The self-aggregation behavior of two amphiphilic poly(ethylene oxide)-poly[(R)-3-hydroxybutyrate]-poly(ethylene oxide) (PEO-PHB-PEO) triblock copolymer samples with nearly identical PHB block lengths but different PEO block lengths, PEO-PHB-PEO(2000-810-2000) and PEO-PHB-PEO(5000-780-5000), was studied with dynamic and static light scattering (DLS and SLS), in combination with fluorescence spectroscopy and transmission electron microscopy (TEM). The formation of polymeric micelles by the two PEO-PHB-PEO triblock copolymers was confirmed with fluorescence technique and TEM. DLS analysis showed that the hydrodynamic radius (R(h)) of the monodistributed polymeric micelles increased with an increase in PEO block length. The relative thermostability of the triblock copolymer micelles was studied by SLS and DLS at different temperatures. The aggregation number and the ratio of the radius of gyration over hydrodynamic radius were found to be independent of temperature, probably due to the strong hydrophobicity of the PHB block. The combination of DLS and SLS studies indicated that the polymeric micelles were composed of a densely packed core of hydrophobic PHB blocks and a corona shell formed by hydrophilic PEO blocks. The aggregation numbers were found to be approximately 53 for PEO-PHB-PEO(2000-810-2000) micelles and approximately 37 for PEO-PHB-PEO(5000-780-5000) micelles. The morphology of PEO-PHB-PEO spherical micelles determined by DLS and SLS measurements was further confirmed by TEM.  相似文献

Block copolymers comprising thermosensitive poly(N-isopropylacrylamide) (PNIPAM) and hydrophobic poly(n-butyl acrylate) (PBA) blocks, were synthesized using the reversible addition-fragmentation chain transfer polymerization (RAFT), their thermosensitive behavior was studied by ultraviolet spectrophotometer (UV) and dynamic light scattering (DLS). The lower critical solution temperature (LCST) was strongly correlated to the hydrophobic/hydrophilic ratio of the copolymers. Their micellization and self-assembly behavior in dilute aqueous solution were studied by surface tension (SFT), DLS and TEM. The resulting block copolymers reversibly formed or deformed micellar assemblies during their LCSTs. The critical micelle concentration (CMC) was controlled by the composition of PBA and PNIPAM, indicating the successful formation of the block copolymers.  相似文献

Summary: Branched poly(L -lactide)-poly(ethylene glycol) (PLLA-PEG) block copolymers were synthesized from trifunctional PLLA and amine functionalized methoxy poly(ethylene glycol)s. The copolymers in water formed hydrogels that showed thermo-responsive behavior. The hydrogels underwent a gel to sol transition with increasing temperature as determined with the vial tilting method and oscillatory rheology. For all copolymers, the transition temperature increased with increasing copolymer concentration. The transition temperature of corresponding branched copolymers also increased with increasing PEG molecular weight, and surprisingly decreased with increasing molecular weight of the PLLA branches. In general, the gel-sol transition is explained by disruption of micellar or aggregate interactions because of partial dehydration and shrinkage of the PEG chains. An increase in the molecular weight of the PLLA branches led to the formation of micelles and aggregates as observed with DLS at low concentrations. It is speculated that the non-uniform size distribution and possible crystallization of longer PLLA blocks may have a negative effect on the formation of micellar packing upon gelation, allowing the disruption of micellar or aggregate interactions to occur at lower temperatures. The transition temperature of the gels could be tuned closely to body temperature by varying the concentration of the solution or the molecular weight of the PEG block and the PLLA blocks, which implies that these polymers may be used as injectable systems for in-situ gel formation.  相似文献

