The synthesis and characterization of three heteronuclear Pt(2)Pd(2) (4, 5) and PtPd(2) (6) complexes of the model nucleobase 9-methyladenine (9-MeA) is reported. The compounds were prepared by reacting [Pt(NH(3))(3)(9-MeA-N7)](ClO(4))(2) (1) with [Pd(en)(H(2)O)(2)](ClO(4))(2) at different ratios r between Pt and Pd, with the goal to probe Pd(II) binding to any of the three available nitrogen atoms, N1, N3, N6 or combinations thereof. Pd(II) coordination occurs at N1 and at the deprotonated N6 positions, yet not at N3. 4 and 5 are isomers of [{(en)Pd}(2){N1,N6-9-MeA(-)-N7)Pt(NH(3))(3)}(2)](ClO(4))(6)·nH(2)O, with a head-head orientation of the two bridging 9-MeA(-) ligands in 4 and a head-tail orientation in 5. 6 is [{(en)Pd}(2)(OH)(N1,N6-9MeA(-)-N7)Pt(NH(3))(3)](ClO(4))(4)·4H(2)O, hence a condensation product between [Pt(NH(3))(3)(9-MeA-N7)](2+) and a μ-OH bridged dinuclear (en)Pd-OH-Pd(en) unit, which connects the N1 and N6 positions of 9-MeA(-) in an intramolecular fashion. 4 and 5, which slowly interconvert in aqueous solution, display distinct structural differences such as significantly different intramolecular Pd···Pd contacts (3.124 0(16) ? in 4; 2.986 6(14) ? in 5), among others. Binding of (en)Pd(II) to the exocyclic N6 atom in 4 and 5 is accompanied by a large movement of Pd(II) out of the 9-MeA(-) plane and a trend to a further shortening of the C6-N6 bond as compared to free 9-MeA. The packing patterns of 4 and 5 reveal substantial anion-π interactions.  相似文献

The (15)N-labeled diammine(mu-oxo)ruthenium complex cis,cis-[(bpy)(2)(H(3)(15)N)Ru(III)ORu(III)((15)NH(3))(bpy)(2)](4+) ((2-(15)N)(4+)) was synthesized from cis,cis-[(bpy)(2)(H(2)O)Ru(III)ORu(III)(H(2)O)(bpy)(2)](4+) by using ((15)NH(4))(2)SO(4) and isolated as its perchlorate salt in 17% yield. A 1:1 mixture of (2-(15)N)(4+) and nonlabeled cis,cis-[(bpy)(2)(H(3)(14)N)Ru(III)ORu(III)((14)NH(3))(bpy)(2)](4+) were electrochemically oxidized in aqueous solution. The gaseous products (14)N(2) and (15)N(2) were formed in equimolar amounts with only a small amount of (14)N(15)N detected. This demonstrates that dinitrogen formation by oxidation of the diammine complex proceeds by intramolecular N---N coupling.  相似文献

The dinuclear hydroxo complex cis-[L(2)Pt(mu-OH)](2)(NO(3))(2) (L = PMePh(2), 1), in CH(2)Cl(2), CH(3)CN, or DMF solution, deprotonates the NH(2) group of 9-methyladenine (9-MeAd) to give the complex cis-[L(2)Pt[9-MeAd(-H)]](3)(NO(3))(3), 2, which was isolated in good yield. The X-ray structure shows that the nucleobase binds symmetrically the metal centers through the N(1),N(6) atoms forming a cyclic trimer with Pt...Pt distances in the range 5.202(1)-5.382(1) A. Dissolution of 2 in DMSO or DMF determines the partial (or total) dissociation of the cyclic structure to form several fragments. A multinuclear NMR analysis of the resulting mixture supports the presence of the mononuclear species cis-[L(2)Pt[9-MeAd(-H)]](+), 3, in which the deprotonated nucleobase chelates the metal center with the N(6),N(7) atoms. Addition of a stoichiometric amount of the nitrato complex cis-[L(2)Pt(ONO(2))(2)] (L = PMePh(2), 4) to a DMSO or DMF solution of 2 affords quantitatively the diplatinated compound cis-[L(2)Pt(ONO(2))[9-MeAd(-H)]PtL(2)](NO(3))(2), 5. The single-crystal X-ray analysis shows that the adenine behaves as a tridentate ligand bridging two cis-L(2)Pt units at the N(1) and N(6),N(7) sites, respectively [Pt(1)-N(1) = 2.109(5) A, Pt(2)-N(6) = 2.095(7) A, Pt(2)-N(7) = 2.126(7) A]. The N(1)-bonded metal center completes the coordination sphere through an oxygen atom of a nitrate group, and its coordination plane is arranged orthogonally with respect the second one. The Pt-O distance [2.109(5) A] is similar to those found in the nitrato complex 4 [2.110 A, average]. The related complex cis-[[L(2)Pt(ONO(2))](2)(9-MeAd)](NO(3))(2), 6, containing the neutral adenine platinated at the N(1),N(7) atoms, was isolated and its stability in solution investigated by NMR spectroscopy. In DMSO, 6 undergoes decomposition forming a mixture of the species 4, 5, and the adenine mono- and bis-adducts cis-[L(2)Pt(9-MeAd)(DMSO)](2+), 7, and cis-[L(2)Pt(9-MeAd)(2)](2+), 8, respectively. This last complex, quantitatively formed upon addition of 9-MeAd (Pt/adenine = 1:2) to the mixture, was also isolated and characterized.  相似文献

