宁波地木耳人工培养条件的研究

摘    要:以宁波野生地木耳为研究对象,针对原植体和藻丝体细胞,利用液体和固体2种培养方法,研究了培养基、温度、光照强度以及摇动等人工培养条件下的生长状态.结果表明:宁波地木耳原植体的最佳培养条件是25℃,光强1500lx.液体培养时,BG11-C培养基最适原植体生长,且摇动培养比静止培养生长状态佳.固体培养下30℃,H1-C培养基最适合无菌细胞系的增殖.

Abstract:Nostoc commune is a kind of edible blue algae with medicinal value. Due to the restraints of Nostoc natural resources and its rigorous grow the conditions, the techniques of Nostoc germplasm cultivation and a large number of artificial expansion have not yet reached the mature point. In this work, Nostoc commune sampled form Ningbo local area is studied. The optimized mediums, the effects of culatre conditions such as temperature, light intensity and turbulence on the growth of the algae are investigated, under the circumstance that Nostoc commune thallus and filaments are cultured in liquid or on agar medium. The conclusion based on the experimental results can be made as follows: The optimal culture conditions for Nostoc commune thallus are 25 ℃, 1 500lx; Optimized medium BG11-C favored the growth of Nostoc commune thallus is considered the best when the algae is cultured in liquid medium; Algae in suspending culture with turbulence grows better than that without turbulence; 30 ℃ and HI-C favored for the Nostoc commune algal filaments growth is identified to be the best conditions when the algae are cultured on agar medium base.
