The NH acidity of pyrrolanthrone in dimethyl sulfoxide and the COOH acidity of pyrrolanthrone-1-carboxylic acid exceed the acidities of indole and indole-2-carboxylic acid by 6 and 2.5 orders of magnitude, respectively; this is due to the electron-acceptor effect of the condensed anthrone ring. A quantitative estimate of the effect of substituents in the 1 position on the NH acidity indicates that the bonds in the pyrrole fragment are localized to a significant degree; the C1-N bond approaches a single bond, and the C1-C10b bond consequently approaches a double bond. A new β-amino acid, trans-3-aminopyran-2-carboxylic acid (APyC), was designed and synthesized from (R)-glyceraldehyde derivative and used in the synthesis of α/β-peptides in a 1 : 1 alternating pattern with d-Ala. The presence of oxygen atom at the Cβ(2)-position in APyC was envisaged to provide opportunity for additional interaction. These hybrid peptides have shown the presence of 9/11-helix through extensive NMR and MD studies. The amide protons of d-Ala, in addition to participating in 9-mr H-bonding with CO of succeeding β-residue, were also involved in additional electrostatic interaction with pyran ring oxygen of preceding β-residue, which facilitated further stabilization to the 9/11-mixed helix. The study thus results in a new 'motif' for a 9/11-helix, and the first example from a cyclic β-amino acid.  相似文献

The mechanism of synthesis of acridones based on treatment of diphenylamine-2-carboxylic acid with phosphorus oxychloride is considered. It is established that triphenylamine-2-carboxylic acid and the anilide of diphenylamine-2-carboxylic acid, when treated with phosphorus oxychloride, give high yield of 10-phenylacridone and 9-anilinoacridone respectively. The hitherto undescribed N-(o-chlorobenzoyl)diphenylamine-2-carboxyanilide is obtained by reacting phenylanthranilanilide with o-chlorobenzoyl chloride, or by heating o-chlorobenzanilide with copper powder in nitrobenzene.For Part XXXVII see [1].  相似文献

3-Methyl-5-nitrobenzofuran ( 2 ) and 3-methyl-5-nitrobenzofuran-2-carboxylic acid ( 3 ) were obtained by heating 2-acetyl-4-nitrophenoxyacetic acid ( 1 ) with various bases in acetic anhydride. It appeared that 3-hydroxy-3-methyl-5-nitro-2,3-dihydrobenzofuran-2-carboxylic acid ( 4 ) was the intermediate in the benzofuran synthesis. The properties of 4 were examined under various conditions. Using strong bases such as triethyl-amine in place of sodium acetate, 3-methyl-5-nitrobenzofuran-2-carboxylic acid ( 3 ) was obtained exclusively. However, in the presence of acetic acid in the reaction mixture 3-methyl-5-nitrobenzofuran ( 2 ) was obtained in good yield. The reaction pathways for the formation of 2 and 3 are discussed.  相似文献

1-Benzenesulfonylindole-2,3-dicarboxylic anhydride was reacted with aniline to give the 2-carbamoylindole-3-carboxylic acid as the sole product, but with N-methylaniline, the 3-carbamoylindole-2-carboxylic acid was the major product, which could be transformed into the 1-benzenesulfonylbenzo-γ-carbolinone in the presence of Pd(OCOCF3)2. The reduction of the benzo-γ-carbolinone with LiAlH4 gave the cryptosanginolentine in high yield.  相似文献

Nine 2,4-dioxo-2,3,4,5-tetrahydro-1H-pyrrolo[3,2-d]pyrimidine-6-carboxylic acid benzyl esters 12 were synthesized in four steps from 4-oxo-N-(PhF)proline benzyl ester 7 by a general method in which elements of molecular diversity were readily added onto the pyrimidine nitrogens. Conversion of 4-oxoproline 7 into the corresponding aminopyrrole 8 using benzyl-, allyl-, and isopropylamine followed by treatment with phenyl, allyl, and ethyl isocyanate gave nine different ureas 9. 4-Ureido-1H-pyrrole-2-carboxylic acid benzyl esters 9 were then converted into the respective pyrrolo[3,2-d]pyrimidines 12 using trichloroacetyl chloride in acetonitrile followed by treatment with Cs(2)CO(3). Crystallization from toluene gave the desired deazapurines in 37-55% overall yield from proline 7.  相似文献

Unprecedented aromatization was observed during N-alkylation reactions of 1-methyl-3,4-dihydro-β-carboline-3-carboxylic acid methyl ester, giving rise to 9-alkyl-1-methyl-β-carboline-3-carboxylic acid methyl esters. Inverse addition of base during a similar reaction resulted in a chemoselective alkylation to form novel 3-butyl-1-methyl-3,4-dihydro-β-carboline-3-carboxylic acid methyl ester as the major product in good yield.  相似文献

