Glucose biosensor enhanced by nanoparticles   总被引:4,自引:0,他引:4  
Glucose biosensors have been formed with glucose oxidase (GOD) immobilized in composite immobilization membrane matrix, which is composed of hydrophobic gold, or hydro-philic gold, or hydrophobic silica nanoparticles, or the combination of gold and silica nanoparticles, and polyvinyl butyral (PVB) by a sol-gel method. The experiments show that nanoparticles can significantly enhance the catalytic activity of the immobilization enzyme. The current response can be increased from tens of nanoamperometer (nA) to thousands of nanoamperometer to the same glucose concentration, and the electrodes respond very quickly, to about 1 min. The function of nanoparticles effect on immobilization enzyme has been discussed.  相似文献

A new approach for a simple electrochemical detection of PAT gene fragment is described. Poly(2,6-pyridinedicarboxylic acid) (PDC) modified glassy carbon electrode (GCE) was prepared by potential scan electropolymerization in an aqueous solution. Mg2 ions were incorporated by immer-sion of the modified electrode in 0.5 mol/L aqueous solution of MgCl2 to complete the preparation of a generic "activated" electrode ready for binding the probe DNA. The ssDNA was linked to the conduct-ing polymer by forming a bidentate complex between the carboxyl groups on the polymer and the phosphate groups of DNA via Mg2 . DNA immobilization and hybridization were characterized with dif-ferential pulse voltammetry (DPV) by using methylene blue (MB) as indicator and electrochemical im-pedance spectroscopy (EIS). The EIS was of higher sensitivity for DNA detection as compared with voltammetric methods in our strategy. The electron transfer resistance (Ret) of the electrode surface in EIS in [Fe(CN)6]3-/4- solution increased after the immobilization of the DNA probe on the Mg/PDC/GCE electrode. The hybridization of the DNA probe with complementary DNA (cDNA) made Ret increase further. The difference between the Ret at ssDNA/Mg/PDC/GCE and that at hybridization DNA modified electrode (dsDNA/Mg/PDC/GCE) was applied to determine the specific sequence related to the target PAT gene with the dynamic range comprised between 1.0 × 10-9 and 1.0 × 10_5 mol/L. A detection limit of 3.4 × 10-10 mol/L of oligonucleotides can be estimated.  相似文献

A photosensitive electrode was prepared by electrodepositing a membrane of chlorophyll a (Chla) on a SnO2 optical transparent electrode,with which and a metal counter electrode a Chla photoelectrochemical cell was formed.Photoinduced current (Ii) and photoinduced voltage (Vi) of the cell were measured.The dependence of Ii on the properties of metal electrodes was obvious,which was illustrated with mechanism of Chla photoelectrical effects Ii in this work was as high as 2×10-5 A·cm-2.  相似文献

In the present study, a disposable electrochemical DNA nano-biosensor is proposed for the rapid detection of genotoxic compounds and bio-analysis of water pollution. The DNA nano-biosensor is prepared by immobilizing DNA on Au nanoparticles and a self-assembled monolayer of cysteamine modified Au electrode. The assembly processes of cysteamine, Au nanoparticles and DNA were characterized by cyclic voltammetry (CV). The Au nanoparticles enhanced DNA immobilization resulting in an increased guanine signal. The interaction of the analyte with the immobilized DNA was measured through the variation of the electrochemical signal of guanine by square wave voltammetry (SWV). The biosensor was able to detect the known genotoxic compounds: 2-anthramine, acridine orange and 2- naphthylamine with detection limits of 2, 3 and 50 nmol/L, respectively. The biosensor was also used to test actual water samples to evaluate the contamination level. Additionally, the comparison of results from the classical genotoxiciw bioassay has confirmed the applicability of the method for real samoles.  相似文献

This work presents an electrochemical extraction of cerium and synthesization of Al–Ce alloy in LiCl–KCl melts on Mo and Al electrodes by chlorination of CeO2 using AlCl3 at 873 K. The cyclic voltammogram on Mo electrodes in LiCl–KCl–CeO2 melt showed no obvious reduction wave other than the reduction of Li(I). After the addition of AlCl3, the signals of the reaction of Ce(ⅡI)/Ce(0) and the synthesization of Al–Ce and Al–Li alloys were investigated by cyclic voltammetry, square-wave voltammetry, open-circuit chronopotentiometry and chronopotentiometry. These results indicated that AlCl3 can chloridize CeO2 and that it is possible to extract cerium and form Al–Ce and Al–Li–Ce alloys in LiCl–KCl–CeO2–AlCl3 melts. According to potentiostatic electrolysis, only the Al4 Ce layer coated the Al electrodes. According to galvanostatic electrolysis, Al–Ce(Al4Ce, Al3 Ce, and Al92Ce8), Al2Li3, and Al phases were formed on Mo electrodes, and the content of cerium in the Al–Li–Ce alloys was more than 17 wt%.  相似文献

