The feasibility of the combination of field-amplified sample injection (FASI) and in-capillary derivatization was explored for improving sensitivity of histamine in capillary electrophoresis (CE). Naphthalene-2,3-dicarboxaldehyde (NDA) was used as derivatization reagent. The reagent and sample was introduced by tandem mode. The derivatization was accomplished by at-inlet mode with standing time of 1.5 min. The combination of FASI and in-capillary derivatization was successfully achieved with about 400-fold concentration sensitivity enhancement compared to pre-capillary derivatization at the same set-up. The detection limit of concentration for histamine reached 1.25 x 10(-11) M by CE and fluorescence detection with S/N = 3. Parameters affecting FASI and in-capillary derivatization process including sample matrix, buffer concentration and reagent injection amount, were investigated.  相似文献

Throughout the separation of chiral basic drugs by capillary electrophoresis (CE) with neutral hydroxypropyl-beta-cyclodextrin (HP-beta-CD) as chiral selector, the sensitivity of detection has been improved by using field-amplified sample injection (FASI). In the present work, this on-line stacking method has been used to detect low ng/mL levels of cationic enantiomers of a new adrenoreceptor antagonist in plasma. A systematic study of the parameters affecting on-line concentration of these enantiomers (nature of the preinjection plug, composition of sample solvent, injection times of water and sample plugs) has been performed enabling the detection sensitivity of antagonist enantiomers to be improved by 180 times compared with usual hydrodynamic injection. The quantification of each adrenoreceptor antagonist enantiomer in plasma samples was then performed in the 2-100 ng/mL (or 8-400 nM) concentration range after a solid-phase extraction step. Using this FASI-CE-UV procedure, the limit of quantification (LOQ) for each enantiomer was in the low ng/mL concentration range (3 ng/mL or 10 nM).  相似文献

An on-line sample stacking method, namely field-amplified sample injection, has been developed for the separation and determination of carnosine, anserine, and homocarnosine by capillary electrophoresis. Using electrokinetic injection, about 130- to 160-fold improvement of sensitivity was achieved without loss of separation efficiency when compared to conventional sample injection. For conventional injection, the samples were dissolved in running buffer and then hydrodynamically injected for 10 s (3.45 kPa). Various parameters affecting separation and sample stacking were optimized. Under optimum conditions, linear responses were obtained over two orders of magnitude and the detection limits (defined as S/N = 3) of carnosine, anserine, and homocarnosine were 1.5 x 10(-8) to 1.6 x 10(-8) mol/L.  相似文献

采用添加乙腈引发的场放大进样与瞬间等速电泳结合的预富集方法,实现了在毛细管内大体积高盐样品中阳离子的有效富集与分离。详细讨论了影响富集的缓冲体系、尾随离子种类、毛细管有效长度、进样时间和等速电泳时间等重要因素。选择在400 mmol/L LiAc-HAc缓冲液(pH 4.5)和400 mmol/L β-丙氨酸-HAc尾随液(pH 4.5)及10 kV下样品和尾随溶液电动注入时间分别为270和90 s的条件下对高盐溶液中两种结构相近的药物普萘洛尔和美托洛尔进行了富集和分离。该方法富集倍数约为常规电动进样的280倍,普萘洛尔和美托洛尔的检出限分别为2×10-3和8×10-3 mg/L。  相似文献

This paper describes the use of reversed-phase, reversed-polarity head-column field-amplified sample stacking (HCFASS) for on-line sample concentration in conventional capillary electrophoresis. The effective stacking efficiency was determined as a function of sodium hydroxide concentration in the sample matrix. Results concur with theoretical predictions where stacking efficiency depends on the conductivity (electric field strength) and electrophoretic mobility in the sample matrix solution. Fluorescein isothiocyanate-derivatized aniline and 2,4-dimethylaniline were dissolved in sodium hydroxide (800 microM), separated in a phosphate running buffer (0.05 M, pH 9.0) and detected utilising laser-induced fluorescence. The use of reversed-phase, reversed-polarity HCFASS with laser-induced fluorescence detection yielded sensitivity improvements with respect to normal injection schemes in excess of three orders of magnitude, and a limit of detection as low as 10(-13) M.  相似文献

Conditions for the spectrophotometric determination of copper with 2,9-dimethyl-l,10-phenan-throline (neocuproine) in the presence of ascorbic acid in a water-ethanol solution (9 : 1) at pH 4.5–5.0 have been found. The detection limit is 3 x 10-6 M. The concentration range is from 4.4 x 10-6 to 3 x 10-4 M. Conditions for the determination of copper(I) with neocuproine by thermal lens spectrometry have been proposed. The detection limit is 4 x 10-7 M. The concentration range is from 7 x 10-7 to 6 x 10-5 M. Iron(II) at concentrations as high asn x 10-4 M does not interfere with the determination of copper. Changes in the conditions for the photometric reaction associated with passing from spectrophotometric measurements to thermal lensing are discussed.  相似文献

