A proficiency testing round was undertaken to assess the performance of laboratories to measure acrylamide in a sample of crispbread. Retail samples of crispbread were ground to a fine powder and after thorough mixing were packed in 40 g units for distribution. Ten samples were selected at random and analyzed in duplicate for acrylamide by liquid chromatography/mass spectrometry (LC/MS). Standard statistical tests showed that the material was homogeneous for the purposes of proficiency testing. Test samples were distributed to 55 laboratories in 16 countries in Europe, North America, Australia, and the Middle East. The results were analyzed by standard proficiency testing statistical procedures, and laboratories were awarded z-scores on the basis of their reported results. Based on a target standard deviation (sigmap value) taken from the Horwitz equation, for a robust mean value of 1.2 mg/kg acrylamide, satisfactory results (z-score within +/- 2 for those between 0.8 and 1.6 mg/kg) were obtained by 86% of the 37 laboratories that returned results. Only 1 laboratory was unsatisfactory and 4 had questionable results. About equal numbers of laboratories used gas chromatography (GC)/MS and LC/MS procedures with about 25% using MS/MS and one using GC with electron capture detection. There was no evident trend in performance or bias in results. GC/MS and LC/MS data were evenly distributed across the population of laboratories reporting results.  相似文献

The European Commission's Directorate General Joint Research Centre has organized several proficiency tests on the determination of acrylamide (AA) in food. This paper presents the results and outcome of a proficiency test that focused on the determination of AA in crispbread samples. One of the goals was the identification of the influence of different parameters such as analyte extraction or instrument calibration on the analytical results. A set of samples, containing 3 different crispbread samples as well as extracts of one crispbread sample and AA standard solutions, was shipped to each participant. A total of 42 European laboratories reported analytical results that were evaluated by applying internationally accepted protocols and procedures. The study found that, for each sample, the results of 4-8 laboratories were outside the range formed by the target value plus or minus the 2-fold of the target standard deviation; thus, they did not perform satisfactorily. In transferring this knowledge to the data of monitoring databases of AA in food, care must be taken that data are quality controlled, as it is likely that some of them may be biased.  相似文献

Simple and rapid gas chromatographic (GC) method employing a high-resolution time-of-flight mass analyzer that enables direct analysis (no derivatization) of acrylamide in various heat-processed foodstuffs has been developed and validated. Co-isolation of acrylamide precursors such as sugars and asparagine, constituting the risk of results overestimation due to additional formation of analyte in hot GC injector, is avoided by the extraction with n-propanol followed by solvent exchange to acetonitrile (MeCN). Introduction of a novel purification strategy, dispersive solid phase extraction, based on addition of primary-secondary amine (PSA) sorbent into deffated extract in MeCN, provides a significant reduction of some abundant matrix co-extracts (mainly free fatty acids). Isotope dilution technique (d3-acrylamide as an internal standard) is employed for compensation of potential target analyte losses and/or matrix-inducted chromatographic response enhancement. Limits of quantifications (LOQs) ranged between 15 and 40 μg kg−1 and recoveries were between 97 and 108% depending on the examined food matrix. The repeatability of measurements (expressed as relative standard deviation, R.S.D.) was as low as 1.9% for potato crisps containing acrylamide at a level of 1 mg kg−1. Slightly higher values (R.S.D. < 4.0%) were achieved for breakfast cereals and crisp bread with approximately 10 times lower content of this processing contaminant. Trueness of results generated by this new method was demonstrated via FAPAS® (Food Analysis Performance Assessment Scheme) interlaboratory proficiency tests.  相似文献

A method for the determination of zearalenone in corn has been developed applying pressurised liquid extraction (PLE) and using environmentally acceptable and less noxious organic solvents. The extracted samples were analysed with liquid chromatography coupled to mass spectrometry (LC-MS) equipped with an electrospray (ESI) ionisation interface. The optimised extraction mixture was isopropanol and an aqueous solution of triethylamine (1%) 50:50 (v/v), which allowed to halve the use of organic solvent compared to the method proposed by ISO. When applying the optimised method to five different naturally contaminated corn samples the obtained concentrations were slightly increased compared to the analysis using the previously used extraction solvent (acetonitrile-methanol). The relative standard deviation (RSD, n = 3) varied between 4 and 10% depending on the concentration level of the target analyte in the test material.  相似文献

