Potassium is the most abundant intracellular metal in the body, playing vital roles in regulating intracellular fluid volume, nutrient transport, and cell-to-cell communication through nerve and muscle contraction. On the other hand, aberrant alterations in K+ homeostasis contribute to a diverse array of diseases spanning cardiovascular and neurological disorders to diabetes to kidney disease to cancer. There is an unmet need for studies of K+ physiology and pathology owing to the large differences in intracellular versus extracellular K+ concentrations ([K+]intra = 150 mM, [K+]extra = 3–5 mM). With a relative dearth of methods to reliably measure dynamic changes in intracellular K+ in biological specimens that meet the dual challenges of low affinity and high selectivity for K+, particularly over Na+, currently available fluorescent K+ sensors are largely optimized with high-affinity receptors that are more amenable for extracellular K+ detection. We report the design, synthesis, and biological evaluation of Ratiometric Potassium Sensor 1 (RPS-1), a dual-fluorophore sensor that enables ratiometric fluorescence imaging of intracellular potassium in living systems. RPS-1 links a potassium-responsive fluorescent sensor fragment (PS525) with a low-affinity, high-selectivity crown ether receptor for K+ to a potassium-insensitive reference fluorophore (Coumarin 343) as an internal calibration standard through ester bonds. Upon intracellular delivery, esterase-directed cleavage splits these two dyes into separate fragments to enable ratiometric detection of K+. RPS-1 responds to K+ in aqueous buffer with high selectivity over competing metal ions and is sensitive to potassium ions at steady-state intracellular levels and can respond to decreases or increases from that basal set point. Moreover, RPS-1 was applied for comparative screening of K+ pools across a panel of different cancer cell lines, revealing elevations in basal intracellular K+ in metastatic breast cancer cell lines vs. normal breast cells. This work provides a unique chemical tool for the study of intracellular potassium dynamics and a starting point for the design of other ratiometric fluorescent sensors based on two-fluorophore approaches that do not rely on FRET or related energy transfer designs.

Cations are critical for the folding and assembly of nucleic acids. In G-quadruplex structures, cations can bind between stacked G-tetrads and coordinate with negatively charged guanine carbonyl oxygens. They usually exchange between binding sites and with the bulk in solution with time constants ranging from sub-millisecond to seconds. Here we report the first observation of extremely long-lived K+ and NH4+ ions, with an exchange time constant on the order of an hour, when coordinated at the center of a left-handed G-quadruplex DNA. A single-base mutation, that switched one half of the structure from left- to right-handed conformation resulting in a right–left hybrid G-quadruplex, was shown to remove this long-lived behaviour of the central cation.

We report four new A-site vacancy ordered thiocyanate double double perovskites, , A = K+, NH4+, CH3(NH3)+ (MeNH3+) and C(NH2)3+ (Gua+), including the first examples of thiocyanate perovskites containing organic A-site cations. We show, using a combination of X-ray and neutron diffraction, that the structure of these frameworks depends on the A-site cation, and that these frameworks possess complex vacancy-ordering patterns and cooperative octahedral tilts distinctly different from atomic perovskites. Density functional theory calculations uncover the energetic origin of these complex orders and allow us to propose a simple rule to predict favoured A-site cation orderings for a given tilt sequence. We use these insights, in combination with symmetry mode analyses, to show that these complex orders suggest a new route to non-centrosymmetric perovskites, and mean this family of materials could contain excellent candidates for piezo- and ferroelectric applications.

The local electronic structure of the metal-active site and the deexcitation pathways of metalloporphyrins are crucial for numerous applications but difficult to access by commonly employed techniques. Here, we applied near-edge X-ray absorption mass spectrometry and quantum-mechanical restricted active space calculations to investigate the electronic structure of the metal-active site of the isolated cobalt(iii) protoporphyrin IX cation (CoPPIX+) and its deexcitation pathways upon resonant absorption at the cobalt L-edge. The experiments were carried out in the gas phase, thus allowing for control over the chemical state and molecular environment of the metalloporphyrin. The obtained mass spectra reveal that resonant excitations of CoPPIX+ at the cobalt L3-edge lead predominantly to the formation of the intact radical dication and doubly charged fragments through losses of charged and neutral side chains from the macrocycle. The comparison between experiment and theory shows that CoPPIX+ is in a 3A2g triplet ground state and that competing excitations to metal-centred non-bonding and antibonding σ* molecular orbitals lead to distinct deexcitation pathways.

