutorial review discusses a new class of colloidal metal nanoparticles that is able to enhance the efficiencies of surface-enhanced Raman scattering (SERS) by as much as 10(14)-10(15) fold. This enormous enhancement allows spectroscopic detection and identification of single molecules located on the nanoparticle surface or at the junction of two particles under ambient conditions. Considerable progress has been made in understanding the enhancement mechanisms, including definitive evidence for the single-molecule origin of fluctuating SERS signals. For applications, SERS nanoparticle tags have been developed based on the use of embedded reporter molecules and a silica or polymer encapsulation layer. The SERS nanoparticle tags are capable of providing detailed spectroscopic information and are much brighter than semiconductor quantum dots in the near-infrared spectral window. These properties have raised new opportunities for multiplexed molecular diagnosis and in vivo Raman spectroscopy and imaging.  相似文献

Sensitive and detailed molecular structural information plays an increasing role in molecular biophysics and molecular medicine. Therefore, vibrational spectroscopic techniques, such as Raman scattering, which provide high structural information content are of growing interest in biophysical and biomedical research. Raman spectroscopy can be revolutionized when the inelastic scattering process takes place in the very close vicinity of metal nanostructures. Under these conditions, strongly increased Raman signals can be obtained due to resonances between optical fields and the collective oscillations of the free electrons in the metal. This effect of surface-enhanced Raman scattering (SERS) allows us to push vibrational spectroscopy to new limits in detection sensitivity, lateral resolution, and molecular structural selectivity. This opens up exciting perspectives also in molecular biospectroscopy. This article highlights three directions where SERS can offer interesting new capabilities. This includes SERS as a technique for detecting and tracking a single molecule, a SERS-based nanosensor for probing the chemical composition and the pH value in a live cell, and the effect of so-called surface-enhanced Raman optical activity, which provides information on the chiral organization of molecules on surfaces.  相似文献

罗丹明6G(Rhodamine 6G,R6G)是单分子表面增强拉曼光谱(SM-SERS)研究中最常用的探针分子之一,对R6G分子在表面吸附行为的研究有助于了解R6G分子和表面的相互作用. 本文应用电化学和电化学表面增强拉曼光谱技术,研究不同电位下R6G的银电极表面的吸附行为. 结果表明,随着电位负移罗丹明6G在银表面上从垂直吸附转为倾斜吸附,该变化和碱性条件下吸附于金纳米粒子上R6G的吸附构象一致. 这说明,在部分单分子实验中所发现的R6G反常光谱其来源是单个R6G分子在表面吸附取向变化. 本研究对后续详细分析SM-SERS研究中单分子SERS谱峰变化的机制有一定的参考价值.  相似文献

The electrochemical interaction of thiourea with a copper electrode in sulphuric acid solution was investigated using Fourier transform Raman and in situ surface-enhanced Raman scattering (SERS) spectroscopy. SERS spectra of thiourea at a copper electrode were obtained in solutions containing greater than 5 ppm thiourea; the spectra obtained were consistent with adsorption of the molecule on the copper electrode via the sulphur atom. The SERS spectra provide evidence of complex formation involving thiourea and sulphate species at the electrode surface.  相似文献

A magnetic capture-based, surface-enhanced Raman scattering (SERS) assay for DNA detection has been developed which utilizes Au-coated paramagnetic nanoparticles (Au@PMPs) as both a SERS substrate and effective bioseparation reagent for the selective removal of target DNAs from solution. Hybridization reactions contained two target DNAs, sequence complementary reporter probes conjugated with spectrally distinct Raman dyes distinct for each target, and Au@PMPs conjugated with sequence complementary capture probes. In this case, target DNAs were derived from the RNA genomes of the Rift Valley Fever virus (RVFV) or West Nile virus (WNV). The hybridization reactions were incubated for a short period and then concentrated within the focus beam of an interrogating laser by magnetic pull-down. The attendant SERS response of each individually captured DNA provided a limit of detection sensitivity in the range 20-100 nM. X-ray diffraction and UV-vis analysis validated both the desired surface plasmon resonance properties and bimetallic composition of synthesized Au@PMPs, and UV-vis spectroscopy confirmed conjugation of the Raman dye compounds malachite green (MG) and erythrosin B (EB) with the RVFV and WNV reporter probes, respectively. Finally, hybridization reactions assembled for multiplexed detection of both targets yielded mixed MG/EB spectra and clearly differentiated peaks which facilitate the quantitative detection of each DNA target. On the basis of the simple design of a single-particle DNA detection assay, the opportunity is provided to develop magnetic capture-based SERS assays that are easily assembled and adapted for high-level multiplex detection using low-cost Raman instrumentation.  相似文献