Several series of amphiphilic diblock copolymers are investigated as macrosurfactants in comparison to reference low-molar-mass and polymeric surfactants. The various copolymers share poly(butyl acrylate) as a common hydrophobic block but are distinguished by six different hydrophilic blocks (one anionic, one cationic, and four nonionic hydrophilic blocks) with various compositions. Dynamic light scattering experiments indicate the presence of micelles over the whole concentration range from 10(-4) to 10 g x L(-1). Accordingly, the critical micellization concentrations are very low. Still, the surface tension of aqueous solutions of block copolymers decreases slowly but continuously with increasing concentration, without exhibiting a plateau. The longer the hydrophobic block, the shorter the hydrophilic block, and the less hydrophilic the monomer of the hydrophilic block is, the lower the surface tension is. However, the effects are small, and the copolymers reduce the surface tension much less than standard low-molar-mass surfactants. Also, the copolymers foam much less and even act as anti-foaming agents in classical foaming systems composed of standard surfactants. The copolymers stabilize O/W emulsions made of methyl palmitate as equally well as standard surfactants but are less efficient for O/W emulsions made of tributyrine. However, the copolymer micelles exhibit a high solubilization power for hydrophobic dyes, probably at their core-corona interface, in dependence on the initial geometry of the micelles and the composition of the block copolymers. Whereas micelles of copolymers with strongly hydrophilic blocks are stable upon solubilization, solubilization-induced micellar growth is observed for copolymers with moderately hydrophilic blocks.  相似文献

以聚(ε-己内酯-b-L-丙交酯)/聚乙二醇单甲醚(P(CL-b-LLA)-b-mPEG)和聚(ε-己内酯-b-D,L-丙交酯)/聚乙二醇单甲醚(P(CL-b-DLLA)-b-mPEG)两种两亲嵌段共聚物为载体,选择了物理状态完全不同、而疏水性相近的吲哚美辛和维生素E为模型药物,研究了药物包载对高分子胶束形态的影响.发现两种药物在高分子胶束内部的增溶均会导致胶束形态发生显著改变,变化行为与胶束内核的结晶性和药物疏水性有关.另外,还研究了两种嵌段共聚物的载药性能,发现非结晶性疏水内核共聚物的药物包载率明显大于可结晶疏水内核的共聚物.  相似文献

Brush-like block copolymers with poly(t-butyl methacrylate) (PBMA) and poly(N-isopropylacrylamide) (PNIPAAm) as side arms, PBMA-b-PNIPAAm, were designed and synthesized via a simple free radical polymerization route. The chemical structure and molecular weight of these polymer brushes were characterized and determined by nuclear magnetic resonance (1H NMR), Fourier transform infrared spectrometry (FTIR) and gel permeation chromatography (GPC). The micellar formation by these polymer brushes in aqueous solutions were detected by a surface tension technique, and the critical micelle concentration (CMC) ranged from 1.53 to 8.06 mg L−1. The morphology and geometry of polymer micelles were investigated by transmission electron microscope (TEM) and dynamic light scattering (DLS). The polymer micelles assume the regularly-spherical core-shell structure with well-dispersed individual nanoparticles, and the particle size was in the range from 36 to 93 nm. The PNIPAAm segments exhibited a thermoreversible phase transition, so the resulting block polymer brushes were temperature-sensitive and the low critical solution temperature (LCST) was determined by UV-vis spectrometer at about 28.82–29.40°C. The characteristic parameters of the polymer micelles such as CMC, micellar size and LCST values were affected by their compositional ratios and the length of hydrophilic or hydrophobic chains. The evaluation for caffeine drug release behavior of the block polymer micelles demonstrated that the self-assembled micelles exhibited thermal-triggered properties in controlled drug release.  相似文献

