摘    要:海藻糖的积累对水稻对干旱、高盐和低温胁迫的耐受性有一定的影响,准确测定其含量对研究相关的功能有重要的意义.本文建立了转基因水稻样品中海藻糖的高效阴离子色谱测定方法,采用美国Dionex公司DX-500型离子色谱仪,Pac PA1离子色谱分离柱,以0.125 moL.L-1NaOH溶液作为流动相,利用Au工作电极、pH参比电极的脉冲安培检测器测定了水稻提取液中的海藻糖.方法的线性范围为0.02~10.0 mg.L-1,检出限为0.012 mg.L-1,标准品和样品测定的相对标准偏差分别为0.83%和1.52%.用该法测定了抗逆转基因水稻中的海藻糖,取得了较为满意的结果,加标回收率为95.3%~106.0%.所建立的方法具有高的灵敏度和精密度,适合于水稻样品中海藻糖的分析.

Abstract:A method based on high-performance anion exchange chromatography and pulsed ampere detector(HPAEC-PAD) for the determination of trehalose content in transgenic rice plants acclimation to the environmental stress was employed.Trehalose was extracted in boiling water and centrifuged,and the supernatant filtered through a 0.45 μm filter unit.Quantitative carbohydrate analysis is carried out by HPAEC with a Carbo-Pak PA1 column using the DX500 HPAEC system,with which the eluent is 0.125 mol·L-1 NaOH,a gold electrode is used as working electrode and Ag-AgCl electrode as reference electrode.Experimental results show that the linear relationship and repeatability are good.And the results are satisfactory.
