| 摘 要： | 建立兔眼房水中两性霉素B含量测定的高效液相色谱方法,研究两性霉素B脂质体药膜的缓释作用。采用Hypersil C18（4.6mm×150mm,5μm）色谱柱,以甲醇：EDTA-2Na（5mmol/L）=80：20（V/V）为流动相,流速：1mL/min;柱温：室温;检测波长：405nm;进样量：20μL。结果表明两性霉素B在22.3—2450ng/mL范围内线性关系良好（r=0.9996）,房水中两性霉素B的平均提取回收率大于95%,日内RSD为1.05%。检出限为4ng/mL。该法简便、准确、可靠,适合两性霉素B的含量测定。

| Abstract: | The method for the determination of amphotericin B（AmB） in aqueous humor of rabbits was established by HPLC to study sustained release of liposome emulsion of AmB,with Hypersil C18（4.6mm×150mm,5μm） column and methanol：EDTA-2Na（5mmol/L）=80：20（V/V） as mobile phase.The flow rate and detection wavelength were 1.0mL/min,405nm,respectively,with sample volume for determination of 20μL.The results showed that AmB had a good linear in the range of 22.3—2450ng/mL（r=0.9996）.The average recovery was above 95% with RSD of 1.05% in a day.The detection limit was 4ng/mL.This method is simple,rapid,accurate and selective for the determination of AmB. |
