Abstract:The Tri-Step identification technique including Fourier Transform Infrared Spectroscopy(FTIR),Secondary Derivative IR Spectra and Two-dimensional Correlation Spectroscopy(2D-IR) was applied to analyze and evaluate the effect of storage time on the different parts of Salvia miltiorrhiza f.alba.The crude drugs of Salvia miltiorrhiza f.alba and their different extracts of the root,stem,leaf and flower were all referred.Similar results were obtained from single dimensional FTIR for the different parts of Salvia miltiorrhiza f.alba stored different time,while the shape and intensity of the peaks in the secondary derivative IR spectra of the different parts showed distinct differences.In the 2D-IR related spectrum range of 1800~1350 cm-1,the number of autopeaks in root and flower stored 0 years were more than that stored 5 years,while number of autopeaks in stem and leaf stored 0 years were less than that stored 5 years.The spectra of water-soluble extracts and ethanol-soluble extracts from different parts of Salvia miltiorrhiza f.alba stored different time was analyzed,and results showed that the main compounds remained invariant,while the percentages of constituents were different.Therefore,Tri-Step identification technique was an express and comprehensive approach to analyze and evaluate the imperceptible differences among different parts of the medicinal herbs stored for years objectively.
