BioCARS, a NIH‐supported national user facility for macromolecular time‐resolved X‐ray crystallography at the Advanced Photon Source (APS), has recently completed commissioning of an upgraded undulator‐based beamline optimized for single‐shot laser‐pump X‐ray‐probe measurements with time resolution as short as 100 ps. The source consists of two in‐line undulators with periods of 23 and 27 mm that together provide high‐flux pink‐beam capability at 12 keV as well as first‐harmonic coverage from 6.8 to 19 keV. A high‐heat‐load chopper reduces the average power load on downstream components, thereby preserving the surface figure of a Kirkpatrick–Baez mirror system capable of focusing the X‐ray beam to a spot size of 90 µm horizontal by 20 µm vertical. A high‐speed chopper isolates single X‐ray pulses at 1 kHz in both hybrid and 24‐bunch modes of the APS storage ring. In hybrid mode each isolated X‐ray pulse delivers up to ~4 × 1010 photons to the sample, thereby achieving a time‐averaged flux approaching that of fourth‐generation X‐FEL sources. A new high‐power picosecond laser system delivers pulses tunable over the wavelength range 450–2000 nm. These pulses are synchronized to the storage‐ring RF clock with long‐term stability better than 10 ps RMS. Monochromatic experimental capability with Biosafety Level 3 certification has been retained.  相似文献

Structural changes of heme‐heme oxygenase‐1 complex following carbon monoxide (CO) dissociation were studied by time‐resolved resonance Raman spectroscopy. We observed temporal changes for resonance Raman bands of the Fe–His stretch and the heme propionate bends in the subnanosecond and microsecond time regimes. These changes suggest structural rearrangements in the Fe–His linkage and the salt bridges of the heme propionates following CO dissociation. The present data supports the model proposed by an X‐ray crystallographic study. The Fe–His stretching mode exhibited an upshift until 30 µs after dissociation as the delay time increased. This is the first example of a CO‐dissociation‐induced strengthening of the Fe‐His linkage in hemeproteins. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

An accurate and simple method, Raman peak‐shift simulation, is proposed to determine the characteristics of a laser‐driven shock wave. Using the principle of the Raman peaks shifting at high pressure and the pressure distribution in the gauge layer, the profile of the Raman peak can be numerically simulated. Combined with time‐resolved Raman spectroscopy, some main characteristics of the shock wave were determined. In the experiment, polycrystalline anthracene was used as the pressure gauge. The pump–probe technique was used to obtain the time‐resolved Raman spectra of anthracene under shock loading. The velocity of the shock wave, the peak pressure and the rise time of the shock front were determined by simulating the experimental spectra numerically. The result shows that the method of Raman peak‐shift simulation is effective in obtaining the characteristics of a laser‐driven shock wave. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

A high‐repetition‐rate pump–probe experiment is presented, based on the asynchronous sampling approach. The low‐α mode at the synchrotron ANKA can be used for a time resolution down to the picosecond limit for the time‐domain sampling of the coherent THz emission as well as for hard X‐ray pump–probe experiments, which probe structural dynamics in the condensed phase. It is shown that a synchronization of better than 1 ps is achieved, and examples of phonon dynamics of semiconductors are presented.  相似文献