This research involves the preparation of a biosensor using silicon oxide for biomedical applications, and its effective use for the detection of target DNA hybridization. An electrochemical DNA biosensor was successfully fabricated by using(3-aminopropyl) tri-ethoxysilane(APTES) as a linker molecule combined with gold nanoparticles(GNPs) on a thermally oxidized SiO_2 thin film. The size of the GNPs was calculated by utilizing UV–vis data with an average calculated particle size within the range of 30±5 nm, and characterization by transmission electron microscopy(TEM) and atomic force microscopy(AFM). The GNP-modified SiO_2 thin films were electrically characterized through the measurement of capacitance, permittivity and conductivity using a low-cost dielectric analyzer. The capacitance, permittivity and conductivity profiles of the fabricated sensor clearly differentiated DNA immobilization and hybridization.  相似文献

An effective procedure for constructing a DNA biosensor is developed based on covalent immobilization of NH_2 labeled,single strand DNA(NH_2-ssDNA) onto a self-assembled diazo-thiourea and gold nanoparticles modified Au electrode(diazo-thiourea/GNM/Au).Gold nano-particles expand the electrode surface area and increase the amount of immobilized thiourea and single stranded DNA(ssDNA) onto the electrode surface.Diazo-thiourea film provides a surface with high conductibility for electron transfer and a bed for the covalent coupling of NH_2-ssDNA onto the electrode surface.The immobilization and hybridization of the probe DNA on the modified electrode is studied by differential pulse voltammetry(DPV) using methylene blue(MB) as a well-known electrochemical hybridization indicator.The linear range for the determination of complementary target ssDNA is from 9.5(±0.1) × 10~(-13) mol/L to1.2(±0.2) x 10~(-9) mol/L with a detection limit of 1.2(±0.1) 10~(-13) mol/L.  相似文献

The electrochemical characteristics of poly(o-phenylendiamine) (POPD) film modified electrodes have been investigated using different electrochemical techniques.The main interest is focused on the effect of potential and film thickness on the electrode process.Good agreement has been found for the apparent diffusion coefficient estimated by chronocoulometry and impedance spec-troscopy.The charge transfer process within POPD films is diffusion processes at negative and positive overpotentials and electron hopping mechanism at formal potential.The POPD film conductivity of the oxidized state is better than that of the reduced state.For all electrode processes,the H+ may penetrate the film/electrolyte interface and take part in charge transfer or protonation-deprotonation of phenazine rings.  相似文献

Surface processes of CO2 reduction on Pt(210), Pt(310), and Pt(510) electrodes were studied by cyclic voltammetry. Different surface structures of these platinum single crystal electrodes were obtained by various treatment conditions. The experimental results illustrated that the electrocatalytic activity of Pt single crystal electrodes towards CO2 reduction is decreased in an order of Pt(210)>Pt(310)>Pt(510), i.e., with the decrease of (110) step density on well-defined surfaces. When the surfaces were reconstructed due to oxygen adsorption, the catalytic activity of all the three electrodes has been enhanced to a cer- tain extent. Although the activity order remains unchanged, the electrocatalytic activity has been en- hanced more significantly as the density of (110) step sites is more intensive on the Pt single crystal surface. It has revealed that the more open the surface structure is, the more active the Pt single crystal electrode will be, and the easier for the electrode to be transformed into a surface structure that exhib- its higher activity under external inductions. However, the relatively ordered surfaces of Pt single crystal electrode are comparatively stable under the same external inductions. The present study has gained knowledge on the interaction between CO2 and Pt single crystal electrode surfaces at a micro- scopic level, and thrown new insight into understanding the surface processes of electrocatalytic re- duction of CO2.  相似文献

The nanoporous TiO2 film electrodes have been prepared by a sol-gel deposition process The photostability of the electrodes in basic solutions has been studied. The results show that the photostability of the electrodes decreases rapidly in strong basic solutions with or without methanol. The reaction of holes to O^2- produces active O^2- atoms and the products O^2- atomsoxidize Ti^3 to Ti^4 on TiO2 film surface and subsurface. This results in the TiO2 film electrodes unstable in basic solutions both without methanol and with too low concentration.  相似文献

