We have previously described the efficient guidance and unidirectional sliding of actin filaments along nanosized tracks with adsorbed heavy meromyosin (HMM; myosin II motor fragment). In those experiments, the tracks were functionalized with trimethylchlorosilane (TMCS) by chemical vapor deposition (CVD) and surrounded by hydrophilic areas. Here we first show, using in vitro motility assays on nonpatterned and micropatterned surfaces, that the quality of HMM function on CVD-TMCS is equivalent to that on standard nitrocellulose substrates. We further examine the influences of physical properties of different surfaces (glass, SiO(2), and TMCS) and chemical properties of the buffer solution on motility. With the presence of methylcellulose in the assay solution, there was HMM-induced actin filament sliding on both glass/SiO(2) and on TMCS, but the velocity was higher on TMCS. This difference in velocity increased with decreasing contact angles of the glass and SiO(2) surfaces in the range of 20-67 degrees (advancing contact angles for water droplets). The corresponding contact angle of CVD-TMCS was 81 degrees. In the absence of methylcellulose, there was high-quality motility on TMCS but no motility on glass/SiO(2). This observation was independent of the contact angle of the glass/SiO(2) surfaces and of HMM incubation concentrations (30-150 microg mL(-)(1)) and ionic strengths of the assay solution (20-50 mM). Complete motility selectivity between TMCS and SiO(2) was observed for both nonpatterned and for micro- and nanopatterned surfaces. Spectrophotometric analysis of HMM depletion during incubation, K/EDTA ATPase measurements, and total internal reflection fluorescence spectroscopy of HMM binding showed only minor differences in HMM surface densities between TMCS and SiO(2)/glass. Thus, the motility contrast between the two surface chemistries seems to be attributable to different modes of HMM binding with the hindrance of actin binding on SiO(2)/glass.  相似文献

A facile method to produce very hydrophobic surface was developed simply using the mixture of TiO(2) nanoparticles and poly(methyl methacrylate) (PMMA). When the volume ratio of TiO(2) to PMMA is between 35 vol.% and 50 vol.%, the mixture of two hydrophilic materials became very hydrophobic. Analysis of the molecular structure by infrared spectroscopy shows that the preferential orientation and attachment of a carbonyl group of a polymer molecule to the surface of TiO(2) nanoparticles play a critical role in the appearance of the strong hydrophobicity. When this composite was exposed to UV light, PMMA on the surface of the mixture film was decomposed through a photocatalytic reaction and the very hydrophobic surface turned to the superhydrophilic one. The superhydrophilic property of UV-irradiated composite lasts as long as several months. This long lasting superhydrophilicity is ascribed to the porous structure, which provides high roughness and strong capillary interaction. Easy transition from strong hydrophobicity to superhydrophilicity in TiO(2)/PMMA composites could be applied to designing high contrast hydrophilic/hydrophobic patterns.  相似文献