In this study, a new method for the determination of organophosphorus pesticides (OPPs) (ethoprophos, diazinon, parathion methyl, fenitrothion, malathion, isocarbophos and quinaphos) in orange juice was developed. Single-drop microextraction (SDME) parameters, such as organic solvent, drop volume, agitation rate, extraction time, and salt concentration were optimized through analysis of OPPs in fortified water. The orange juice was simply centrifuged and diluted with water, extracted by SDME and analyzed by gas chromatography (GC) equipped with a flame photometric detection (FPD). Fortification tests were conducted for concentrations between 10 and 500 microg/L; mean relative recoveries for each pesticide were all above 76.2% and below 108.0%. Limits of detection of the method for orange juice were below 5 microg/L for all target pesticides. The repeatability of the proposed method, expressed as relative standard deviation varied between 4.6 and 14.1% (n=5). The proposed method is acceptable in the analysis of OPPs pesticides in juice matrices.  相似文献

A method using liquid chromatography tandem mass spectrometry (LC-MS-MS) with electrospray for the analysis of acrylamide in foods is reported. The method comprises the addition of deuterium-labelled acrylamide-d3, extraction with water, mixed mode solid phase extraction, ultrafiltration and a graphitised carbon column for chromatography. The transitions m/z 72 > 55, 72 > 54, 72 > 44, 72 > 27, 72 > 72 and 75 > 58 were recorded in multiple reaction monitoring mode for identification and quantification. In-house validation data for products from potatoes and cereals (30 to 10,000 microg kg(-1)) are presented (accuracy 91 to 102%, relative standard deviation 3 to 21%). Interlaboratory validation data (comparison with gas chromatography mass spectrometry, 25 to 2000 microg kg(-1)) showed excellent results (r2 = 0.998).  相似文献

An improved method was validated for the determination of acrylamide in foods using ultra-performance liquid chromatography (UPLC) coupled to eletrospray ionization tandem mass spectrometry (MS/MS) in the present study. This improved method supplies a rapid quantitative procedure of acrylamide with a run time of only 3 min. Results showed a good repeatability (RSD< or =4.5%) with 50, 100, 250, 500 and 1000 microg/kg spiked concentrations in potato crisps in within-day (n=5) and day-to-day (n=10) precision tests. Meanwhile, good recoveries (81.6-99.0%) were obtained with the same spiked concentrations in acrylamide-free cereal samples (n=3). The excellent method validation data and proficiency test results (Z-score: -0.1) of the official Food Analysis Performance Assessment Scheme (FAPAS) suggested that the present quantitative method could be applied for rapid determination of acrylamide in many investigations.  相似文献

A pyrolyzer gas chromatography/mass spectrometry (GC/MS) method eliminates toxic solvents that burden our environment and can address the crucial problem of the solvent extraction GC/MS method. The purpose of this study is to establish an efficient quantitative analysis method for 10 phthalates that are regulated by the several governments. A change of concentrations over time for phthalates and internal standards was measured to verify the feasibility of using an auto sampler that facilitates analyzing multiple samples. Both standards maintained constant concentrations over the appropriate time for analysis. A certified reference material under the auspices of the Korea Research Institute of Standards and Science was used to verify the calibration curve obtained by the pyrolyzer GC/MS method, and a deviation was considered similar to the solvent extraction GC/MS method. Then, the limit of detection and limit of quantitation values were confirmed for various consumer products. To verify the reliability of the method, a comparative test with several accredited testing institutes was conducted, and the results were within the standard deviations of the results provided by the institutes. These results indicate that the pyrolyzer GC/MS method can be used in not only screening but also in accurate quantitative analysis.  相似文献

