摘    要:将反相悬浮交联法制备的壳聚糖微球树脂与三嗪染料活性蓝F3GA(Cibacron Blue F3GA)反应,制得亲和色谱填料,并采用扫描电镜、红外光谱、染料泄漏率测定等对此填料进行鉴定与表征.该填料具有良好的色谱性能,在pH为7.0的磷酸盐缓冲液体系下,对过氧化氢酶的饱和吸附容量为24.6 mg/g,改变pH值及离子强度对结合量有明显影响.

Abstract:Chitosan microsphere was prepared by phase-inversion method as the support matrices. Cibacron blue F3GA was covalently attached onto the chitosan microspheres. The chitosan microsphere structure was characterized by scanning electron microscopy. These dye-carrying chitosan microspheres were used in the catalase adsorption studies. The non-specific adsorption value was 2. 758 mg/g. Cibacron blue F3GA attachment increased the adsorption capacity up to 24.6 mg/g. Langmuir adsorption model was found to be applicable in interpreting catalase adsorption by Cibacron blue F3GA-attached chitosan microspheres. The effect of pH value and ionic strength on cata- lase adsorption were investigated. It showed that F3GA attached chitosan microspheres could be applied in adsorption and purification of catalase without significant denaturation.