Treatment of the bridging bidentate 1,Z-bis(aminopropyl)-1,Z-dicarba-closo-dodecaborane(12)(1,Z-bis(aminopropyl)-1,Z-carborane) ligands of the type 1,Z-[H(2)N(CH(2))(3)](2)-1,Z-C(2)B(10)H(10)(L(1), Z= 7, 5) or (L(2), Z= 12, 6) with two equivalents of trans-[PtClI(2)(NH(3))](-), followed by halogen ligand metathesis with AgOTf and HCl((aq)) afforded the novel diplatinum(II)-amine species cis-[[PtCl(2)(NH(3))](2)L(n)](7(n= 1) or 8(n= 2), respectively). Similarly, the reaction of L(1) or L(2) with the labile trans-[PtCl(dmf)(NH(3))(2)](+) afforded trans-[[PtCl(NH(3))(2)](2)L(n)](OTf)(2)(9(n= 1) or 10(n= 2), respectively) in good yield and purity. However, isolation of the analogous 1,2-carborane complexes was not possible owing to decomposition reactions that led to extensive degradation of the carborane cage and reduction of the metal centre. The mixed dinuclear complex [cis-[PtCl(2)(NH(3))]-L(1)-trans-[PtCl(NH(3))(2)]]OTf (19) was prepared by treatment of the Boc-protected amine ligand 1-[(Boc)(2)N(CH(2))(3)]-7-[H(2)N(CH(2))(3)]-1,7-C(2)B(10)H(10)(L(3), 15) with trans-[PtCl(dmf)(NH(3))(2)](+) to yield trans-[PtCl(NH(3))(2)L(3)]OTf (16), followed by acid deprotection of the pendant amine group, complexation with trans-[PtClI(2)(NH(3))](-), and halogen ligand metathesis using AgOTf and HCl((aq)). A novel trinuclear species containing 5 was prepared by the addition of two equivalents of 15 to the labile precursor cis-[Pt(dmf)(2)(NH(3))(2)](2+) followed by acid deprotection of the pendant amine groups. Further complexation with two equivalents of trans-[PtClI(2)(NH(3))](-) followed by halogen ligand metathesis using AgOTf and HCl((aq)) afforded the triplatinum(II)-amine species [cis-[Pt(NH(3))(2)(L(1))(2)]-cis-[PtCl(2)(NH(3))](2)](OTf)(2)(23). Complexes 7-10, 19 and 23 represent the first examples of multinuclear platinum(ii)-amine derivatives containing carborane cages. Preliminary in vitro cytotoxicity studies for selected complexes are also reported.  相似文献

