深绿短肠蕨抗氧化、降糖、抗炎活性研究

摘    要:取干燥深绿短肠蕨Allantodia viridissima根状茎粉末适量,乙醇回流浸提法得本实验所用样品.采用紫外-可见分光光度法对深绿短肠蕨中多酚含量进行测定;分别对样品的乙醇提取部分(AVH),石油醚萃取物(AVP),乙酸乙酯萃取物(AVE)和正丁醇萃取物(AVB)进行DHHP法和ABTS~+法抗氧化活性研究;采用体外α-葡萄糖苷酶活性抑制模型对其进行降糖活性研究;并采用脂多糖(LPS)刺激的RAW264.7细胞炎症模型法研究其抗炎活性.结果表明,深绿短肠蕨乙醇提取物中多酚类物质含量为(172.8±0.7)mg没食子酸/g提取物;抗氧化活性最强部分为AVE,EC_(50)分别为16.2μg/mL和22.3μg/mL;AVH有较好的降糖和抗炎活性.本研究为深绿短肠蕨的深度研究和利用提供理论依据.

On antioxidant activity,hypoglycemic activity and anti-inflammatory effects of Allantodia viridissima

Abstract:Extraction of Rhizomes of Allantodia viridissima were obtained by ethanol reflux extraction. The total polyphenols components, as well as the clearance rate of DPPH radical and ABTS radical were detected with UV-Visible spectrophotometric methods; Inhibition of hypoglycemic effect by in vitro α-glucosidase activity inhibitor method; detection of NO content by Griess method for anti-inflammatory effect. The results showed that the total phenolics content of the methanol extracts was(172.8±0.7)mg gallic acid/g extract, and the ethyl acetate extracts had certain scavenging ability on DPPH and ABTS free radicals, with the EC_(50) of 16.2 and 22.3 μg/mL, respectively, followed by ethanol, n-butanol and petroleum ether extracts; AV ethanol extract has significant hypoglycemic and anti-inflammatory activity. The study provides a basis research on A. viridissima.
