地中海伞藻叶绿体生物节律基因的分子克隆及限制酶图谱分析

摘    要:本用分离纯化的地中海伞藻叶绿体DNA,以pcos2 EMBL为载体构建了cosmid分子克隆库。并用果蝇的与生物节律控制相关的DNA顺序为探针,从地中海伞藻叶绿体cosmid分子克隆库中筛选到一组含有同源顺序的克隆,经印迹杂交和限制酶分析,这些克隆都含有与果蝇生物节律探针强烈同DNA顺序,且各克隆的限制酶图谱都相互重叠。这一结果表明,在地中海伞藻叶绿体基因组中可能也存在与果蝇相同或基本相同的与生

Abstract:Biological Rhythm is a physiological phenomenon and widely exist in living nature,it exist in the Acetabularia Mediterranea also. To understand the mechanism of the gene regulation for the phenomeno,the chloroplast genome cosmid library of A. mediterranea was constructed by using cosmid pcos2EMBL as vector. A set of cosmid clones were obtained from cosmid library using DNA prob from Drosophila concern biological rhythm. These cosmid clones contain a DNA sequence homologous with Drosophila DNA prob and overlaped each other by the analysis of blot hybridization and restriction mapping. It is shown that the gene in the Actabularia mediterranea may be the same or the same mainly with Drosophila,and the gene is a single copy sequence located on a single locus. The remaining research on the expression and regulation of the gene are cary on.
