Base-catalyzed tandem carbon-carbon followed by nitrogen-nitrogen bond formations quantitatively converted N-alkyl-2-nitro-N-(2-oxo-2-aryl-ethyl)-benzenesulfonamides to 2H-indazoles 1-oxides under mild conditions. A rapid and efficient synthesis of 2H-indazoles has been developed using a [3 + 2] dipolar cycloaddition of sydnones and arynes. A series of 2H-indazoles have been prepared in good to excellent yields using this protocol, and subsequent Pd-catalyzed coupling reactions can be applied to the halogenated products to generate a structurally diverse library of indazoles.  相似文献

While N-(2-nitrobenzylidene)anilines produced mixtures of 2,1-benzisoxazoles and 3-anilino-2-aryl-2H-indazoles in the presence of indium and iodine in MeOH, N-(2-nitrobenzylidene)anilines were transformed into 3-anilino-2-aryl-2H-indazoles as the predominant major product through the change of the solvent from protic MeOH to aprotic THF. In an indium-mediated one-pot reductive reaction, 2-benzaldehydes and anilines in THF were also successfully transformed into the corresponding indazoles.  相似文献

A new route to substituted 2-phenyl-2H-indazoles through the cyclization of (2-ethynylphenyl)phenyldiazenes is presented. A coarctate reaction pathway forms the isoindazole carbene under neutral conditions, at moderate temperatures, and without the requirement of a carbene stabilizer. A wide variety of previously unknown diazene precursors was synthesized and cyclized. Trapping of the carbene with a silyl alcohol followed by deprotection affords the 3-hydroxymethyl-2-phenyl-2H-indazoles in good overall yield. The free carbene could also be trapped as a [2 + 1] cycloadduct with 2,3-dimethyl-2-butene.  相似文献

An efficient Bi(OTf)3-catalyzed synthesis of 3-alkenyl-2-pyrrolidine-2H-indazoles from triazenylaryl allylic alcohols via the intramolecular direct amination process is reported. Compared with the dodecyl benzenesulfonic acid (DBSA)-catalyzed method, the new method is more efficient and gives greater yields and functionality tolerance. Additionally, the 3-alkenyl-2-pyrrolidine-2H-indazoles can be transformed to a series of new products under different reaction conditions.  相似文献

[reaction: see text] A variety of 2-aryl-2H-indazoles were synthesized by the palladium-catalyzed intramolecular amination of the corresponding N-aryl-N(o-bromobenzyl)hydrazines. Of several sets of reaction conditions surveyed, the combination of Pd(OAc)2/dppf/tBuONa gave the best results. This method applies to a wide scope of substrates containing electron-donating and electron-withdrawing substituents.  相似文献

A one-step heterocyclization of o-nitrobenzylamines to 3-alkoxy-2H-indazoles is reported. The electronic nature of the nitrophenyl group, the steric and electronic nature of the R1-functionalized benzylic amine, and the nature of the alcoholic solvent affect the efficiency of this heterocyclization reaction (approximately 40-90%).  相似文献

2-Bromobenzaldehydes react with arylhydrazines in toluene at 100 [degree]C in the presence of a catalytic amount of a palladium catalyst and phosphorus chelating ligands such as 1,1[prime or minute]-bis(diphenylphosphino)ferrocene and 1,3-bis(diphenylphosphino)propane along with NaO-t-Bu to afford 1-aryl-1H-indazoles in good yields.  相似文献

A variety of six-membered-ring annulated 2H-indazoles and 1,2,3- and 1,2,4-triazoles were synthesized in good to excellent yields from the corresponding bromoethyl azoles and aryl iodides. The annulation process involves a one-pot norbornene-mediated palladium-catalyzed sequence whereby an alkyl-aryl bond and an aryl-heteroaryl bond are successively formed through two C-H bond activations. Subsequent functionalizations of the resulting polycyclic through cross-coupling reactions are also presented.  相似文献

A one-pot-three-step method has been developed for the conversion of oxazolino-2H-indazoles into triazolotriazepinoindazolones with three points of diversity. Step one of this process involves a propargyl bromide-initiated ring opening of the oxazolino-2H-indazole (available by the Davis-Beirut reaction) to give an N(1)-(propargyl)-N(2)-(2-bromoethyl)-disubstituted indazolone, which then undergoes -CH(2)Br → -CH(2)N(3) displacement (step two) followed by an uncatalyzed intramolecular azide-alkyne 1,3-dipolar cycloaddition (step three) to form the target heterocycle. Employing 7-bromooxazolino-2H-indazole allows for further diversification through, for example, palladium-catalyzed coupling chemistry, as reported here.  相似文献

