Zusammenfassung 2,3,6,7,8-Pentachlorimidazo[1,2—b]pyridazin und 3,6,7,8-Tetrachlor-s-triazolo[4,3—b]pyridazin wurden aus unsubstituierten Verbindungen und einigen Chlor-Derivaten synthetisiert. Die Reihenfolge der Substitution von Wasserstoffatomen durch Chloratome konnte beim Imidazo[1,2—b]pyridazin als 3>2, 7>8>6 ermittelt werden, während beis-Triazolo-[4,3—b]pyridazin der Verlauf als 3>8>7>6 sichergestellt wurde. Nukleophile Substitutionen an diesen perchlorierten Systemen verlaufen am leichtesten in Stellung 8.
Pyridazines. XLVIII: Syntheses and Reactivity of 2,3,6,7,8-Pentachloroimidazo[1,2—b]pyridazine and 3,6,7,8-Tetrachloro-s-triazolo[4,3—b]pyridazine
2,3,6,7,8-Pentachloroimidazo[1,2—b]pyridazine and 3,6,7,8-Tetrachloro-s-triazolo[4,3—b]pyridazine were synthesized from the parent compounds and some of their chloro derivatives. It could be established that in the case of the first condensed system the stepwise introduction of chlorine atoms followed the order of 3>2, 7>8>6 and in the case of the second system this appears to be 3>8>7>6. Nucleophilic substitutions on these perchloro compounds proceed most easily at position 8.

The novel ring system, 6H-1,2,4-triazino[4,3—b]1,2,4-triazolo[3,4—f]-pyridazine was prepared either by ring closure of1-(1-ethoxycarbonylethylene)-2-(1,2,4-triazolo[4,3—b]pyridazinyl-6)-hydrazine derivatives (4, 5, 7) in polyphosphoric acid or of a hydrazine15 derived from pyridazino-1,2,4-triazine under the action of triethyl orthoformate. Compound8 showed a positive inotropic effect.

Zusammenfassung Das neue Ringsystem 6H-1,2,4-Triazino[4,3—b]1,2,4-triazolo[3,4—f]-pyridazin wurde entweder durch Ringschluß der1-(1-Ethoxycarbonylethylen)-2-(1,2,4-triazolo[4,3—b]pyridazinyl-6)-hydrazin-Derivate(4, 5, 7) 4, 5, 7 in Polyphosphorsäure, oder aus einem von Pyridazino-1,2,4-triazin abgeleiteten Hydrazin15 durch Ringschluß mit Orthoameisensäuretriethylester hergestellt. Verbindung8 zeigte einen positiven inotropen Effekt.

s-Triazolo[4,3-b]pyridazine (I) reacted with cyclohexene under the influence of ultraviolet light to yield 4a,5,7,8,8a,9-hexahydro-9-methylene-6H-s-triazolo[1,5-a]indole (IV) and 9-cyanomethyl-4a,5,7,8,8a,9-hexahydro-6H-s-triazolo[1,5-a]indole (V). These products were formed by the addition of the alkene to the 1,8 positions of I with a concurrent cleavage of the N4? N5 bond. Similar additions were observed with cyclopentene and 2,3-dimethyl-1,3-butadiene. The isomeric s-triazolo[2,3-b]pyridazine (III) reacted with cyclohexene to form an isomer of IV, 4a,5,7,8,8a,9-hexahydro-9-methylene-6H-s-triazolo[4,3-a]indole (XV) and two [2 + 2] cycloadducts (XVI and XVII).  相似文献

6-Azidotetrazolo[1,5-b]pyridazine (III) has been prepared by two different routes and is easily transformed into the 6-amino derivative (IV). The attempted cyclization of 6-hydrazinotetrazolo[1,5-b] pyridazine (II) into the postulated tricycle has been shown to result in the formation of 6-azido-s-triazolo [4,3-b]pyridazine (VI), obtained also in a separate experiment from VII. The azido structure of VI has been confirmed from spectroscopic data and from its conversion into 6-amino-s-triazolo [4,3-b]pyridazine (VIII), obtained in another experiment from VII. Similarly, cyclization of II with cyanogen bromide resulted in the simultaneous formation of the s-triazolo ring and ring opening of the fused tetrazolo ring, giving IX. For 6-azido-2-phenylimidazo[1,2-b]pyridazine (XII) the expected azide structure proved to be correct.  相似文献

Zusammenfassung Es wird über den Deuteriumaustausch bei einigen Imidazo[1.2-b]-,s-Triazolo[4.3-b]- und Tetrazolo[1.5-b]pyridazines unter neutralen, sauren und basischen Reaktionsbedingungen berichtet. Die Austauschgeschwindigkeiten und Halbwertszeiten wurden durch NMR-spektroskopische Messungen bestimmt.

Hydrogen-deuterium exchange reactions in some imidazo[1.2-b]-,s-Triazolo[4.3-b], and tetrazolo[1.5-b]pyridazines under neutral, acid or base-catalyzed conditions are reported. The exchange rates and half-lives were determined by nmrspectroscopy.

