摘    要:通过薄层层析、高效液相色谱等方法对大肠杆菌、醇洒酵母、栗酒裂殖酵母和烟草等四种不同生物所含的泛醌(即辅酶Q)进行了定性分析,确定了各种生物所含的泛醌种类,并利用表达泛醌合成基因的大肠杆菌培养过程澡泛醌含量的变化以及添加前体物质对提高大肠杆菌泛醌合成量进行了初步研究,确定了稳定生长前中期是生物体中泛醌含量最高时期引进泛醌合成的外源基因以及在培养其中添加0.1mM前体物质一对羟甲酸可以提高生物体内泛醌

Abstract:Ubiquinone, extracted from \$Escherichia coli, Saccharomyces cerevisiae, Schizosaccharomyces pombe\$ and \$Nicotiana tabacum\$, were analyzed by thin\|layer chromatography (TLC) and high performance liquid chromatography (HPLC), and the ubiquinone species of them were confirmed. Then, ubiquinone biosynthetic genes were expressed in \$E. coli\$. Effect of cultivative time and precursor \$p\$\|hydroxybenzoate on ubiquinone yield was investigated. The results show that the higher level of ubiquinone is produced in the stationary phage of growth period, and over\|expressing exogenous genes as well as adding 0.1mM \$p\$\|hydroxybenzoate may increase ubiquinone yield of organism.