New biodegradable/biocompatible ABC block copolymers, poly(ethylene oxide)‐b‐poly(glycidol)‐b‐poly(L ,L ‐lactide) (PEO‐PGly‐PLLA), were synthesized. First, PEO‐b‐poly(1‐ethoxyethylglycidol)‐b‐PLLA was synthesized by a successive anionic ring‐opening copolymerization of ethylene oxide, 1‐ethoxyethylglycidyl ether, and L ,L ‐lactide initiated with potassium 2‐methoxyethanolate. In the second step, the 1‐ethoxyethyl blocking groups of 1‐ethoxyethylglycidyl ether were removed at weakly acidic conditions leaving other blocks intact. The resulting copolymers were composed of hydrophilic and hydrophobic segments joined by short polyglycidol blocks with one hydroxyl group in each monomeric unit. These hydroxyl groups may be used for further copolymer transformations. The PEO‐PGly‐PLLA copolymers with a molecular weight of PLLA blocks below 5000 were water‐soluble. Above the critical micellar concentration (ranging from 0.05 to1.0 g/L, depending on the composition of copolymer), copolymers formed macromolecular micelles with a hydrophobic PLLA core and hydrophilic PEO shell. Thermosensitive polymer micelles are generally obtained with block copolymers in which one block exhibits a lower critical solution temperature in aqueous solution. We investigate a different design that is based on the use of one block bearing a thermally labile side group, whose hydrolysis upon heating shifts the hydrophilic-hydrophobic balance toward the destabilization of block copolymer micelles. Atom transfer radical polymerization was utilized to synthesize a series of diblock copolymers composed of hydrophilic poly(ethylene oxide) (PEO) and hydrophobic poly(2-tetrahydropyranyl methacrylate) (PTHPMA). We show that micelles of PEO-b-PTHPMA in aqueous solution can be destabilized as a result of the thermosensitive hydrolytic cleavage of tetrahydropyranyl (THP) groups that transforms PTHPMA into hydrophilic poly(methacrylic acid). The three related processes occurring in aqueous solution, namely, hydrolytic cleavage of THP, destabilization of micelles, and release of loaded Nile Red (NR), were investigated simultaneously using 1H NMR, dynamic light scattering, and fluorescence spectroscopy, respectively. At 80 degrees C, the results suggest that the three events proceed with a similar kinetics. Although slower than at elevated temperatures, the disruption of PEO-b-PTHPMA micelles can take place at the body temperature (approximately 37 degrees C), and the release kinetics of NR can be adjusted by changing the relative lengths of the two blocks or the pH of the solution.  相似文献

Four-arm star block polymers consisting of hydrophobic poly(?-caprolactone) (PCL) block and hydrophilic poly(2-(diethylamino) ethyl methacrylate)) (PDEAEMA) block were successfully synthesized by ring opening polymerization (ROP) and atom transfer radical polymerization (ATRP). Chain lengths of PDEAEMA segments were varied to obtain a series of star copolymers with different hydrophilic/hydrophobic ratio, which were desired for self-assembly study. Dynamic light scattering (DLS) and transmission electron microscopic (TEM) were used to study their self-assembly behavior. In the PBS solution with different pH value, the star polymers formed micelles or nanoparticles. Furthermore, the morphologies of the micelles were also pH-dependent. Critical micelle concentrations of star copolymers changed from 5.0 to 17.5 mg/L with the increase of hydrophilic block length or the pH decrease. Moreover, a steady increase was found on the micelles diameters when the pH decreased from 7.0 to 3.0. The low CMC value and slight changes on micelle diameter indicated that the micelle remained stable under the changing external stimulus.  相似文献

A series of poly(styrene‐blocktert‐butyl acrylate) heteroatom star block copolymers having various block lengths were prepared by atom transfer radical polymerization (ATRP), using an “as synthesized” cynurate modified trifunctional initiator. The structure of the star polymers was confirmed by the characterization of the individual arms resulting from hydrolysis. Amphiphilic poly(styrene‐block‐acrylic acid) star copolymers were further synthesized by hydrolyzing PtBA blocks using anhydrous trifluoroacetic acid. The characterization data are reported from analyses using gel permeation chromatography, infrared, 1H and 13C NMR spectroscopies. The stable micelle solution was prepared by dialyzing the solution of these polymers in N,N‐dimethylformamide against deionized water. The temperature‐induced associating behavior of these amphiphilic star polymers were studied using dynamic laser light scattering spectroscopy. The hydrodynamic diameter of both micelles and unassociated chains were obtained in the same solution using light scattering cumulant's calculation method. The homogeneity and the size distribution of the micelle population in the solution were determined using centrifuge/sedimentation particle size distribution analyzer. Field emission scanning electron microscope was used to visualize the size of the micelles formed and the micellar aggregates. The influence of the temperature on the viscosity of the micelle solution was studied using an Ubbelohde viscometer. The interaction of amphiphilic block copolymers comprising an anionic block (polyacrylate or polymethacrylate) and a hydrophobic block (polystyrene, poly(butyl acrylate) or polyisobutylene) with lightly crosslinked poly(N,N-diallyl-N,N-dimethylammonium chloride) is studied for the first time. It is shown that the cationic hydrogel can sorb anionic amphiphilic block copolymers via electrostatic interaction with the corona of block copolymer micelles. The rate of sorption of block copolymer polyelectrolytes is significantly lower than the rate of sorption of linear polyions and is controlled by the lengths of the hydrophilic and hydrophobic blocks and the flexibility of the latter blocks. The sorption of amphiphilic block copolymers is accompanied by their self-assembly in the polycomplex gel and formation of a continuous hydrophobic layer impermeable to water and the low-molecular-mass salt dissolved in it.  相似文献