The reaction of cis-[Ru(NO)(CH(3)CN)(bpy)(2)](3+) (bpy = 2,2'-bipyridine) in H(2)O at room temperature proceeded to afford two new nitrosylruthenium complexes. These complexes have been identified as nitrosylruthenium complexes containing the N-bound methylcarboxyimidato ligand, cis-[Ru(NO)(NH=C(O)CH(3))(bpy)(2)](2+), and methylcarboxyimido acid ligand, cis-[Ru(NO)(NH=C(OH)CH(3))(bpy)(2)](3+), formed by an electrophilic reaction at the nitrile carbon of the acetonitrile coordinated to the ruthenium ion. The X-ray structure analysis on a single crystal obtained from CH(3)CN-H(2)O solution of cis-[Ru(NO)(NH=C(O)CH(3))(bpy)(2)](PF(6))(3) has been performed: C(22)H(20.5)N(6)O(2)P(2.5)F(15)Ru, orthorhombic, Pccn, a = 15.966(1) A, b = 31.839(1) A, c = 11.707(1) A, V = 5950.8(4) A(3), and Z = 8. The structural results revealed that the single crystal consisted of 1:1 mixture of cis-[Ru(NO)(NH=C(O)CH(3))(bpy)(2)](2+) and cis-[Ru(NO)(NH=C(OH)CH(3))(bpy)(2)](3+) and the structural formula of this single crystal was thus [Ru(NO)(NH=C(OH(0.5))CH(3))(bpy)(2)](PF(6))(2.5). The reaction of cis-[Ru(NO)(CH(3)CN)(bpy)(2)](3+) in dry CH(3)OH-CH(3)CN at room temperature afforded a nitrosylruthenium complex containing the methyl methylcarboxyimidate ligand, cis-[Ru(NO)(NH=C(OCH(3))CH(3))(bpy)(2)](3+). The structure has been determined by X-ray structure analysis: C(25)H(29)N(8)O(18)Cl(3)Ru, monoclinic, P2(1)/c, a = 13.129(1) A, b = 17.053(1) A, c = 15.711(1) A, beta = 90.876(5) degrees, V = 3517.3(4) A(3), and Z = 4.  相似文献

The mechanism of the formation of dinuclear platinum(II) mu-hydroxo complexes from cisplatin hydrolysis products, their interconversion, decomposition, and reactions with biomolecules has been explored using a combined DFT/CDM approach. All activation barriers for the formation of [cis-{Pt(NH(3))(2)(X)}-(mu-OH)-cis-{Pt(NH(3))(2)(Y)}](n)()(+) (X, Y = Cl, OH(2), OH) via nucleophilic attack of a hydroxo complex on an aqua complex are lower than the activation barriers for cisplatin hydrolysis. Considering therapeutic Pt(II) concentrations in tumors, however, only the reaction between two molecules of cis-[Pt(NH(3))(2)(OH(2))(OH)](+) (E) yielding [cis-{Pt(NH(3))(2)(OH(2))}-(mu-OH)-cis-{Pt(NH(3))(2)(OH)}](2+) (5) remains kinetically superior to cisplatin hydrolysis. 5 is strongly stabilized by intramolecular hydrogen bonding between the terminal aqua and hydroxo ligands, resulting in an unusually high pK(a) of 5 and a low pK(a) of its conjugate acid. Unimolecular cyclization of 5 yields the dimers [cis-{Pt(NH(3))(2)}(mu-OH)](2)(2+) (7a with antiperiplanar OH groups and 7b with synperiplanar OH groups). The electronic structure of several diplatinum(II) complexes has been analyzed to clarify whether there are metal-metal interactions. The overall reactivity to guanine (Gua) and dimethyl sulfide (Met, representing the thioether functional group of methionine) increases in the order 5 < 7a approximately 7b < mononuclear complexes, whereas the kinetic selectivity to Gua relative to Met increases in the order 7a approximately 5 < 7b approximately monocationic mononuclear complexes < dicationic mononuclear complex. The results of this work (i) help assess whether dinuclear metabolites play a role in cisplatin chemotherapy, (ii) elucidate the toxicity and pharmacological inactivity of [cis-{Pt(NH(3))(2)}(mu-OH)](2)(2+), and (iii) suggest future investigations of dinuclear anticancer complexes that contain one mu-hydroxo ligand.  相似文献

