The synthesis of two naturally-occurring isomers of 3,4-dihydroxyproline is reported. l-2,3-cis-3,4-trans-3,4-Dihydroxyproline was synthesized from l-arabinose in 10 steps and 31% overall yield. The same series of reactions was employed to convert l-xylose to l-2,3-trans-3,4-trans-3,4-dihydroxyproline. Orthogonally protected versions of these amino acids were produced on gram scale, en route to the free amino acids, and these will serve as versatile intermediates in peptide synthesis. This synthetic strategy involved -Fmoc protection and protection of the C3 and C4 secondary alcohols as methoxyethoxymethyl (MEM) ethers.  相似文献

The synthesis of four UDP-GalNAc analogues (1-4) is described. The 3-, 4- and 6-deoxygenated analogues 1-3 were obtained by way of a divergent strategy starting from a 3,6-di-O-pivaloyl GlcNAc derivative as a common precursor. Analogue 4 bearing a N-trifluoroacetamido group was prepared from the trifluoromethylated oxazoline 24 as key intermediate. These compounds were designed to probe the substrate specificity of polypeptide-α-GalNAc transferases.  相似文献

These studies describe the chemoselective deprotection of trans-fused 2N,3O-oxazolidinone derivatives of N-acetyl-β-d-glucosamine. Selective opening of the oxazolidinone ring or N-deacetylation without ring opening is demonstrated. Certain amines are shown to efficiently afford C-2 ureido sugars under mild conditions. This work demonstrates the high degree of chemoselective manipulation possible with ring-fused 2N,3O-oxazolidinone derivatives of N-acetyl-d-glucosamine.  相似文献

Deoxyfluorination of alcohols was carried out using N,N-diethyl-α,α-difluoro-(m-methylbenzyl)amine (DFMBA). Primary alcohols were effectively converted to fluorides under microwave irradiation or conventional heating. Deoxyfluorination of an anomeric hydroxy group in sugars by DFMBA proceeded at below room temperature and glycosyl fluorides could be obtained in good yields. The deoxyfluorination reaction chemoselectively proceeded and various protecting groups on the sugar can survive under the reaction conditions.  相似文献

This paper reports the synthesis of new 4-phospho-d-erythronic acid derivatives, namely 4-phospho-d-erythronohydroxamic acid (1), 4-phospho-d-erythronohydrazide (2), and 4-phospho-d-erythronamide (3), and their kinetic evaluation as new competitive inhibitors of the isomerization reaction between d-ribose 5-phosphate and d-ribulose 5-phosphate catalyzed by spinach ribose-5-phosphate isomerase (RPI). By comparison to the only known RPI inhibitor, 4-phospho-d-erythronate (4, Ki=28 μM, Km/Ki=270), the hydroxamic acid 1, obtained by an eight-step synthesis from d-arabinose, appears as a new potent high-energy intermediate analogue inhibitor of the isomerization reaction (Ki=29 μM, Km/Ki=260).  相似文献

A general and efficient one-pot method for the synthesis of carbohydrate thionolactones from the corresponding 1-thiosugar is described involving the formation of an intermediate glycosyl S-tert-butyl thiosulfinate in situ by treatment of a thiol with commercially available tert-butylsulfinyl chloride in toluene at room temperature, followed by thermolysis. The method can also be used to generate reactive thioaldehydes and thioketones directly from thiols, which can be trapped in situ with a suitable diene.  相似文献

The glycosylation of alcohols by the new 2-O-MEM-substituted d-galactal-derived allyl epoxide affords the corresponding alkyl 2-O-MEM-3-deoxy-β-d-threo-hex-2-enopyranosides through a completely 1,4-regio- and a highly to completely substrate-dependent stereoselective glycosylation processes. The glycosides obtained can be regioselectively transformed into corresponding 3-deoxy-β-O-glycosides, 3-deoxy-β-d-threo-hexopyranosid-2-uloses, and 3,4-dideoxy-β-d-glycero-hex-3-enopyranosid-2-uloses, which are useful synthetic tools for further transformations.  相似文献

The glycosylation of alcohols, phenol, and partially protected monosaccharides with the diastereoisomeric d-allal and d-galactal-derived N-nosyl aziridines and leads to the corresponding 4-N-(nosylamino)-2,3-unsaturated-α-O- () and β-O-glycosides and disaccharides (), respectively, in a stereospecific substrate-dependent O-glycosylation process. The N-(nosylamino) group of and  can easily be deprotected to give the corresponding 4-amino-2,3-unsaturated-O-glycosides and , with an increased value to our glycosylation protocol.  相似文献

We studied the effects of storage temperature on the stability of dehydrated POPC (1-palmitoyl-2-oleoyl-phosphatidylcholine) mixed with sucrose, trehalose, or a sucrose/raffinose mixture. We used DSC to measure the gel-to-fluid phase transition temperature (T m) of POPC after incubation either below or near the glass transition temperature (T g) of the sugars in the mixture. Glass formation by the sugars around fluid-phase POPC led to the lowering ofT m below that of the fully hydrated lipid. Phospholipid phase behavior did not change during storage belowT g. In some samples stored aboveT g, trehalose crystallized completely; in these samples, theT g of POPC increased to that of the partially dehydrated phospholipid. Melting the crystalline sugar re-established its ability to lower POPC'sT m. We conclude that prevention of complete sugar crystallization was important for stability in the dry state, and that storage belowT g conferred long-term stability to the dehydrated sugar-lipid mixtures.  相似文献