β-Cyclodextrin 1 was directly oxidized to the corresponding monoaldehyde 2 on their primary faces by cyclized 2-iodoxybenzoic acid(IBX) in DMSO, followed by the synthesis of β-cyclodextrin derivatives bearing Schiff-base group 3. A new chiral stationary phase(BCDS 6) was then prepared by immobilization of β-cyclodextrin derivative with α-Schiff base group onto the surface of sillica gel. A series of compounds with amino groups were readily separated using this CSE Methanol and acetonitrile were tested as the mobile phase while the influence of temperature and the addition of aqueous triethylammonium acetate buffer to the mobile phase was also innvestigated. A novel disulphide derivatised deuteroporphyrin 2,7,12,18-tetramethyl-13,17-(propionylaminoethyldithioethyl amino-formy-lethyl) -29,34-bis-(methoxyformyl)porphyrin(PDTEP,3) and its cobalt(Ⅱ) complex(Co(II)PDTEP,4) were conveniently synthesized. The disulphide functional group of 4 allowed its stable immobilization on gold electrodes.The modified electrode was characterized by IR and confirmed electrochemically and showed good stability and catalytic activity toward the electro-catalyzed reduction of hydrogen peroxide.  相似文献

Single-wall carbon nanotubes(SWNTs) modified gold electrodes were prepared by using two different methods.The electrochemical behavior of cytochrome c on the modified gold electrodes was investigated.The first kind of SWNT-modified electrode (noted as SWNT/Au electrode)was prepared by the adsorption of carboxylterminated SWNTs from DMF dispersion on the gold electrode.The oxidatively processed SWNT tips were covalently modified by coupling with amines (AET) to form amide linkage.Via Au-S chemical bonding,the self-assembled monolayer of thiol-unctionalized nanotubes on gold surface was fabricated so as to prepare the others SWNT-modified electrode (noted as SWNT/AET/Au electrode).It was shown from cyclic voltammetry cxperiments that cytochrome c exhibited direct electrochemical responses on the both electrodes, but only the current of controlled diffusion existed on the SWNT/Au electrode while both the currents of controlled diffusion and adsorption of cytochrome c occurred on the SWNT/AET/Au electrode.Photoelastic Modulation Infared Reflection Absorpthion Spectroscopy (PEM-IRRAS) and Quartz Crystal Microbalance (QCM) were employed to verify the adsorption of SWNTs on the gold electrodes.The results proved that SWNTs could enhance the direct electron transfer proecss between the electrodes and redox proteins.  相似文献

Lead dioxide electrodes on Ti substrates were prepared by thermal-deposition or electro-deposition. The amount of hydroxyl radicals generated at the electrodes prepared by the above-mentioned two methods was compared with that at the electrodes mingled with Bi or La prepared by electro-deposition. The experimental results indicate that the highest concentration of hydroxyl radicals generated by thermal-deposition, electro-deposition mingled with nothing, electro-deposition mingled with Bi or La was 0.781, 1.048, 1.838 or 2.044 μmol/L, respectively. When phenol was electrolyzed on the four electrodes at a current density of 30 mA/cm2, the removal efficiency of phenol after electrolysis for 1.5 h was 87.30%, 93.55%, 97.95% or 98.70%, TOC removal efficiency after electrolysis for 5 h was 86.76%, 94.26%, 98.53% or 99.60%, respectively. Through the degradation experiments of phenol, the amount of hydroxyl radicals was responsible for the removal efficiency of phenol. The electro-catalytic characteristics were investigated by SEM, the generation amount of hydroxyl radicals, the degradation degree of phenol and the stability and conductivity of the electrodes were also investigated. The experimental results indicate that the four electrodes all show good electro-catalytic characteristics; the electro-catalytic characteristics of the electrode mingled with La were superior to those of the other three ones, and the electrochemical degradation of phenol followed one-step reaction dynamics.  相似文献

