We report the synthesis of a dense dendrimer ended by the PTA ligand (1,3,5-triaza-7-phosphaadamantane), and the use of the corresponding Rh and Ru complexes for catalysis. The catalytic properties of these dendrimers are compared with those of two other dendrimers: a dendrimer of the same generation but having half the number of ligands, and a dendrimer of the next generation, but having the same number of ligands. The positive influence of the density of catalytic sites on the surface of these dendrimers for alcohol isomerization in water has been evidenced.  相似文献

Amphiphilic polyester-co-polyether (PEPE) dendrimers synthesized from poly(ethylene glycol) (PEG) were examined to understand the influence of alterations in the architecture of dendrimers on their conformation at interfaces and distribution of various groups on their surface. Effect of changes in the number of branching points, type of terminal functional groups and generation of dendrimer was primarily evaluated. Dendrimers were deposited on mica by spin coating at 0.1 mg/mL. Tapping mode atomic force microscopy (AFM) was employed for the visualization of dendrimer topographies while, X-ray photoelectron spectroscopy (XPS), AFM phase and force imaging were used as the tools for characterization of their surfaces. Individual dendrimer molecules could be imaged by AFM, which showed that they are round or oval in topography. Dendrimers were also flattened on mica but the extent of flattening differed with the chemical structure; for instance, third generation dendrimers were more flattened than second generation dendrimers whereas, dendrimers with higher number of branches had greater height above the mica surface. Hydrophilic and hydrophobic groups present towards the aerial interface existed in distinct zones rather than being distributed randomly, except in dendrimer with higher number of branches. The percentage of various hydrophobic groups on the surface of dendrimer was enhanced by increase in the number of branches but, was lowered by the presence of hydroxyl groups as the pendant terminal groups. Furthermore, the core of dendrimers was not always located towards the centre, its position was found to be altered by the number of branching points, type of terminal functional groups and the generation of dendrimer.  相似文献

We have investigated the fluorescence properties of dendrimers (Gn is the dendrimer generation number) containing four different luminophores, namely terphenyl (T), dansyl (D), stilbenyl (S), and eosin (E). In the case of T, the dendrimers contain a single p-terphenyl fluorescent unit as a core with appended sulfonimide branches of different size and n-octyl chains. In the cases of D and S, multiple fluorescent units are appended in the periphery of poly(propylene amine) dendritic structures. In the case of E, the investigated luminophore is noncovalently linked to the dendritic scaffold, but is encapsulated in cavities of a low luminescent dendrimer. Depending on the photophysical properties of the fluorescent units and the structures of the dendrimers, different mechanisms of fluorescence depolarization have been observed: (i) global rotation for GnT dendrimers; (ii) global rotation and local motions of the dansyl units at the periphery of GnD dendrimers; (iii) energy migration among stylbenyl units in G2S; and (iv) restricted motion when E is encapsulated inside a dendrimer, coupled to energy migration if the dendrimer hosts more than one eosin molecule.  相似文献

Although it is known that noncollagenous proteins of dental origin bind to the hydroxyapatite crystal surfaces, no measure of their binding strength has been calculated. This experiment used -COOH-capped generation 7 PAMAM dendrimers as nanoprobes of the biological hydroxyapatite nanorod surfaces. Dendrimer distribution was characterized using AFM. The results showed dendrimers to be spaced at intervals along the c-axis of the crystals. From these observations and assuming a fully ionized -COOH dendrimer, a mathematical model of the binding capacity of the crystal surface with the dendrimer was developed. The Monte Carlo method was used to simulate the binding process between the dendrimer and crystal surface, and the binding strength of the -COOH groups to the surface was calculated to be 90 +/- 20 kJ/mol. These results support the CFM studies which have described alternating bands of charge domains on the crystal surface and that the binding strength will be dependent on both the intensity of the charge on the protein and the crystal surface.  相似文献

