长江鳙遗传多样性的研究

摘    要:运用40 个10 碱基随机引物对长江中游鳙(Aristchthysnobilis)的遗传多样性进行了随机扩增多态DNA(RAPD)分析. 所用引物中,有10 个能对鳙不同个体扩增出多态DNA 带,占总引物数的25% . 据所取长江中游自然繁殖的18条鳙样品的RAPD谱带的分析结果表明,鳙任意两个体间的遗传相似度在0.938 6~0.985 7之间,平均值为0.966 1;遗传变异度则在0.014 3~0.061 4 之间,平均值为0.043 9;所分析样本的Shannon 表型多样性指数(Ho)为7.285 8,Shannon 多样性值(H)为0.053 2. 人工繁殖鳙的遗传变异度和Shannon 表型多样性指数均明显低于长江自然繁殖群体.

Abstract:Genetic diversity analysis of bighead carp (Aristichthys nobilis) from the Yangtze River was carried out by RAPD technigue using 40 random oligonucleotide primers (10 mer each). Total 137 bands amplified with 36 effective primers were shown on electrophoretogram and 10 of total 40 primers (25%) produced polymorphic bands in different individuals. Based on RAPD data, the genetic similarity (S) and variability\|diversity (D) between individuals in the population are 0.966 1 (S) and 0.043 9 (D) on average, ranging from 0.938 6 to 0.985 7 (S) and from 0.014 3 to 0.061 4 (D) respectively. The Shannon's index ( H o) and value (H) of phenotypic diversity of the populations are 7.285 8 ( H o) and 0.053 2(H) separatively. The genetic diversity of bighead carp in the Yangtze River and its significance were disscussed.