The synthesis of s-triazolo[4,3-b] pyridazine 5-oxides is reported. To our knowledge, they are the first example of N-oxides of heteroaromatic azoloazines with a bridgehead nitrogen. They are easily rearranged, in particular when irradiated with ultraviolet light, into 6-hydroxy-s-triazolo[4,3-b]pyridazines.  相似文献

Zusammenfassung Durch stufenweisen Austausch von Chloratomen im 2,4,6-Trichlor-1,3,5-triazin wurden verschiedene unsymmetrisch substituierte Derivate des 1,3,5-Triazins synthetisiert. Von einigen Hydrazinotriazinen ausgehend gelang die Synthese von substituiertens-Triazolo[4,3-a]-1,3,5-triazinen.
By a successive replacement of the chlorine atoms in 2.4.6-trichloro-1.3.5-triazine it was possible to synthesize different unsymmetrically substituted derivatives of 1.3.5-triazine. Some hydrazino triazines were used as starting material for the synthesis of substituteds-triazolo[4.3-a]-1.3.5-triazines.

Imidazo[1,2-b]pyridazine, s-triazolo[4,3-b]pyridazine and tetrazolo[1,5-b]pyridazine and some derivatives thereof were reduced with sodium borohydride to give the corresponding 5,6,7,8-tetrahydro derivatives. A mechanism for these reductions is proposed and reduction at the C7-C8 bond occurs before the reduction of the C6? N5 bond. Substituents at position 7 and/or 8 cause a significant decrease in the extent of reduction or lead to a 5,6-dihydro derivative by competitive attack at the 6 position.  相似文献

Carbon-13 nmr techniques have been employed to study imidazo[1,2-a]pyridine, imidazo-[ 1,2-b]pyridazine, s-triazolo[4,3-b]pyridazine, and s-triazolo[ 1,5-b]pyridazine and their N-1 quaternized derivatives. The data on quaternized derivatives have been analyzed to determine the tautomeric structures possible in the protonated derivatives. The data suggest that protonation at the bridgehead nitrogen does not occur in these compounds and, of the possible tautomerie forms, the N-1 tautomer predominates.  相似文献

Open-chain C-nucleosides, 1-(s-triazolo[4,3-x]azinyl-3)polyols 13–18 were prepared by one-pot synthesis from hydrazinoazines 20a,c and various D-aldoses 1–6. No protective groups were required for these transformations. 1-(s-Triazolo[4,3-b]pyridazinyl-3)-D-xylo-tetritol (15a) was isolated and characterised in the form of its 4-O-triphenylmethyl derivative 19. Reaction of hydrazinopyridazines 20a,b with methyl 2,3-di-O-acetyl-L-threuronate ( 22 ), followed by treatment with bromine, gave the corresponding (2R,3S)-2,3-diacetoxy-3-(s-triazolo[4,3-b]pyridazinyl-3)propanoic acid methyl esters 24a,b. Acetonisation of 1-(6-chloro-s-triazolo[4,3-b]pyridazinyl-3)-D-gluco-pentitol ( 17a ) gave a mixture of isomeric bis-acetonides 25 and 26 , while acetonisation of 1-(6-chloro-s-triazolo[4,3-b]pyridazinyl-3)-D-manno-pentitol ( 18a ) gave acetonide 27 as a single isomer.  相似文献

Zusammenfassung Es wird über Ringöffnungen von Tetrazolo[1,5—a]pyridinen, Imidazo[1,2—b]pyridazinen, s-Triazolo[4,3—b]pyridazinen und Pyrimido[1,2—b]pyridazinen berichtet. Die Reaktion verläuft stets am Sechserring der Azoloazine und bei den entstandenen monocyclischen Verbindungen kann nachfolgend einecis—trans-Isomerisierung stattfinden, oder es kommt zu einem neuen Ringschluß.

Ring opening reactions of tetrazolo[1.5—a]pyridines, imidazo[1.2—b]pyridazines, s-triazolo[4.3—b]pyridazines and pyrimido[1.2—b]pyridazines are reported. The reaction takes place at the six-membered ring of azoloazines and the formed monocyclic compounds may subsequently isomerize at the double bonds or new cycles may be formed.