A capillary electrophoresis method using carboxymethyl-beta-cyclodextrin as the chiral selector and mixture of methanol and ethanol as the organic additive was successfully developed for the simultaneous enantioseparation and determination of six beta-blockers, namely, carteolol, atenolol, sotalol, metoprolol, esmolol and propranolol in this paper. The most suitable running buffer for enantioseparation was found to be the solution of 20 mmol/L NaH(2)PO(4)-Na(2)HOP(4) (pH 5.5) containing 1.5% w/v carboxymethyl-beta-cyclodextrin, 5% v/v methanol and 5% v/v ethanol. Furthermore, field-amplified sample injection as an on-line sample stacking method was developed in order to increase the detection sensitivity. The experimental conditions for both simultaneous enantioseparation and the field-amplified sample injection method had been investigated in detail. Under the optimum conditions, the detection limits (defined as S/N=3) of this method were 0.01, 0.05, 0.05, 0.05, 0.05 and 0.5 microg /mL for (+/-) carteolol, (+/-) atenolol, (+/-) sotalol, (+/-) metoprolol, (+/-) esmolol and (+/-) propranolol, respectively, which were much lower than those of the conventional methods. The enhancement factors were greatly improved by 25-fold for the enantiomers of the beta-blockers except five-fold for (+/-) propranolol. Eventually, the proposed method has been applied for the analysis of human serum sample.  相似文献

The copper-mercury film electrode has been suggested for the determination of Se(IV) in a wide range of concentration from 1x10(-9) to 1x10(-6) mol L(-1)by square-wave cathodic stripping voltammetry. Insufficient reproducibility and sensitivity of the mercury film electrode have been overcome by using copper(II) ions during the plating procedure. Copper(II) has been found to be reduced and form a reproducible copper-mercury film on a glassy carbon electrode surface. The plating potential and time, the concentration of copper(II) and the concentration of the supporting electrolyte have been optimised. Microscopy has been used for a study of the morphology of the copper-mercury film. It has been found that it is the same as for the mercury one. The preconcentration step consists in electrodeposition of copper selenide on the copper-mercury film. The relative standard deviation is 4.3% for 1x10(-6) mol L(-1) of Se(IV). The limit of detection is 8x10(-10) mol L(-1) for 5 min of accumulation.  相似文献

In order to improve the sensitivity of CE-ESI-MS for the analysis of brain-gut peptides, a solid-phase extraction combined with field-amplified sample injection was used for the pre-concentration of the brain-gut peptides. Compared with the conventional pressure injection method, the sensitivity in the detection of brain-gut peptides was improved more than 100-fold. The possible factors affecting sample stacking, such as the sample matrix, the composition and the length of the water column, the types and the volumes of eluent, have been investigated in detail. Under the optimum conditions, the detectable concentration of brain-gut peptides was found to be as low as 0.02?μM. A linear response concentration for the detection was developed in the range of 0.08-25?μmol/L. A real sample of human cerebrospinal fluids, which was spiked with brain-gut peptides, was also examined in order to evaluate the reliability of the proposed approach. The recovery of the method was in a range from 69.2 to 85.4%. The method was found to be reliable, accurate and potentially applicable for clinical drug analysis.  相似文献

Electrodes for the dopamine (DA) determination in biological samples have been developed with improved selectivity and sensitivity in an excess of ascorbic acid (AA). Negatively charged Ni(II) complex was synthesized and electropolymerized on the glassy carbon electrode to impart the surface with anionic characteristics that could act both as a catalyst and as a discriminating layer against AA based on the electrostatic interaction. Thus prepared electrodes enabled selective determination of DA even in a large excess of AA by differential pulse voltammetry at physiological pH. Linear response was found down to 1.0 x 10(-7) M with 5.0 x 10(-9) M of LOD (Limit of Detection). In a flow injection analysis performed in an amperometric mode, the detection limit was lowered by two orders of magnitude down to 1.0 x 10(-9) M with a linear range of 1.0 x 10(-9) to 1.0 x 10(-6) M. The relative standard deviation was found to be 3.36% from 25 independent measurements for 1.0 x 10(-5) M of DA. Stable oxidation current of DA was observed even after 30 days storage in air. The recoveries of DA in the 100-fold diluted human urine samples were 97.7% for 4 measurements. The rate constant for the DA oxidation was 1.3 x 10(-3) cm s(-1) from hydrodynamic experiments using a rotating disk electrode.  相似文献