Dendrimers are highly branched and monodisperse macromolecules that display an exact and large number of functional groups distributed with unprecedented control on the dendritic framework. Based on their globular structure, compared to linear polymers of the same molecular weight, dendrimers are foreseen to deliver extraordinary features for applications in areas such as cancer therapy, biosensors for diagnostics and light harvesting scaffolds. Of the large number of reports on dendrimer synthesis only a few have reached commercial availability. This limitation can be traced back to challenges in the synthetic paths including a large number of reaction steps required to obtain dendritic structures with desired features. Along with an increased number of reaction steps come not only increased waste of chemical and valuable starting materials but also an increased probability to introduce structural defects in the dendritic framework. This tutorial review briefly covers traditional growth approaches to dendrimers and mainly highlights accelerated approaches to dendrimers. A special focus capitalizes on the impact of the click chemistry concept on dendrimer synthesis and the promise it has to successfully accomplish highly sophisticated dendrimers, both traditional as well as heterofunctional, in a minimum number of chemical steps. It is clear that accelerated synthetic approaches are of greatest importance as these will encourage the scientific community to synthesize and access dendrimers for specific applications. The final goal of accelerated synthesis is to deliver economically justified dendritic materials for future applications without compromising the environmental perspective.  相似文献

Cobalt(II) has been utilized as an external paramagnetic (1)H NMR probe for the study of the structure of dendrimers that possess specifically located metal recognition sites. The hyperfine-shifted (1)H NMR signals of the Co(II) complexes of several 2,6-diamidopyridine-containing dendrimers have been fully assigned by means of 1D and 2D NMR techniques, including NOE difference, EXSY, COSY, and TOCSY. Temperature-dependent T(1) values of the hyperfine-shifted signals were used to conclude that the Co(II)-dendrimer complexes are in the "liquidlike" regime, indicative of a shell-like structure instead of a "dense-core" structure. The presence of sizable cavities within the dendrimers was observed including a loosely packed conformation for the 2,6-diamidopyridino moiety to bind to potential guest molecules. Cooperativity among the dendritic arms in metal binding is also observed, whereby two dendritic arms bind to the metal center at the same time. In the case of dendrimers with the metal binding site located near the surface of the molecule, such binding cooperativity is still observed despite the large degree of freedom of the metal-binding moiety. Cooperativity among the dendritic arms can thus be considered an intrinsic property, which has to be taken into consideration in future design of functional dendrimers for the purpose of specific recognition and catalysis. The hydrodynamic radii of these dendrimers have been determined by means of nuclear Overhouser effect at low temperature. The study offers a method for the study of the dynamics of dendrimers in solution under different conditions and upon ligand binding and recognition. The study also provides a tool for monitoring systematic variation of the metal binding site in different dendrimer frameworks for specific applications, such as catalysis and molecular recognition.  相似文献

In this paper, we report synthesis of new alkyldiphenyl phosphine ligand modified poly(ether imine) dendrimers up to the third generation. The phosphinated dendrimers were obtained by functional group transformations of the alcohols present at the periphery of the dendrimers to chloride, followed by phosphination using LiPPh2. The modification at the peripheries of the dendrimers was performed successfully to obtain up to 16 alkyl diphenylphosphines in the case of a third generation dendrimer, in good yields for each individual step. After phosphination, dendritic ligands were complexed with Pd(COD)Cl2 to give dendritic phosphine-PdII complexes. Both the ligands and the metal complexes were characterized by spectroscopic and spectrometric techniques including high-resolution mass spectral analysis for the lower generations. Evaluation of the catalytic efficacies of the dendrimer-PdII metal complexes in mediating a prototypical C-C bond forming reaction, namely the Heck reaction, was performed using various olefin substrates. While the substrate conversion lowered with catalyst in the order from monomer to third generation dendrimer, the second and third generation dendrimers themselves were found to exhibit significantly better catalytic activities than the monomer and the first generation dendrimer.  相似文献

Several new series of dendrimers containing a single redox-active 4,4'-bipyridinium (viologen) residue were synthesized and characterized. In these dendrimers, the viologen group is covalently attached to the apical position of a Newkome- or Frechet-type dendron, ranging in size from first to third generation of growth. The half-wave potentials corresponding to the two consecutive one-electron reductions of the viologen residue are affected by the size of the dendritic component. The size effects are more pronounced in the Newkome-type dendrimers and seem to result from the polarity contrast between the microenvironments provided by the solution and the internal phase of the dendrimer. Unlike in many other dendrimers having a redox-active core, the voltammetric behavior remains fast (reversible) even in third generation dendrimers. Pulse gradient stimulated echo NMR diffusion coefficient measurements on the Newkome-type dendrimers reveal that their hydrodynamic radii are relatively invariant in solvents of widely different polarities (dichloromethane to dimethyl sulfoxide). The host-guest binding interactions between the viologen residue in these dendrimers and the crown ether host bis-p-phenylene-34-crown-8 were also investigated. While in Newkome-type dendrimers the growth of the dendron caused a substantial attenuation of the binding constant values, this size effect was not observed in the Frechet-type dendrimers. These electrochemical and binding measurements underscore some of the structural differences between these two common types of dendritic architectures.  相似文献