Organic acidurias are a large group of inherited metabolic disorders (IMDs), commonly diagnosed by GC-MS analysis of organic acids in urine after acidic extraction and trimethylsilylation. In this study, a GC×GC-ToF-MS method has been optimized for the analysis of pathological metabolites in urine. An automated data processing strategy based on the use of mass spectra and GC retention times for the target search and quantification of pathological metabolites has been developed. Using this procedure, each unknown sample is automatically examined for the presence of markers of several diseases at the same time. The method has been applied for the analysis of 6 challenging proficiency testing samples from patients with IMDs (thymidine phosphorylase deficiency, mevalonic aciduria, hawkinsinuria, aromatic l-amino acid decarboxylase deficiency, propionic acidemia and medium-chain acyl-CoA dehydrogenase deficiency). Using the GC×GC-ToF-MS method, we were able to determine complete sets of markers for all the IMDs. The quality of the mass spectral matches for the pathological markers was higher than 800 (out of 1000).  相似文献

This paper proposes a new method for quantitative analysis of acrylamide in cereal-based foods and potato chips. The method uses reaction with trifluoroacetic anhydride, and analyses the resulting derivative by use of gas chromatography with electron-capture detection (GC–ECD). The effects of derivatization conditions, including temperature, reaction time, and catalyst, on the acylation reaction were evaluated. Chromatographic analysis was performed on an SE-54 capillary column. Under the optimum conditions, good retention and peak response were achieved for the acrylamide derivative. The analytical method was fully validated by assessment of LODs and LOQs (1 ng g?1 and 25 ng g?1, with relative standard deviations (RSD) 2.1 and 3.6, respectively), linearity (R?=?0.9935 over the range 0.03–10 μg g?1), and extraction recovery (>96 %, with RSD below 2.0, for acrylamide spiked at 1, 20, 50, and 100 ng g?1; 99.8 % for acrylamide content >1000 ng g?1). The method requires no clean-up of the acrylamide derivative before injection. The method has been successfully used to determine acrylamide levels in different commercial cereal-based foods, French fries, and potato chips.

The objective of this project was to conduct an aflatoxin proficiency test program in government, academia, and industry laboratories in Thailand. Aflatoxin-free corn and peanuts and corn and peanuts naturally contaminated with aflatoxins diluted to approximately 25 micrograms/kg were analyzed. Homogeneity of prepared, naturally contaminated test samples was checked on multiple replicates. The test was conducted according to the ISO/IUPAC/AOAC INTERNATIONAL Harmonized Protocol with z scores indicating laboratory performance. The participants used 3 methods: enzyme-linked immunosorbent assay, thin-layer chromatography, and the minicolumn. Of 19 laboratories that reported results for aflatoxins in naturally contaminated corn, 13 (68%) performed satisfactorily, on the basis of the mean obtained by an expert laboratory, a calculated target value for standard deviation, and the z score. Of 21 laboratories that reported results for aflatoxins in naturally contaminated peanuts, 10 (48%) performed satisfactorily. For aflatoxin-free corn, 6 laboratories reported finding aflatoxins at > or = 10 ng/g, chiefly by the minicolumn method; for aflatoxin-free peanuts, 1 laboratory reported finding aflatoxins at > 10 ng/g. Subsequently, a workshop of lectures and laboratory sessions was conducted to improve performance. A new and simple successive outlier removal procedure applied to the same data removed the same laboratories as did the use of z scores.  相似文献

The use of rapid and inexpensive nonlaboratory-based screening tests for drug quality assessments is recommended as a component of a drug quality assurance program in poor resource settings. We have established routine Minilab test procedures to screen product quality and a proficiency testing program to determine the competency of the inspectors and reliability of results. Samples for the proficiency testing were prepared by pulverizing a standard reference tablet of the appropriate drug and making serial dilutions with starch to obtain concentrations of 0, 40, and 100%. The samples, which were labeled only with the drug name and an identifying letter, were given to inspectors for quality screening using Minilab procedures. In round 1 of the proficiency test, only 3 of 28 substandard samples were correctly identified. Round 2 of the proficiency test, which was administered after a performance qualification test for the analytical method, showed much improvement: 19 of 27 substandard drugs were correctly identified, while 5 out of 9 inspectors made the correct inference on the quality of 45 samples. However, in both rounds, 2 inspectors failed to identify substandard samples, indicating that their technical competencies need to be improved for the reliability of the results. Although the thin-layer chromatography screening methods provide a rapid means for drug quality assessment, they need to be put in the hands of competent users. The inclusion of a proficiency test in the screening program provides a measure of determining competency of the personnel and reliability of the results.  相似文献