Adenine acidification as a consequence of simultaneous PtII binding to N1 and N7 facilitates deprotonation of the exocyclic N(6)H2 group and permits PtII migration from N1 to N6 under mild conditions. Starting from the trinuclear complex cis-[(NH3)2Pt(N1-9-MeA-N7)2{Pt(NH3)3)}2]6+ (3), stepwise migration of cis-(NH3)2PtII takes place in the alkaline aqueous solution to give initially cis-[(NH3)2Pt(N1-9-MeA-N7)(N6-9-MeA--N7){Pt(NH3)3}2]5+ (4) and eventually cis-[(NH3)2Pt(N6-9-MeA--N7)2{Pt(NH3)3}2]4+ (5) (with 9-MeA = neutral 9-methyladenine, 9-MeA- = 9-methyl-adenine monoanion, deprotonated at N6). The migration process has been studied by 1H NMR spectroscopy, and relevant acid-base equilibria have been determined. 5 has been crystallized as its nitrate salt and has been characterized by X-ray crystallography. The precursor of 3, [(NH3)3Pt (9-MeA-N7)]Cl2.2H2O (2) has likewise been studied by X-ray analysis.  相似文献

cis-[PtCl2(NH3)(2-picoline)] (AMD473) is a sterically-hindered anticancer complex with a profile of chemical and biological activity that differs significantly from that of cisplatin. Adducts of AMD473 with neutral 9-ethylguanine (9-EtGH) and anionic (N1-deprotonated) 9-ethylguanine (9-EtG) as perchlorate and nitrate salts, and also a nitrate salt of the trans isomer (AMD443), were prepared and their structures determined by X-ray crystallography: cis-[Pt(NH3)(2-pic)(9-EtGH)2](ClO4)2 (1).2H(2)OMe(2)CO, cis-[Pt(NH3)(2-pic)(9-EtGH)2](NO3)2 (2).2H2O, cis-[Pt(NH3)(2-pic)(9-EtGH)(9-EtG)]NO3 (3),3.5 H2O, trans-[Pt(NH3)(2-pic)(9-EtGH)(9-EtG)]NO3 (4).8H2O. In all cases, platinum coordination is through N7 of neutral (1, 2) and anionic (3, 4) guanine. In each complex, the guanine bases are arranged in the head-to-tail conformation. In complex 1, there is an infinite array of six-molecule cycles, based on both hydrogen bonding and pi-pi stacking of the 2-picoline and guanine rings. Platinum(II) coordinated at N7 acidifies the N1 proton of neutral 9-ethylguanine (pKa = 9.57) to give pKa1 = 8.40 and pKa2 = 8.75 for complex 2, and pKa1 = 7.77 and pKa2 = 9.00 for complex 4. In complexes 3 and 4, three intermolecular hydrogen bonds are formed between neutral and deprotonated guanine ligands involving O6, N1 and N2 sites. Unusually, both of the platinated guanine bases of complexes 3 and 4 participate in this triple G triple bond G hydrogen bonding. This is the first report of X-ray crystal structures of nucleobase adducts of the promising anticancer drug AMD473.  相似文献

The first triethylphosphine-stabilized Pt-Au cluster compounds, [Pt(AuPEt(3))(10)](2+) (2) and [Pt(AuPEt(3))(9)](3+) (3), were prepared by the direct reaction of Pt(PEt(3))(3) with AuPEt(3)NO(3) under a dihydrogen atmosphere. Cluster 2 is the highest-nuclearity homoleptic Pt(AuPR(3))(n)() cluster yet prepared. The reactivity and structures of these clusters are in agreement with the well-established electron-counting arguments. The 18-electron cluster 2 was converted into the 16-electron cluster 3 by oxidation with 2 equiv of ferricinium ion [Fe(eta(5)-C(5)H(5))(2)](+). Cluster 3 was converted into 2 by reduction with H(2) in the presence of [AuPEt(3)](+). Cluster 3 was also observed to cleanly add the 2-electron donors CO and PEt(3) to form the 18-electron clusters [(CO)Pt(AuPEt(3))(9)](3+) (4) and [(PEt(3))Pt(AuPEt(3))(9)](3+) (5), respectively. Single-crystal X-ray diffraction results show that 3 has a flattened, toroidal structure in which the PtAu(9) framework has a Pt-centered, tricapped trigonal prismatic geometry. Crystal data for [Pt(AuPEt(3))(9)](NO(3))(3) is as follows: hexagonal P6(3)/m, a = 15.134(5) ?, c = 23.48(1) ?, V = 4657 ?(3), Z = 2, residuals R = 0.056, and R(w)() = 0.053 for 1489 observed reflections and 81 variables, Mo Kalpha radiation. Compound 3 was found to reversibly add H(2) in solution to form the dihydride cluster [(H)(2)Pt(AuPEt(3))(9)](3+) (6). The equilibrium constant for this addition reaction is 1.1 x 10(3) M(-)(1) (CD(2)Cl(2) solution, 25 degrees C), slightly smaller than that for [Pt(AuPPh(3))(8)](2+). The rate of the addition is also slower than that with [Pt(AuPPh(3))(8)](2+). Cluster 3 is an excellent homogeneous catalyst for H(2)-D(2) equilibration giving a turnover rate for HD production of 0.13 s(-)(1) (nitrobenzene solvent, 30 degrees C, 1 atm). The PEt(3)-containing clusters give similar rates and follow the same general trends previously observed with PPh(3)-ligated clusters. The chemistry of these new clusters is explained by consideration of the steric and electronic properties of the PEt(3) ligand. These new compounds will be useful as models for hydrogen activation by Pt-Au clusters and as precursors for supported Pt-Au catalysts.  相似文献

