苯基-硅胶色谱介质的合成及其在紫杉醇提纯中的应用

摘    要: 以硅胶为基质 ,γ 氨丙基三乙氧基硅烷 (APTS)为连接臂 ,用 4种方法合成了苯基反相液相色谱介质。探讨了APTS 硅胶的合成条件 ,比较了这 4种介质用于紫杉醇纯化的分离效果 ,其中气相法合成的介质可使紫杉醇的纯度达到 77 4% ,回收率在 90 %以上。

Abstract:Phenyl-silica gels were synthesized with four methods of reaction on gamma-aminopropyltriethoxysilane (APTS) silica gel. The synthesized gels were used as stationary phase of normal pressure chromatography to separate and purify taxol. The comparison of these four silica gels was carried out. It was found that the gel synthesized by gas-phase reaction gave the best results. The purity of taxol was 77.4%, and the recovery was about 90%.