A new class of poly(benzyl ether) dendrimers, decorated in their cores with N‐Boc‐protected 1,2‐diphenylethylenediamine groups, were synthesized and fully characterized. It was found that the gelation capability of these dendrimers was highly dependent on dendrimer generation, and the second‐generation dendrimer (R,R)‐G2DPENBoc proved to be a highly efficient organogelator. A number of experiments (SEM, TEM, FTIR spectroscopy, 1H NMR spectroscopy, rheological measurements, UV/Vis absorption spectroscopy, CD, and XRD) revealed that these dendritic molecules self‐assembled into elastically interpenetrating one‐dimensional nanostructures in organogels. The hydrogen bonding, π–π, and solvophobic interactions were found to be the main driving forces for formation of the gels. Most interestingly, these dendritic organogels exhibited smart multiple‐stimulus‐responsive behavior upon exposure to environmental stimuli such as temperature, anions, and mechanical stress.  相似文献

We report the development of dendritic siRNA nanostructures that are able to penetrate even difficult to transfect cells such as neurons with the help of a special receptor ligand. The nanoparticles elicit strong siRNA responses, despite the dendritic structure. An siRNA dendrimer directed against the crucial rabies virus (RABV) nucleoprotein (N protein) and phosphoprotein (P protein) allowed the suppression of the virus titer in neurons below the detection limit. The cell‐penetrating siRNA dendrimers, which were assembled using click chemistry, open up new avenues toward finding novel molecules able to cure this deadly disease.  相似文献

Dissipative particle dynamics simulations are used to study the specific binding structures of polyamidoamine (PAMAM) dendrimers on amphiphilic membranes and the permeation mechanisms. Mutually consistent coarse-grained (CG) models both for PAMAM dendrimers and for dimyristoylphosphatidylcholine (DMPC) lipid molecules are constructed. The PAMAM CG model describes correctly the conformational behavior of the dendrimers, and the DMPC CG model can properly give the surface tension of the amphiphilic membrane. A series of systematic simulations is performed to investigate the binding structures of the dendrimers on membranes with varied length of the hydrophobic tails of amphiphiles. The permeability of dendrimers across membranes is enhanced upon increasing the dendrimer size (generation). The length of the hydrophobic tails of amphiphiles in turn affects the dendrimer conformation, as well as the binding structure of the dendrimer-membrane complexes. The negative curvature of the membrane formed in the dendrimer-membrane complexes is related to dendrimer concentration. Higher dendrimer concentration together with increased dendrimer generation is observed to enhance the permeability of dendrimers across the amphiphilic membranes.  相似文献

Dendrimers and dendrons offer an excellent platform for developing novel drug delivery systems and medicines. The rational design and further development of these repetitively branched systems are restricted by difficulties in scalable synthesis and structural determination, which can be overcome by judicious use of molecular modelling and molecular simulations. A major difficulty to utilise in silico studies to design dendrimers lies in the laborious generation of their structures. Current modelling tools utilise automated assembly of simpler dendrimers or the inefficient manual assembly of monomer precursors to generate more complicated dendrimer structures. Herein we describe two novel graphical user interface toolkits written in Python that provide an improved degree of automation for rapid assembly of dendrimers and generation of their 2D and 3D structures. Our first toolkit uses the RDkit library, SMILES nomenclature of monomers and SMARTS reaction nomenclature to generate SMILES and mol files of dendrimers without 3D coordinates. These files are used for simple graphical representations and storing their structures in databases. The second toolkit assembles complex topology dendrimers from monomers to construct 3D dendrimer structures to be used as starting points for simulation using existing and widely available software and force fields. Both tools were validated for ease-of-use to prototype dendrimer structure and the second toolkit was especially relevant for dendrimers of high complexity and size.  相似文献

