To obtain 5-deazaflavins exhibiting red-shifted light absorption spectra, a number of C(8) substituted 5-deazaisoalloxazines were synthesized. This was accomplished a) by the oxidative cyclization of 5,5′-arylmethylenebis(6-methylaminouracil) derivatives and b) by the cyclization of N-methylanilinouracil derivatives with a one-carbon reagent. The latter method led to the formation of impure products. Condensation and oxidation reactions with the π-electron deficient C(8) methyl group in 5-deazalumiflavin did not occur. Introduction of substituents at the C(8) position caused a bathochromic shift that varied between 10 and 40 nm.  相似文献

Abstract

Methyl [2-(trimethylsilyl)ethyl 5-acetamido-3,5-dideoxy-L-glycero-α-D-galacto-2-nonulopyranosid]onate (6) and its 8-epi-N-acetylneuraminic acid derivatives were synthesized from methyl [2-(trimethylsilyl)ethyl 5-acetamido-4,7-di-O-acetyl-3,5-dideoxy-D-glycero-α-D-galacto-2-nonulo-pyranosid]onate (1) and methyl [2-(trimethylsilyl)ethyl 5-acetamido-3,5-dideoxy-4,7-di-O-2-(trimethylsilyl)ethoxymethyl-D-glycero-α-D-galacto-2-nonulopyranosid]onate (2).  相似文献

Four types of carboeyclic analogs of 8-azahypoxanthine and 8-azaadenine nucleosides have been prepared. This group of analogs is comprised of derivatives having the (±)-as-3-(hydroxy-methyl)cyclopentyl,(±)-trans-3-hydroxy-cis-4-(hydroxymethyl)cyclopentyl), (±)-trans-2-hydroxy-cis-4-(hydroxymethyl)cyclopentyl, and (±)-trans-2, trans-3-dihydroxy-cis-4-(hydroxymethyl)-cyelopentyl groups at position 3 of 3,6-dihydro-7H-v-triazolo[4,5-d]pyrimidm-7-one and of 7-amino-3H-v-triazolo[4,5-d]pyrimidine. Diazotization of (5-amino-6-chloropyrimidin-4-yl-amino)eyclopentane derivatives and acidic hydrolysis, without isolation of the resulting 7-chloro-3H-v-triazolo[4,5-d]pyrimidines yielded the 8-azahypoxanthine derivatives (III). Treatment of unpurified 7-chloro-3H-v-triazolo[4,5-d]pyrimidines with anhydrous ammonia gave the 8-azaadenine derivatives (IV). The cyclopentane analogs of 8-azaadenylic acid and of 8-aza-adenosine 3′,5′ -cyclic monophosphate were prepared from the 8-azaadenosine analog.  相似文献

Derivatives of 8-methylquinoline-5-carboxylic acid were synthesized. Alcoholysis of 8-methyl-5-cyanoquinoline gave methyl and ethyl 8-methylquinoline-5-carboxylates, which were converted to the corresponding 8-bromomethyl derivatives. The latter were used for the introduction of a methylamino group and the synthesis of N-acyl derivatives. The reaction of N‐(2‐R1‐oxyphenyl)benzimidoyl chlorides with cyanamide derivatives in the presence of titanium tetrachloride as a catalyst has yielded some new 4‐amino‐8‐R1‐oxy‐2‐phenylquinazolines. pKa values have been determined for these compounds and the influence of substituents at the basicity of the parent system has been discussed. Some investigations on the methyl‐quinazolinyl ether cleavage in different medium have been done yielding the appropriate hydroxyquinazoline derivatives. In those cases when the deprotection of 4‐amino‐8‐methoxy‐2‐phenylquinazoline was carried in aqueous acidic solutions, the formation of the hydrolysis products 3,4‐dihydro‐2‐phenyl‐4‐quinazolone derivatives was observed as well.  相似文献

Reactions of different chloronitrobenzenes and chloronitropyrimidines with monosubstituted hydrazines, R-NHNH2, where R is methyl, carbomethoxy and phenyl, are described. The reactivity of these hydrazines in displacement-ortho substituent cyclizations varies substantially, depending on the nature of the aromatic substrate and the hydrazine R group. New routes to 8-azapurine and benzopyrazole derivatives are decribed.  相似文献