The diblock copolymer poly(methyl methacrylate)‐b‐poly(sodium sulfonated glycidyl methacrylate) (PMMA‐b‐PSGMA) was end‐attached by its hydrophobic block (PMMA) onto mica hydrophobized by a stearic trimethylammonium iodide (STAI) layer, to form a polyelectrolyte brush immersed in water. With a surface force balance (SFB), we extended earlier measurements between two such brush layers for the case of normal and shear forces at different shear rates, surface separation, and compressions between one mica surface coated with STAI or a STAI‐diblock layer against a bare mica surface. After coating one of the surfaces with STAI, a long range attraction that results in a jump into an adhesive flat contact between the hydrophobic and hydrophilic surfaces was observed. A very different behavior was seen after forming the polyelectrolyte brush on the STAI‐coated surface. The long range attraction was replaced by repulsion, accompanied by very low friction during shear (ca. three orders of magnitude lower than with adsorbed polyelectrolytes). On further compression, a weak attraction to the adhesive contact was observed. From the final surface–surface contact separation, we deduce that most of the polyelectrolyte diblock brush layer was squeezed out from the gap, leaving the STAI layer and a small amount of the polymer attached to the surface. Stick‐sliding behavior was seen while applying shear, suggesting a dissipation mechanism caused by the trapped polyelectrolyte. © 2004 Wiley Periodicals, Inc. The in vitro motility assay is valuable for fundamental studies of actomyosin function and has recently been combined with nanostructuring techniques for the development of nanotechnological applications. However, the limited understanding of the interaction mechanisms between myosin motor fragments (heavy meromyosin, HMM) and artificial surfaces hampers the development as well as the interpretation of fundamental studies. Here we elucidate the HMM-surface interaction mechanisms for a range of negatively charged surfaces (silanized glass and SiO2), which is relevant both to nanotechnology and fundamental studies. The results show that the HMM-propelled actin filament sliding speed (after a single injection of HMM, 120 microg/mL) increased with the contact angle of the surfaces (in the range of 20-80 degrees). However, quartz crystal microbalance (QCM) studies suggested a reduction in the adsorption of HMM (with coupled water) under these conditions. This result and actin filament binding data, together with previous measurements of the HMM density (Sundberg, M.; Balaz, M.; Bunk, R.; Rosengren-Holmberg, J. P.; Montelius, L.; Nicholls, I. A.; Omling, P.; T?gerud, S.; M?nsson, A. Langmuir 2006, 22, 7302-7312. Balaz, M.; Sundberg, M.; Persson, M.; Kvassman, J.; M?nsson, A. Biochemistry 2007, 46, 7233-7251), are consistent with (1) an HMM monolayer and (2) different HMM configurations at different contact angles of the surface. More specifically, the QCM and in vitro motility assay data are consistent with a model where the molecules are adsorbed either via their flexible C-terminal tail part (HMMC) or via their positively charged N-terminal motor domain (HMMN) without other surface contact points. Measurements of zeta potentials suggest that an increased contact angle is correlated with a reduced negative charge of the surfaces. As a consequence, the HMMC configuration would be the dominant configuration at high contact angles but would be supplemented with electrostatically adsorbed HMM molecules (HMMN configuration) at low contact angles. This would explain the higher initial HMM adsorption (from probability arguments) under the latter conditions. Furthermore, because the HMMN mode would have no actin binding it would also account for the lower sliding velocity at low contact angles. The results are compared to previous studies of the microtubule-kinesin system and are also discussed in relation to fundamental studies of actomyosin and nanotechnological developments and applications.  相似文献

The ability of polystyrene nanoparticles to facilitate the froth flotation of glass beads was correlated to the hydrophobicity of the nanoparticles. Contact angle measurements were used to probe the hydrophobicity of hydrophilic glass surfaces decorated with hydrophobic nanoparticles. Both sessile water drop advancing angles, θ(a), and attached air bubble receding angle measurements, θ(r), were performed. For glass surfaces saturated with adsorbed nanoparticles, flotation recovery, a measure of flotation efficiency, increased with increasing values of each type of contact angle. As expected, the advancing water contact angle on nanoparticle-decorated, dry glass surfaces increased with surface coverage, the area fraction of glass covered with nanoparticles. However, the nanoparticles were far more effective at raising the contact angle than the Cassie-Baxter prediction, suggesting that with higher nanoparticle coverages the water did not completely wet the glass surfaces between the nanoparticles. A series of polystyrene nanoparticles was prepared to cover a range of surface energies. Water contact angle measurements, θ(np), on smooth polymer films formed from organic solutions of dissolved nanoparticles were used to rank the nanoparticles in terms of hydrophobicity. Glass spheres were saturated with adsorbed nanoparticles and were isolated by flotation. The minimum nanoparticle water contact angle to give high flotation recovery was in the range of 51° < θ(np(min)) ≤ 85°.  相似文献

This paper presents a simple method to change the hydrophilic nature of the glass surface in a poly(dimethylsiloxane) (PDMS)-glass hybrid microfluidic device to hydrophobic by an extra-heating step during the fabrication process. Glass substrates bonded to a native or oxygen plasma-treated PDMS chip having microchambers (12.5 mm diameter, 110 μm height) were heated at 200°C for 3 h, and then the hydrophobicity of the glass surfaces on the substrate was evaluated by measuring the contact angle of water. By the extra-heating process, the glass surfaces became hydrophobic, and its contact angle was around 109°, which is nearly the same as native PDMS surfaces. To demonstrate the usefulness of this surface modification method, a PDMS-glass hybrid microfluidic device equipped with microcapillary vent structures for pneumatic manipulation of droplets was fabricated. The feasibility of the microcapillary vent structures on the device with the hydrophobic glass surfaces are confirmed in practical use through leakage tests of the vent structures and liquid handling for the electrophoretic separation of DNA molecules.  相似文献