建立了毛细管电泳-场强放大样品堆积测定染发剂中4,4′-二氨基二苯甲烷、苯胺、邻甲氧基苯胺、对氨基苯甲醚、3,4-二甲基苯胺、间氨基苯酚、1-萘胺7种苯胺类物质的分析方法。在优化的缓冲溶液体系(0.15 mol/L NaH2PO4,0.015 mol/L 三乙醇胺, pH 2.3)下7种分析物在6.5 min内实现基线分离。考察了样品中添加的磷酸浓度和乙腈浓度、水柱长度、电动进样时间与电压对场强放大富集效率及重现性的影响。最佳的富集条件为: 水柱注入3.45 kPa(0.5 psi)×6 s,样品中添加40%(v/v)乙腈和0.6×10~3mol/L磷酸,进样电压与进样时间为10 kV×10 s。线性范围为3~1000 μg/L(R2>0.996),检出限为0.26~2.75 μg/L,将已有方法的检测灵敏度提高了1~3个数量级。在2种市售黑色染发剂中均检测到间氨基苯酚,含量分别为7.32 mg/g和1.34 mg/g。平均加标回收率为74%~108%。该方法灵敏度高、快速、重现性好、成本低,可供多种样品基质中痕量苯胺类污染物及其他阳离子物质的测定借鉴使用。  相似文献

Capillary electrophoresis (CE) with Ru(bpy)3(2+) electrochemiluminescence (ECL) detection system was established to the determination of contamination of banknotes with controlled drugs and a high efficiency on-column field-amplified sample stacking (FASS) technique was also optimized to increase the ECL intensity. The method was illustrated using heroin and cocaine, which are two typical and popular illicit drugs. Highest sample stacking was obtained when 0.01 mM acetic acid was chosen for sample dissolution with electrokinetical injection for 6 s at 17 kV. Under the optimized conditions: ECL detection at 1.2 V, separation voltage 10.0 kV, 20 mM phosphate-acetate (pH 7.2) as running buffer, 5 mM Ru(bpy)3(2+) with 50 mM phosphate-acetate (pH 7.2) in the detection cell, the standard curves were linear in the range of 7.50x10(-8) to 1.00x10(-5) M for heroin and 2.50x10(-7) to 1.00x10(-4) M for cocaine and detection limits of 50 nM for heroin and 60 nM for cocaine were achieved (S/N = 3), respectively. Relative standard derivations of the ECL intensity and the migration time were 3.50 and 0.51% for heroin and 4.44 and 0.12% for cocaine, respectively. The developed method was successfully applied to the determination of heroin and cocaine on illicit drug contaminated banknotes without any damage of the paper currency. A baseline resolution for heroin and cocaine was achieved within 6 min.  相似文献

A capillary electrophoresis method, using field-amplified sample injection (FASI), was developed for separation and determination of some beta 2-agonists, such as cimaterol, clenbuterol and salbutamol. The optimum conditions for this system had been investigated in detail. The precision of the migration time, peak height and accuracy were determined in both intra-day (n = 5) and inter-day (n = 15) assays. Under the optimum conditions, the detection limits (defined as S/N = 3) of this method were found to be lower than 2.0 ng/mL for all of these three beta 2-agonists, which were much lower than that of the conventional electro-migration injection method, the enhancement factors were greatly improved to be 30-40-fold. Such lower detection limit lets this method to be suitable for determination of above-mentioned beta 2-agonists in the urine sample. The mean recoveries in urine were higher than 96.2%, 95.6% and 95.3% for cimaterol, clenbuterol and salbutamol, respectively, with relative standard deviations lower than 3.5%.  相似文献

A polarographic investigation of the copper-3-hydroxy-1-p-sulphonatophenyl-3-phenyltriazene (HSPT) complex in 0.05 M sodium tetraborate medium is described and a simple and sensitive single-sweep polarographic method for the determination of trace amounts of copper in biological samples is proposed. The complex was shown to be Cu(HSPT)2 with log beta' = 11.38. The polarographic wave is caused by the reduction of copper(II) in the adsorbed complex to copper amalgam on the surface of a mercury electrode. The current peak is directly proportional to the concentration of copper in the range 8.0 x 10(-9)-4.0 x 10(-6) M and the detection limit is 5.0 x 10(-9) M.  相似文献

Ion-exchanger colorimetry for the microdetermination of bismuth(III) in water samples is described. The yellow bismuth-iodide complexes are specifically sorbed and concentrated on an anion-exchange resin in the sulphate form. The resin-phase absorbances at 492 and 700 nm are measured directly. Bismuth in the ppb-ppm range can be determined without interference, except in presence of a large amount of copper(II), silver(I) and lead(II). The detection limit is 6.4 x 10(-9)M, i.e., 1.3 ppb. The distribution ratio is larger than 10(7). It is, therefore, possible to enhance the sensitivity by increasing the sample volume. The method is useful for the determination of bismuth in natural waters and industrial effluents.  相似文献

