A reliable micellar electrokinetic chromatographic method for the determination of oxcarbazepine and its two main metabolites, 10-hydroxycarbamazepine and 10,11-trans-dihydroxy-10,11-dihydroxycarbamazepine, in human plasma was developed. The separation and determination of the analytes was achieved using a system consisting of 60 mM SDS in phosphate buffer (30 mM, pH 8.0), to which 20% (v/v) methanol was added. Separation was carried out in an uncoated fused-silica capillary with a separation voltage of 25 kV and currents typically less than 40 microA. Spectrophotometric detection was at 205 nm. Isolation of oxcarbazepine and its metabolites from plasma was accomplished by a solid-phase extraction procedure. The mean extraction yield of the analytes from plasma was higher than 94%. The linear correlation coefficients were better than 0.994 for all analytes. The limit of detection was 0.05 microg/mL, the limit of quantitation 0.15 microg/mL. The repeatability for the spiked blank plasma samples was lower than 1.9% and the intermediate precision lower than 2.1%, both expressed as RSD%. The results obtained analysing real plasma samples from epileptic patients under therapy with Tolep were satisfactory in terms of precision, accuracy and detectability.  相似文献

A rapid, sensitive and simple-to-operate high performance liquid chromatographic method for the simulataneous determination of oxcarbazepine, 10-hydroxycarbazepine and 10,11-dihydro-10,11-trans-dihydroxy-carbamazepine in plasma is described. The drug and its metabolites were extracted from plasma using commercially available reversed phase octadecylsilane bonded-silica columns (Bond Elut C18, 1 mL capacity). Chromatographic separation of oxcarbazepine and its metabolites was achieved using a mobile phase consisting of acetonitrile/methanol/water (13:25:62 by volume) at a flow rate of 1.2 mL/min in conjunction with a Waters Associates Nova-Pak C18 column. The analytical column, in Radial-Pak cartridge form, was used in combination with a LiChrospher 5 microns C18 guard column. By measuring the UV absorbance at 214 nm, plasma levels in the region of 50-100 ng/mL for the drug and its metabolites can be detected with only 100 microL of plasma. The method has been applied to pharmacokinetic studies of oxcarbazepine and its metabolites in children with epilepsy; preliminary pharmacokinetic findings in two patients at steady-state are presented.  相似文献

A new chelating polymeric sorbent is developed using Merrifield chloromethylated resin anchored with calix[4]arene-o-vanillinsemicarbazone for simultaneous separation and solid phase extractive preconcentration of U(VI) and Th(IV). The “upper-rim” functionalized calix[4]arene-o-vanillinsemicarbazone was covalently linked to Merrifield resin and characterized by FT-IR and elemental analysis. The synthesized chelating polymeric sorbent shows superior binding affinity towards U(VI) and Th(IV) under selective pH conditions. Various physico-chemical parameters that influence the quantitative extraction of metal ions were optimized. The optimum pH range and flow rates for U(VI) and Th(IV) were 6.0-7.0 and 1.0-4.0 ml min−1 and 3.5-4.5 and 1.5-4.0 ml min−1, respectively. The total sorption capacity found for U(VI) and Th(IV) was 48734 and 41175 μg g−1, respectively. Interference studies carried out in the presence of diverse ions and electrolyte species showed quantitative analyte recovery (98-98.5%) with lower limits of detection, 6.14 and 4.29 μg l−1 and high preconcentration factors, 143 and 153 for U(VI) and Th(IV), respectively. The uptake and stripping of these metal ions on the resin were fast, indicating a better accessibility of the metal ions towards the chelating sites. The analytical applicability of the synthesized polymeric sorbent was tested with some synthetic mixtures for the separation of U(VI) and Th(IV) from each other and also from La(III), Cu(II) and Pb(II) by varying the pH and sequential acidic elution. The validity of the proposed method was checked by analyzing these metal ions in natural water samples, monazite sand and standard geological materials.  相似文献