A novel active surface-enhanced Raman scattering (SERS) platform for dynamic on-demand generation of SERS active sites based on optoelectrofluidics is presented in this paper. When a laser source is projected into a sample solution containing metal nanoparticles in an optoelectrofluidic device and an alternating current (ac) electric field is applied, the metal nanoparticles are spontaneously concentrated and assembled within the laser spot, form SERS-active sites, and enhance the Raman signal significantly, allowing dynamic and more sensitive SERS detection. In this simple platform, in which a glass slide-like optoelectrofluidic device is integrated into a conventional SERS detection system, both dynamic concentration of metal nanoparticles and in situ detection of SERS signal are simultaneously possible with only a single laser source. This optoelectrofluidic SERS spectroscopy allows on-demand generation of 'hot spots' at specific regions of interest, and highly sensitive, reliable, and stable SERS measurements of the target molecules in a tiny volume (~500 nL) of liquid sample without any fluidic components and complicated systems.  相似文献

After over 30 years of development, surface-enhanced Raman spectroscopy (SERS) is now facing a very important stage in its history. The explosive development of nanoscience and nanotechnology has assisted the rapid development of SERS, especially during the last 5 years. Further development of surface-enhanced Raman spectroscopy is mainly limited by the reproducible preparation of clean and highly surface enhanced Raman scattering (SERS) active substrates. This review deals with some substrate-related issues. Various methods will be introduced for preparing SERS substrates of Ag and Au for analytical purposes, from SERS substrates prepared by electrochemical or vacuum methods, to well-dispersed Au or Ag nanoparticle sols, to nanoparticle thin film substrates, and finally to ordered nanostructured substrates. Emphasis is placed on the analysis of the advantages and weaknesses of different methods in preparing SERS substrates. Closely related to the application of SERS in the analysis of trace sample and unknown systems, the existing cleaning methods for SERS substrates are analyzed and a combined chemical adsorption and electrochemical oxidation method is proposed to eliminate the interference of contaminants. A defocusing method is proposed to deal with the laser-induced sample decomposition problem frequently met in SERS measurement to obtain strong signals. The existing methods to estimate the surface enhancement factor, a criterion to characterize the SERS activity of a substrate, are analyzed and some guidelines are proposed to obtain the correct enhancement factor.  相似文献

The progress of surface-enhanced Raman scattering (SERS) microscopy and spectroscopy on individual nanostructured materials has been reviewed in this feature article. After a brief introduction on individual nanomaterial SERS, we provide a systematic overview on the fabrication and SERS studies of individual nanoparticulates, nano-junctions and hierarchical nano-aggregate. These SERS-active nanomaterials have great potential in designing novel highly sensitive SERS substrates for the development of SERS-based sensing devices with a broad range of applications.  相似文献

Recently, Raman spectroscopy become a popular and potential analytical technique for the analysis of pharmaceuticals as a result of its advancement. The innovation of laser technology, Fourier Transform-Raman spectrometers with charge coupled device (CCD) detectors, ease of sample preparation and handling, mitigation of sub-sampling problems using different geometric laser irradiance patterns and invention of different optical components of Raman spectrometers are contributors of the advancement of Raman spectroscopy. Transmission Raman Spectroscopy is a useful tool in pharmaceutical analysis to address the problems related with sub-sampling in conventional Raman back scattering. More importantly, the development of surface-enhanced Raman scattering (SERS) has been a prominent advancement for Raman spectroscopy to be applied for pharmaceuticals analysis as it avoids the inherent insensitivity and fluorescence problems. As the active pharmaceutical ingredients (APIs) contain aromatic or conjugated domains with strong Raman scattering activity, Raman spectroscopy is an attractive alternative conventional analytical method for pharmaceuticals. Coupling of Raman spectroscopy with separation techniques is also another advancement applied to reduce or avoid possible spectral interferences. Therefore, in this review, transmission Raman spectroscopy, SERS, and SERS coupled with various separation techniques for pharmaceutical analysis are presented.  相似文献

