一种纯天然产物白藜芦醇用作辣根过氧化物酶(HRP)底物。对其化学性质的研究证实,白藜芦醇在空气中较稳定,对HRP、H2O2的电化学响应性能优于传统HRP底物,对人体无毒害。白藜芦醇在HRP催化下可被H2O2氧化成醌,产物醌在电极上于-376 mV处可被还原,其电流的大小与HRP的浓试在一定浓度范围内呈线性相关。将兔布氏杆菌抗原包埋在石墨-石蜡基质中制备了测定兔布氏杆菌抗体的电化学酶联免疫传感器,该传感器测定兔布氏杆菌抗体的线性范围为3×10-4~1.65×10-2g/L;检出限为1×10-4g/L;RSD为4.6%。本方法制备免疫传感器的电化学性能稳定,抗原活性保持良好。  相似文献

为探索用电化学方法检测生物样品中氯霉素残留的高灵敏度技术,本实验研究了阳离子表面活性剂十六烷基三甲基溴化铵(CTMAB)对氯霉素在玻碳电极上伏安行为的影响,实验结果表明,以0.02 mol/L高氯酸为支持电解质,在0.40~-0.60 V的范围内进行伏安扫描时,2×10-5mol/L CTMAB能显著提高氯霉素在-0.41 V处的还原峰电流。利用这种改进的电化学方法检测氯霉素的线性范围为0.0026~8 mg/L,检出限达到0.83μg/L。研究了缓冲液种类及其酸碱度及其它表面离子活性剂等对测试氯霉素的影响。  相似文献

An indirect catalytic method for the separate microdetermination of oxalate, citrate, and fluoride ions is described. The method is based on the inhibition action of oxalate, citrate, and fluoride ions on the catalytic oxidation reaction of 2,4-diaminophenol-hydrogen peroxide by iron(III).Procedures for the determination of 1.76 × 10−2 to 17.6 × 10−2 μg/ml for oxalate ion, 3.78 × 10−2 to 30.24 × 10−2 μg/ml for citrate ion, and 0.38 to 4.18 μg/ml for fluoride ion are given.Quantities of 1.76 × 10−2 to 17.6 × 10−2 μg/ml for oxalate ion, 3.78 × 10−2 to 30.24 × 10−2 μg/ml for citrate ion, and 0.38 to 4.18 μg/ml for fluoride ion could be determinated with a relative error of about 1–3.5% for oxalate and citrate ions and 1–2% for fluoride ion.  相似文献

Semiautomatic methods are described for the catalytic titrimetric determination of microamounts of silver and mercury(II) using a chloramine-T-selective electrode as monitor. The methods are based on the inhibitory effect of Ag(I) and Hg(II) on the iodide-catalyzed chloramine-T-arsenite and chloramine-T-H2O2 reactions. Microamounts of silver in the range 0.2–200 μg (1 × 10−7−1 × 10−4 M) and of mercury(II) in the range 0.1–200 μg (2.5 × 10−8−5 × 10−5 M) were determined using the chloramine-T-As(III) indicator reaction. Mercury(II) in the range 4–2000 μg (1 × 10−6−5 × 10−4 M) was also determined using the chloramine-T-H2O2 indicator reaction. The accuracy and precision were in the range 0.1–1%.  相似文献

毛细管电泳-安培法测定复方磺胺甲噁唑片中的有效成分   总被引:1,自引:0,他引:1  
采用毛细管电泳-安培法(CE-AD)同时分离测定了磺胺甲噁唑(sulfamethoxazole,SMZ)、磺胺嘧啶(sulfadiazine,SD)和抗菌增效剂甲氧苄胺嘧啶(trimethoprim,TMP)3种常用磺胺类抗菌药物成分,考察了实验参数对分离、检测体系的影响。在优化实验条件下,以300μm碳圆盘电极作为工作电极,检测电位为1050mV(vs.SCE),在Na2B4O7(13mmol/L)-KH2PO4(18mmol/L)(pH5.8)的缓冲溶液中,分离电压18kV,进样6s,3组分在14min内可实现基线分离。上述3组分浓度分别在5×10-4~5×10-2、5×10-4~0.1和5×10-4~5×10-2g/L范围内与其峰电流强度呈线性关系,检出限达5.1×10-5~8.0×10-5g/L(S/N=3)。该方法已成功应用于复方磺胺甲噁唑片中抗菌活性成分的含量测定,结果令人满意。  相似文献