Well‐resolved Raman spectra of samples of nitrogen‐containing compounds were detected using a portable Raman instrument (Ahura First Defender XL) outdoors at a low ambient temperature of −15 °C and at an altitude of 2860 m (Pitztall, Austria). The portable Raman spectrometer tested here is equipped with a 785‐nm diode laser and a fixed frontal probe. Solid form of formamide, urea, 3‐methylpyridine, aniline, indene, 1‐(2‐aminoethyl)piperazine, indoline and benzofuran were detected unambiguously under high‐mountain field conditions. The main Raman features (strong, medium and partially weak bands) were observed at the correct wavenumber positions (with a spectral resolution 7–10 cm−1) in the wavenumber range 200–1600 cm−1. The results obtained demonstrate the possibility of employing a miniaturised Raman spectrometer as a key instrument for investigating the presence of nitrogen‐containing organic compounds and biomolecules outdoors under low temperature conditions. Within the payload designed by European Space Agency (ESA) and National Aeronautics and Space Administration (NASA) for future missions, focussing not only on Mars, Raman spectroscopy represents an important instrumentation for the detection of organic nitrogen‐containing compounds relevant to life detection on planetary surfaces or near sub‐surfaces. This article reports an efficient method to simulate time and frequency resolved coherent anti‐Stokes Raman scattering spectra measured with picosecond pump and probe fields and ultrashort Stokes pulses. A systematic comparison of measured and simulated time and frequency dependent data is presented for carbon tetrachloride, chloroform, cyclohexane, octane, and poly(methyl methacrylate). While the first compound exhibits no Raman active modes in the considered spectral region of the CH‐stretch vibrations, the other ones show Raman spectra of increasing complexity. Vibrational frequencies and homogeneous dephasing rates are extracted by fitting explicit analytical formulas to the recorded data. Interference between nonresonant and resonant contributions to the nonlinear polarization is taken fully into account. The ability to measure the influence of inhomogeneous broadening is discussed. The SOLEIL synchrotron radiation source is regularly operated in special filling modes dedicated to pump–probe experiments. Among others, the low‐α mode operation is characterized by shorter pulse duration and represents the natural bridge between 50 ps synchrotron pulses and femtosecond experiments. Here, the capabilities in low‐α mode of the experimental set‐ups developed at the TEMPO beamline to perform pump–probe experiments with soft X‐rays based on photoelectron or photon detection are presented. A 282 kHz repetition‐rate femtosecond laser is synchronized with the synchrotron radiation time structure to induce fast electronic and/or magnetic excitations. Detection is performed using a two‐dimensional space resolution plus time resolution detector based on microchannel plates equipped with a delay line. Results of time‐resolved photoelectron spectroscopy, circular dichroism and magnetic scattering experiments are reported, and their respective advantages and limitations in the framework of high‐time‐resolution pump–probe experiments compared and discussed.  相似文献

Time‐resolved Raman spectroscopy, spatially offset Raman spectroscopy and time‐resolved spatially offset Raman spectroscopy (TR‐SORS) have proven their capability for the non‐invasive profiling of deep layers of a sample. Recent studies have indicated that TR‐SORS exhibits an enhanced selectivity toward the deep layers of a sample. However, the enhanced depth profiling efficiency of TR‐SORS, in comparison with time‐resolved Raman spectroscopy and spatially offset Raman spectroscopy, is yet to be assessed and explained in accordance to the synergistic effects of spatial and temporal resolutions. This study provides a critical investigation of the depth profiling efficiency of the three deep Raman techniques. The study compares the efficiency of the various deep Raman spectroscopy techniques for the stand‐off detection of explosive precursors hidden in highly fluorescing packaging. The study explains for the first time the synergistic effects of spatial and temporal resolutions in the deep Raman techniques and their impact on the acquired spectral data. Non‐degenerate second‐order scattering due to interaction of infrared and ultraviolet pulses is observed in picosecond infrared‐pump/anti‐Stokes Raman‐probe experiments under electronic resonance conditions. We detected resonance hyper‐Rayleigh scattering at the sum frequency of the pulses as well as the corresponding frequency‐down‐shifted resonance hyper‐Raman lines. Nearly coinciding resonance hyper‐Raman and one‐photon resonance Raman spectra indicate conditions of A‐term resonance Raman scattering. Second‐order scattering is distinguished from transient anti‐Stokes Raman scattering of v = 1 to v = 0 transitions and v = 1 to v′ = 1 combination transitions by taking into account their different spectral and temporal behaviour. A new analysis method for the short excited‐state lifetime measurement of photosensitive species in crystals is described. Based on photocrystallographic techniques, this method is an alternative to spectroscopic methods and is also valid for non‐luminescent excited species. Two different approaches are described depending on the magnitude of the lifetime τ. For very short lifetimes below the width of the synchrotron pulse, an estimated τ can be obtained from the occurrence of the maximal system response as a function of the pump–probe delay time Δt. More precise estimates for both short and longer lifetimes can be achieved by a refinement of a model of the response as a function of the pump–probe delay time. The method also offers the possibility of the structure determination of excited species with lifetimes in the 40–100 ps range.  相似文献

The recent developments in X‐ray detectors have opened new possibilities in the area of time‐resolved pump/probe X‐ray experiments; this article presents the novel use of a PILATUS detector to achieve X‐ray pulse duration limited time‐resolution at the Advanced Photon Source (APS), USA. The capability of the gated PILATUS detector to selectively detect the signal from a given X‐ray pulse in 24 bunch mode at the APS storage ring is demonstrated. A test experiment performed on polycrystalline organic thin films of α‐perylene illustrates the possibility of reaching an X‐ray pulse duration limited time‐resolution of 60 ps using the gated PILATUS detector. This is the first demonstration of X‐ray pulse duration limited data recorded using an area detector without the use of a mechanical chopper array at the beamline.  相似文献

