摘    要:在多肽固相合成中,分别采用酰氯法和活化酯法,研究了20种Fmoc氨基酸与Wang树脂的连接反应.除精氨酸外,其他氨基酸与树脂的连接效率均能达到80%以上.酰氯法反应速率快、操作简便,优于活化酯法.在酰氯法中,对于精氨酸,选择弱极法溶剂如DCM,延长反应时间至20h,连接效率接近80%.

Abstract:The reaction of twenty kinds of Fmoc- AA -OH linked to Wang resin using methods of acyl chloride or active ester was studied in solid-phase peptide synthesis. Expect for arginine, the yield of other amino acid linked to Wang resin was over 80%. The method of acyl chloride was better than the method of active ester for its high efficiency and convenient operation. For arginine, the yield was improved to nearly 80% by using weakly polar solvent such as DCM and simultaneously increasing the reaction time to 20 h in the method of acyl chloride.
