产腈水合酶菌Nocardia sp.HD9611超高诱导及发酵工艺改进

摘    要:研究了金属离子和诱导因子对Nocardia sp.HD9611产腈水合酶的促进作用.结果表明,培养基中加入Co^2 和脲将会对腈水合酶的诱导激活起关键作用,并且发现诱导剂A和B对腈水合酶的产生具有较强的诱导作用,当它们的质量分数分别为0.8%和0.2%时,酶活分别提高了49.4%和104.3%.新的发酵工艺采用诱导剂分批加入方式,不仅可以有效解除诱导剂对细胞生长抑制,并且酶活也有较大的提高。

Abstract:The effects of some metal ions and inducing factors on production of nitrile hydratase in Nocardia sp.HD9611 were studied. The results show that it is necessary to add Co~(2+ )and urea into the culture for inducing the production of nitrile hydratase. And inducer A and B have power activation for expression of nitrile hydratase. The enzyme activities have individual increasing of 0.8% and 0.2% when the mass present of inducer A was 0.8% and B was 0.2%.The new stage addition mode of inducer can not only effectively relieve inhibition but also improve the enzyme activity when fermentation method is modified.