1,2-cis-Fused bicyclic sugar thiocarbamates of gluco and manno configurations have been prepared by treatment of the corresponding O-unprotected amino sugars and glycopyranosyl amines with thiophosgene. The reactivity of these compounds towards amines has been studied in order to determine whether these compounds could act as latent isothiocyanates; it is shown that 1,2-cis-fused bicyclic sugar thiocarbamates are more stable than their trans analogues, and are not transformed into thioureas upon treatment with amines. An unprecedented isomerization of a peracetylated glucopyranoso[2,1-d]oxazolidine-2-thione into a glucopyranoso[2,1-d]thiazolidin-2-one in DMF is also reported. The structure of this thiazolidin-2-one was confirmed by X-ray crystallography.  相似文献

Purified lipases (via interfacial activation on hydrophobic supports) from different microbial extracts have been evaluated in the regio-selective hydrolysis of peracetylated sugars (peracetylated glucose, ribose and sucrose). Among the enzymes tested, lipases from Candida rugosa (CRL) and from Pseudomonas fluorescens (PFL) exhibited the best properties in these reactions.Then, we have prepared two different immobilized lipase preparations obtained by interfacial activation on hydrophobic supports or by covalent attachment on glutaraldehyde agarose. Interfacially activated lipases exhibited a higher activity than covalently attached enzymes (even by a 100-fold factor), giving the higher yields of mono deacetylated sugars (in some instances by more than a threefold factor) in short reaction times. In the hydrolysis of 1,2,3,5-tetra-O-acetyl-β-d-ribofuranose catalyzed by PFL adsorbed on octyl agarosa, hydrolyzed mainly the 3 position (30% of yield) while the CRL gave the hydrolysis only in position 5 (about 50% of yield).Depending on the enzyme immobilized preparation, we have been able also to obtain selective hydrolysis of 1,2,3,4,6-penta-O-acetyl-α/β-d-glucopyranose obtaining a free hydroxyl group in position 1, 4 or 6. Moreover, selective hydrolysis in the 4′ position of peracetylated sucrose was achieved when the hydrolysis is performed with CRL immobilized on octyl-agarose (yield was 77%).  相似文献

A route has been developed for the synthesis of enantiomerically pure trihydroxylated pyrrolizidines starting from l-erythrose glycosylhydroxylamine. The latter acts as a masked acyclic nitrone and reacts diastereoselectively from its Re-face with methyl acrylate to give the corresponding isoxazolidines, which after reductive N-O cleavage are recyclized to trihydroxypyrrolizidines via a Mitsunobu condensation.  相似文献

The stereoselective transformation of the cyclopropyl derivative 2, stereoselectively obtained with the Simmons-Smith reaction of methyl 2,6-di-O-benzyl-α-l-threo-hex-4-enopyranoside (1), into gabosines and deoxy-carbahexoses is described. The treatment of 2 with mercuric trifluoroacetate in dry methanol gives the organomercuric chloride 3, which by demercuration with lithium aluminum hydride affords the 4-C-deoxy-4-methyl-1,5-bis-glycoside 4. The acid hydrolysis of 4 produces a mixture of the two diastereoisomeric 2-methyl-cyclohex-2-enones 6 and 7, catalytically reduced to carba-sugars 10 and 11. Structures and stereochemistry of all isolated compounds were determined by 1D and 2D NMR experiments.  相似文献

The structure previously reported for coyolosa, a hypoglycemic substance isolated from natural sources, as a 6,6-ether linked pseudo-disaccharide derived from allose, is incorrect since 6-O-(6-deoxy-d-allos-6-yl)-d-allose 10, synthesised by an unequivocal route, and its derived peracetate 11, both have spectral properties, which differ from those reported for coyolosa and its peracetate. A versatile synthesis has been developed for this type of compound, containing two carbohydrate moieties joined through an ether linkage at their primary centres, which involves the displacement of a primary carbohydrate triflate with a carbohydrate alkoxide, and it allows the preparation of both symmetrical and unsymmetrical representatives of this class of compound.  相似文献

The purpose of this work was to investigate the physicochemical changes occurring during the thermal-based production of água-mel, a traditional Portuguese honey-related food product. The refractive index, color parameters (hue angle, H°; chroma, C*), and the content of total reducing sugars, glucose, fructose, total brown pigments, and 5-hydroxymethylfurfural were monitored along the entire production process, and their evolution was kinetically modelled. Thermal processing caused a gradual decrease in sugars, which was accompanied by the formation of brown pigments and 5-hydroxymethylfurfural, increased concentration of soluble solids as evaluated through refractive index measurements, as well as the appearance of darker colors. In particular, a zero-order kinetic model could explain the changes in H° and reducing sugars, while the evolution of refractive index, brown pigments, 5-hydroxymethylfurfural, C*, fructose, and glucose were best fitted using a first-order kinetics model.  相似文献