A liquid chromatographic method for the simultaneous analysis of marker pteridins and biopterin reduced forms, in urine samples is proposed. A Zorbax Eclipse XDB-C18 column was used for the chromatographic separation, using a 98/2 (v/v), citrate buffer (pH 5.5)-acetonitrile mobile phase, in isocratic mode. A post-column photoderivatization was carried out with an on-line photoreactor, located between a diode array detector (DAD) and a fast scanning fluorescence detector (FSFD). Neopterin (NEO), biopterin (BIO), pterin (PT) and dihydrobiopterin (BH2) were determined by measuring native fluorescence, using the photoreactor in OFF-mode, and tetrahydrobiopterin (BH4) was determined by measuring of the induced fluorescence of the generated photoproducts, using the photoreactor in ON-mode. In addition, Creatinine (CREA), as a reference of metabolites excrection in urine, was simultaneously determined using the DAD detector. Detection limits were 0.2, 13.0, 0.3, 0.3 and 3.5 ng mL−1, for NEO, BH2, BIO, PT and BH4, respectively, and 0.4 μg mL−1 for CREA. Ratio values for NEO/CREA, PT/CREA, BH4/CREA, BH2/CREA, NEO/BIO and BIOtotal/CREA, in urine samples, of healthy children and adults, phenylketonuric children and infected mononucleosis children, are reported. A comparative study, about the mean values obtained for each of the compounds, by the present procedure and by the classical iodine oxidation method (Fukushimás method), has been performed, in urine samples belonging to healthy volunteers. The values obtained were BH4/CREA: 0.41, BH2/CREA: 0.31 and BIOtotal/CREA: 0.73, by the proposed method, and BH4/CREA: 0.35, BH2/CREA: 0.20 and BIOtotal/CREA: 0.48, by iodine oxidation method.  相似文献

A mass spectrometric (MS) method for the identification of iron protoporphyrin (IX) (FePTP, heme b) in marine particulate material and phytoplankton is described. Electrospray ionisation of FePTP produced the molecular Fe(III)PTP+ ion (m/z = 616) or the pseudomolecular [Fe(II)PTP + H]+ ion (m/= 617), depending on the oxidation state of the central iron ion. Collision induced dissociation (CID) in the ion trap mass spectrometer resulted in a single detected product ion (m/z = 557) indicative of loss of ethanoic acid from a carboxylic acid side chain. Widening the isolation width to 616 ± 3 resulted in production of a mass spectrum demonstrating the distinctive isotopic ratio of the iron containing fragment, further increasing the specificity of the analysis. Selective reactant monitoring (SRM) of the fragment ion (m/z = 557) was applied to the detection of FePTP after chromatography of ammoniacal OGP extracts of marine samples. The detection limit for FePTP analysed by SRM after chromatography was 1.2 ± 0.5 fmol. For phytoplankton samples, reasonably good agreement was achieved between results obtained with SRM and those obtained by monitoring absorbance at λ = 400 nm using a diode array detector (DAD). Use of SRM for analysis of particulate material obtained from the high latitude North Atlantic allowed for the analysis of FePTP in the presence of a co-eluting compound that interfered with detection by DAD. Simultaneous collection of mass spectra from m/z = 300 to 1500 resulted in identification of the pseudomolecular ion for the interfering compound. The CID fragmentation pattern and UV–visible mass spectra indicated that the interfering compound was a previously unidentified chlorin type compound. Comparison of FePTP determined by SRM and DAD on samples where this compound could not be detected showed that results collected using the two methods correlated. The use of both MS and DAD results in a powerful tool for quantifying this important biogenic component of the particulate iron pool.  相似文献

A sensitive and robust high-performance liquid chromatography-electrospray ionization tandem mass spectrometry method to analyze 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) and its five metabolites in one passage was developed and validated. The method achieved excellent reproducibility and accuracy. Linearity was observed for all six compounds (R2 = 0.999) with detection limits (S/N ≥ 3) ranging from 0.2 to 2.4 pg on column and 0.01-0.12 ng ml−1 in samples injected. Average intra-day and inter-day variations (% R.S.D.) were 1.2 and 3.5%, respectively. A sample preparation method involving C8 and C18 solid phase extraction provided satisfactory recovery of the analytes in mouse urine. Each NNK metabolite was identified by its chromatographic retention time and specific fragmentation pattern. Since the carcinogenicity of NNK is related to its metabolism, the method described in this report should facilitate toxicological investigations into the carcinogenesis due to NNK exposure in the environment.  相似文献