The molten globule (MG) state can aid in the folding of a protein to a functional structure and is loosely defined as an increase in structural disorder with conservation of the ensemble secondary structure content. Simultaneous observation of persistent secondary structure content with increased disorder has remained experimentally problematic. As a consequence, modeling how the MG state remains stable and how it facilitates proper folding remains difficult due to a lack of amenable spectroscopic techniques to characterize this class of partially unfolded proteins. Previously, deep‐UV resonance Raman (dUVRR) spectroscopy has proven useful in the resolution of global and local structural fluctuations in the secondary structure of proteins. In this work, dUVRR was employed to study the MG to ordered transition of a model four‐helix bundle protein, HP7. Both the average ensemble secondary structure and types of local disorder were monitored, without perturbation of the solvent, pH, or temperature. The MG to ordered transition is induced by stepwise coordination of two heme molecules. Persistent dUVRR spectral features in the amide III region at 1295–1301 and 1335–1338 cm−1 confirm previous observations that HP7 remains predominantly helical in the MG versus the fully ordered state. Additionally, these spectra represent the first demonstration of conserved helical content in a MG protein. With successive heme binding, significant losses are observed in the spectral intensity of the amide III3 and S regions (1230–1260 and 1390 cm−1, respectively), which are known to be sensitive to local disorder. These observations indicate that there is a decrease in the structural populations able to explore various extended conformations with successive heme binding events. DUVRR spectra indicate that the first heme coordination between two helical segments diminishes exploration of more elongated backbone structural conformations in the inter‐helical regions. This paper reviews our results on femtosecond time‐resolved spectroscopy (transient absorption, transient‐grating and fluorescence spectroscopy) to study the photophysics and photochemistry of the two very important biological photoreceptor chromophores phycocyanobilin (PCB) and protochlorophyllide a (PChla). The compound PCB serves as a model chromophore for the photoreceptor phytochrome. By means of transient‐grating spectroscopy where the excitation wavelength was varied ove r the spectral region of the S0S1‐absorption the ultrafast processes were studied upon excitation with varying excess energy delivered to the system. On the basis of the results obtained, both the rate of the photoreaction in PCB and the rate of the decay of different excited‐state species via different decay channels depend on the excitation wavelength. Furthermore, transient absorption experiments illuminating the excited‐state dynamics of PChla, a porphyrin‐like compound and, as substrate of the NADPH/protochlorophyllide oxidoreductase (POR), a precursor of the chlorophyll biosynthesis are presented. In addition to pump‐energy‐dependent measurements performed with PChla dissolved in methanol, the excited‐state dynamics of PChla was interrogated in different solvents that were chosen to mimic different environmental conditions. In addition to the femtosecond time‐resolved absorption experiments the picosecond time‐resolved fluorescence of the system was studied. The transient absorption and tim e‐resolved fluorescence data allow suggesting a detailed model for the excited‐state relaxation of PChla describing the excited‐state processes in terms of a branching of the initially excited state population into a reactive and nonreactive path. Thus, the excited‐state potential energy surface exhibits two distinct S1 and Sx minima separated from the Franck–Condon region along two most likely orthogonal reaction coordinates. Finally, the model derived is related to models suggested to acco unt for the reduction of PChla to chlorophyllide a within the natural enzymatic environment of POR.  相似文献

A new set‐up is presented to measure element‐selective magnetization dynamics using the ALICE chamber [Grabis et al. (2003), Rev. Sci. Instrum. 74 , 4048–4051] at the BESSY II synchrotron at the Helmholtz‐Zentrum Berlin. A magnetic‐field pulse serves as excitation, and the magnetization precession is probed by element‐selective X‐ray resonant magnetic scattering. With the use of single‐bunch‐generated X‐rays a temporal resolution well below 100 ps is reached. The ALICE diffractometer environment enables investigations of thin films, described here, multilayers and laterally structured samples in reflection or diffuse scattering geometry. The combination of the time‐resolved set‐up with a cryostat in the ALICE chamber will allow temperature‐dependent studies of precessional magnetization dynamics and of damping constants to be conducted over a large temperature range and for a large variety of systems in reflection geometry.  相似文献