Imidazo[1.2-b]-,s-Triazolo[4.3-b]- ands-Triazolo-[2.3-b]pyridazines and their derivatives were submitted to homolytic phenylation and the reaction mixtures obtained separated by chromatography into individual components which were then identified. Homolytic phenylation takes place preferentially at position 8 of all systems investigated, whereas a lower selectivity was observed for positions 7 and 3.  相似文献

The synthesis of 1,2,4-triazolo[4,3-b]pyridazines and the unknown ring system, pyrazino[2,3-d]-1,2,4-triazolo[4,3-b]pyridazine, has been achieved. The preparation of the new tricyclic 1,2,4-triazole was accomplished first by ring closure of the triazole ring followed by formation of the pyrazine ring. Substitution of the pyrazino[2,3-d]-1,2,4-triazolo[4,3-b]pyridazine ring system was carried out in order to provide information of its reactivity and to provide a variety of interesting compounds for biological testing.  相似文献

A one step synthesis of s-triazolo[4,3-b]pyridazine using 3-chloro-6-hydrazinopyridazine and an appropriate thioformimidate is described. Applying this procedure to 5-O-benzoyl-1-benzylthio-1-formimidate-D-ribofuranose, 5′ benzoyl-6-chloro-3β-D-ribofuranosyl-s-triazolo[4,3-b]pyridazine was obtained. Substitutions of chlorine by nucleophilic reagents afforded some derivatives of a new series of C-nucleo-sides. Structural determination including anomeric configuration assignment is discussed based mainly on 1H nmr spectroscopy.  相似文献

s-Triazolo[4,3-b]pyridazine (I) photochemically reacted with cyclooctene, cyclododecene, bicyclo[2.2.2]oct-2-ene, bicyclo[2.2.1]hept-2-ene and indene to form tri and tetracyclic triazoles (II-X). Generally, two or more products were formed. For example the reaction with cyclooctene gave 4a,5,7,8,9,10,10a,11-octahydro-11-methylene-6H-cycloocta[4,5]-pyrrole[1,2-b]-s-triazole (II) and the corresponding 11-cyanomethyl product (III). When I was reacted with open chain alkenes, various isomers of the substituted pyrrolo[1,2-b]triazoles (XI-XV) were formed. Since the reaction was not stereospecific, cycloaddition must be a two step process.  相似文献

Cyclization of N-acyl-N′-(6-chloropyrid-2-yl)hydrazines ( 2a-2e ) with phosphorus oxychloride has produced several 5-chloro-s-triazolo[4,3-a]pyridines ( 3a-3e ). Nucleophilic displacement of the chlorosubstituent of 5-chloro-s-triazolo[4,3-a]pyridine ( 3a ) availed the 5-ethoxy ( 4a ) and 5-thioethoxy ( 4b ) derivatives and di(s-triazolo[4,3-a]pyrid-5-yl)sulfide ( 8 ) while reaction of 5-ethylsulfonyl-s-triazolo[4,3-a]pyridine ( 4d ) with potassium hydroxide yielded the 5-hydroxy/5-one system ( 4c or 6 ). Further reaction of 3a with bromine to give 3-bromo-5-chloro-s-triazolo-[4,3-a]pyridine ( 3g ) has provided the corresponding 3-cyano- and 3-carboxamido-5-chloro-s-triazolo[4,3-a]pyridine derivatives ( 3h and 3i ). Treatment of 6-chloro-2-hydrazinopyridine ( 1 ) with cyanogen bromide has provided 3-amino-5-chloro-s-triazolo[4,3-a]pyridine ( 3f ) which, with bromoacetaldehyde dimethyl acetal, transformed into 7-chloroimidazo[1,2-b]-s-triazolo[4,3-a]-pyridine ( 7 ). Finally, attempts at cyclizing N-oxalyl-N′-(6-chloropyrid-2-yl)hydrazine derivatives ( 2g-2i ) with intentions of preparing various 3-acyl-5-chloro-s-triazolo[4,3-a]pyridines for entry into other 3,5-disubstituted systems were unsuccessful.  相似文献

Many other alkenes, particularly the phenyl ethylenes, did not react with compound 1. An unequivocal synthesis of 5-chloro-7-methyl- (8) and 7-methyl-1,2,4-triazolo[1,5-a]pyrimidine (10) from 2-amino-4-chloro-6-methylpyrimidine (5) through the corresponding amidine6 and formamide oxime7 was developed. It was unambigously shown by comparison of the chemical shifts and the magnitude of coupling constants that the compounds obtained by condensation of 3-amino-1,2,4-triazole (12) and ethyl acetoacetate (13) and some further transformations are isomeric 5-methyl substituted 1,2,4-triazolo[1,5-a]pyrimidines1,9, and11.
Eine eindeutige Synthese einiger substituierter 1,2,4-Triazolo[1,5-a]pyrimidine
Zusammenfassung Es wurde ein eindeutiger Syntheseweg für 5-Chlor-7-methyl- (8) und 7-Methyl-1,2,4-triazolo[1,5-a]pyrimidin (10) ausgehend von 2-Amino-4-chlor-6-methylpyrimidin (5) über das entsprechende Amidin6 und das Formamidoxim7 entwickelt. Durch Vergleich von chemischen Verschiebungen und Kopplungskonstanten konnte eindeutig gezeigt werden, daß die Verbindungen, die bei der Kondensation von 3-Amino-1,2,4-triazol (12) and Ethylacetoacetat (13), sowie einige weitere Transformationsprodukte, isomere 5-Methylsubstituierte 1,2,4-Triazolo[1,5-a]pyrimidine sind (1,9,11).