Alkali ion intercalation is fundamental to battery technologies for a wide spectrum of potential applications that permeate our modern lifestyle, including portable electronics, electric vehicles, and the electric grid. In spite of its importance, the Nernstian nature of the charge transfer process describing lithiation of carbon has not been described previously. Here we use the ultrathin few-layer graphene (FLG) with micron-sized grains as a powerful platform for exploring intercalation and co-intercalation mechanisms of alkali ions with high versatility. Using voltammetric and chronoamperometric methods and bolstered by density functional theory (DFT) calculations, we show the kinetically facile co-intercalation of Li+ and K+ within an ultrathin FLG electrode. While changes in the solution concentration of Li+ lead to a displacement of the staging voltammetric signature with characteristic slopes ca. 54–58 mV per decade, modification of the K+/Li+ ratio in the electrolyte leads to distinct shifts in the voltammetric peaks for (de)intercalation, with a changing slope as low as ca. 30 mV per decade. Bulk ion diffusion coefficients in the carbon host, as measured using the potentiometric intermittent titration technique (PITT) were similarly sensitive to solution composition. DFT results showed that co-intercalation of Li+ and K+ within the same layer in FLG can form thermodynamically favorable systems. Calculated binding energies for co-intercalation systems increased with respect to the area of Li+-only domains and decreased with respect to the concentration of –K–Li– phases. While previous studies of co-intercalation on a graphitic anode typically focus on co-intercalation of solvents and one particular alkali ion, this is to the best of our knowledge the first study elucidating the intercalation behavior of two monovalent alkali ions. This study establishes ultrathin graphitic electrodes as an enabling electroanalytical platform to uncover thermodynamic and kinetic processes of ion intercalation with high versatility.

Functional nanoporous materials are widely explored for CO2 separation, in particular, small-pore aluminosilicate zeolites having a “trapdoor” effect. Such an effect allows the specific adsorbate to push away the sited cations inside the window followed by exclusive admission to the zeolite pores, which is more advantageous for highly selective CO2 separation. Herein, we demonstrated that the protonated organic structure-directing agent in the small-pore silicoaluminophosphate (SAPO) RHO zeolite can be directly exchanged with Na+, K+, or Cs+ and that the Na+ form of SAPO-RHO exhibited unprecedented separation for CO2/CH4, superior to all of the nanoporous materials reported to date. Rietveld refinement revealed that Na+ is sited in the center of the single eight-membered ring (s8r), while K+ and Cs+ are sited in the center of the double 8-rings (d8rs). Theoretical calculations showed that the interaction between Na+ and the s8r in SAPO-RHO was stronger than that in aluminosilicate RHO, giving an enhanced “trapdoor” effect and record high selectivity for CO2 with the separation factor of 2196 for CO2/CH4 (0.02/0.98 bar). The separation factor of Na-SAPO-RHO for CO2/N2 was 196, which was the top level among zeolitic materials. This work opens a new avenue for gas separation by using diverse silicoaluminophosphate zeolites in terms of the cation-tailored “trapdoor” effect.

Potassium channels modulate various cellular functions through efficient and selective conduction of K+ ions. The mechanism of ion conduction in potassium channels has recently emerged as a topic of debate. Crystal structures of potassium channels show four K+ ions bound to adjacent binding sites in the selectivity filter, while chemical intuition and molecular modeling suggest that the direct ion contacts are unstable. Molecular dynamics (MD) simulations have been instrumental in the study of conduction and gating mechanisms of ion channels. Based on MD simulations, two hypotheses have been proposed, in which the four-ion configuration is an artifact due to either averaged structures or low temperature in crystallographic experiments. The two hypotheses have been supported or challenged by different experiments. Here, MD simulations with polarizable force fields validated by ab initio calculations were used to investigate the ion binding thermodynamics. Contrary to previous beliefs, the four-ion configuration was predicted to be thermodynamically stable after accounting for the complex electrostatic interactions and dielectric screening. Polarization plays a critical role in the thermodynamic stabilities. As a result, the ion conduction likely operates through a simple single-vacancy and water-free mechanism. The simulations explained crystal structures, ion binding experiments and recent controversial mutagenesis experiments. This work provides a clear view of the mechanism underlying the efficient ion conduction and demonstrates the importance of polarization in ion channel simulations.