Micellization in dilute solutions of diblock copolymers with a polyelectrolyte and a hydrophilic nonionic blocks and oppositely charged polyions is studied using mean-field theory. In aqueous solutions the micelle core consists of the polyelectrolyte complex (PEC) while the corona is formed by hydrophilic blocks of the block copolymers. Describing PEC as a globule in the framework of the Lifshitz [Zh. Eksp. Teor. Fiz. 55, 2408 (1968)] globule theory we calculate the surface tension of the micellar core/solvent interface as a function of the polyion degree of ionization, solvent quality, and concentration of low-molecular-mass salt. The equilibrium aggregation number of starlike micelles formed by block copolymers and homopolymers of opposite charge at stoichiometric mixture compositions is found as a function of the system parameters. It is shown that micelles disintegrate upon addition of salt.  相似文献

Histidine functional block copolymers are thermally self‐assembled into polymer micelles with poly‐N‐isopropylacrylamide in the core and the histidine functionality in the corona. The thermally induced self‐assemblies are reversible until treated with Cu2+ ions at 50 °C. Upon treatment with 0.5 equivalents of Cu2+ relative to the histidine moieties, metal‐ion coordination locks the self‐assemblies. The self‐assembly behavior of histidine functional block copolymers is explored at different values of pH using DLS and 1H NMR. Metal‐ion coordination locking of the histidine functional micelles is also explored at different pH values, with stable micelles forming at pH 9, observed by DLS and imaged by atomic force microscopy. The thermal self‐assembly of glycine functional block copolymers at pH 5, 7, and 9 is similar to the histidine functional materials; however, the self‐assemblies do not become stable after the addition of Cu2+, indicating that the imidazole plays a crucial role in metal‐ion coordination that locks the micelles. The reversibility of the histidine‐copper complex locking mechanism is demonstrated by the addition of acid to protonate the imidazole and destabilize the polymer self‐assemblies. © 2019 Wiley Periodicals, Inc. A series of ABA amphiphilic triblock copolymers possessing polystyrene (PS) central hydrophobic blocks, one group with “short” PS blocks (DP = 54–86) and one with “long” PS blocks (DP = 183–204) were synthesized by atom transfer radical polymerization. The outer hydrophilic blocks were various lengths of poly(oligoethylene glycol methyl ether) methacrylate, a comb‐like polymer. The critical aggregation concentrations were recorded for certain block copolymer samples and were found to be in the range circa 10−9 mol L−1 for short PS blocks and circa 10−12 mol L−1 for long PS blocks. Dilute aqueous solutions were analyzed by transmission electron microscopy (TEM) and demonstrated that the short PS block copolymers formed spherical micelles and the long PS block copolymers formed predominantly spherical micelles with smaller proportions of cylindrical and Y‐branched cylindrical micelles. Dynamic light scattering analysis results agreed with the TEM observations demonstrating variations in micelle size with PS and POEGMA chain length: the hydrodynamic diameters (DH) of the shorter PS block copolymer micelles increased with increasing POEGMA block lengths while maintaining similar PS micellar core diameters (DC); in contrast the values of DH and DC for the longer PS block copolymer micelles decreased. Surface‐pressure isotherms were recorded for two of the samples and these indicated close packing of a short PS block copolymer at the air–water interface. The aggregate solutions were demonstrated to be stable over a 38‐day period with no change in aggregate size or noticeable precipitation. The cloud point temperatures of certain block copolymer aggregate solutions were measured and found to be in the range 76–93 °C; significantly these were ∼11 °C higher in temperature than those of POEGMA homopolymer samples with similar chain lengths. © 2008 Wiley Periodicals, Inc. A range of poly(ε-caprolactone)/poly(N-vinyl-2-pyrrolidone) amphiphilic block copolymers with well-de ned hydrophilic chain length were synthesized by the living/controlled reversible addition fragmentation chain transfer polymerization method. The composition and struc-ture of the targeted resultants were characterized with 1H NMR, 13C NMR, FT-IR spec-troscopy and gel permeation chromatography. The various block copolymers were success-fully employed to fabricate the spherical micelle with core-shell morphological structure. The poly(N-vinyl-2-pyrrolidone) block-dependent characteristics of the copolymeric micelles were investigated by uorescence spectroscopy, dynamic light scattering, and transmission electron microscopy. The solubilization of the hydrophobic ibuprofen as a model drug in the micelle solution was also explored. It was found that the drug loading contents are related to the micellar morphology structure determined by hydrophilic chain length in the copolymer.  相似文献