A method is reported for the quantification of isoorientin (using a standard addition method) and total flavonoids (expressed as rutin, using the external standard method) in passion fruit pulp (Passiflora edulis Sims f. flavicarpa Degener, Passifloraceae). Extraction of flavonoids was optimized by experimental design methodology, and quantitative analysis was performed by high-performance liquid chromatography with photo-diode array detection (HPLC-UV/DAD). The method was developed and validated according to ICH requirements for specificity, linearity, accuracy, precision (repeatability and intermediate precision), LOD and LOQ. Rutin was chosen as standard for the quantification of total flavonoids in order to propose a HPLC method feasible for routine analysis of the flavonoids in the passion fruit pulp. The passion fruit pulp contained 16.226 ± 0.050 mg L− 1 of isoorientin and 158.037 ± 0.602 mg L− 1 of total flavonoid, suggesting that P. edulis fruits may be comparable with other flavonoid food sources such as orange juice or sugarcane juice.  相似文献

In this work, a new stir cake sorptive extraction (SCSE) using polymeric ionic liquid monolith as sorbent was prepared. The sorbent was obtained by in situ copolymerization of an ionic liquid, 1-allyl-3-methylimidazolium bis[(trifluoro methyl)sulfonyl]imide (AMII) and divinylbenzene (DB) in the presence of N,N-dimethylformamide. The influence of the content of ionic liquid and the porogen in the polymerization mixture on extraction performance was studied thoroughly. The physicochemical properties of the polymeric ionic liquid were characterized by infrared spectroscopy, elemental analysis, scanning electron microscopy and mercury intrusion porosimetry. The usefulness of SCSE–AMIIDB was demonstrated by the enrichment of trace benzimidazole anthelmintics. Several parameters affecting the extraction efficiency were investigated, and under the optimized conditions, a simple and effective method for the determination of trace benzimidazoles residues in water, milk and honey samples was established by coupling SCSE–AMIIDB with high performance liquid chromatography/diode array detection (SCSE–AMIIDB–HPLC/DAD). Results indicated that the limits of detection (S/N = 3) for target compounds were 0.020–0.072 μg L−1, 0.035–0.10 μg L−1 and 0.026–0.076 μg L−1 in water, milk and honey samples, respectively. In addition, an acceptable reproducibility was achieved by evaluating the repeatability and intermediate precision with relative standard deviations (RSD) of less than 9% and 11%, respectively. Finally, the established AMII–SCSE–HPLC/DAD method was successfully applied for the determination of benzimidazoles residues in milk, honey and environmental water samples. Recoveries obtained for the determination of benzimidazole anthelmintics in spiking samples ranged from 70.2% to 117.6%, with RSD below 12% in all cases.  相似文献

A rapid and sensitive ultraperformance liquid chromatography tandem mass spectrometry assay was developed for the simultaneous analysis of oxcarbazepine and its main metabolite in human plasma. The assay involves a simple solid‐phase extraction procedure of 0.3 mL of human plasma and analysis was performed on a triple‐quadrupole tandem mass spectrometer in multiple reaction monitoring mode via electrospray ionization. Separation was achieved on an Acquity UPLC™ BEH C18 column (50 × 2.1 mm, i.d., 1.7 µm) with isocratic elution at a flow‐rate of 0.25 mL/min and imipramine was used as the internal standard. The standard calibration curve was linear over the range 9.580–5070.205 ng/mL for oxcarbazepine (OXC) and 19.444–10290.800 ng/mL for 10,11‐dihydro‐10‐hydroxycarbamazepine (MHD), expressed by the linear correlation coefficient r2, which was better than 0.995 for OXC and MHD. The intra‐ and inter‐day precision and accuracy of the quality control samples were within 10.0%. The recoveries were 81.0, 89.6 and 66.6% for OXC, MHD and imipramine, respectively. The total run time was 1.5 min only for each sample, which makes it possible to analyze more than 350 samples per day. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