A selective and sensitive stripping voltammetric method for the determination of trace amounts of copper(II) with cyclopentanone thiosemicarbazone (CPTSC) is presented. The method is based on the adsorptive accumulation of the resulting copper-CPTSC complex on a hanging mercury drop electrode, followed by the stripping voltammetric measurements at the reduction current of the adsorbed complex at -0.37 V vs. Ag/AgCl. The optimal conditions for the stripping analysis of copper include pH 9.3, deposition time of 120 s, and a deposition potential of -0.1 V (vs. Ag/AgCl). The peak current is linearly proportional to the copper concentration over a range 3.14 x 10(-9) M to 1.57 x 10(-6) M with a limit of detection of 1.57 x 10(-9) M. The technique has been applied to the determination of copper in biological samples, like urine and whole blood.  相似文献

In sample-stacking techniques, the detection limit cannot be improved by simply increasing the length of the sample solution, because the individual electrophoretic parameters must be optimized. In an attempt to increase the amount of sample injected, as well as to focus them onto a small zone, two novel methods are proposed. One of these employs an "ultra-high conductivity zone", which was inserted between the sample zone and background solution to build an unequal conductivity gradient. The other employs a "low temperature bath". A portion of the capillary (near the junction between the sample solution and the background solution) was immersed in a low temperature bath, which served as a "pseudo-high-conductivity zone" due to the fact that conductivity would increases when the temperature is decreased. As a result, a large volume of sample injection can be achieved. Using 3,4-methylenedioxymethamphetamine as a model compound, the detection limit was determined to be 1.6 x 10(-6) M (S/N = 3) by means of normal non-aqueous capillary electrophoresis (NACE). This could be improved to 3.0 x 10(-8) M, 4.8 x 10(-9) M and 5.0 x 10(-9) M, respectively, when the normal stacking, ultra-high conductivity zone NACE-stacking and the low-temperature zone NACE-stacking methods were applied.  相似文献

This report describes a novel online field-amplified sample stacking (FASS) procedure to analyze 16 chlorinated acid herbicides. By using a poly(vinyl alcohol) (PVA)-coated capillary to reduce electroosmotic flow and introducing a methanol-water plug before sample loading, the sample injection time could be very long without loss of sample and separation efficiency. Under the optimized condition, the FASS procedure could provide great sensitivity enhancement (5000-10 000-fold) and satisfactory reproducibility (relative standard deviations of migration times less than 2.4%, relative standard deviations of peak areas less than 8.0%). Combined with cationic surfactant-assisted solid-phase extraction (CSA-SPE), the limit of detection of the herbicides ranged from 0.269 to 20.3 ppt, which are two orders lower than those of the US Environmental Protection Agency standard method 515.1. The CSA-SPE-FASS-CE method was successfully applied to the analysis of local pond water.  相似文献

A new design of a continuous flow system applied to the simultaneous determination of the concentration of zinc(II), cadmium(II), lead(II), copper(II), nickel(II), cobalt(II) and chromium(VI) in river water is described. A flow cell made in the laboratory, which has been patented, based on a 'wall-jet' configuration with a three-electrode system is described. Optimum conditions for the determination of the metal ions are reported. The detection limits and relative standard deviation values were 4.01x10(-9) M and 0.078 for Zn(II), 1.76x10(-10) M and 0.056 for Cd(II), 4.69x10(-10) M and 0.134 for Pb(II), 2.29x10(-10) M and 0.138 for Cu(II), 1.61x10(-9) M and 0.093 for Ni(II), 1.91x10(-9) M and 0.113 for Co(II), and 1.35x10(-9) M and 0.081 for Cr(VI). The procedure was applied to a sample of water from the Arlanzón river and the results were compared with inductively coupled mass plasma spectrometry (ICP-MS) as reference method. The final aim of this work is to design a flow system, which can be automated.  相似文献

A new chemiluminescence system was developed for the determination of cysteine by flow injection system. This method is based on the reaction of L-cysteine with Ru(phen)3(2+) and Ce(IV) to produce chemiluminescence. The calibration curve was linear over the range 8.0x10(-7) to 4.0x10(-5) and 4.0x10(-5) to 1.0x10(-3) M with a detection limit of 7.0x10(-7) M (S/N=3). The relative standard deviation of 4.0x10(-6) M cysteine was found 3.5% (n=10). The influence of potential interfering substances was studied. The proposed method was successfully applied for the flow injection determination of cysteine in the real samples with minimum sampling rate of 90 sample/h.  相似文献