We report the interaction between B-type tri-lacunary heteropolyoxotungstate anions and actinyl(V) cations in aqueous solution, yielding a greater understanding of the stability of the O≡An≡O(1+) linear dioxo actinide moiety. Previously we reported that B-α-[BiW(9)O(33)](9-) and B-α-[SbW(9)O(33)](9-) will react with NpO(2)(1+) to yield [(Np(3)W(4)O(15))(H(2)O)(3)(MW(9)O(33))(3)](18-) (M = Bi, or Sb). Single crystal structural characterisation of salts of these complexes revealed a core in which three Np(V) atoms interact with a central W(VI) atom through bridging oxo groups. These bridging oxygen atoms come from one of the two axial oxygens in O≡Np≡O(1+) and represent a highly unusual interaction for a discrete molecular species. In this study visible/near infra-red spectroscopy indicates that [(Np(3)W(4)O(15))(H(2)O)(3)(BiW(9)O(33))(3)](18-) could be readily stabilized in solution at near neutral pH for several months, with (NH(4))(14)Na(4)[(Np(3)W(4)O(15))(H(2)O)(39)BiW(9)O(33))(3)]·62H(2)O crystallising from solution in high yield. At lower pH and [BiW(9)O(33)](9-) : NpO(2)(1+) ratios additional Np(V) species could be observed in solution. Stabilization of [(Np(3)W(4)O(15))(H(2)O)(3)(SbW(9)O(33))(3)](18-) in solution proved more challenging, with several distinctive Np(V) near infra-red transitions observed in solution. Slow complexation kinetics and reduction to Np(IV) was also observed. High [SbW(9)O(33)](9-) : NpO(2)(1+) molar ratios and careful control of solution pH was required to prepare solutions in which [(Np(3)W(4)O(15))(H(2)O)(3)(SbW(9)O(33))(3)](18-) was the only neptunium containing species. In stark contrast to the NpO(2)(1+) chemistry, [BiW(9)O(33)](9-) readily oxidizes PuO(2)(1+) to PuO(2)(2+) yielding further evidence of the decreased stability of Pu(V)vs. Np(V). Np L(II)-edge XAFS measurement revealed very good agreement with single crystal diffraction data for the Np structural environment for [(Np(3)W(4)O(15))(H(2)O)(3)(MW(9)O(33))(3)](18-) (M = Bi, or Sb) in the solid state. There was also good agreement between coordination shells for [(Np(3)W(4)O(15))(H(2)O)(3)(BiW(9)O(33))(3)](18-) in the solid state and in solution, yielding further confirmation of the high stability of this particular cluster.  相似文献

A new organically templated layered uranium phosphate fluoride, [(CH(3))(2)NH(CH(2))(2)NH(CH(3))(2)][(UO(2))(2)F(2)(HPO(4))(2)] has been synthesized by hydrothermal reaction of UO(3), H(3)PO(4), HF, and (CH(3))(2)NCH(2)CH(2)N(CH(3))(2) at 140 degrees C. [(CH(3))(2)NH(CH(2))(2)NH(CH(3))(2)][(UO(2))(2)F(2)(HPO(4))(2)] has a layered crystal structure consisting of seven-coordinated UO(5)F(2) pentagonal bipyramids and four-coordinated HPO(4) tetrahedra. Each anionic layer containing three-, four-, and six-membered rings is separated by [(CH(3))(2)NH(CH(2))(2)NH(CH(3))(2)](2+) cations. The [(CH(3))(2)NH(CH(2))(2)NH(CH(3))(2)](2+) cations may be readily exchanged with the M(2+) ions (M = Ba, Sr and Ca) in water to give high crystalline AE(UO(2))(2)(PO(4))(2).6H(2)O (AE = Ca, Sr, Ba).  相似文献