A novel sorbent was prepared by the functionalization of an inorganic support material, MCM-41, with N-methylglucamine for the uptake of boron from aqueous solutions prior to its determination by inductively coupled plasma optical emission spectrometry (ICP-OES). Characterization of the newly synthesized material was performed using BET, XRD, TEM, SEM and DRIFTS techniques, in addition to its C and N elemental content. Sorption behavior of the novel sorbent for boron was also investigated and found to obey Freundlich and Dubinin-Radushkevich (D-R) isotherm models. The maximum amount of B (as H3BO3) that can be sorbed by the sorbent was calculated from the D-R isotherm and was found to be 0.8 mmol B g−1 of sorbent. The applicability of the new sorbent for the removal/preconcentration of boron from aqueous samples was examined by batch method. It was found that the sorbent can take up 85% of boron in 5 min whereas quantitative sorption is obtained in 30 min. Any pH greater than 6 can be used for sorption. Desorption from the sorbent was carried out using 1.0 M HNO3. The sorption efficiency of the new sorbent was also compared to that of Amberlite IRA 743, a commercial resin with N-methylglucamine functional groups. Within the experimental conditions employed, the new sorbent was found to have higher sorption efficiency than the commercial resin. For method validation, spike recovery tests were performed at various concentration levels in different water types and were found to be between 83-95 and 75-92% for ultra pure water and geothermal water, respectively.  相似文献

A novel method has been developed for simultaneous analysis for three pairs of trans- and cis-isomers of 2-glucosyloxycinnamic acids, along with their biogenic metabolites (three coumarin derivatives including scopoletin, scoparone and ayapin) in a Chinese medicinal herb Dendrobium thyrsiflorum by using high-performance liquid chromatography (HPLC)-photodiode array detection (DAD)-electrospray ionization (ESI)-tandem mass spectrometry (MS). The method was carried out by using a Polaris C18 column with a gradient solvent system of 0.5% acetic acid aqueous solution-acetonitrile. Seven target analytes including isodensifloside, isothyrsifloside, densifloside, thyrsifloside, scoparone, ayapin and scopoletin were exclusively identified by comparing their retention behaviors, UV and MS spectra with the authentic standards, and their contents in D. thyrsiflorum were simultaneous determined by employing UV detection at 342 nm. In addition, another pair of isomers of 2-glucosyloxycinnamic acids was putatively elucidated mainly based on the MS fragmentation. The method was validated and found to be satisfactorily linear, selective and robust. Recoveries ranged from 95.56 to 97.94% for all compounds at three different spiking levels. The limits of detection (LOD) and quantitation (LOQ) ranged, respectively, from 0.02 to 0.13 μg mL−1 and 0.07 to 0.39 μg mL−1 depending on various compounds. The established quality evaluation method was successfully used for evaluating the quality of D. thyrsiflorum samples of different organs and collections.  相似文献

A high-performance liquid chromatographic method has been developed for the determination in human plasma of the specific serotonin reuptake inhibitor (SSRI) antidepressant paroxetine and its three main metabolites (M1, M2, M3). Fluorescence detection was used, exciting at λ = 294 nm and monitoring emission at λ = 330 nm for paroxetine (λexc = 280 nm, λem = 330 nm for M1 and M2; λexc = 268 nm, λem = 290 nm for M3). Separation was obtained on a reversed-phase C18 column using a mobile phase composed of 66.7% aqueous phosphate at pH 2.5 and 33.3% acetonitrile. Imipramine (λexc = 252 nm, λem = 390 nm) was used as the internal standard. A careful pre-treatment of plasma samples was developed, using solid-phase extraction with C8 cartridges (50 mg, 1 mL). The calibration curves were linear over a working range of 2.5-100 ng mL−1 for paroxetine and of 5-100 ng mL−1 for all metabolites. The limit of detection (LOD) was 1.2 ng mL−1 for PRX and 2.0 ng mL−1 for the metabolites. The method was applied with success to plasma samples from depressed patients undergoing treatment with paroxetine. Hence, the method seems to be suitable for the therapeutic drug monitoring of paroxetine and its main metabolites in depressed patients’ plasma.  相似文献