Three-component condensation of quinolin-5-amine with methyl 2,2-dimethyl-4,6-dioxocyclohexane-1-carboxylate and aromatic (heteroaromatic) aldehydes or cyclohex-3-ene-1-carbaldehyde gave previously unknown 1,7-phenanthroline derivatives. The reactions in butan-1ol were characterized by 100% regioselectivity but low stereoselectivity, and mixtures of cis- and trans-isomeric methyl 7-aryl(hetaryl, cyclohexenyl)-10,10-dimethyl-8-oxo-7,8,9,10,11,12-hexahydrobenzo[b][1,7]phenanthroline-9-carboxylates were obtained at a ratio of ∼45: 55.  相似文献

Treatment of 2‐hydroxy‐, 2‐mercapto‐, and 2‐ethoxycarbonylamino‐benzonitriles 12 with 2‐fluoro‐ or 2‐nitrophenacylbromides 13 under alkaline conditions provided the corresponding benzofuran, benzothiophene, and indole intermediates 10 , respectivelly. Nucleophilic cyclization of these compounds led to the corresponding tetracyclic quinolinones 7a, 7b , and 3. Denitrocyclization reaction of compounds 10 (R = NO2) was found especially useful. Compounds 7a, 7b , and 3 were converted to their chloro derivatives 14a‐c , which were reduced with hydrogen and a catalyst to the corresponding compounds 8a, 8b , and 2. The presented pathway represents a new method of preparation of quindoline 2 and its O and S analogs 8. Chloro derivatives 14 are reactive enough to provide the corresponding methoxy derivatives 15 and dimethylamino derivatives 16. Methylation of compounds 7a and 7b with iodomethane providing mixtures of major N‐methyl derivatives 17 and minor O‐methyl derivatives 15 were also studied.  相似文献

(8-Quinolylthiomethyl)trimethoxy- and methyl(8-quinolylthiomethyl)dimethoxysilanes were synthesized and transformed into the corresponding organyltrifluoro- and methyldifluorosilanes. IR spectroscopy revealed the absence of NSi donor-acceptor interaction in (8-quinolylthiomethyl)trifluorosilane and methyl(8-quinolylthiomethyl)difluorosilane. Transesterification of (8-quinolylthiomethyl)trimethoxysilane with triethanolamine yielded 1-(8-quinolylthiomethyl)silatrane; the crystal and molecular structures of the latter were determined by X-ray study.Translated fromIzvestiya Akademii Nauk. Title compounds were obtained starting from the key imidazole intermediate, 5‐amino‐1‐phenyl‐methyl‐2‐mercapto‐1H‐imidazole‐4‐carboxylic acid amide 5 , readily derived from the base catalyzed rearrangement of a thiazole, 5‐amino‐2‐phenylmethylaminothiazole‐4‐carboxylic acid amide 4 . Alkylation of the thiol function on 5 with phenylmethyl and allylic chlorides gave compounds 6 and 7 respectively. Cyclization of 6 with a variety of esters afforded 8‐phenylmethylthiohypoxanthines, 8–11 . Similarly, 7 was cyclized to 8‐allylthiohypoxanthines, 20–21 . Compound 5 was also cyclized, but formed 8‐mercaptohypox‐anthines, 22–24 . Alkylation of 8‐mercaptohypoxanthines afforded 8‐alkylthiohypoxanthines, 8, 9,25 and 26 (see Scheme 2). Chlorination of 9–11 afforded 16–18 ; adenine 19 was derived from 16 . Oxidation of hypox‐anthines 8–11 with m‐chloroperbenzoic acid gave the corresponding 8‐phenylmethylsulfonyl derivatives 12 ‐ 15 . These derivatives proved resistant to nucleophilic displacement reactions with primary amines.  相似文献