An intramolecular charge transfer fluorescence probe of 4′-N,N-dimethylamino-4-amino-chalcone(DMAC) exhibits characteristics clearly correlated with the polarity of solvents. The interaction of this fluorescence probe with calf thymus DNA has been investigated. Generally, DMAC bound to DNA shows marked changes in fluorescence and absorbance properties compared to the spectral characteristics of the free form in solution phase. In the presence of DNA the fluorescence intensity of DMAC is greatly increased with a large bathochromic shift of excitation and emission wavelengths. A hypochromism in absorption spectrum was also observed. The absorption and fluorescence spectra, salt concentration effect, and KI quenching experiments demonstrate that DMAC molecule as an intercalator is inserted into the base-stacking domain of DNA double helix, and the interaction of the nucleobases with DMAC molecule causes the increase of fluorescence intensity and hypochromism in absorption spectrum. The intrinsic binding constant and the binding site number were estimated to be 7.04 × 106 mol L−1 in base pairs and 0.065, respectively. The I/I0 vs DNA concentration plot shows a linear range covering 1.98 × 10−6 to 2.08 × 10−4 mol L−1 in base pairs which can be used for determining DNA with a detection limit of 6.0 × 10−7 mol L−1 in base pairs (0.6 μg ml−1).  相似文献

A catalytic for determination of nanomolar concentrations of Co(II), i.e., oxidation of -adrenaline hydrochloride with H2O2 in alkaline medium, is proposed. The reaction gives a low limit of detection of 2.5 × 10 −9 M Co(II) in the reaction mixture, good reproducibility with a relative standard deviation (R.S.D.) of 4−5% in the Co(II) concentration range 8.0 × 10−9−8.0 × 10−8M and good selectivity. On the basis of this indicator reaction, a catalytic-spectrophotometric method for the determination of cobalt in small urine samples (5.00 ml) was elaborated. The analysis of 17 urine samples, taken from healthy persons of different ages, gave cobalt concentrations in the range 0.20–1.50 μmol 1−1. The R.S.D. for ten replicate analyses of a urine sample with an average cobalt content of 0.63 μmol 1−1 was 5.6%. The reliability of the method was verified by a comparative photometric method (r = 0.9755) and by a determination based on known additions of cobalt (r = 0.9894).  相似文献

A new electroanalytical methodology was developed for the quantification of the phytohormone indole-3-acetic acid (IAA), using a graphite–polyurethane composite electrode (GPU) and the square wave voltammetry (SWV), in 0.1 mol L− 1 phosphoric acid solution (pH 1.6). Analytical curves were constructed under optimized conditions (f = 100 s− 1, a = 50 mV, Ei = 5 mV) and the reached detection and quantification limits were 26 μg L− 1 and 0.2 mg L− 1, respectively. The developed methodology is simple and accurate for the routine determination of IAA. In order to verify the application of the electroanalytical methodology in fortified soil samples without previous treatment, an IAA assay was performed without serious interferences of the soil constituents.  相似文献

A highly sensitive fluorescence quenching method has been developed for selective determination of hydrogen peroxide based on the catalytic effect of hemin on theo-hydroxyphenylfluorone (a new fluorogenic substrate) and hydrogen peroxide system. Under optimum conditions, the calibration graph was linear over the range 0–1.0 × 10−6mol/liter hydrogen peroxide, with a limit of detection of 8.0 × 10−9mol/liter in a 10-min reaction period. It can easily be incorporated into the determination of biochemical substances that produce hydrogen peroxide under catalytic oxidation by their oxidase. This possibility has been tested for the determination of glucose in human sera as an example.  相似文献

