摘    要:采用反相高效液相色谱-质谱(HPLC-MS)联用技术分析了烟草提取液中的5种糖苷类化合物.将烟叶烘干、磨碎后用95%乙醇提取,提取液用正己烷-乙醚(体积比1∶1)除去油脂,水层经大孔树脂分离除杂后,进HPLC-MS分析,获得了5种糖苷类化合物的结构信息.通过对相对分子质量及碎片离子结构信息的解析,推测5种糖苷类化合物分别为:α-紫罗兰醇-β-D-葡萄糖苷、3-氧代-α-紫罗兰醇-β-D-葡萄糖苷、4-羟基-α-紫罗兰醇-β-D-葡萄糖苷、3-羟基-β-大马酮-β-D-葡萄糖苷和地黄普内酯-β-D-葡萄糖苷.

Abstract:A high performance liquid chromatography-electrospray ionization-mass spectrometry method(HPLC-ESI MS) was used for analysis and identification of 5 glycosides in tobacco extraction.After being dried by oven for four hours and grinded into powder,the tobacco leaves were distilled by 95%ethanol.The extract was removed lipin by hexane-ether(1∶1,by volume),and cleaned up on a AmberliteXAD-2 column.The analysis of extraction was performed by HPLC-MS with gradient elution using methanol-water as mobile phase.The retention time, molecular weight and structural information of compounds were obtained.On the basis of analysis of the molecular weight and fragmentation information,five glycoside compounds,including α-ionol-β-D-glucopyranoside,3-oxo-α-ionol-β-D-glucopyranoside,4-OH-α-ionol-β-D-glucopyranoside,3-OH-β-damascenone-β-D-glucopyranoside and lolidide-β-D-glucopyranoside were identified.The information derived from this work was useful for the quality control and study of tobacco.