The generation of dendrimers is a powerful tool in the control of the size and biodistribution of polyion complexes (PIC). Using a combinatorial screening of six dendrimers (18–243 terminal groups) and five oppositely charged PEGylated copolymers, a dendrimer‐to‐PIC hierarchical transfer of structural information was revealed with PIC diameters that increased from 80 to 500 nm on decreasing the dendrimer generation. This rise in size, which was also accompanied by a micelle‐to‐vesicle transition, is interpreted according to a cone‐ to rod‐shaped progression in the architecture of the unit PIC (uPIC). This precise size tuning enabled dendritic PICs to act as nanorulers for controlled biodistribution. Overall, a domino‐like control of the size and biological properties of PIC that is not attainable with linear polymers is feasible through dendrimer generation.  相似文献

Functionalization of multivalent structures such as dendrimers and monolayer passivated nanoparticles with catalytically active groups results in very potent catalysts, a phenomenon described as the positive dendritic effect. Here, we describe a series of peptide dendrons and dendrimers of increasing generation functionalized at the periphery with triazacyclononane, a ligand able to form a strong complex with Zn(II). Kinetic studies show that these metallodendrimers very efficiently catalyze the cleavage of the RNA model compound HPNPP, with dendrimer D32 exhibiting a rate acceleration of around 80,000 (kcat/k(uncat)) operating at a concentration of 600 nM. A theoretical model was developed to explain the positive dendritic effect displayed by multivalent catalysts in general. A detailed analysis of the saturation profile and the Michaelis-Menten parameters kcat and KM shows that it is not necessary to ascribe the positive dendritic effect to, for instance, changes in the catalytic site, increased substrate binding constant, or changes in the microenvironment. Rather it appears that the efficient catalytic behavior of multivalent catalysts is mainly determined by two factors: the number of catalytic sites occupied by substrate molecules under saturation conditions, and the efficiency of the multivalent system to generate catalytic sites in which multiple catalytic units act cooperatively on the substrate.  相似文献

The novel iron(III) porphyrin dendrimers of generation zero ([ 1 ⋅FeIII]Cl), one ([ 2 ⋅FeIII]Cl), and two ([ 3 ⋅FeIII]Cl) (Fig. 1) were prepared (Schemes 1 and 3) as models of heme monooxygenases. They feature controlled axial ligation at the Fe center by one imidazole tethered to the porphyrin core and possess a vacant coordination site available for ligand binding and catalysis. The high purity of the dendrimers and the absence of structural defects was demonstrated by matrix‐assisted laser‐desorption‐ionization time‐of‐flight (MALDI‐TOF) mass spectrometry (Fig. 3). The electronic properties of the FeIII porphyrin dendrimers and comparison compounds [ 4 ⋅FeIII]Cl and [ 12 ⋅FeIII(1,2‐Me2Im)]Cl (1,2‐Me2Im=1,2‐dimethylimidazole) were investigated by UV/VIS and EPR (electronic paramagnetic resonance) spectroscopy, as well as by measurements of the magnetic moments by the Evans‐Scheffold method. Epoxidation of olefins and oxidation of sulfides to sulfoxides, catalyzed by the new dendritic metalloporphyrins, were investigated in CH2Cl2 with iodosylbenzene as the oxidant (Tables 1 and 2). The total turnover numbers were found to increase with the size of the dendrimer, due to improved catalyst stability at higher dendritic generations (Figs. 4 and 5). The second‐generation complex [ 3 ⋅FeIII]Cl was, therefore, the most efficient catalyst in the series, despite the fact that its active site is considerably hindered by the encapsulation inside the sterically demanding, fluctuating dendritic wedges. Very high product selectivities were observed in all oxidation reactions, regardless of dendrimer generation.  相似文献

Detection and separation of polyamidoamine dendrimers possessing various cores and surface groups was studied by polyacrylamide gel electrophoresis. Although many dyes and staining techniques were able to detect dendrimers on polyacrylamide gels, Coomassie Blue was found to be the most sensitive and convenient. Amine and hydroxyl terminated dendrimers were best separated under acidic conditions, while dendrimers with carboxyl surfaces required alkaline buffers. Some dendrimers were very susceptible to diffusion that could occur during their separation, staining or destaining steps. In the absence of an appropriate fixation step, dendrimers could be resolved by using small pore size gels and low voltage or current. Increasing core lengths did not significantly affect migration of a given dendrimer generation but exhibited improved separation and staining characteristics. Polyacrylamide gel electrophoresis was found to be a rapid, inexpensive, and reliable procedure to characterize many different water-soluble dendritic macromolecules.  相似文献