The NLRP3 inflammasome regulates production of the pro-inflammatory cytokines interleukin-1β (IL-1β) and IL-18, and contributes to inflammation exacerbating disease. Fenamate non-steroidal anti-inflammatory drugs (NSAIDs) were recently described as NLRP3 inflammasome inhibitors via chloride channel inhibition. Fenamate NSAIDs inhibit cyclooxygenase (COX) enzymes, limiting their potential as therapeutics for NLRP3-associated diseases due to established side effects. The aim here was to develop properties of the fenamates that inhibit NLRP3, and at the same time to reduce COX inhibition. We synthesised a library of analogues, with feedback from in silico COX docking potential, and IL-1β release inhibitory activity. Through iterative screening and rational chemical design, we established a collection of chloride channel inhibiting active lead molecules with potent activity at the canonical NLRP3 inflammasome and no activity at COX enzymes, but only in response to stimuli that activated NLRP3 by a K+ efflux-dependent mechanism. This study identifies a model for the isolation and removal of unwanted off-target effects, with the enhancement of desired activity, and establishes a new chemical motif for the further development of NLRP3 inflammasome inhibitors.

Dye-sensitised photoanodes modified with a water oxidation catalyst allow for solar-driven O2 evolution in photoelectrochemical cells. However, organic chromophores are generally considered unsuitable to drive the thermodynamically demanding water oxidation reaction, mainly due to their lack of stability upon photoexcitation. Here, the synthesis of a dyad photocatalyst (DPP-Ru) consisting of a diketopyrrolopyrrole chromophore (DPPdye) and ruthenium-based water oxidation catalyst (RuWOC) is described. The DPP-Ru dyad features a cyanoacrylic acid anchoring group for immobilisation on metal oxides, strong absorption in the visible region of the electromagnetic spectrum, and photoinduced hole transfer from the dye to the catalyst unit. Immobilisation of the dyad on a mesoporous TiO2 scaffold was optimised, including the use of a TiCl4 pretreatment method as well as employing chenodeoxycholic acid as a co-adsorbent, and the assembled dyad-sensitised photoanode achieved O2 evolution using visible light (100 mW cm−2, AM 1.5G, λ > 420 nm). An initial photocurrent of 140 μA cm−2 was generated in aqueous electrolyte solution (pH 5.6) under an applied potential of +0.2 V vs. NHE. The production of O2 has been confirmed by controlled potential electrolysis with a faradaic efficiency of 44%. This study demonstrates that metal-free dyes are suitable light absorbers in dyadic systems for the assembly of water oxidising photoanodes.

We report the reactivity between the water stable Lewis acidic trioxatriangulenium ion (TOTA+) and a series of Lewis bases such as phosphines and N-heterocyclic carbene (NHC). The nature of the Lewis acid–base interaction was analyzed via variable temperature (VT) NMR spectroscopy, single-crystal X-ray diffraction, UV-visible spectroscopy, and DFT calculations. While small and strongly nucleophilic phosphines, such as PMe3, led to the formation of a Lewis acid–base adduct, frustrated Lewis pairs (FLPs) were observed for sterically hindered bases such as P(tBu)3. The TOTA+–P(tBu)3 FLP was characterized as an encounter complex, and found to promote the heterolytic cleavage of disulfide bonds, formaldehyde fixation, dehydrogenation of 1,4-cyclohexadiene, heterolytic cleavage of the C–Br bonds, and interception of Staudinger reaction intermediates. Moreover, TOTA+ and NHC were found to first undergo single-electron transfer (SET) to form [TOTA]·[NHC]˙+, which was confirmed via electron paramagnetic resonance (EPR) spectroscopy, and subsequently form a [TOTA–NHC]+ adduct or a mixture of products depending the reaction conditions used.

A cationic nickel complex of the bis(8-quinolyl)(3,5-di-tert-butylphenoxy)phosphine (NPN) ligand, [(NPN)NiCl]+, is a precursor to efficient catalysts for the hydrosilation of alkenes with a variety of hydrosilanes under mild conditions and low catalyst loadings. DFT studies reveal the presence of two coupled catalytic cycles based on [(NPN)NiH]+ and [(NPN)NiSiR3]+ active species, with the latter being more efficient for producing the product. The preferred silyl-based catalysis is not due to a more facile insertion of alkene into the Ni–Si (vs. Ni–H) bond, but by consistent and efficient conversions of the hydride to the silyl complex.