An improved sample preparation method was developed to enhance acrylamide recovery in high‐fat foods. Prior to concentration, distilled deionized water was added to protect acrylamide from degradation, resulting in a higher acrylamide recovery rate from fried potato chips. A Chrome‐Matrix C18 column (2.6 μm, 2.1 × 100 mm) was used for the first time to analyze acrylamide levels using ultra high performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry, displaying good separation of acrylamide from interference. A solid‐phase extraction procedure was avoided, and an average recovery of >89.00% was achieved from different food matrices for three different acrylamide spiking levels. Good reproducibility was observed, with an intraday relative standard deviation of 0.04–2.38%, and an interday relative standard deviation of 2.34–3.26%. Thus, combining the improved sample preparation method for acrylamide analysis with the separation on a Chrome‐Matrix C18 column (2.6 μm, 2.1 × 100 mm) using ultra high performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry is highly useful for analyzing acrylamide levels in complex food matrices.  相似文献

A procedure for the analysis of over 100 pesticides that only contain combinations of carbon, hydrogen, nitrogen, sulfur, and oxygen (i.e., no phosphorous or halogen atoms) has been developed. The procedure employs gas chromatography with a mass selective detector (GC/MSD), electron impact ionization, and selected ion monitoring. This GC/MSD procedure provided lower limits of quantitation and increased confirmational data compared to the traditional element-selective GC procedures that are commonly used for the detection of this "class" of pesticides. These analytical improvements were demonstrated by 26 pesticides that were detected at ng/g levels in a variety of fruit and vegetable matrixes using this procedure; these pesticides were missed by the traditional element-selective GC procedures. Validation of the procedure was performed using acetone extraction with solid-phase extraction cleanup. Twenty representative target pesticides were used to demonstrate repeatability and linearity of the chromatographic response and recovery from fruit and vegetable matrixes.  相似文献

The state-of-the-art of analysis for acrylamide in food is reviewed. The majority of analytical methods adopts a similar approach: addition of internal standard to the specimen, extraction with water, purification of extract using a solid-phase extraction cartridge, and then determination using either gas chromatography coupled to mass spectroscopy (GC/MS) after bromination, or direct measurement with liquid chromatography coupled to mass spectroscopy (LC/MS). The available methods generally show good agreement and are likely to be accurate. However, improvements in precision (within-laboratory) and repeatability (between-laboratory) are needed by particular data users.  相似文献

In early 2002, Swedish National Food Administration (SNFA) and University of Stockholm together announced that certain foods that are processed or cooked at high temperature contain relatively high levels of acrylamide. The occurrence of acrylamide is derived from heat-induced reactions between the amino group of asparagine and the carbonyl group of reducing sugars during baking and frying. Corresponding chromatographic methods are used to determine various structural groups present during this process. Gas chromatography (GC)-mass spectrometry (MS) and liquid chromatography with tandem mass spectrometry (LC-MS/MS) analysis are both acknowledged as the major useful and authoritative methods for the acrylamide determination and other chromatographic methods are also briefly introduced. The aim of this review is to summarize the state-of-the-art about the occurrence, analytical methods, and extraction and clean-up procedures of acrylamide. Special attention is given to chromatographic techniques applied for the occurrence and determination of acrylamide.  相似文献