A C3-symmetric tridentate hexahydroxamate ligand molecule was specially synthesized and used for coordination self-assembly of branched multilayers on Au surfaces precoated with a self-assembled monolayer (SAM) of ligand anchors. Layer-by-layer (LbL) growth of multilayers via metal-organic coordination using Zr4+ ions proceeds with high regularity, adding one molecular layer in each step, as shown by ellipsometry, wettability, UV-vis spectroscopy, and atomic force microscopy (AFM). The branched multilayer films display improved stiffness, as well as a unique defect self-repair capability, attributed to cross-linking in the layers and lateral expansion over defects during multilayer growth. Transmetalation, i.e., exposure of Zr4+-based assemblies to Hf4+ ions, was used to evaluate the cross-linking. Conductive atomic force microscopy (AFM) was used to probe the electrical properties of the multilayers, revealing excellent dielectric behavior. The special properties of the branched layers were emphasized by comparison with analogous multilayers prepared similarly using linear (tetrahydroxamate) ligand molecules. The process of defect annihilation by bridging over defective areas, attributed to lateral expansion via the excess bishydroxamate groups, was demonstrated by introduction of artificial defects in the anchor monolayer, followed by assembly of two layers of either the linear or the branched molecule. Analysis of selective binding of Au nanoparticles (NPs) to unblocked defects emphasized the superior repair mechanism in the branched layers with respect to the linear ones.  相似文献

Generation 5 ethylenediamine (EDA)-cored poly(amidoamine) (PAMAM) dendrimers (E5, E denotes the EDA core and 5 the generation number) with different degrees of acetylation and carboxylation were synthesized and used as a model system to investigate the effect of charge and the influence of dendrimer surface modifications on electrophoretic mobility (EM) and molecular distribution. The surface-modified dendrimers were characterized by size-exclusion chromatography, 1H NMR, MALDI-TOF-MS, PAGE, and CE. The focus of our study was to determine how EM changes as a function of particle charge and molecular mass, and how the molecular distribution changes due to surface modifications. We demonstrate that partially modified dendrimers have much broader migration peaks than those of fully surface functionalized or unmodified E5 dendrimers due to variations in the substitution of individual dendrimer surfaces. EM decreased nonlinearly with increases in surface acetylation for both PAMAM acetamides and PAMAM succinamic acids, indicating a complex migration activity in CE separations that is not solely due to charge/mass ratio changes. These studies provide new insights into dendrimer properties under an electric field, as well as into the characterization of dendrimer-based materials being developed for medical applications.  相似文献

NOESY-HSQC 3D-NMR and NOESY 2D-NMR techniques have been used on a 750 MHz spectrometer to study the chain conformations of different generation DAB dendrimers (poly[propylene imine] dendrimers) in chloroform and benzene solutions. The high-field multidimensional NMR techniques provided the chemical shift dispersion needed to resolve all of the unique resonances in the dendrimers. By studying the NOE interactions among dendritic chain protons, information about through space interactions between protons on different parts of the dendrimer chain is obtained, which is directly related to the conformation of the dendrimer. These experiments also give further proof of the chemical shift assignments obtained from the HMQC-TOCSY 2D and 3D NMR experiments. The concentration effects on chemical shifts have also been observed, revealing information about the interactions between solvent and different parts of dendrimer molecules. These studies clearly show for DAB dendrimers, that folded chain conformations can occur in nonpolar solvents such as benzene and extended chain conformations are predominant in polar solvents such as chloroform.  相似文献

As a monomeric ligand for a soluble 1D coordination polymer, a benzyl-ether based dendrimer having a rigid 4,4'-bispyridine ligand at the focal point has been synthesized and the coordination chemistry with Pd(II) investigated by nuclear magnetic resonance, ultraviolet-visible and fluorescence spectroscopies, gel permeation chromatography measurement, and X-ray photoelectron spectroscopy. As a result, it was found that the synthesized dendrimer forms a stable, soluble Pd(II) coordination polymer with rough estimation of degree of polymerization of 10 in organic solvents. Furthermore, through the coordination polymer we attempted to link fourth-generation poly(amidoamine) dendrimers (PAMAM) individually immobilized on mica and confirmed the interconnection of the PAMAM through coordination polymers by atomic force microscopy.  相似文献