The unexpected phenomenon and mechanism of the alkali metal involved NH3 selective catalysis are reported. Incorporation of K+ (4.22 wt %) in the tunnels of α‐MnO2 greatly improved its activity at low temperature (50–200 °C, 100 % conversion of NOx vs. 50.6 % conversion over pristine α‐MnO2 at 150 °C). Experiment and theory demonstrated the atomic role of incorporated K+ in α‐MnO2. Results showed that K+ in the tunnels could form a stable coordination with eight nearby O atoms. The columbic interaction between the trapped K+ and O atoms can rearrange the charge population of nearby Mn and O atoms, thus making the topmost five‐coordinated unsaturated Mn cations (Mn5c, the Lewis acid sites) more positive. Therefore, the more positively charged Mn5c can better chemically adsorb and activate the NH3 molecules compared with its pristine counterpart, which is crucial for subsequent reactions.  相似文献

Methylalumoxane (MAO), a perennially useful activator for olefin polymerization precatalysts, is famously intractable to structural elucidation, consisting as it does of a complex mixture of oligomers generated from hydrolysis of pyrophoric trimethylaluminum (TMA). Electrospray ionization mass spectrometry (ESI-MS) is capable of studying those oligomers that become charged during the activation process. We have exploited that ability to probe the synthesis of MAO in real time, starting less than a minute after the mixing of H2O and TMA and tracking the first half hour of reactivity. We find that the process does not involve an incremental build-up of oligomers; instead, oligomerization to species containing 12–15 aluminum atoms happens within a minute, with slower aggregation to higher molecular weight ions. The principal activated product of the benchtop synthesis is the same as that observed in industrial samples, namely [(MeAlO)16(Me3Al)6Me], and we have computationally located a new sheet structure for this ion 94 kJ mol−1 lower in Gibbs free energy than any previously calculated.

The activator methylaluminoxane is made by hydrolysis of trimethylaluminum. Analysis using ESI-MS reveals rapid formation of small oligomers is followed by slower aggregation to the larger precursors most capable of releasing [Me2Al]+.  相似文献

We present the results of acid–base experiments performed at the single ion (H+ or OH) limit in ∼6 aL volume nanopores incorporating electrochemical zero-mode waveguides (E-ZMWs). At pH 3 each E-ZMW nanopore contains ca. 3600H+ ions, and application of a negative electrochemical potential to the gold working electrode/optical cladding layer reduces H+ to H2, thereby depleting H+ and increasing the local pH within the nanopore. The change in pH was quantified by tracking the intensity of fluorescein, a pH-responsive fluorophore whose intensity increases with pH. This behavior was translated to the single ion limit by changing the initial pH of the electrolyte solution to pH 6, at which the average pore occupancy 〈npore ∼3.6H+/nanopore. Application of an electrochemical potential sufficiently negative to change the local pH to pH 7 reduces the proton nanopore occupancy to 〈npore ∼0.36H+/nanopore, demonstrating that the approach is sensitive to single H+ manipulations, as evidenced by clear potential-dependent changes in fluorescein emission intensity. In addition, at high overpotential, the observed fluorescence intensity exceeded the value predicted from the fluorescence intensity-pH calibration, an observation attributed to the nucleation of H2 nanobubbles as confirmed both by calculations and the behavior of non-pH responsive Alexa 488 fluorophore. Apart from enhancing fundamental understanding, the approach described here opens the door to applications requiring ultrasensitive ion sensing, based on the optical detection of H+ population at the single ion limit.

Heterocyclic orifices in cage-opened fullerene derivatives are regarded as potential ligands toward metals or ions, being reminiscent of truncated fullerenes as a hypothetical class of macrocycles with spherical π-conjugation. Among a number of cage-opened examples reported thus far, the coordination ability and dynamic behavior in solution still remained unclear due to difficulties in structural determination with multiple coordination sites on the macrocycles. Herein, we present the detailed solution dynamics of a cage-opened C60 derivative bearing a diketo bis(hemiketal) moiety in the presence of alkali metal ions. The NMR spectroscopy disclosed the coordination behavior which is identified as a two-step process with a 1 : 2 stoichiometry. Upon coordination to the Li+ ion, the macrocycle largely varies its properties, i.e., increased absorption coefficients in the visible region due to weakly-allowed charge transfer transitions as well as the inner potential field from neutral to positive by the charge delocalization along with the spherical π-surface. The Li+-complexes formed in situ underwent unprecedented selective dehydroxyhydrogenation under high-pressure conditions. These findings would facilitate further studies on fullerene-based macrocycles as metal sensors, bulky ligands in organic reactions, and ion carriers in batteries and biosystems.