Time-dependent bacterial adhesion forces of four strains of Staphylococcus epidermidis to hydrophobic and hydrophilic surfaces were investigated. Initial adhesion forces differed significantly between the two surfaces and hovered around -0.4 nN. No unambiguous effect of substratum surface hydrophobicity on initial adhesion forces for the four different S. epidermidis strains was observed. Over time, strengthening of the adhesion forces was virtually absent on hydrophobic dimethyldichlorosilane (DDS)-coated glass, although in a few cases multiple adhesion peaks developed in the retract curves. Bond-strengthening on hydrophilic glass occurred within 5-35 s to maximum adhesion forces of -1.9 +/- 0.7 nN and was concurrent with the development of multiple adhesion peaks upon retract. Poisson analysis of the multiple adhesion peaks allowed separation of contributions of hydrogen bonding from other nonspecific interaction forces and revealed a force contribution of -0.8 nN for hydrogen bonding and +0.3 nN for other nonspecific interaction forces. Time-dependent bacterial adhesion forces were comparable for all four staphylococcal strains. It is concluded that, on DDS-coated glass, the hydrophobic effect causes instantaneous adhesion, while strengthening of the bonds on hydrophilic glass is dominated by noninstantaneous hydrogen bond formation.  相似文献

The S-layer present at the outermost cell surface of some lactobacillus species is known to convey hydrophobicity to the lactobacillus cell surface. Yet, it is commonly found that adhesion of lactobacilli to solid substrata does not proceed according to expectations based on cell surface hydrophobicity. In this paper, the role of cell surface hydrophobicity of two lactobacillus strains with and without a surface layer protein (SLP) layer has been investigated with regard to their adhesion to hydrophobically or hydrophilically functionalized glass surfaces under well-defined flow conditions and in low and high ionic strength suspensions. Similarly, the interaction of the lactobacilli with similarly functionalized atomic force microscope (AFM) tips was measured. In a low ionic strength suspension, both lactobacillus strains show higher initial deposition rates to hydrophobic glass than to hydrophilic glass, whereas in a high ionic strength suspension no clear influence of cell surface hydrophobicity on adhesion is observed. Independent of ionic strength, however, AFM detects stronger interaction forces when both bacteria and tip are hydrophobic or hydrophilic than when bacteria and tip have opposite hydrophobicities. This suggest that the interaction develops in a different way when a bacterium is forced into contact with the tip surface, like in AFM, as compared with contacts developing between a cell surface and a macroscopic substratum under flow. In addition, the distance dependence of the total Gibbs energy of interaction could only be qualitatively correlated with bacterial deposition and desorption in the parallel plate flow chamber.  相似文献

To enhance our understanding of liquids in contact with rough surfaces, a systematic study has been carried out in which water contact angle measurements were performed on a wide variety of rough surfaces with precisely controlled surface chemistry. Surface morphologies consisted of sandblasted glass slides as well as replicas of acid-etched, sandblasted titanium, lotus leaves, and photolithographically manufactured golf-tee shaped micropillars (GTMs). The GTMs display an extraordinarily stable, Cassie-type hydrophobicity, even in the presence of hydrophilic surface chemistry. Due to pinning effects, contact angles on hydrophilic rough surfaces are shifted to more hydrophobic values, unless roughness or surface energy are such that capillary forces become significant, leading to complete wetting. The observed hydrophobicity is thus not consistent with the well-known Wenzel equation. We have shown that the pinning strength of a surface is independent of the surface chemistry, provided that neither capillary forces nor air enclosure are involved. In addition, pinning strength can be described by the axis intercept of the cosine-cosine plot of contact angles for rough versus flat surfaces with the same surface chemistries.  相似文献

