摘    要:本文的目的是建立直接耐晒宝石蓝的急性毒性微量快速分析方法。通过用基于电阻法细胞计数原理的手持式细胞计数器测定由密度约为1.0×104细胞/ml的酿酒酵母悬液20μl和系列浓度待测染料溶液130μl与2.2倍YPD液体培养基110μl所组成的体系在30℃、150 r/min恒温摇床上震荡培养6 h后的细胞数目,求算出该染料的EC50。结果显示直接耐晒宝石蓝对酿酒酵母的EC50为4995 mg/l,与文献报道的小鼠经口LC50=5 g/kg一致。表明本法可作为传统急性毒性测定的替代方法。

Abstract:This study was aimed to establish a quick acute toxicity analysis for Direct Fast Turquoise Blue GL(DFTB).Yeast(Saccharomyces cerevisiae Meyen ex Hansen) cell suspension which density was about 1.0×104 cells/ml was preparation.Then a series of concentrations of the dye solution was preparation.The suspension systems were composed of 20 μl of the yeast suspension,130 μl of the series of concentrations dye solution and 110 μl of 2.2-fold YPD liquid medium.Yeast cells were grown in the suspension system on a shaker with 150 rpm at 30°C.After incubated for 6 h,cell numbers were counted with a hand-held cell counter(Millipore Scepter).EC50 was calculated.The results suggest that the EC50 of DFTB was 4995 mg/l and the results were satisfactory.Conclusion: This method can be used as an alternative to the traditional LD50 to acute toxicity determined in mice.