A Johann‐type spectrometer for the study of high‐energy resolution fluorescence‐detected X‐ray absorption spectroscopy, X‐ray emission spectroscopy and resonant inelastic X‐ray scattering has been developed at BL14W1 X‐ray absorption fine structure spectroscopy beamline of Shanghai Synchrotron Radiation Facility. The spectrometer consists of three crystal analyzers mounted on a vertical motion stage. The instrument is scanned vertically and covers the Bragg angle range of 71.5–88°. The energy resolution of the spectrometer ranges from sub‐eV to a few eV. The spectrometer has a solid angle of about 1.87 × 0?3 of 4π sr, and the overall photons acquired by the detector could be 105 counts per second for the standard sample. The performances of the spectrometer are illustrated by the three experiments that are difficult to perform with the conventional absorption or emission spectroscopy. We describe a versatile infrared laser system capable of generating picosecond pulses continuously tunable over the wavelength range 3.3–8.4 μm, with peak powers of 1–20 MW. The IR pulse duration is ~1.3 ps with a time-bandwidth product of ~0.3. The system uses stimulated electronic Raman scattering (SERS) in caesium vapour to shift the frequency of a picosecond dye laser into the infrared. The quantum efficiency of the Raman conversion process exceeds 40% over almost the entire tuning range. This technique can be extended to other tuning ranges in the IR by using different atomic vapours and SERS transitions.  相似文献

We demonstrate a new technique that combines polarization sensitivity of the coherent anti‐Stokes Raman scattering (CARS) response with heterodyne amplification for background‐free detection of CARS signals. In this heterodyne interferometric polarization CARS (HIP‐CARS), the major drawbacks of polarization and heterodyne CARS are rectified. Using a home‐built picosecond optical parametric oscillator, we are able to address vibrational stretches between 600 and 1650 cm−1 and record continuous high‐resolution Raman equivalent HIP‐CARS spectra. The travelling-wave parametric process allows the generation of intense picosecond pulses tunable between 2700 and 17,000 cm−1. Angle- and temperature tuning was investigated using LiNbO3 crystals of different cuts. Various factors determining the bandwidth of the new pulses are evaluated and experimental spectra are presented. Frequency doubling readily leads to light pulses up to 32,000 cm−1. The jitter of two parametric systems operating in parallel is less than 1 ps for pump pulses of 7 ps.  相似文献

Time‐resolved X‐ray scattering patterns from photoexcited molecules in solution are in many cases anisotropic at the ultrafast time scales accessible at X‐ray free‐electron lasers (XFELs). This anisotropy arises from the interaction of a linearly polarized UV–Vis pump laser pulse with the sample, which induces anisotropic structural changes that can be captured by femtosecond X‐ray pulses. In this work, a method for quantitative analysis of the anisotropic scattering signal arising from an ensemble of molecules is described, and it is demonstrated how its use can enhance the structural sensitivity of the time‐resolved X‐ray scattering experiment. This method is applied on time‐resolved X‐ray scattering patterns measured upon photoexcitation of a solvated di‐platinum complex at an XFEL, and the key parameters involved are explored. It is shown that a combined analysis of the anisotropic and isotropic difference scattering signals in this experiment allows a more precise determination of the main photoinduced structural change in the solute, i.e. the change in Pt—Pt bond length, and yields more information on the excitation channels than the analysis of the isotropic scattering only. Finally, it is discussed how the anisotropic transient response of the solvent can enable the determination of key experimental parameters such as the instrument response function.  相似文献

The technical implementation of a multi‐MHz data acquisition scheme for laser–X‐ray pump–probe experiments with pulse limited temporal resolution (100 ps) is presented. Such techniques are very attractive to benefit from the high‐repetition rates of X‐ray pulses delivered from advanced synchrotron radiation sources. Exploiting a synchronized 3.9 MHz laser excitation source, experiments in 60‐bunch mode (7.8 MHz) at beamline P01 of the PETRA III storage ring are performed. Hereby molecular systems in liquid solutions are excited by the pulsed laser source and the total X‐ray fluorescence yield (TFY) from the sample is recorded using silicon avalanche photodiode detectors (APDs). The subsequent digitizer card samples the APD signal traces in 0.5 ns steps with 12‐bit resolution. These traces are then processed to deliver an integrated value for each recorded single X‐ray pulse intensity and sorted into bins according to whether the laser excited the sample or not. For each subgroup the recorded single‐shot values are averaged over ~107 pulses to deliver a mean TFY value with its standard error for each data point, e.g. at a given X‐ray probe energy. The sensitivity reaches down to the shot‐noise limit, and signal‐to‐noise ratios approaching 1000 are achievable in only a few seconds collection time per data point. The dynamic range covers 100 photons pulse?1 and is only technically limited by the utilized APD.  相似文献