While individual water molecules adsorb strongly on a talc surface (hydrophilic behavior), a droplet of water beads up on the same surface (hydrophobic behavior). To rationalize this dichotomy, we investigated the influence of the microscopic structure of the surface and the strength of adhesive (surface-water) interactions on surface hydrophobicity. We have shown that at low relative humidity, the competition between adhesion and the favorable entropy of being in the vapor phase determines the surface coverage. However, at saturation, it is the competition between adhesion and cohesion (water-water interactions) that determines the surface hydrophobicity. The adhesive interactions in talc are strong enough to overcome the unfavorable entropy, and water adsorbs strongly on talc surfaces. However, they are too weak to overcome the cohesive interactions, and water thus beads up on talc surfaces. Surprisingly, even talc-like surfaces that are highly adhesive do not fully wet at saturation. Instead, a water droplet forms on top of a strongly adsorbed monolayer of water. Our results imply that the interior of hydrophobic zeolites suspended in water may contain adsorbed water molecules at pressures much lower than the intrusion pressure.  相似文献

The adsorption behaviors of extended anionic surfactants linear sodium dodecyl(polyoxyisopropene)4 sulfate (L-C12PO4S), branched sodium dodecyl(polyoxyisopropene)4 sulfate (G-C12PO4S), and branched sodium hexadecyl(polyoxyisopropene)4 sulfate (G-C16PO4S) on polymethylmethacrylate (PMMA) surface have been studied. The effect of branched alkyl chain on the wettability of the PMMA surface has been explored. To obtain the adsorption parameters such as the adhesional tension and PMMA-solution interfacial tension, the surface tension and contact angles were measured. The experimental results demonstrate that the special properties of polyoxypropene (PO) groups improve the polar interactions and allow the extended surfactant molecules to gradually adsorb on the PMMA surface by polar heads. Therefore, the hydrophobic chains will point to water and the solid surface is modified to be hydrophobic. Besides, the adsorption amounts of the three extended anionic surfactants at the PMMA–liquid interface are all about 1/3 of those at the air–liquid interface before the critical micelle concentration (CMC). However, these extended surfactants will transform their original adsorption behavior after CMC. The surfactant molecules will interact with the PMMA surface with the hydrophilic heads towards water and are prone to form aggregations at the PMMA–liquid interface. Therefore, the PMMA surface will be more hydrophilic after CMC. In the three surfactants, the branched G-C16PO4S with two long alkyl chains exhibits the strongest hydrophobic modification capacity. The linear L-C12PO4S is more likely to densely adsorb at the PMMA–liquid interface than the branched surfactants, thus L-C12PO4S possesses the strongest hydrophilic modification ability and shows smaller contact angles on PMMA surface at high concentrations.  相似文献

利用座滴法研究了阳离子表面活性剂十六烷基醚羟丙基季铵盐(C16PC)、十六烷基聚氧乙烯醚羟丙基季铵盐(C16(EO)3PC)和两性离子表面活性剂十六烷基醚羟丙基羧酸甜菜碱(C16PB)、十六烷基聚氧乙烯醚羟丙基羧酸甜菜碱(C16(EO)3PB)溶液在聚甲基丙烯酸甲酯(PMMA)表面上的润湿性质, 考察了表面活性剂类型及浓度对接触角的影响趋势. 研究发现: 低浓度条件下表面活性剂分子可能以平躺的方式吸附到固体界面, 且亲水基团靠近固体界面, PMMA表面被轻微疏水化; 在高浓度时则通过Lifshitz-van der Waals 作用吸附, 亲水基团在外, PMMA表面被亲水改性. 聚氧乙烯基团(EO基团)的引入对阳离子表面活性剂的接触角影响不大; 而含有聚氧乙烯基团的两性离子表面活性剂在PMMA界面上以类似半胶束的聚集体吸附, 大幅度降低接触角.  相似文献