A new type of crystalline solid, termed “solvate sponge crystal”, is presented, and the chemical basis of its properties are explained for a melt- and press-castable solid sodium ion conductor. X-ray crystallography and atomistic simulations reveal details of atomic interactions and clustering in (DMF)3NaClO4 and (DMF)2NaClO4 (DMF = N-N′-dimethylformamide). External pressure or heating results in reversible expulsion of liquid DMF from (DMF)3NaClO4 to generate (DMF)2NaClO4. The process reverses upon the release of pressure or cooling. Simulations reveal the mechanism of crystal “juicing,” as well as melting. In particular, cation–solvent clusters form a chain of octahedrally coordinated Na+–DMF networks, which have perchlorate ions present in a separate sublattice space in 3 : 1 stoichiometry. Upon heating and/or pressing, the Na+⋯DMF chains break and the replacement of a DMF molecule with a ClO4 anion per Na+ ion leads to the conversion of the 3 : 1 stoichiometry to a 2 : 1 stoichiometry. The simulations reveal the anisotropic nature of pressure induced stoichiometric conversion. The results provide molecular level understanding of a solvate sponge crystal with novel and desirable physical castability properties for device fabrication.

The ring-opening oxidative amination of methylenecyclopropanes (MCPs) with diazenes catalyzed by py3TiCl2(NR) complexes is reported. This reaction selectively generates branched α-methylene imines as opposed to linear α,β-unsaturated imines, which are difficult to access via other methods. Products can be isolated as the imine or hydrolyzed to the corresponding ketone in good yields. Mechanistic investigation via density functional theory suggests that the regioselectivity of these products results from a Curtin–Hammett kinetic scenario, where reversible β-carbon elimination of a spirocyclic [2 + 2] azatitanacyclobutene intermediate is followed by selectivity-determining β-hydrogen elimination of the resulting metallacycle. Further functionalizations of these branched α-methylene imine products are explored, demonstrating their utility as building blocks.

Loading Ag and Co dual cocatalysts on Al-doped SrTiO3 (AgCo/Al-SrTiO3) led to a significantly improved CO-formation rate and extremely high selectivity toward CO evolution (99.8%) using H2O as an electron donor when irradiated with light at wavelengths above 300 nm. Furthermore, the CO-formation rate over AgCo/Al-SrTiO3 (52.7 μmol h−1) was a dozen times higher than that over Ag/Al-SrTiO3 (4.7 μmol h−1). The apparent quantum efficiency for CO evolution over AgCo/Al-SrTiO3 was about 0.03% when photoirradiated at a wavelength at 365 nm, with a CO-evolution selectivity of 98.6% (7.4 μmol h−1). The Ag and Co cocatalysts were found to function as reduction and oxidation sites for promoting the generation of CO and O2, respectively, on the Al-SrTiO3 surface.

The development of photocatalysts is an essential task for clean energy generation and establishing a sustainable society. This paper describes the aggregation-induced photocatalytic activity (AI-PCA) of amphiphilic rhodamines and photocatalytic functions of the supramolecular assemblies. The supramolecular assemblies consisting of amphiphilic rhodamines with octadecyl alkyl chains exhibited significant photocatalytic activity under visible light irradiation in water, while the corresponding monomeric rhodamines did not exhibit photocatalytic activity. The studies on the photocatalytic mechanism by spectroscopic and microscopic analyses clearly demonstrated the AI-PCA of the rhodamines. Moreover, the supramolecular assemblies of the rhodamines exhibited excellent photocatalytic hydrogen evolution rates (up to 5.9 mmol g−1 h−1).

Simple amphiphilic rhodamines formed supramolecular assemblies and exhibited aggregation-induced photocatalytic activity and hydrogen evolution in water.  相似文献