Biosensors based on electronic conducting polymers appear particularly well suited to the requirements of modern biological analysis—multiparametric assays, high information density, and miniaturization. We describe a new methodology for the preparation of addressed DNA matrices. The process includes an electrochemically directed copolymerization of pyrrole and oligonucleotides bearing on their 5′ end a pyrrole moiety. The resulting polymer film deposited on the addressed electrode consists of pyrrole chains bearing covalently linked oligonucleotides (ODN). An oligonucleotide array was constructed on a silicon device bearing a matrix of 48 addressable 50 × 50 μm gold microelectrodes. This technology was successfully applied to the genotyping of hepatitis C virus in blood samples. Fluorescence detection results show good sensitivity and a high degree of spatial resolution. In addition, gravimetric studies carried out by the quartz crystal microbalance technique provide quantitative data on the amount of surface-immobilized species. In the case of ODN, it allows discrimination between hybridization and nonspecific adsorption. The need for versatile processes for the immobilization of biological species on surfaces led us to extend our methodology. A biotinylated surface was obtained by coelectropolymerization of pyrrole and biotin-pyrrole monomers. The efficiency for recognition (and consequently immobilization) of R-phycoerythrin-avidin was demonstrated by fluorescence detection. Copolymerization of decreasing ratios of pyrrole-biotin over pyrrole allowed us to obtain a decreasing scale of fluorescence.  相似文献

A multianalyte biosensor for the simultaneous determination of glucose and galactose was developed by immobilizing glucose oxidase (GOD) and galactose oxidase (GAO) on Nation-modified thin film platinum disk electrodes. The dual Pt working electrodes with disk shape and the surrounding ring shaped counter electrode were fabricated by thin film technology,which were integrated onto the same microchip. The response of the designed biosensor for glucose and galactose were linear up to 6.0mmol/L and 3.5mmol/L with sensitivities of 0.3μA/mmol/L and 0.12μA/mmol/L, respectively. No cross-talking effect was observed.  相似文献

A selective DNA sensing with zeptomole detection level is developed based on coulometric measurement of gold nanoparticle (AuNPs)-mediated electron transfer (ET) across a self-assembled monolayer on the gold electrode. After immobilization of a thiolated hairpin-structured DNA probe, an alkanethiol monolayer was self-assembled on the resultant electrode to block [Fe(CN)6 ]-3-/4in a solution from accessing the electrode. In the presence of DNA target, hybridization between the DNA probe and the DNA target breaks the stem duplex of DNA probe. Consequently, stem moiety at the 3′-end of the DNA probes was removed from the electrode surface and made available for hybridization with the reporter DNA-AuNPs conjugates (reporter DNA-AuNPs). The thiolated reporter DNA matches the stem moiety at the 3′-end of the DNA probe. AuNPs were then enlarged by immersing the electrode in a growth solution containing HAuCl 4 and H2O2 after the reporter DNA-AuNPs bound onto the electrode surface. The enlarged AuNPs on the electrode restored the ET between the electrode and the [Fe(CN)6]3 -/4- , as a result, amplified signals were achieved for DNA target detection using the coulometric measurement of Fe(CN)6 3- electro-reduction by prolonging the electrolysis time. The quantities of ET on the DNA sensor increased with the increase in DNA target concentration through a linear range of 3.0 fM to 1.0 pM when electrolysis time was set to 300 s, and the detection limit was 1.0 fM. Correspondingly, thousands of DNA (zeptomole) copies were detected in 10L samples. Furthermore, the DNA sensor showed excellent differentiation ability for single-base mismatch.  相似文献

A nanostructured Ni/Pd-Ni catalyst with high activity for methanol oxidation in alkaline solution was prepared by electrodeposition followed by galvanic replacement, that is, electrodeposition of Ni-Zn on a Ni coating with subsequent replacement of the Zn by Pd at the open circuit potential in a Pd-containing alkaline solution. The surface morphology and composition of the coatings were examined by energy dispersive X-ray spectroscopy and scanning electron microscopy. The Ni/Pd-Ni coatings were porous and were composed of discrete Pd nanoparticles of about 58 nm. The electrocatalytic activity of the Ni/Pd-Ni electrodes for the oxidation of methanol was examined by cyclic voltammetry and electrochemical impedance spectroscopy. The onset potentials for methanol oxidation on Ni/Pd-Ni were 0.077 V and 0.884 V, which were lower than those for flat Pd and smooth Ni electrodes, respectively. The anodic peak current densities of these electrodes were 4.33 and 8.34 times higher than those of flat Pd (58.4 mA/cm2 vs 13.47 mA/cm2) and smooth Ni (58.4 mA/cm2 vs 7 mA/cm2). The nanostructured Ni/Pd-Ni electrode is a promising catalyst for methanol oxidation in alkaline media for fuel cell application.  相似文献

The research work on preparation process of active materials of the rare earth ion selective electrodes and the valence of the electrode membrane were reported. The active materials were characterized by infrared spectrum, elemental analysis and XPS.We had studied the rare earth ion selective electrodes in which the active materials consist of functional polymers[1-3].