The preparation and electrochemical characterization of a carbon paste electrode modified with copper(II) hexacyanoferrate(III) (CuHCF) as well as its behavior as electrocatalyst toward the oxidation of N-acetylcysteine were investigated. The electrochemical behavior of the modified electrode and the electrooxidation of N-acetylcysteine were explored using sweep linear voltammetry. The best voltammetric response was observed for a paste composition of 20% (w/w) copper(II) hexacyanoferrate(III) complex, acetate buffer solution at pH of 6.0 as the electrolyte and scan rate of 10 mV s− 1. A linear voltammetric response for N-acetylcysteine was obtained in the concentration range from 1.2 × 10− 4 to 8.3 × 10− 4 mol L− 1, with a detection limit of 6.3 × 10− 5 mol L− 1. The proposed electrode is useful for the quality control and routine analysis of N-acetylcysteine in pharmaceutical formulations.  相似文献

The use of rice husks as an alternative adsorbent in an on-line preconcentration system for Cd (II) and Pb (II) determination by flame atomic absorption spectrometry (FAAS) is described. The potential of rice husks as a natural adsorbent was evaluated as a material modified with 0.75 mol l−1 NaOH solution and in the unmodified form. For this task, several techniques such as spectroscopy and thermogravimetry were used for elucidation of possible functional groups responsible for the uptake of Cd (II) and Pb (II). Furthermore, based on adsorption studies and adsorption isotherms applied to the Langmüir model, it was possible to verify that modified rice husks present a higher adsorption capacity for both metals. After establishing this material as a promising natural adsorbent, it was used for on-line preconcentration of Cd (II) and Pb (II) metals. The multivariate optimisation of chemical and flow variables was performed by using a full factorial design (24) including the following factors: preconcentration time, preconcentration flow rate, concentration and volume of eluent. The optimum pH values used for on-line preconcentration were taken from prior univariate experiments. Under optimised conditions for Cd (II) determination (4 min of preconcentration at a 6 ml min−1 preconcentration flow rate, in which comprises 24 ml of preconcentration volume, 200 μl elution volume and 1.0 mol l−1 HNO3 solution as eluent), the system achieved a detection limit of 1.14 μg l−1 and an enrichment factor of 72.4. Similar conditions were used for Pb (II) determination (4 min of preconcentration, 6 ml min−1 preconcentration flow rate, 300 μl elution volume and 1.0 mol l−1 HNO3 solution as eluent) from which a detection limit of 14.1 μg l−1 and enrichment factor of 46.0 were achieved. Also, rice husks have been shown to be a homogeneous and stable adsorbent in which more than 100 preconcentration/elution cycles provide a relative standard deviation (RSD) of less than 6.0% on the analytical signal. The satisfactory accuracy of the method developed was obtained by using spiked water samples (mineral water and lake water) and spiked red wine samples. These values were confirmed by electrothermal atomic absorption spectrometry (ETAAS). The certified reference material [pig kidney (CRM 186)] and the reference material [beech leaves (CRM 100)] were also used.  相似文献

The electrochemical behavior of myoglobin (Mb) and hemoglobin (Hb) was investigated with a boron-doped diamond (BDD) electrode by cyclic voltammetry. In acetate buffer solutions, the oxygen reduction at the BDD electrode showed a very high overpotential while the reduction of Mb or Hb was observed in the more positive potential region. Owing to the electrocatalytic reaction of O2 and the participation of H+ following the electrochemical reduction of ferric proteins, the voltammetric responses for Mb and Hb on the BDD electrode in the negative going scans became remarkable in acidic buffer solutions in air. The peak current was linearly proportional to the concentration of Mb in the range 1×10−6–2×10−5 M or the concentration of Hb from 1×10−6 to 1×10−5 M.  相似文献