The synthesis of 1H-indazoles is achieved from o-aminobenzoximes by the selective activation of the oxime in the presence of the amino group. The reaction occurs with a variety of substituted o-aminobenzoximes using a slight excess of methanesulfonyl chloride and triethylamine at 0-23 degrees C and is amenable to scale-up. The synthesis of 1H-indazoles under these conditions is extremely mild compared with previous synthetic approaches and affords the desired compounds in good to excellent yields.  相似文献

[reaction: see text] A general method for the one-step regioselective synthesis of 1-alkyl- or 1-aryl-1H-indazoles from ortho-halogenated alkanoylphenones, benzophenones, and arylcarboxylic acids, via copper-catalyzed amination, was developed by using 0.2% mol of CuO in the presence of K(2)CO(3). The reaction involves amination followed by intramolecular dehydration. Different functionalized alkyl aryl ketones, diaryl ketones, and benzoic acid derivatives were efficiently coupled with several hydrazines. Ligands commonly employed as catalysts for intermolecular amination were shown to be ineffective for this cyclization.  相似文献

A series of alkyne-linked bis-2H-indazoles has been prepared by the double cyclization of ethyne- or butadiyne-linked phenyltriazene or phenyldiazene moieties. Even though there are two five-membered ring cyclizations and several triple bond shifts involved, the reactions proceed rapidly under neutral conditions with mild heating, affording the heterocycles in excellent yields. DFT calculations, in agreement with experimental observations, indicate that the reactions: (1) occur via a very short-lived carbene intermediate, (2) are concerted via an asymmetrical transition state, or (3) are even synchronous, with as many as 16 bonds that are made or broken simultaneously. The biscyclizations presented herein strikingly illustrate the concept of coarctate reactions, the stabilization of transition states by coarctate M?bius aromaticity, the ethynologation principle, and the stereochemical rules.  相似文献

Palladium-catalyzed cyclization of arylhydrazones of 2-bromoaldehydes and 2-bromoacetophenones to give 1-aryl-1H-indazoles has been studied in detail. The cyclization of arylhydrazone of 2-bromobenzaldehydes can be performed with good to high yields using Pd(dba)2 and chelating phosphines, of which the most effective are rac-BINAP, DPEphos, and dppf, in the presence of Cs2CO3 or K3PO4 as a base. Electron-rich, bulky ligands commonly employed for intermolecular amination such as PtBu3 and o-PhC6H4PtBu2 were shown to be ineffective for cyclization and to lead instead to extensive oligomerization and tarring. The method developed is applicable for preparation of a wide scope of indazoles bearing electron-donating or electron-withdrawing substituents, among them, unprotected carboxyl, as well as various indazole heteroanalogues. The cyclization of arylhydrazones of less reactive halides such as 2-chlorobenzaldehyde, as well as 2-bromoacetophenone and bromotetralone, has been achieved. The purity of the starting hydrazone has been shown to be a critical parameter, as various impurities inhibit the cyclization.  相似文献

On Dialkali Metal Dichalcogenides β-Na2S2, K2S2, α-Rb2S2, β-Rb2S2, K2Se2, Rb2Se2, α-K2Te2, β-K2Te2 and Rb2Te2 The first presentation of pure samples of α- and β-Rb2S2, α- and β-K2Te2, and Rb2Te2 is described. Using single crystals of K2S2 and K2Se2, received by ammonothermal synthesis, the structure of the Na2O2 type and by using single crystals of β-Na2S2 and β-K2Te2 the Li2O2 type structure will be refined. By combined investigations with temperature-dependent Guinier-, neutron diffraction-, thermal analysis, and Raman-spectroscopy the nature of the monotropic phase transition from the Na2O2 type to the Li2O2 type will be explained by means of the examples α-/β-Na2S2 and α-/β-K2Te2. A further case of dimorphic condition as well as the monotropic phase transition of α- and β-Rb2S2 is presented. The existing areas of the structure fields of the dialkali metal dichalcogenides are limited by the model of the polar covalence.  相似文献