A method for preparing 15,16-epoxylabda-8(9),13(16),14-trien-18-oic (phlomisoic) acid was proposed. Its structure was confirmed by an XSA. N-containing derivatives of phlomisoic acid that contained amines, hydrazides, and methyl esters of amino acids on the C-18 atom in addition to (2-oxo-2-aminoacetyl)-substituted derivatives of the C-16 methyl ester of phlomisoic acid were prepared.  相似文献

The total synthesis of 6-amino-1,2,3-triazolo[4,5-c]pyridin-4(5H)one (8-aza-3-deazaguanine, 3 ) and 6-amino-1-(β-D-ribofuranosyl)-1,2,3-triazolo[4,5-c]pyridin-4(5H)one (8-aza-3-deazaguano-sine, 22 ) has been described for the first time by a novel base-catalyzed ring closure of 4(5)-cyanomethyl-1,2,3-triazole-5(4)carboxamide (14) and methyl 5-cyanomethyl-1-(2,3,5-tri-O-ben-zoyl-β-D-ribofuranosyl)-1,2,3-triazole-4-carboxylate (17) , respectively. Under the catalysis of DBU, 2,4-dinitrophenylhydrazone of dimethyl 1,3-acetonedicarboxylate (7) was converted to methyl 5-methoxycarbonylmethyl-1-(2,4-dinitroanilino)-1,2,3-triazole-4-carboxylate (12) via dimethyl 2-diazo-3-iminoglutarate (8) . Catalytic reduction of 12 gave methyl 4(5)methoxycar-bonylmethyl-1,2,3-triazole-5(4)carboxylate (11) from which methyl 4(5)carbamoylmethyl-1,2,3-triazole-5(4)carboxylate (10) was obtained by ammonolysis. Dehydration of 10 provided methyl 4(5)cyanomethyl-1,2,3-triazole-5(4)carboxylate (13) which on amination gave 14 . The 1,2,3-triazole nucleosides 17, 18 and 19 were obtained from the stannic chloride-catalyzed condensation of the trimethylsilyl 13 and a fully acylated β-D-ribofuranose. The yield and ratio of the ribofuranosyl derivatives of 13 markedly depends on the ratio of stannic chloride used. The structures of the nucleosides 22 and 23 were established by a combination of NOE, 1H-nmr and 13C-nmr spectroscopy.  相似文献

A method was elaborated for the synthesis of polyfunctional compoonds of the drimane series,viz., drim-8(9)-en-7-one, drima-5,8(9)-dien-7-one, and their 11,12-dibromo derivatives, potential intermediates for the synthesis of natural biologically active drimane sesquiterpenoids starting from norambreinolide. In this study, methyl 2‐(quinolin‐8‐yloxy) acetate ( 2 ) obtained by reaction of 8‐hydroxyquinoline ( 1 ) with methyl chloroacetate was condensed with hydrazine hydrate to afford the carbohydrazide ( 3 ). Thio/semicarbazide derivatives ( 4a , 4b , 4c , 4d , 4e , 4f , 4g ) were obtained by treatment of the 3 with substituted phenyl iso/thioisocyanates. The 4a , 4b , 4c , 4d , 4e , 4f , 4g on acidic and basic intramolecular cyclization led to N‐(aryl)‐5‐((quinolin‐8‐yloxy)methyl)‐1,3,4‐oxa/thiadiazol‐2‐amines ( 5a , 5b , 5c , 5d , 5e , 5f , 5g ) and 4‐aryl‐5‐((quinolin‐8‐yloxy)methyl)‐2H‐1,2,4‐triazole‐3(4H)‐thiones ( 6a , 6b , 6c , 6d , 6e , 6f , 6g ), respectively. All the synthesized compounds were characterized by spectroscopic techniques and elemental analyses. The thiosemicarbazide ( 4c ) was also confirmed by X‐ray crystallography.  相似文献