Amphiphilic diblock copolymers with various block compositions were synthesized on poly(2‐ethyl‐2‐oxazoline) (PEtOz) as a hydrophilic block and poly(4‐methyl‐ε‐caprolactone) (PMCL) or poly(4‐phenyl‐ε‐caprolactone) (PBCL) as a hydrophobic block. These PEtOz‐b‐PMCL and PEtOz‐b‐PBCL copolymers consisting of soft domains of amorphous PEtOz and PM(B)CL had no melting endothermal peaks but displayed Tg. The lower critical solution temperature (LCST) values for the PEtOz‐b‐PMCL, and the PEtOz‐b‐PBCL aqueous solution were observed to shift to lower temperature than PEtOz homopolymers. Their aqueous solutions were characterized using fluorescence techniques and dynamic light scattering (DLS). The block copolymers formed micelles with critical micelle concentrations (CMCs) in the range 0.6–11.1 mg L?1 in an aqueous phase. As the length of the hydrophobic PMCL or PBCL blocks elongated, lower CMC values were generated. The mean diameters of the micelles were between 127 and 318 nm, with PDI in the range of 0.06–0.21, suggesting nearly monodisperse size distributions. The drug entrapment efficiency and drug‐loading content of micelles depend on block polymer compositions. In vitro cell viability assay showed that PEtOz‐b‐PMCL has low cytotoxicity. Doxorubicin hydrochloride (DOX)‐loaded micelles facilitated human cervical cancer (HeLa) cell uptake of DOX; uptake was completed within 2 h, and DOX was able to reach intracellular compartments and enter the nuclei by endocytosis. © 2013 Wiley Periodicals, Inc. We prepared well‐defined diblock copolymers of thermoresponsive poly(N‐isopropylacrylamide‐coN,N‐dimethylacrylamide) blocks and biodegradable poly(D ,L ‐lactide) blocks by combination of reversible addition‐fragmentation chain transfer radical (RAFT) polymerization and ring‐opening polymerization. α‐Hydroxyl, ω‐dithiobenzoate thermoresponsive polymers were synthesized by RAFT polymerization using hydroxyl RAFT agents. Biodegradable blocks were prepared by ring‐opening polymerization of D ,L ‐lactide initiated by α‐hydroxyl groups of thermoresponsive polymers, which inhibit the thermal decomposition of ω‐dithioester groups. Terminal dithiobenzoate (DTBz) groups of thermoresponsive blocks were easily reduced to thiol groups and reacted with maleimide (Mal). In aqueous media, diblock copolymer products formed surface‐functionalized thermoresponsive micelles. These polymeric micelles had a low critical micelle concentration of 22 μg/L. In thermoresponsive studies of the micelles, hydrophobic DTBz‐surface micelles demonstrated a significant shift in lower critical solution temperature (LCST) to a lower temperature of 30.7 °C than that for Mal‐surface micelles (40.0 °C). In addition, micellar LCST was controlled by changing bulk mixture ratios of respective heterogeneous end‐functional diblock copolymers. Micellar disruption at acidic condition (pH 5.0) was completed within 5 days due to hydrolytic degradation of PLA cores, regardless of showing a slow disruption rate at physiological condition. Furthermore, we successfully improved water‐solubility of hydrophobic drug, paclitaxel by incorporating into the micellar cores. © Wiley Periodicals, Inc. The self-association characteristics of very short and well-defined poly(butyl acrylate)-b-poly(acrylic acid) (PBA-b-PAA) block copolymers in water have been studied. The diblocks are asymmetric with the PBA block longer than the PAA block, giving rise to hollow sphere morphology. This is affirmed by experimental data and theoretical evaluations of the hydrophilic and hydrophobic domain sizes, as well as a value close to 1 for the ratio of the hydrodynamic to the gyration radius of the micelles. Besides, the untypically short PBA blocks (polymerization number around 15) render the micelles dynamic. Indications in support include among others the following: the CMC (critical micellar concentration) values depend, together with the aggregation numbers and the micellar sizes, on the block lengths, as predicted by theory; above the CMC their sizes are concentration-independent, while the micelles disappear below CMC. A comparison was also made with a random PBA-co-PAA copolymer of similar length, which self-associates at an apparent CMC 1 order of magnitude larger than those of the block copolymers, but the size of the formed micelles depends on the concentration.  相似文献