We have constructed a dark‐field light scattering microscope using a very low‐cost digital camera to investigate the adsorption of gold nanoparticles (AuNPs) on four different substrates at various pH values. The substrates used are glass, polycarbonate (PC), poly(dimethylsiloxane) (PDMS), and poly(methyl methacrylate) (PMMA). The coverage of AuNPs on hydrophobic substrates such as PDMS is greater than that on hydrophilic substrates like glass. The adsorption and aggregation of AuNPs on a particular substrate increased upon decreasing the pH (from 9.0 to 4.0). A greater coverage percentage of AuNPs, but less aggregation, occurs on glass treated with poly(diallyldimethylammonium) (PDDA) than on bare glass. The scattering intensity increases upon increasing the number of layers of adsorbed AuNPs on glass that was treated sequentially with AuNPs and PDDA. When compared to UV‐Vis absorption, dark‐field microscope provides greater sensitivity and qualitative surface information.  相似文献

A simple and cost-effective successive ionic layer adsorption and reaction (SILAR) method was adopted to fabricate hydrophobic ZnO nanostructured surfaces on transparent indium-tin oxide (ITO), glass and polyethylene terephthalate (PET) substrates. ZnO films deposited on different substrates show hierarchical structures like spindle, flower and spherical shape with diameters ranging from 30 to 300 nm. The photo-induced switching behaviors of ZnO film surfaces between hydrophobic and hydrophilic states were examined by water contact angle and X-ray photoelectron spectroscopy (XPS) analysis. ZnO nanostructured films had contact angles of ~140° and 160°±2 on glass and PET substrates, respectively, exhibiting hydrophobic behavior without any surface modification or treatment. Upon exposure to ultraviolet (UV) illumination, the films showed hydrophilic behavior (contact angle: 15°±2), which upon low thermal stimuli revert back to its original hydrophobic nature. Such reversible and repeatable switching behaviors were observed upon cyclical exposure to ultraviolet radiation. These biomimetic ZnO surfaces exhibit good anti-reflective properties with lower reflectance of 9% for PET substrates. Thus, the present work is significant in terms of its potential application in switching devices, solar coatings and self-cleaning smart windows.  相似文献

Hydrophilic silica (SiO2) nanoparticles were dispersed in solutions of poly(methyl methacrylate) (PMMA) and in solutions of a commercial poly(alkyl siloxane) (Rhodorsil 224), and the suspensions were sprayed on glass surfaces. The effect of the particle concentration on the hydrophobic character of PMMA-SiO2 and Rhodorsil-SiO2 films was investigated and showed the following: (i) Static contact angles (theta s), measured on surfaces that were prepared from dilute dispersions (particle concentration <1% w/v), increase rapidly with particle concentration and reach maximum values (154 and 164 degrees for PMMA-SiO2 and siloxane-SiO2, respectively). Further increases in particle concentration do not have any effect on theta s. (ii) The effect of particle concentration on the contact angle hysteresis (thetaAlpha - thetaR) is more complicated: as the particle concentration increases, we first notice an increase in hysteresis, which then decreases and finally becomes constant at elevated particle concentrations. The lowest thetaAlpha - thetaR values were 5 degrees for PMMA-SiO2 and 3 degrees for siloxane-SiO2, respectively. (iii) SEM and AFM images show that a two-length-scale hierarchical structure is formed on the surface of the superhydrophobic films. It is demonstrated that superhydrophobicity can be achieved using various hydrophilic nanoparticles (alumina and tin oxide nanoparticles were successfully tested) and that the substrate has almost no effect on the hydrophobic character of the applied coatings, which were produced on silicon, concrete, aluminum, silk, wood, marble, and of course glass. The results are discussed in light of Wenzel and Cassie-Baxter models.  相似文献

The manipulation of colloidal nanoparticles (NPs) in a drying droplet has critical importance not only for several industrial applications but also their assembly into patterns on surfaces. The influence of a tip with hydrophilic or hydrophobic surfaces dipped into a drying droplet on hydrophilic or hydrophobic surfaces on the behavior of 98 nm latex NPs was investigated. The formation of concentric rings on hydrophilic glass surfaces regardless of the surface chemistry of the dipped tip was observed. On the other hand, no pattern formation on hydrophobic surfaces was observed with the insertion of the tip. With a hydrophilic tip, the concentric rings were formed due to stick-slip motion of the solvent contact line resulting from competition between pinning and capillary forces while the capillary effect was not effective until the surface of the tip was changed by adherent NPs making the tip surface available for water adherence with a hydrophobic tip, which results in the pulling of droplet towards the tip. It is also found that the tip thickness and suspension concentration significantly influences the formation of concentric rings on surfaces. This simple procedure can be used to influence the distribution or assembly of NPs in the droplet area.  相似文献

