Abstract: | A novel fluorimetric method is described for the determination of ethanol by means of alcohol dehydrogenase immobilized on a stirrer. Ethanol is oxidized by alcohol dehydrogenase to aldehyde, with simultaneous conversion of NAD to NADH; the increase in fluorescence of NADH is measured at 465 nm (λex = 370 nm). The calibration curve is linear for ethanol concentrations up to . Deproteinization is required prior to the assay of blood samples; recoveries are 97–102.7%. Enzyme stability, reproducibility and selectivity are discussed. |