Although surface-enhanced Raman spectroscopy (SERS) has been applied for gathering fingerprint information, even in single molecule analysis, the decayed Raman signals in aqueous solutions largely obstruct the on-site insight reaction process. In this study, large-scaled semiconductor films with multi-walled (TiO2/WO3/TiO2) nanopore distribution are fabricated by combining electrochemical anodization and sputtering technique, and then employed as the SERS substrates for detection of molecules at the solid/liquid interfaces. Given the remarkably improved electrochromic property of the multi-walled film, such SERS substrates were endowed with tunable oxygen vacancy (VO) density and distribution via simply applying electrochemical bias voltage, which enabled one to achieve an enhanced charge transfer efficiency and thus a remarkably increased Raman signal even in solution. The VO-rich SERS substrate is highly repeatable, thus providing a reliable platform for in-situ monitoring of the target molecules or intermediates at the solid/liquid interfaces.  相似文献

Two different silver colloids were prepared by chemical reduction of silver nitrate with trisodium citrate and hydroxylamine hydrochloride to compare their characteristics in relation to their possible use in surface-enhanced Raman scattering (SERS) spectroscopy. The morphology and plasmon resonance of the single nanoparticles and aggregates integrating these colloids were characterized by means of UV-vis absortion spectroscopy and scanning electron microscopy, revealing important differences between each type of nanoparticle as concerns their physical properties. These metallic systems also manifested differences in the aggregation and the adherence to glass surfaces, revealing significant differences in the chemical surface properties of these nanoparticles. SERS and surface-enhanced IR also indicated the presence of interference bands which can overlap the spectra of the analyte, mainly in the case of the citrate colloid. All these differences have an important influence on the applicability of these nanostructured systems in SERS. In fact, the enhancement factor and spectral pattern of the SERS obtained by using alizarin as a molecule probe are different.  相似文献

We investigated the cellular uptake behavior of non-fluorescent metal nanoparticles (NPs) by use of surface-enhanced Raman scattering (SERS) combined with dark-field microscopy (DFM). The uptake of Au NPs inside a single cell could also be identified by DFM first and then confirmed by z-depth-dependent SERS at micrometer resolution. Guided by DFM for the location of Au NPs, an intracellular distribution assay was possible using Raman dyes with unique vibrational marker bands in order to identify the three-dimensional location inside the single cell by obtaining specific spectral features. Au NPs modified by 4-mercaptobenzoic acid (MBA) bearing its –COOH surface functional group were used to conjugate transferrin (Tf) protein using the 1-ethyl-3-[3-dimethylaminopropyl]carbodiimide hydrochloride (EDC) reaction. The protein conjugation reaction on Au surfaces was examined by means of color change, absorption spectroscopy, and SERS. Our results demonstrate that DFM techniques combined with SERS may have great potential for monitoring biological processes with protein conjugation at the single-cell level.  相似文献

Although surface-enhanced Raman spectroscopy (SERS) is probably one of the most sensitive surface spectroscopic techniques, most of the previously electrochemical SERS studies have been carried out in steady state. Recently, much attention has been attracted to combine time-dependent SERS with electrochemical transient techniques to investigate the electrode/solution interfacial dynamics at molecular level[1,2]. Our previous studies indicate that thiourea (TU) can coadsorb with ClO4- at Au electrodes in steady state. In the present work, time-dependent SERS has been used to study the structural change with the potential step for the coadsorption system.  相似文献

Track it down: A recognized surface-enhanced Raman scattering (SERS) nanotag signal was monitored from a thin, dispersed layer of bisphosphonate-functionalized nanotags on a bone sample, through a 20?mm thick specimen of porcine muscle tissue by surface-enhanced spatial offset Raman spectroscopy (SESORS; see picture). 对硝基苯硫酚是表面增强拉曼光谱研究中最常用的探针分子之一,对硝基苯硫酚在电极表面电化学还原反应的研究有助于对芳香族硝基化合物还原机理的认识. 本文应用暂态电化学-表面增强拉曼光谱技术,研究了对硝基苯硫酚在循环伏安和计时电流法过程中的表面增强拉曼光谱. 结果表明,实验实现了完全与电化学检测时间分辨率同步的表面增强拉曼光谱检测,以最快5毫秒的时间分辨率研究了对硝基苯硫酚分子在金电极表面的还原过程. 结果分析推测其此反应过程极快,在5毫秒的时间分辨率下仍难以捕获其中间物种. 本研究为人们更深层次研究和认识硝基苯类化合物电化学还原过程提供了参考和方向.  相似文献