Luminescent [(NH(3))(4)Pt][Au(CN)(2)](2).1.5(H(2)O), which forms from aqueous solutions of [(NH(3))(4)Pt]Cl(2) and K[Au(CN)(2)], crystallizes with extended chains of the two ions with multiple close Pt...Au (3.2804(4) and 3.2794(4) A) and Au...Au (3.2902(5), 3.3312(5), and 3.1902(4) A) contacts. Nonluminescent [(NH(3))(4)Pt][Ag(CN)(2)](2).1.4(H(2)O) is isostructural with [(NH(3))(4)Pt][Au(CN)(2)](2).1.5(H(2)O). Treatment of [(NH(3))(6)Ni]Cl(2) with K[Au(CN)(2)] forms [(NH(3))(2)Ni][Au(CN)(2)](2) in which the [Au(CN)(2)](-) ions function as nitrile ligands toward nickel, which assumes a six-coordinate structure with trans NH(3) ligands. The [Au(CN)(2)](-) ions self-associate into linear columns with close Au...Au contacts of 3.0830(5) A, and pairs of gold ions in these chains make additional but longer (3.4246(5) A) contacts with other gold ions.  相似文献

Oxidation of the acetate-bridged half-lantern platinum(II) complex cis-[Pt(II)(NH(3))(2)(μ-OAc)(2)Pt(II)(NH(3))(2)](NO(3))(2), [1](NO(3))(2), with iodobenzene dichloride or bromine generates the halide-capped platinum(III) species cis-[XPt(III)(NH(3))(2)(μ-OAc)(2)Pt(III)(NH(3))(2)X](NO(3))(2), where X is Cl in [2](NO(3))(2) or Br in [3](NO(3))(2), respectively. These three complexes, characterized structurally by X-ray crystallography, feature short (≈2.6 ?) Pt-Pt separations, consistent with formation of a formal metal-metal bond upon oxidation. Elongated axial Pt-X distances occur, reflecting the strong trans influence of the metal-metal bond. The three structures are compared to those of other known dinuclear platinum complexes. A combination of (1)H, (13)C, (14)N, and (195)Pt NMR spectroscopy was used to characterize [1](2+)-[3](2+) in solution. All resonances shift downfield upon oxidation of [1](2+) to [2](2+) and [3](2+). For the platinum(III) complexes, the (14)N and (195)Pt resonances exhibit decreased line widths by comparison to those of [1](2+). Density functional theory calculations suggest that the decrease in the (14)N line width arises from a diminished electric field gradient at the (14)N nuclei in the higher valent compounds. The oxidation of [1](NO(3))(2) with the alternative oxidizing agent bis(trifluoroacetoxy)iodobenzene affords the novel tetranuclear complex cis-[(O(2)CCF(3))Pt(III)(NH(3))(2)(μ-OAc)(2)Pt(III)(NH(3))(μ-NH(2))](2)(NO(3))(4), [4](NO(3))(4), also characterized structurally by X-ray crystallography. In solution, this complex exists as a mixture of species, the identities of which are proposed.  相似文献