Gold nanorods (GNRs) with suitable aspect ratio were synthesized with a template technique and then dispersed in a saturated sodium citrate solution by ultrasonication to form a GNR suspension. A GNR-modified electrode was fabricated using the GNR suspension. The oxidation of dopamine at the GNR/GC electrode exhibited surprisingly high electrocatalytic activity and adsorption-controlled characteristics. Square-wave voltammetry was used to detect dopamine. At the GNR/GC electrode, the linear concentration range of DA is from 1 × 10−8 M to 1 × 10−7 M and the detection limit (s/n = 3) is as low as 5.5 × 10−9 M. The current sensitivity is 3.280 μA/μM, and 1000-fold ascorbic acid (AA) cannot interfere with the determination of DA. All these performances are greatly superior to those of the bare GC electrode.  相似文献

A voltammetric method of Cr(VI) determination in a flow system based on the combination of selective accumulation of the product of Cr(VI) reduction on hanging mercury drop electrode and a very sensitive method of chromium determination in the presence of cupferron previously described is proposed. The calibration graphs were linear from 3 × 10−9 to 3 × 10−8 and from 5 × 10−10 to 5 × 10−9 mol L−1 for accumulation times of 120 and 600 s, respectively. The detection limit for the accumulation time of 600 s was 9 × 10−11 mol L−1. The relative standard deviation was 5.1% (n = 5) for Cr(VI) concentration 1 × 10−8 mol L−1 and the accumulation time of 120 s. The influence of foreign ions commonly present in water samples is presented. The validation of the method was made by studying the recovery of Cr(VI) from spiked natural water samples.  相似文献

An automated on-line pre-reduction of arsenate, monomethylarsonate (MMA) and dimethylarsinate (DMA) using flow injection hydride generation atomic absorption spectrometry (FI-HGAAS) is feasible. The kinetics of pre-reduction and complexation depend strongly on the concentration of -cysteine and on the temperature in the following increasing order: inorganic As(V)<DMA<MMA. Arsenate is pre-reduced/complexed within less than 50 s at 70–100°C compared to 1 h at room temperature, while MMA and DMA require 1.5–2 min at 70–100°C and up to 1–2 h at room temperature. The characteristic masses and concentrations for 100 μl injections are 0.01 ng and 0.1 μg l−1 in integrated absorbance and 0.2 ng and 2 μg l−1 in peak height measurements, and the limits of detection are ca. 0.5 ng and 5 μg l−1, respectively. In a high-performance liquid chromatography (HPLC)–HGAAS system, the -cysteine complexes of inorganic As(III), MMA and DMA are best separated within 7 min by HPLC on a strongly acidic cation exchange column such as Spherisorb S SCX 120×4 mm (5 μm) with a mobile phase containing 12 mmol l−1 phosphate buffer (KH2PO4/H3PO4)–2.5 mmol l−1 -cysteine, pH 3.3–3.5. Upon dilution to -cysteine levels below 10 mmol l−1, which are compatible with HPLC separations, the DMA–cysteine complex is unstable on storage. No baseline separations are possible with anion exchange and reverse phase C18 HPLC columns. The limits of detection with 50 μl injections in peak area mode are ca. 0.5 ng and 10 μg l−1, respectively.  相似文献

The present paper proposes an on-line pre-concentration procedure for lead determination in drinking water and saline waste from oil refinery by flame atomic absorption spectrometry (FAAS). It is based on the sorption of lead (II) ions in a minicolumn of polyurethane foam loaded with 4-(2-pyridylazo)-resorcinol (PAR) reagent. The optimization step was performed using Doehlert matrix involving the variables: sampling flow rate (SR), buffer concentration (BC), pH and eluent concentration (EC). The validation process was performed considering the parameters: linearity and other characteristics of the calibration curve, analytical features of on-line system, precision, robustness, effect of other ions in the pre-concentration system and accuracy. Using the established experimental conditions, the procedure allows lead determination with detection limit (3δ/S) of 0.4 μg l−1, quantification limit (10δ/S) of 1.4 μg l−1, and a precision, calculated as relative standard deviation (RSD) of 5.7 (n=8) and 2.1% (n=8) for lead concentration of 5 and 50 μg l−1, respectively. The pre-concentration factor (PF) considering the ratio among the slopes of the analytical curves with and without pre-concentration is 51. The achieved recovery for lead determination in presence of several cations demonstrated that this procedure could be applied for analysis of water samples. The accuracy was confirmed by analysis of the standard reference material NIST 1640 Trace elements in natural water. The sorption process was characterized by the Langmuir isotherm. The method was applied for lead determination in drinking water collected in Salvador City, Brazil and in saline effluent samples from oil refinery. The lead content for 16 samples of drinking water analyzed varied from 0.77 to 6.98 μg l−1.  相似文献