A simple and rapid gas chromatography/mass spectrometry (GC/MS) analysis method was developed for the determination of essential oils in the crude extract of Schizonepeta tenuifolia Briq (Sch.t.Briq). Five major volatiles (menthone, pulegone, 2-hydroxy-2-isopropenyl-5-methylcylohexanone, cis-pulegone oxide, and schizonal) were extracted and isolated from Sch.t.Briq as marker compounds for use in the quality control of herbal medicines. Various extraction techniques, such as solvent immersion, mechanical shaking, and sonication, were evaluated, and the greatest efficiency was observed with sonication extraction using petroleum ether. The dynamic range of the GC/MS method depended on the specific analyte; acceptable quantification was obtained between 10 and 1000 μg/mL for menthone and pulegone, and between 2.5 and 75.0 μg/mL for 2-hydroxy-2-isopropenyl-5-methylcylohexanone, cis-pulegone oxide, and schizonal. The method was deemed satisfactory by inter- and intra-day validation and exhibited both high accuracy and precision, with a relative standard deviation < 10%. Overall limits of detection were approximately 0.40–0.50 μg/mL, with a standard deviation (σ)-to-calibration slope (s) ratio (σ/s) of 3. The limit of quantitation in our experiments was approximately 2.5 μg/mL at a σ/s of 10. On the basement of method validation, 21 samples of Sch.t.Briq collected from markets in Korea were monitored for the quality control. In addition, principal component analysis (PCA) and hierarchical cluster analysis (HCA) were performed on the analytical data of 21 different Sch.t.Briq samples in order to classify samples that were collected from different regions.  相似文献

The objective of the present study was to develop and validate a rapid, highly sensitive, and reliable extraction method to determine acrylamide in water samples. The method was based on the derivatisation of the acrylamide in the presence of KBr, HBr and saturated Br2 solution into 2,3-dibromopropionamide and dispersive liquid–liquid microextraction (DLLME) followed by gas chromatography–electron capture detection (GC–ECD) of the analyte. Different parameters that affect the DLLME process such as types and volumes of disperser solvent, ionic strength of aqueous solution and extraction time were investigated and optimised. Under optimal conditions, excellent linearity was obtained between concentration of acrylamide and the response of ECD with correlation of determination (R2) of 0.9999. The precision of the method, which was determined by calculating the relative standard deviations (RSD) of the at least three replicate measurements, was 3.6%. The method presented in this study is sensitive enough for the determination of acrylamide in different water samples with the limit of detection (LOD) value of 1?ng?L?1. The mean percentage recoveries exceeded 91% for all of spiking levels in the real water samples. The results obtained from DLLME method are validated by EPA method 8032A.  相似文献

In the present work, an analytical multiresidue method has been developed for the analysis of 32 organochlorine, organophosphorus and organonitrogen pesticides at microg kg(-1) levels in virgin olive oil. The method consists of the extraction of the pesticides with acetonitrile saturated in n-hexane followed by a clean-up process based on gel permeation chromatography (GPC) with ethyl acetate-ciclohexane (1:1) as mobile phase to separate the low-molecular mass pesticides from the high-molecular mass fat constituents of the oil. The target compounds were determined in the final extract by gas chromatography (GC) using thermoionic specific (TSD) and electron-capture (ECD) detection. In the case of positive samples, the amounts found were confirmed by GC-MS/MS, being the results in good agreement. Recoveries and RSDs (n = 10) values were 91-124% and 1-8% (GC-ECD), 82-100% and 9-20% (GC-TSD), and 89-105% and 4-14% (GC-MS/MS), respectively. The three proposed methods were applied to samples collected directly in two olive mills located in the Jaén province (Spain). Specifically, 24 samples of virgin olive oil were collected. The most frequently pesticide residues found were the herbicides terbuthylazine and diuron and endosulfan sulfate, a degradation product of the insecticide endosulfan. The herbicide concentration was higher in those oil samples obtained from olives which were collected from the ground after they had fallen down than in those oil samples from olives harvested directly from the tree. The GC-MS/MS developed method was also applied to the analysis of an olive oil sample from a proficiency test spiked with organochlorine pesticides and all the values obtained were within the specified "satisfactory" range.  相似文献