A reliable, sensitive, rapid and environmentally friendly analysis procedure for the simultaneous determination of the analytes with a wide range of polarity in the environmental water was developed by coupling dispersive magnetic solid-phase extraction (d-MSPE) with high-performance liquid chromatography (HPLC)–diode array detector (DAD) and ultra-high pressure liquid chromatography (UHPLC)-triple quadrupole mass spectrometer (MS/MS), in this work. Magnetic ionic liquid modified multi-walled carbon nanotubes (m-IL-MWCNTs) were prepared by spontaneous assembly of magnetic nanoparticles and imidazolium-modified carbon nanotubes, and used as the sorbent of d-MSPE to simultaneously extract aryloxyphenoxy-propionate herbicides (AOPPs) and their polar acid metabolites due to the excellent π–π electron donor–acceptor interactions and anion exchange ability. The factors, including the amount of sorbent, pH of the sample solution, extraction time and the volume of elution solvent were investigated. Under the optimized conditions, the proposed d-MSPE coupling to HPLC–DAD system had a satisfactory performance, the limits of detection (LODs, defined as the signal to noise ratio of 3) and the limits of quantification (LOQs, defined as the signal to noise ratio of 10) for analytes in Milli-Q water were in the range of 2.8–14.3 and 9.8–43.2 μg L−1 respectively. Calibration curves were linear (r2 > 0.998) over the concentration range from 0.02 to 1 mg L−1. The recoveries of the eight analytes ranged from 66.1 to 89.6% with the RSDs less than 8.6%. In order to extend the method in extremely low concentration analysis, d-MSPE-UHPLC–MS/MS was investigated, which showed better performance in terms of limit of detection and analysis time.  相似文献

A new method that utilizes 1-(2-aminoethyl)-3-phenylurea-modified silica gel as a solid-phase extractant has been developed for preconcentration of trace Sc(III) prior to the measurement by inductively coupled plasma atomic emission spectrometry (ICP-AES). Experimental conditions for effective adsorption of trace level of Sc(III) were optimized using batch and column procedures in detail. The optimum pH value for the separation of Sc(III) on the new sorbent was 4 and complete elution of Sc(III) from the sorbent surface was carried out using 1.0 mL of 0.1 mol L−1 HCl. Common coexisting ions did not interfere with the separation and determination of the analyte. The maximum static adsorption capacity of the sorbent at optimum conditions was found to be 32.5 mg g−1 while the time of 95% adsorption was less than 2 min. The detection limit of present method was found to be 0.091 μg g−1, and the relative standard deviation (RSD) was lower than 3.0% (n = 8). The method was successfully applied for the preconcentration of trace Sc(III) in the environmental samples with satisfactory results.  相似文献

Continuous flow injection and UV spectrophotometric detection have been proposed for simultaneous determination of the two binary mixtures, butylated hydroxytoluene (BHT)/n-propyl gallate (n-PG) and butylated hydroxytoluene (BHT)/butylated hydroxyanisole (BHA), in food and cosmetics samples. The method is based on the different residence times of each antioxidant when the flow cell is packed to a height of 25 mm with silica C18 using methanol-water 50:50% (v/v) as a carrier with a flow rate of 1.25 and 1.10 ml min−1, respectively. The determination of each antioxidant is based on the measurement of its absorbance at its maximum wavelengths using a DAD detector at 30 and 180 s for the mixture n-PG-BHT and 90 and 220 s for BHA-BHT. Calibration graphs were linear over the range 10.0-300.0 μg ml−1 for each antioxidant in both mixtures. The relative standard deviations were 2.5% for BHT and 2.0% for the co-existing antioxidant. Resolution of the n-PG-BHT mixture in ratios between 1:8 and 8:1 and the BHA-BHT mixture in ratios between 1:10 and 10:1 is possible. The method was applied to the determination of both antioxidants in fat foods and cosmetics samples with recoveries ranging between 101 and 105%.  相似文献

This work presents a new methodology to quantify ethyl carbamate (EC) in fortified wines. The presented approach combines the microextraction by packed sorbent (MEPS), using a hand-held automated analytical syringe, with one-dimensional gas chromatography coupled with mass spectrometry detection (GC–MS). The performance of different MEPS sorbent materials was tested, namely SIL, C2, C8, C18, and M1. Also, several extraction solvents and the matrix effect were evaluated. Experimental data showed that C8 and dichloromethane were the best sorbent/solvent pair to extract EC. Concerning solvent and sample volumes optimization used in MEPS extraction an experimental design (DoE) was carried out. The best extraction yield was achieved passing 300 μL of sample and 100 μL of dichloromethane. The method validation was performed using a matrix-matched calibration using both sweet and dry fortified wines, to minimize the matrix effect. The proposed methodology presented good linearity (R2 = 0.9999) and high sensitivity, with quite low limits of detection (LOD) and quantification (LOQ), 1.5 μg L−1 and 4.5 μg L−1, respectively. The recoveries varied between 97% and 106%, while the method precision (repeatability and reproducibility) was lower than 7%. The applicability of the methodology was confirmed through the analysis of 16 fortified wines, with values ranging between 7.3 and 206 μg L−1. All chromatograms showed good peak resolution, confirming its selectivity. The developed MEPS/GC–MS methodology arises as an important tool to quantify EC in fortified wines, combining efficiency and effectiveness, with simpler, faster and affordable analytical procedures that provide great sensitivity without using sophisticated and expensive equipment.  相似文献