During the condensation of 2,3-dichloroquinizarine with methyl pivaloylacetate in the presence of potassium carbonate in dimethyl sulfoxide the main reaction products are derivatives of angular 3-pivaloylanthra[1,2-b]furan-2,6,11(3H)-trione (about 70%) and anthra[2,1-d][1,3]dioxole-6,11-dione (15%), whereas the yield of the targeted linear methyl 2-tert-butyl-4,11-dihydroxyanthra[2,3-b]furan-5,10-dione-3-carboxylate is only 2%. Methods are developed for modification of the obtained 3-pivaloylanthra[1,2-b]furan-2,6,11(3H)-trione, making it possible to use it for the synthesis of the tert-butyl derivatives of linear anthra[2,3-b]furan-5,10-dione or angular anthra[1,2-b]furan-6,11-dione. *For Communication 7 see [1]. Summary.  Reaction of 6-Amino-2-thiouracil with hydrazonoyl halides yielded regioselectively 7-amino-1,3-disubstituted-1,2,4-triazolo[4,3-a]pyrimidine derivatives. Upon treatment with methyl (Z)-2-benzoylamino-3-dimethylaminopropenoate, the corresponding methyl (Z)-2-benzoylamino-3-([1,2, 4]triazolo[4,3-a]pyrimidin-7-yl)-amino propenoates were obtained which cyclized in the presence of sodium ethoxide to afford novel derivatives of pyrido[2,3-d][1,2,4]triazolo[4,3-a]pyrimidine-5,6-(1H,8H)-diones. In order to define the influence of an allenic type conjugation, the NMR spectra of vinylallene and of some of its methyl derivatives are analysed The signs of the coupling constants are also determined. These compounds are characterised by a planar s-trans structure of the two noncumulated double bonds. A discussion of the parameters, and especially of the methyl replacement effects, evidences the importance of the π type contributions. The theoretical calculations of the coupling constants by the methods of finite perturbation CNDO and INDO corroborate this interpretation.  相似文献

The preferred conformation of 8-methyl-cis-thiahydrindane has been both estimated by 13C NMR chemical shifts and determined by low temperature 13C NMR spectroscopy to be the conformer with the methyl group equatorial with respect to the cyclohexane ring. This result is in disagreement with the interpretation of the temperature dependence of the CD spectra of (+) and (?) 8-methyl-cis-2-thiahydrindane, whereby the conformation with the methyl group axial with respect to the cyclohexane ring was claimed to be the preferred conformation. The preferred conformation of the related oxygen heterocycle, 8-methyl-cis-2-oxahydrindane, has been estimated by 13C NMR chemical shifts to be the conformer with the methyl group axial with respect to the cyclohexane ring. Possible reasons for these observations are discussed.  相似文献

Synthesis of Evernin Two syntheses of the depside evernin 6 are described. Condensation of methyl acetoacetate and methyl crotonate followed by aromatization and reduction with Raney-Ni led to methyl orsellinate (3) . The condensation of everninic acid (4) , obtained by partial methylation of 3 and saponification of the methyl ester, with methyl 2, 4-dihydroxy-3, 6-dimethylbenzoate (methyl β-orcin carboxylate) (5) in presence of cyclohexylcarbodiimide gave evernin ( 6 ). In a second syntheis methyl dihydroorsellinate (1) was regiospecifically converted into its 4-methyl enol ether and aromatized via the benzene selenenyl derivative to yield methyl evernate (7) . Benzylation followed by saponification gave the free acid 8 . Methyl β-orcin carboxylate (5) was synthesized in an analogous way from methyl 3,6-dimethyl-2,4-dioxocyclohexanecarboxylate. Condensation of 8 with the methyl ester 5 by treatment with trifluoroacetic anhydride in toluene yielded 9 , which could be converted into evernin ( 6 ) by hydrogenolysis of the benzyl ether.  相似文献

New 6‐chloro‐ and 6,7‐dichloro‐4‐(4‐aminophenyl)‐2‐(N‐alkylcarbamoyl)‐1,2‐dihydrophthalazines ( 12,18 ) were synthesized starting from 6‐chloro‐ and 6,7‐dichloro‐4‐(4‐nitrophenyl)phthalazine ( 8 ). Routes to 1‐unsubstituted ( 12 ) and 1‐methyl ( 18 ) derivatives are different. Key intermediates 8 were prepared from 4‐chloro‐ and 3,4‐dichlorotoluene via ortho‐formylbenzophenone derivatives 7 .  相似文献
