| 摘 要： | 控制提取工艺需要测定柯柯豆碱母液中柯柯豆碱、咖啡因的含量。采用高效液相色谱法测定柯柯豆碱母液中柯柯豆碱、咖啡因的含量 ,以 C8柱为色谱柱 ,以 V(体积分数为 0 .0 0 4 5 %的高氯酸 )∶ V(乙腈 ) =80∶ 2 0为流动相 ,检测波长为 2 80 nm,柯柯豆碱与咖啡因色谱峰的分离度大于 4 ,线性范围为 10～ 80μg/ m L ,相关系数大于0 .999,回收率大于 99.0 %。

| Abstract: | The assay of theobromine and caffeine from the mother solution of theobromine is determined by HPLC.The chromatography column is C 8.The mobile phase∶0.004 5% perchloride acid∶acetamide(V/V)= 80∶20 .The sample can be detected at UV 280 nm.The assay of theobromine and caffeine can be separately determined.The limit of separation is >4.The linear range is from 10 μg/mL to 80 μg/mL.The relativity is above 0.999.The recovery is above 99.9%. |