Two molecules of indole derivative, e.g. indole-5-carboxylic acid, reacted with one molecule of thiol, e.g. 1,2-ethanedithiol, in the presence of trifluoroacetic acid to yield adducts such as 3-[2-(2-amino-5-carboxyphenyl)-1-(2-mercaptoethylthio)ethyl]-1Hindole-5-carboxylic acid. Parallel formation of dimers, such as 2,3-dihydro-1H,1'H-2,3'-biindole-5,5'-dicarboxylic acid and trimers, such as 3,3'-[2-(2-amino-5-carboxyphenyl) ethane-1,1-diyl]bis(1H-indole-5-carboxylic acid) of the indole derivatives was also observed. Reaction of a mixture of indole and indole-5-carboxylic acid with 2-phenylethanethiol proceeded in a regioselective way, affording 3-[2-(2-aminophenyl)-1-(phenethylthio)ethyl]-1H-indole-5-carboxylic acid. An additional product of this reaction was 3-[2-(2-aminophenyl)-1-(phenethylthio)ethyl]-2,3-dihydro-1H,1'H-2,3'-biindole-5'-carboxylic acid, which upon standing in DMSO-d6 solution gave 3-[2-(2-aminophenyl)-1-(phenethylthio)ethyl]-1H,1'H-2,3'-biindole-5'-carboxylic acid. Structures of all compounds were elucidated by NMR, and a mechanism for their formation was suggested.  相似文献

We have recently described a new synthetic route which has been used in our laboratory for the preparation of several novel heterocyclic systems comprising, the I,l-dimethylthiazolidino[3,4-o)piperazine-5,8- dione-3-carboxylic acid and the 2,2-dimethylthiazolidino[3,2-a]piper- azine-5,8-dione-3-carboxylic acid (8). The former was obtained with quan- titative yield and the latter with 71% yield. As a development of our studies on novel 2,5-piperazinedione systems, we describe in this report, the synthesis of the (2,5-dioxopiperazinyl)2-isobutyric acid (9), then-butyl(2-isopropyl)3,6-dioxopiperazinyl-l) acetate (1 l), and another synthetic route, illustrated in Scheme I, by which the compound 8 was obtained with quantitative yield. Spectrometric data were obtained and their interpretation confirms the proposed structure of the new compounds.  相似文献

A short and practical synthesis of 2′-deoxymugineic acid (DMA) has been developed via reductive alkylation with the aldehyde intermediates. No protection of azetidine-2-carboxylic acid was required and the presence of free carboxylic acid function facilitated purification by simple acid and base extractions. Furthermore, the intermediates were conveniently purified by HPLC due to the presence of chromophoric benzyl ester protecting group(s). Hydrogenolysis of the benzyl protecting groups in the final step furnished DMA in overall good yield.  相似文献

A series of esters of R-tetrahydrothiazo-2-thione-4-carboxylic acid[R-TTCA] was synthesized by direct esterification of R-TTCA with alcohols(CH3OH,C2H5OH,n-C3H7OH,i-C3H7OH,n-C4H9OH,sec-C4H9OH)in the presence of TiCl4 as the catalyst at room temperature without using any other solvent or dehydrant in high yields,91.6%-99.1% for primary alcohols and 55%- 80% for secondary alcohols.The catalyst has a strong chemoselective activity for the esterification of primary alcohols with R-TTCA in the presence of secondary alcohols.Owing to high yield,high chemoselectivity,and mild conditions used,this is an efficient method for the esterification of primary alcohols with R-TTCA.  相似文献

A series of substituted 1,2-dihydro-6-oxo-pyrrolo[3,2,1-ij]quinoline-5-carboxylic acids for the treatment of systemic infections was synthesized via 7-bromo-3-ethylthio-4,5-difluoro-2-methylindole (3), which was prepared by Gassman's indole synthesis in excellent yield. The synthesized pyrroloquinolines were tested for their antibacterial activities. 8-Fluoro-1,2-dihydro-2-methyl-9-(4-methyl-1-piperazinyl)-6-oxo-6H- pyrrolo[3,2,1-ij]quinoline-5-carboxylic acid showed a potent antibacterial activity against gram-positive and gram-negative bacteria.  相似文献