A thermospray flame furnace atomic absorption spectrometer (TS-FF-AAS) was employed for Co determination in biological materials. Cobalt presents a high atomization temperature and consequently poor sensitivity is obtained without changing its thermochemical behavior. The effect of different complexing agents on sensitivity was evaluated based on the formation of Co volatile compounds. A cloud point procedure was optimized for Co preconcentration for further improvement of sensitivity. Samples were treated with 1 mol l− 1 hydrochloric acid solution for quantitative extraction of Co without simultaneous extraction of Fe, since it is a strong interferent. After the extraction and preconcentration steps, a sample volume of 150 μl was introduced into the hot Ni tube using air as carrier at a flow-rate of 0.4 ml min− 1. The best sensitivity was attained using ammonium pyrrolidinedithiocarbamate (APDC) and Triton X-114 was employed for implementation of the cloud point procedure. The detection limit obtained for Co was 2.1 μg l− 1 and the standard deviation was 5.8% for a solution containing 100 μg l− 1 (n = 10). Accuracy was checked using two certified reference materials (tomato leaves and bovine liver) and results were in agreement with certified values at a 95% confidence level. Employing the developed procedure, Co were quantified in different biological materials (plant and animal tissues). The proposed method presents suitable sensitivity for cobalt determination in the quality control of foods.  相似文献

A rubidium-selective electrode has been developed by coating a graphite rod with a PVC membrane containing decylidene bis(4′-benzo-15-crown-5) ether, which is used as a neutral carrier. This electrode exhibited a near Nernstian response to rubidium in the range 1.0 × 10−1to 5.0 × 10−5mol/liter with a slope of 52 mV/pC. The detection limit was 2.0 × 10−5mol/liter. This decylidene bis(4′-benzo-15-crown-5) ether is a new compound prepared by Sichuan University and has never been used to prepare a rubidium electrode.  相似文献

A method to determine iodide in infant formula samples by indirect atomic absorption spectrometry (IAAS) was developed. The iodide in solution resulting from an alkaline digestion (Na2CO3–NaOH) of the sample is precipitated with silver; the precipitate is redissolved by adding cyanide solution, and this solution is subjected to GF-AAS. Temperatures of 1000 and 2100°C were selected for the ashing and atomization steps, respectively, using a mixture of Pd and Mg(NO3)2 as a matrix modifier (at concentrations of 36 and 16 μg ml−1, respectively). The sensitivity, LOD, LOQ and characteristic mass obtained were, respectively, 1.12×10−2 l μg−1, 3.1 μg g−1 and 10.4 μg g−1 and 7.3 pg, referred to sample. The linear interval of concentrations extends up to 10 μg l−1 of iodide, with no need to use the standard addition method; the mean R.S.D. of data within this range is 3.4%, with 2.9% over the whole procedure. No interfering effects were observed among the foreign ions studied, and 100.0% was the mean analytical recovery achieved within the linear range of concentrations. The application of the method to seven real samples gave a mean content of 12.8 μg g−1 of iodide, as well as less than 3.1 μg g−1 in eight other samples.  相似文献

A reaction rate method is described for the microdetermination of iron. The method is based on the catalytic action of iron on the reaction of 2,4-diaminophenol with hydrogen peroxide. The effect of reagent concentration is studied and the maximum tolerable amounts of interfering ions are determined. Procedures for the determination of 2.8 × 10−3 to 2.8 × 10−2 μg/ml are given.Quantities of 2.8 × 10−3 to 2.8 × 10−2 μg/ml could be determinated with a relative error of about 2%.  相似文献