A novel adsorbent of thiacalix[4]arene tetracarboxylate derivative modified mesoporous TiO2 was prepared and was used as a packing material for flow injection (FI) micro-column (20 mm × 4.0 mm i.d.) separation/preconcentration on-line coupled to inductively coupled plasma optical emission spectrometry (ICP-OES) simultaneous determination of trace metals (V, Cu, Pb, Cr) in environmental water samples. The experimental conditions for modified mesoporous TiO2 packed micro-column separation/preconcentration of the target metals were optimized and the interference of commonly coexisting ions was examined. The adsorption capacities of thiacalix[4]arene tetracarboxylate derivative modified mesoporous TiO2 for V, Cu, Pb and Cr were found to be 14.0, 11.7, 17.7 and 14.5 mg g− 1, respectively. The detection limits of the method were 0.09, 0.23, 0.50 and 0.15 µg L− 1 for V, Cu, Pb and Cr, respectively, with a preconcentration factor of 20. The precision of this method were 1.7% (V), 3.9% (Cu), 4.6% (Pb) and 2.9% (Cr) (n = 7, C = 5 µg L− 1), respectively. The developed method was applied to the determination of trace heavy metals in real samples and the recoveries for spiked samples were found to be in the range of 88.7-107.1%. For validation, a certified reference material of GSBZ50009-88 environmental water sample was analyzed and the determined values were in good agreement with the certified values.  相似文献

New S-thiocarbamates possessing a 10,11-dihydro-5H-dibenzo[a,d]cycloheptene moiety were obtained unexpectedly in the process of preparing new O-thiocarbamates starting from 5-dibenzosuberol (10,11-dihydro-5H-dibenzo[a,d]cyclohepten-5-ol) and aroyl-isothiocyanates. These compounds were obtained via a benzylic Newman–Kwart rearrangement mechanism. The structures of the new thiocarbamates were confirmed by elemental analysis, IR, and NMR spectroscopy and by X-ray crystallography.  相似文献

(−)-Xanthorrhizol (1) isolated from the rhizomes of Curcuma xanthorrhiza has been transformed to several bisabolane-type sesquiterpenoids, in a stereoselective manner. 10R- and 10S-10,11-dihydro-10,11-dihydroxyxanthorrhizols (2, 3), (−)-curcuquinone (4), (−)-curcuhydroquinone (5), helibisabonol A (7) and allylic alcohol 8 have been prepared from xanthorrhizol in optically active forms. A novel flow injection analysis (FIA) system based on liquid-liquid microextraction and fluorimetric determination was developed for the determination of traces of the Zn2+ ion using 5-(8-hydroxy-2-quinolinylmethyl)-2,8-dithia-5-aza-2,6-pyridinophane (L) as a sensitive and selective fluorimetric sensor, with λex = 373 nm and λem = 530 nm, and hexanol as the extracting organic solvent. In the designed FIA system, the phase separation takes place via gravitation forces in the absence of any segmenter. The influence of pH and ionic strength of the solution, amount of ligand, nature of counter ion, volume of organic solvent, extraction time and coil length was investigated. Under optimized experimental conditions, the calibration curve found to be liner over a concentration range of 0.025-4.53 μg mL−1 (R2 = 0.9951) with a limit of detection of 2.3 ng mL−1. The enrichment factor was 45 and relative standard deviation for 7 replicate determinations was 2.43%. The method is very fast and uses low levels of organic solvents. The proposed method was applied successfully to the determination of zinc(II) in human hair, human serum and two inorganic sludge samples.  相似文献