An investigation of energetic properties of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) layers deposited on glass, mica, and PMMA (poly(methyl methacrylate)) surfaces was carried out by means of contact angles measurements (advancing and receding) for three probe liquids (diiodomethane, water, and formamide). DPPC was deposited on the surfaces from water (on glass and mica) or methanol (on PMMA) solutions. The topography of the tested surfaces was determined with a help of scanning electron microscopy (SEM) and atomic force microscopy (AFM). Using the measured contact angles, the total apparent surface free energy and its components of the studied layers were determined from van Oss et al.'s (Lifshitz-van der Waals and acid-base components, LWAB) and contact angle hysteresis (CAH) approaches. It allowed us to learn about changes in the surface free energy of the layers (hydrophobicity/hydrophilicity) depending on their number and kind of support. It was found that the changes in the energy greatly depended on the surface properties of the substrate as well as the statistical number of monolayers of DPPC. The spreading and recoiling of water drops on several flat and macroscopically smooth model surfaces and on sized paper surfaces were studied over a range of drop impaction velocities using a high-speed CCD camera. The water drop spreading and recoiling results on several model hydrophobic and hydrophilic surfaces were found to be in agreement with observations reported in the literature. The maximum drop spreading diameter for those model surfaces at impact was found to be dependent upon the initial drop kinetic energy and the degree of hydrophobicity/hydrophilicity of the surface. The extent of the maximum drop recoiling was found to be much weaker for hydrophilic substrates than for hydrophobic substrates. Sized papers, however, showed an interesting switch of behaviour in the process of water drop impaction. They behave like a hydrophobic substrate when a water drop impacts on it, but like a hydrophilic substrate when water drop recoils. Although the contact angle between water and hydrophilic or hydrophobic non-porous surfaces changes from advancing to receding as reported in literature, the change of contact angle during water impact on paper surface is unique in that the level of sizing was found to have a smaller than expected influence on the degree of recoil. Atomic force microscopy (AFM) was used to probe fibres on a sized filter paper surface under water. The AFM data showed that water interacted strongly with the fibre even though the paper was heavily sized. Implications of this phenomenon were discussed in the context of inkjet print quality and of the surface conditions of sized papers. Results of this study are very useful in the understanding of inkjet ink droplet impaction on paper surfaces which sets the initial condition for ink penetration into paper after impaction.  相似文献

Surfaces grafted with poly(methyl methacrylate) (PMMA) and streptavidin were synthesized through click chemistry to investigate the role of surface stiffness on protein adsorption as the hydrophilic and hydrophobic surface coverage of the substituents vary. Surface topographies coupled with the nanoindentation results indicated that, with the appropriate selections of polymer coverage and chain length, the extent of non-specific protein adhesion could be controlled by the hydrophobic interactions between PMMA, biotin, and streptavidin. It was shown that, when the molecular weight and stiffness of PMMA was close to that of streptavidin, patchy PMMA morphologies were obtained, which help inhibit the non-specific adsorption of streptavidin.  相似文献

The surfaces of poly(methyl methacrylate) (PMMA) films modified by O2H2O and H2O radio-frequency glow discharge plasmas were studied using electron spectroscopy for chemical analysis (ESCA or XPS), low energy ion scattering (LEISS or ISS), Fourier transform IR spectroscopy (FTIR) with attenuated total reflectance (ATR) sampling, and critical surface energy from contact angle measurements. The extent and nature of modification with respect to promotion of a hydrophilic surface compared to the hydrophobic surface of the unmodified PMMA has been probed. Results show drastic decreases in C/O ratio at the near surface, which increases to that of the unmodified PMMA as deeper cross sections are analyzed. In addition peak fitting of ESCA data correlated with FTIR functional group information allows for the qualitative and quantitative analysis of the resulting bonding and structure of the modified layer. From these results combined with the polarity and surface energy differences obtained from contact angle measurements, the structural changes are discussed with respect to plasma reaction mechanisms and differences in the structure of the modified polymer films.  相似文献
