摘    要:本文报道了N,N·二甲基甲酸胺(DMF)对冻存人外周血淋巴细胞,具有底温保护作用.实验结果表明,淋巴细胞在含有lob DMF和40%人AB型血清的1640液内冻存40至90天后,获得很高的存活率,并且超微结构完整,细胞凝集也较少.虽然在室温时,DMF对淋巴细胞有一定的毒性,DNA合成能力有所降低,但仍是一种优良的细胞低温保护剂.

Abstract:The cryoprotective effects of N,N-Dimethyl Formamide (DMF) on human peripheral blood lymphocytes are investigated. The samples are suspended in RPMI-1640 media containing 10% DMF and 40% human AB serum as cryoprotectants.The experimental results show that the cell viability is very high after cryopreservation for 40, 60 and 90 days. No significant aggregation is found in the cryopreserved cells and the ultrastructure of frozen lymphocytes remains unaltered. Although the DMF has toxic effects on lymphocytes at room temperature and the synthesis of DNA is slightly reduced, it seems that the DMF is an excellent cryoprotective agent for cell cryopreservation.