The reactions of the dinuclear platinum(II) complexes, [[cis-Pt(NH(3))(2)](2)(mu-OH)(mu-pz)](NO(3))(2) (1, pz = pyrazolate), [[cis-Pt(NH(3))(2)](2)(mu-OH)(mu-1,2,3-ta-N1,N2)](NO(3))(2) (2, 1,2,3-ta = 1,2,3-triazolate), and a newly prepared [[cis-Pt(NH(3))(2)](2)(mu-OH)(mu-4-phe-1,2,3-ta-N1,N2)](NO(3))(2) (3, 4-phe-1,2,3-ta = 4-phenyl-1,2,3-triazolate), whose crystal structure was determined, with 9-ethylguanine (9EtG) have been monitored in aqueous solution at 310 K by means of (1)H NMR spectroscopy. The dinuclear platinum(II) complexes 1-3 each react with 9EtG in a bifunctional way to form 1:2 complexes, [[cis-Pt(NH(3))(2)(9EtG-N7)](2)(mu-pz)](3+) (4), [[cis-Pt(NH(3))(2)(9EtG-N7)](2)(mu-1,2,3-ta-N1,N3)](3+) (5), and [[cis-Pt(NH(3))(2)(9EtG-N7)](2)(mu-4-phe-1,2,3-ta-N1,N3)](3+) (6). The reactions of 2 and 3 involve a novel isomerization, in which the Pt atom, initially bound to N2 on the 1,2,3-ta, migrates to N3 after the first substitution by N7 of 9EtG. This isomerization reaction has been unambiguously characterized by 1D and 2D NMR spectroscopy and pH titration. The reactions of 2 and 3 with 9EtG show faster kinetics, and the second-order rate constants (k) for the reactions of 1-3are 1.57 x 10(-4), 2.53 x 10(-4), and 2.56 x 10(-4) M(-1) s(-1), respectively. The pK(a) values at the N1H site of 9EtG were determined for 4-6 from the pH titration curves. Cytotoxicity assays of 1-3 were performed in L1210 murine leukemia cell lines, respectively sensitive and resistant to cisplatin. In the parent cell line, 2 and 3 exhibit higher cytotoxicity compared to cisplatin, especially, 2 is 10 times as active as cisplatin. 1 was found to be less cytotoxic than cisplatin, but still in the active range and more active than cisplatin in a cisplatin-resistant cell line.  相似文献

The ligand-loss photochemistry of cis-[Ru(bpy)(2)(NH(3))(2)](2+) (bpy = 2,2'-bipyridine) was investigated in water and in the presence of added ligands such as bipyridine and chloride. Irradiation of the complex results in the covalent binding to 9-methyl- and 9-ethylguanine, as well as to single-stranded and double-stranded DNA. This photoinduced DNA binding is not observed for the control complex [Ru(bpy)(2)(en)](2+) (en = ethylenediamine) under similar irradiation conditions. The results presented here show that octahedral Ru(II) complexes with photolabile ligands may prove useful as photoactivated cisplatin analogs.  相似文献

Complexes [Pt(mu-N,S-8-TT)(PPh(3))(2)](2) (1), [Pt(mu-S,N-8-TT)(PTA)(2)](2) (2), [Pt(8-TTH)(terpy)]BF(4) (3), cis-[PtCl(8-MTT)(PPh(3))(2)] (4), cis-[Pt(8-MTT)(2)(PPh(3))(2)] (5), cis-[Pt(8-MTT)(8-TTH)(PPh(3))(2)] (6), cis-[PtCl(8-MTT)(PTA)(2)] (7), cis-[Pt(8-MTT)(2)(PTA)(2)] (8), and trans-[Pt(8-MTT)(2)(py)(2)] (9) (8-TTH(2) = 8-thiotheophylline; 8-MTTH = 8-(methylthio)theophylline; PTA = 1,3,5-triaza-7-phosphaadamantane) are presented and studied by IR and multinuclear ((1)H, (31)P[(1)H]) NMR spectroscopy. The solid-state structure of 4 and 9 has been authenticated by X-ray crystallography. Growth inhibition of the cancer cells T2 and SKOV3 induced by the above new thiopurine platinum complexes has been investigated. The activity shown by complexes 4 and 9 was comparable with cisplatin on T2. Remarkably, 4 and 9 displayed also a valuable activity on cisplatin-resistant SKOV3 cancer cells.  相似文献