Amphilic triblock copolymers with varying ratios of hydrophilic poly[bis (methoxyethoxyethoxy)phosphazene] (MEEP) and relatively hydrophobic poly(propylene glycol) (PPG) blocks were synthesized via the controlled cationic‐induced living polymerization of a phosphoranimine (Cl3P?NSiMe3) at ambient temperature. A PPG block can function as either a classical hydrophobic block or a less hydrophobic component by varying the nature of a phosphazene block. The aqueous phase behavior of MEEP‐PPG‐MEEP block copolymers was investigated using fluorescence techniques, TEM, and dynamic light scattering (DLS). The critical micelle concentrations (cmcs) of MEEP‐PPG‐MEEP block copolymers were determined to be in the range of 3.7–16.8 mg/L. The mean diameters of MEEP‐PPG‐MEEP polymeric micelles, measured by DLS, were between 31 and 44 nm. Nonionic water-soluble poly(acrylamide)s and poly(acrylate)s were synthesized by RAFT and ATRP methods. Similar to the synthesized poly(N-isopropylacrylamide) and poly(N-acryloylpyrrolidine), aqueous solutions of statistical acrylate copolymers bearing two different oligo(ethylene oxide) side chains showed a sharp clouding transition upon heating beyond characteristic temperatures. The temperature of the cloud point can be easily fine tuned by the copolymer composition. As for poly(N-isopropylacrylamide) and poly(N-acryloylpyrrolidine), the cloud-point temperatures of these statistical copolymers are rather insensitive to changes in the molar mass or the NaCl content of the solutions. Also, ternary triblock copolymers containing one permanently hydrophilic block and two different thermoresponsive blocks were synthesized, varying the block sequence systematically. Their aggregation in aqueous solution was followed by turbidimetry and dynamic light scattering. Depending on the heating process and the triblock sequence, micellar aggregates of 40 to 600 nm size were found. The thermally induced aggregation behavior depends sensitively on the block sequence but is also subject to major kinetic effects. For certain block sequences, a thermally induced two-step association is observed when heating beyond the first and second cloud points of the thermoresponsive blocks. However, the thermal-transition temperatures of the block polymers can differ from the thermal-transition temperatures of the individual homopolymers. This may be caused by end-group effects but also by mutual interactions of the different blocks in solution, as physical mixtures of the homopolymers exhibit deviations from a purely additive thermal behavior.  相似文献