Indan-1-one (1a), 1-tetralone (1b), fluorenone (1c), and anthrone (1d) reacted with mercaptoacetic acid in toluene in the presence of p-toluenesulfonic acid to give spiro[indan-1,2′-[1′,3′]oxathialan]-5′-one (2a), spiro[tetrahydro-naphthalene-1,2′-[1,3′]oxathialan]-5′-one (2b), spiro[fluorene9,2′-[1′,3′]-oxathialan]-5′-one (2c), and spiro[anthracene-9(10H)-2′-[1′,3′]-oxathialan]-5′-one (2d), respectively. Compounds 2a–d reacted with arenes in the presence of aluminum chloride to yield spiro[polycyclic-1′-isothiochroman]-4′-one derivatives 3a–t. The mechanisms of these reactions are discussed. All the synthesized spiroheterocycle derivatives were identified by conventional methods (IR, 1H-NMR spectroscopy) and elemental analyses. © 1996 John Wiley & Sons, Inc.  相似文献

Bromination of (E)-1-[4-(2-carboxy-vinyl)phenyl]-[1,2,3]triazole-4-carboxylic acid ethyl ester, which was synthesized in 90% yield by a Huisgen-type [3 + 2]-cycloaddition reaction between 3-(4-azidophenyl) acrylic acid and ethyl propiolate, in CHCl3 followed by a debrominative decarboxylation reaction with Et3N in DMF under microwave irradiation condition afforded stereoselective (Z)-1-(4-(2-bromovinyl)phenyl)-1,2,3-triazole-4-carboxylic acid ethyl ester in 94% yield. Treatment of (Z)-1-(4-(2-bromovinyl)phenyl)-1,2,3-triazole-4-carboxylic acid ethyl ester with EtONa in DMF afforded 1-(4-ethynylphenyl)-1,2,3-triazole-4-carboxylic acid ethyl ester in a yield of 90%.  相似文献

3-Trifluoroacetyl indoles are formed by the action of trifluoroacetic acid on indole-2-carboxylic acid and its benzo-substituted derivatives. New chiral (Z)-alpha,beta-didehydroamino acid (DDAA) derivatives with 3,5-dihydro-2H-1,4-oxazin-2-one structure 11a-f have been stereoselectively prepared after condensation of chiral glycine equivalent 7 with aldehydes in the presence of K(2)CO(3) under mild solid-liquid phase-transfer catalysis reaction conditions. These new systems have been used in diastereoselective cyclopropanation reactions using Corey's ylide for the asymmetric synthesis of 1-aminocyclopropane-1-carboxylic acids (ACCs) such as allo-corononamic and allo-norcoronamic acids. The hydrogenation reaction of these systems at ambient pressure in the presence of formaldehyde affords saturated oxazinones and N-methylated oxazinones which have been transformed into the N-methyl-alpha-amino acids (N-MAAs) (S)-2-(methylamino)butanoic acid and (S)-N-methylleucine. In addition, the parent alpha, beta-didehydroalanine derivative 11g has been prepared by a direct aminomethylation-elimination sequence from 7 and Eschenmoser's salt and has been used in Diels-Alder cycloaddition with endo selectivity for the synthesis of the enantiomerically pure bicyclic alpha-amino acids (-)-2-aminobicyclo[2.2.1]heptane-2-carboxylic and (-)-2-aminobicyclo[2.2.2]octane-2-carboxylic acids.  相似文献

(S)-Indoline-2-carboxylic acid was synthesized by use of a nitro amination approach with l-phenylalanine as chiral pool. The first step of the synthesis was nitration of l-phenylalanine, with urea nitrate (UN)/H2SO4 as nitrating reagent, to give 2,4-dinitro-l-phenylalanine in 75.7 % yield in one-pot synthesis and 69.1 % yield by step-wise nitration. Intramolecular nitro amination of 2,4-dinitro-l-phenylalanine gave (S)-6-nitro-indoline-2-carboxylic acid in 65.7 % yield and more than 99.5 % enantiomeric excess (ee). The title compound, (S)-indoline-2-carboxylic acid, was obtained in 85.9 % yield and high ee by one-pot transformation of (S)-6-nitroindoline-2-carboxylic acid. The total synthesis consisted of three operations and gave the title compound in 42 % yield and more than 99.5 % ee.  相似文献

The pyridine-containing central domain of the amythiamicin group of thiopeptide antibiotics is prepared in protected form in 9 steps, 93%ee and 18% overall yield from (S)-2-[1-(tert-butoxycarbonylamino)-2-methylpropyl]thiazole-4-carboxylic acid by Michael addition-cyclodehydration of a 2-(2-thiazolyl)enamine and 1-(2-thiazolyl)propynone.  相似文献