Three protonated forms of 7-methylguanine (7-MeGH, 1) with different counter ions, [7-MeGH(2)]X (X = NO(3), 1a; ClO(4), 1b; BF(4), 1c) and two Pt(II) complexes, trans-[Pt(NH(3))(2)(7-MeGH-N9)(2)](ClO(4))(2) (4) and trans-[Pt(NH(3))(2)(7-MeGH-N9)(7-MeGH-N3)](ClO(4))(2)·3H(2)O (5) are described and their X-ray crystal structures are reported. 1a-1c form infinite ribbons via pairs of intermolecular hydrogen bonds between N1H···O6 and N3···N2H(2) sites, with anions connecting individual ribbons, thereby generating extended sheets. 4 and 5 do not display unusual features, except that 5 represents a rare case of a bis(nucleobase) complex of Pt(II) in which linkage isomers occur. Unlike in a previously reported compound, [Pt(dien)(7-MeGH-N9)](NO(3))(ClO(4)), the Pt coordination planes and the 7-MeGH planes are not coplanar in 4 and 5. The hydrogen bonding behaviour of 7-MeGH, free and when platinated at N9 (complex 4), was studied in Me(2)SO-d(6). It revealed the following: (i) there is no detectable self-association of 1 in Me(2)SO solution. (ii) 1 and 1-methylcytosine (1-MeC) form Watson-Crick pairs. (iii) 4 does not self-associate. (iv) 4 associates with 1-MeC in the Watson-Crick fashion. (v) 4 and 1 interact in solution, but no model can be proposed at present. (vi) Remarkable interaction shifts between 4 and 1 occur when NH(3) is liberated from trans-(NH(3))(2)Pt(II) to give NH(4)(+) in Me(2)SO-d(6). Feasible models, which imply the presence of deprotonated 7-MeG(-) species are proposed. Finally, DFT calculations were carried out to qualitatively estimate the effect of 7-MeGH acidity in [Pt(dien)(7-MeGH-N9)](2+) in dependence of the dihedral angle between the Pt coordination plane and the nucleobase.  相似文献

The tridentate dianionic ligand 2-[2'-(hydroxyisopropoxyphosphoryl)phenylsulfanyl]benzoate (L(2-)) reacts with cis-[Pt(NH(3))(2)(H(2)O)(2)](2+) to form an S,O-chelate in which the O-coordinated group is either carboxylate or phosphonate, depending on the degree of protonation of the complex. Carboxylate appears to be the stronger ligand, and the stoichiometric reaction between cis-[Pt(NH(3))(2)(H(2)O)(2)](2+) and L(2-) yields the neutral species [Pt(L)(NH(3))(2)], with L bound by sulfanyl and carboxylate groups, both in solution and in the solid state. Upon protonation of [Pt(L)(NH(3))(2)], the stronger basicity of the carboxylate causes the Pt coordination to switch from carboxylate to phosphonate, and the uncoordinated carboxylate group becomes protonated. In methanolic solution, the first-order kinetics of this rearrangement could be observed by (31)P NMR spectroscopy. Both complexes-the carboxylate-bound neutral complex [Pt(L)(NH(3))(2)].H(2)O (triclinic, P1 (no. 2), a=9.529(6), b=9.766(6), c=12.299(7) angstroms, alpha=106.91(2), beta=101.71(2), gamma=102.05(2) degrees, Z=2) and the perchlorate salt of the phosphonate-bound complex [Pt(LH)(NH(3))(2)]ClO(4).H(2)O (monoclinic, P2(1)/c (no. 14), a=12.095(2), b=14.046(2), c=14.448(2) angstroms, beta=95.55(2) degrees, Z=4)-were characterized by X-ray crystallography.  相似文献

A heteronuclear complex of composition trans-[(NH(3))(2)Pt(N4-1-MeC(-)-N3)(2)Cu(H(2)O)(2)](ClO(4))(2) (1a), with 1-MeC(-) = 1-methylcytosinate, has been prepared and characterized by X-ray crystallography. 1a (Cu,Pt) is a linkage isomer of a previously described compound with the two metals inverted (Pt,Cu). The intermetallic distances are significantly different in the two types of compounds, 2.6109(9) A in 1a, yet 2.49-2.56 A in several forms of the linkage isomer. When heated in water in the presence of air, 1a is converted in low yield into diplatinum(III) compounds [(H(3)N)Pt(1-MeC(-)-N3,N4)(4)Pt(NH(3))](2+) (2a) and [(H(2)O)Pt(1-MeC(-)-N3,N4)(4)Pt(NH(3))](2+) (2b), which were crystallized as ClO(4)(-) salts. In a modified procedure a third representative of this group of diplatinum(III) compounds, [(O(2)N)Pt(1-MeC(-)-N3,N4)(4)Pt](+) (2c) was isolated. All three compounds contain the four bridging 1-MeC ligands in a head,tail,head,tail arrangement with Pt-Pt distances (2.4516(7)-2.4976(9) A) that are the shortest ones among diplatinum(III) compounds containing nucleobases.  相似文献
